Complete Genome Sequence Analysis of Two Divergent Groups of Sweet potato chlorotic fleck virus Isolates Collected from Korea

  • The Sweet potato chlorotic fleck virus (SPCFV), of the genus Carlavirus (family Betaflexiviridae), was first detected as one of several viruses infecting sweet potatoes (Ipomea batatas L.) in Korea. Out of 154 sweet potato samples collected in 2012 that were showing virus-like symptoms, 47 (31%) were infected with SPCFV, along with other viruses. The complete genome sequences of four SPCFV isolates were determined and analyzed using previously reported genome sequences. The complete genomes were found to contain 9,104-9,108 nucleotides, excluding the poly-A tail, containing six putative open reading frames (ORFs). Further, the SPCFV Korean isolates were divided into two groups (Group I and Group II) by phylogenetic analysis based on the complete nucleotide sequences; Group I and Group II had low nucleotide sequence identities of about 73%. For the first time, we determined the complete genome sequence for the Group II SPCFV isolates. The amino acid sequence identity in coat proteins (CP) between the two groups was over 90%, whereas the amino acid sequence identity in other proteins was less than 80%. In addition, SPCFV Korean isolates had a low amino acid sequence identity (61% CPs and 47% in the nucleotide-binding protein [NaBp] region) to that of Melon yellowing-associated virus (MYaV), a typical Carlavirus.

Consolidation and Adhesion of Cellulose Nitrate of Folklore Artifacts in the 19~20th Century (19~20세기 생활민속자료에 사용된 셀룰로오스 나이트레이트의 강화와 접착 연구)

  • Cellulose nitrates were used for folklore artifacts(ornamental beads and pipes in hatstrings, frames of eyeglasses, ornamental eyeglass cases, headband ornaments, and jeogori buttons) between the 19th and 20th centuries; however, they are susceptible to cracking, crazing, embrittlement, and crumbling due to deterioration. To consolidate and adhere deteriorated cellulose nitrate folklore artifacts, water-soluble acrylic emulsion adhesives were investigated. For consolidation, Plextol D 498, which has the lowest viscosity in low concentrations, was used. In adhesive films whose glass transition temperature(Tg) is lower than room temperature, the tensile stress and modulus decreased and the strain increased; therefore, the flexibility was high. The Plextol D 498 and Plextol D 498 and Dispersion K 52 films maintained their adhesiveness and flexibility after artificial-sunlight-accelerated ageing, and Plextol D 498 and Dispersion K 52 films hardly caused yellowing. Plextol D 498 was the most stable for accelerating ageing. A low concentration of Plextol D 498 emulsion resulted in the best permeability on the surface of cellulose nitrate, compared with other acrylic emulsions. To prevent ornamental hatstrings from cracking, crazing, embrittlement, and crumbling, a Plextol D 498 emulsion was used. After applying low concentrations(1%, 3%) of the emulsion to consolidate the fragments and high concentration to adhere the fragments, the ornamental hatstrings were protected from crumbling by deterioration, and their fragments were well-adhered. To preserve it from deterioration by oxygen and humidity, the treated ornament was sealed with an oxygen-barrier film using a low-humidity oxygen scavenger.

The Effects of Storage Temperature and pH on Color Change in Garlic Puree (마늘 퓨레의 변색에 관여하는 저장온도와 pH의 영향)

  • This study was carried out to how the effects of storage temperature of garlic bulbs, pH and temperature of garlic puree on its color change. Color change was severely appeared in puree made of garlics stored at pH 4.0, 2$^{\circ}C$ and pH 4.0, 25$^{\circ}C$. The color of garlic puree was changed from green to blue during storage. The yellowing occured by the oxidation at high temperature of puree. Garlic puree absorbed the wavelengths of 486nm. 580nm, and 656nm. The precursors of pigments produced by enzymatic reaction were seemed to be the reasons of color change because there was no my change of color in puree when the enzymes including alliinase were inactivated. However, the color changes of puree at low storage temperature should be studied further in the future.

Studies on the Leaf Vase Life of Hosta Cultivars (Hosta 품종의 절엽 수명에 관한 연구)

  • This experiment was conducted to select optimal cultivars of long leaf vase life for flower arrangements among 21 Hosta cultivars that were selected based on their ornamental value. As a result of the investigation, the cultivar with the longest leaf vase life was 'June' with 29 days record. The cultivars with a more than 10 days record were 'Remember Me', H. lancifolia var. grandiflora, 'Grand Marquee', 'Dress Blue', 'Blue Dimples', 'Frosted Dimples', 'Birchwood Park's Gold', 'Krossa Regal', and 'Paradise Power'. These 10 cultivars had a high chlorophyll content, and had low chlorophyll content changes over time and were high in water absorption. The 10 selected cultivars had equal characteristics of increase in chlorophyll content within a certain period of time. These cultivars have a longer vase life when compared to others. This indicates the potential to develop better materials be use with flower arrangements.

  • We fabricated urethane material based on properties of reversible and unfading for antic-ceramics restoration. This material with low viscosity was made hardness control possible that user want. And it have very strong adhesion and shear strength properties and is shown the best properties for pigment filling, anti-contractibility, coloring as like epoxy system materials. Particularly, the yellowing and ir-reversibility problem in epoxy restoration material were finally solved. So, there is guarantee in the eternity and stabilization of restoration for antic-celamics. And in order to show the reversible state of the restoration, we successfully dissolve this urethane materials in solvent after perfect restoring subsequently.

Effect of Heat Treatment Temperature and Coating Thickness on Conversion Lens for White LED (백색 LED용 색변환 렌즈의 열처리 온도 및 코팅 두께에 따른 영향)

  • Today, silicon and epoxy resin are used as materials of conversion lenses for white LEDs on the basis of their good bonding and transparency in LED packages. But these materials give rise to long-term performance problems such as reaction with water, yellowing transition, and shrinkage by heat. These problems are major factors underlying performance deterioration of LEDs. In this study, in order to address these problems, we fabricated a conversion lenses using glass, which has good chemical durability and is stable to heat. The fabricated conversion lenses were applied to a remote phosphor type. In this experiment, the conversion lens for white LED was coated on a glass substrate by a screen printing method using paste. The thickness of the coated conversion lens was controlled during 2 or 3 iterations of coating. The conversion lens fabricated under high heat treatment temperature and with a thin coating showed higher luminance efficiency and CCT closer to white light than fabricated lenses under low heat treatment temperature or a thick coating. The conversion lens with $32{\mu}m$ coating thickness showed the best optical properties: the measured values of the CCT, CRI, and luminance efficiency were 4468 K, 68, and 142.22 lm/w in 20 wt% glass frit, 80 wt% phosphor with sintering at $800^{\circ}C$.

Resistance Analysis of Cultivars and Occurrence Survey of Fusarium Wilt on Strawberry (국내 딸기 시들음병 발생실태와 품종별 저항성 분석)

  • The occurrence of Fusarium wilt in strawberry fields in Korea was assessed from 2001 to 2003. Fusarium wilt was found from June to August in nursery beds, from September to October after planting in production beds, and from January to March during harvesting period. The symptoms were root rots, discolored vascular tissue in the crown and deformation and yellowing of central leaflets. The disease occurred in up to 30% of plants in 37 of 214 fields surveyed. Fusarium wilt occurred from cvs. ‘Dochiodome’, ‘Maehyang’, ‘Redpearl’, ‘Samaberry ’ and ‘Akihime’ and more severe from cvs. ‘Samaberry’, ‘Maehyang’ and ‘Dochiodome’. Infested soils had high salt concentrations, high nitrogen, phosphate concentrations and low pH. The results of pathogenicity test showed that Fo47 and Fo79 isolated from cvs. ‘Dochiodome’ and ‘Samaberry’ were strong pathogenic to all of four cultivars, and ‘Dochiodome’, ‘Redpearl’, ‘Maehyang’ were relatively susceptible to the all isolates.

Ammonium Excess Promotes Proline Synthesis but Inhibits Glutathione Synthesis in Oilseed Rape (Brassica napus L.)

  • Ammonium (NH4+) serves as a nitrogen source, but its elevated levels can hinder plant growth and production. Excess NH4+ with α-ketoglutarate is assimilated into glutamate, a precursor of proline and glutathione (GSH). This study aimed to investigate the effects of excessive NH4+ on the regulation of proline and GSH synthesis. Detached leaves from oilseed rape (Brassica napus L.) were fed with 0, 50, 100, 500, and 1000 mM NH4Cl for 16 h. As the NH4+ concentrations increased, the leaves exhibited progressive wilting and yellowing. Furthermore, total carotenoid and chlorophyll concentrations declined in response to all NH4+ treatments, with the lowest levels observed in 1000 mM NH4+ treatment. Hydrogen peroxide (H2O2) concentration showed a minor increase at low NH4+ concentration (50 and 100 mM) treatments but a significant increase at high NH4+ (500 and 1000 mM), which was consistent with the localization of H2O2. Amino acid concentrations increased with increasing in NH4+ concentration, while the protein concentration displayed the opposite trend. Proline and cysteine concentrations exhibited a gradual increase in response to increasing NH4+ concentrations. However, GSH concentrations rose only in the 50 mM NH4+ treatment and decreased in the 500 and 1000 mM NH4+ treatments. These results indicate that excessive NH4+ is primarily assimilated into proline, while GSH synthesis is adversely affected.

A study on the development and the physical properties of Epoxy Putty for earthenware restoration (토기 복원용 Epoxy Putty 개발 및 물성에 관한 연구)

  • This study aimed to develop materials to compensate for problems of restoration for lost parts and material problems in the conservation treatment. First, there are several problems with existing materials as follows: secondary damage due to the high shrinkage rate and low adhesive strength, sense of difference due to the severe yellowing, remelting due to irreversibility of materials, processability due to the high strength, sag due to the prolonged setting time in the work process and surface contamination of artifacts due to tools or gloves. In order to solve these problems, this study set developmental goals after understanding the types and physical properties based on epoxy resin among the currently used restoration materials of pottery and earthenware. The developed epoxy resin is epoxy putty, which is cured within 5 minutes, for earthenware restoration. In the earthenware restoration method, the epoxy putty enhanced the workability by quickly curing in paste form and compensated disadvantages such as surface contamination. In addition, the use of white micro-balloon for the epoxy stock solution made coloring easier and weight lighter, and a restoration material with low shrinkage and superior processability was developed.

A Study on Applicability of Neem Resin as a Fixative on the Painting Layer of Mural Paintings from Payathonzu Temple in Bagan, Myanmar (미얀마 바간유적 파야톤주 사원벽화의 채색층 고착처리를 위한 님(Neem) 수지 적용 가능성 연구)

  • The painting layer of Payathonzu temple mural paintings in the ruins of Bagan Myanmar has been damaged due to various reasons. In this study, the applicability of Neem resin, a traditional Myanmar adhesive, as a fixative on the painting layer was examined. Cow glue and Paraloid B-72 were selected as fixatives in the comparison group, and pseudo-specimens with conditions similar to the original mural paintings were produced to examine the changes before and after applying the fixatives and according to the deterioration experiments. As a result of conducting the experiments and comparing the fixatives, it was found that changes on the surface such as smudge, yellowing and gloss are greater with the application of higher concentration of Neem resin than with other fixatives. However, such changes were relatively small under the condition of 4% concentration. It was also confirmed that chromaticity and glossiness vary greatly between before- and after-application of the fixatives but that such discrepancies tend to decrease at 4% concentration compared to other concentrations. As for fixation strength, it was found that the fixation capacity of Neem resin on the base and painting layers is overall higher than other fixatives as the concentration is increased. Therefore, the applicability of 4% concentration of Neem resin as a fixative on the painting layer was confirmed considering the low surface changes according to environmental factors, low color discrepancy and glossiness, and characteristics of excellent fixation strength. It is believed that the findings of this study could be used as basic data for the preservation of Payathonzu temple mural paintings in the future.
