Characteristics and response of mouse bone marrow derived novel low adherent mesenchymal stem cells acquired by quantification of extracellular matrix

  • PURPOSE. The aim of present study was to identify characteristic and response of mouse bone marrow (BM) derived low-adherent bone marrow mesenchymal stem cells (BMMSCs) obtained by quantification of extracellular matrix (ECM). MATERIALS AND METHODS. Non-adherent cells acquired by ECM coated dishes were termed low-adherent BMMSCs and these cells were analyzed by in vitro and in vivo methods, including colony forming unit fibroblast (CFU-f), bromodeoxyuridine (BrdU), multi-potential differentiation, flow cytometry and transplantation into nude mouse to measure the bone formation ability of these low-adherent BMMSCs. Titanium (Ti) discs with machined and anodized surfaces were prepared. Adherent and low-adherent BMMSCs were cultured on the Ti discs for testing their proliferation. RESULTS. The amount of CFU-f cells was significantly higher when non-adherent cells were cultured on ECM coated dishes, which was made by 7 days culturing of adherent BMMSCs. Low-adherent BMMSCs had proliferation and differentiation potential as adherent BMMSCs in vitro. The mean amount bone formation of adherent and low-adherent BMMSCs was also investigated in vivo. There was higher cell proliferation appearance in adherent and low-adherent BMMSCs seeded on anodized Ti discs than machined Ti discs by time. CONCLUSION. Low-adherent BMMSCs acquired by ECM from non-adherent cell populations maintained potential characteristic similar to those of the adherent BMMSCs and therefore could be used effectively as adherent BMMSCs in clinic.

I/O Scheme of Hybrid Hard Disk Drive for Low Power Consumption and Effective Response Time (저전력과 응답시간 향상을 위한 하이브리드 하드디스크의 입출력 기법)

  • Recently, Solid state disk is mainly used because this device has lower power consumption as well as higher response time. But it features higher price and lower performance at delete and write operations compared with HDD. To compensate this defect, Hybrid hard disk with internal non-volatile flash memory was issued. This NVCache is used as a kind of cache for disk blocks. In this paper, an I/O scheme for H-HDD is proposed for improving low power consumption as well as response time. Our method is to use this NVCache as read cache mainly and write cache when write requests are concentrated. In read cache operation, disk blocks with higher priority determined on basis of time as well as spatial localities are prefetched, which can improve response time. The write operation is conducted only at write peak time as disk spindle up costs higher battery power as well as response time. Experiments results show that the suggested method can improve response time of H-HDD and lower the power consumption.

Clinical outcomes and prognostic factors in patients with mycosis fungoides who underwent radiation therapy in a single institution

  • Purpose: We aimed to evaluate clinical outcomes including progression-free survival (PFS), overall survival (OS), partial response, and complete response in patients who underwent radiation therapy (RT) for mycosis fungoides (MF). Also, we sought to find prognostic factors for clinical outcomes. Materials and Methods: Total 19 patients confirmed with MF between 1999-2015 were retrospectively reviewed. Clinical and treatment characteristics, clinical outcomes, and and toxicities were analyzed. Results: Eleven patients were treated with total skin electron beam radiotherapy (TSEBT) and 8 patients with involved field radiation therapy (IFRT) with median dose of 30 Gy, respectively. The median time interval from diagnosis to RT was 2.6 months (range, 0.4 to 87.3 months). The overall response rate was 100%; 11 patients (57.9%) had a complete response and 8 patients (42.1%) a partial response. The presence of positive lymph node at the time of consultation of RT was associated with lower OS (p = 0.043). In multivariate analysis, PFS was significantly lower for patients with increased previous therapies experienced following RT (p = 0.019) and for patients showing PR during RT (p = 0.044). There were no reported grade 3 or more skin toxicities related with RT. Conclusion: Both IFRT and TSEBT are effective treatment for MF patients. Patients with short disease course before RT or complete response during RT are expected to have longer PFS. Positive lymph node status at the initiation of RT was associated woth poor OS, suggesting other treatment modalities such as low-dose RT for patients with low life-expectancy.

DIFFERENTIAL EXPRESSION OF RADIATION RESPONSE GENES IN SPLEEN, LUNG, AND LIVER OF RATS FOLLOWING ACUTE OR CHRONIC RADIATION EXPOSURE

  • We analyzed the differential effects of histopathology, apoptosis and expression of radiation response genes after chronic low dose rate (LDR) and acute high dose rate (HDR) radiation exposure in spleen, lung and liver of rats. Female 6-week-old Sprague-Dawley rats were used. For chronic low-dose whole body irradiation, rats were maintained for 14 days in a $^{60}Co$ gamma ray irradiated room and received a cumulative dose of 2 Gy or 5 Gy. Rats in the acute whole body exposure group were exposed to an equal dose of radiation delivered as a single pulse ($^{137}Cs$-gamma). At 24 hours after exposure, spleen, lung and liver tissues were extracted for histopathologic examination, western blotting and RT-PCR analysis. 1. The spleen showed the most dramatic differential response to acute and chronic exposure, with the induction of substantial tissue damage by HDR but not by LDR radiation. Effects of LDR radiation on the lung were only apparent at the higher dose (5 Gy), but not at lower dose (2 Gy). In the liver, HDR and LDR exposure induced a similar damage response at both doses. RT-PCR analysis identified cyclin G1 as a LDR-responsive gene in the spleen of rats exposed to 2 Gy and 5 Gy gamma radiation and in the lung of animals irradiated with 5 Gy. 2. The effects of LDR radiation differed among lung, liver, and spleen tissues. The spleen showed the greatest differential effect between HDR and LDR. The response to LDR radiation may involve expression of cyclin G1.

  • Low-power radio frequency (RF) chip transceiver technology and the associated structural health monitoring platforms have matured recently to enable high-rate, lossless transmission of measurement data across large-scale sensor networks. The intrinsic value of these advanced capabilities is the allowance for high-quality, rapid operational modal analysis of in-service structures using distributed accelerometers to experimentally characterize the dynamic response. From the analysis afforded through these dynamic data sets, structural identification techniques can then be utilized to develop a well calibrated finite element (FE) model of the structure for baseline development, extended analytical structural evaluation, and load response assessment. This paper presents a case study in which operational modal analysis is performed on a three-span prestressed reinforced concrete bridge using a wireless sensor network. The low-power wireless platform deployed supported a high-rate, lossless transmission protocol enabling real-time remote acquisition of the vibration response as recorded by twenty-nine accelerometers at a 256 Sps sampling rate. Several instrumentation layouts were utilized to assess the global multi-span response using a stationary sensor array as well as the spatially refined response of a single span using roving sensors and reference-based techniques. Subsequent structural identification using FE modeling and iterative updating through comparison with the experimental analysis is then documented to demonstrate the inherent value in dynamic response measurement across structural systems using high-rate wireless sensor networks.

The Comparative Study of Perception of the Public and Stakeholder to Climate Change Adaptation (일반국민 및 이해관계자의 기후변화 적응 인식 비교 연구)

  • The way to respond to climate change is divided into mitigation and adaptation. So far, the response of climate change has been focused on mitigation. The perception of response of climate change also has been focused on mitigation. However, climate change mitigation and adaptation should be highlighted in a comparable concept as a response to climate change. On the other hand, perception of the public on climate change affects the support or opposition of the general public about climate change-related policies. The purpose of this study is to assess awareness of the general public and stakeholders on the subject of climate change adaptation and to present a basis for a future national climate change adaptation policy. Questionnaire on awareness of climate change adaptation were composed of the climate change and climate change response (mitigation, adaptation) understanding and importance, the seriousness of climate change-related adverse climate event, the need for climate change adaptation measures, the degree of implementation, and the level of government effort for climate change response. Climate change adaptation-related awareness survey was conducted for the general public 1,011 people and stakeholders 101 people by telephone interviews. In this study, the perception survey has a difference in aspect of the contents and methodology. And We were able to present the three characteristics of the general public and stakeholders aware of the subject of climate change adaptation. First, the relative importance of climate change adaptation was relatively low. Second level of awareness about climate change response of the public was significantly lower than the stakeholders. Finally, the need to prepare the implementation of the sectoral level, the first adaptation measures was relatively low.

Studies on Digitalis Receptor Desensitization in Rat Ventricle (쥐 심실에서 Digitalis Receptor Desensitization에 관한 연구)

  • [$^3$H]Ouabain binding parameters ( $K_{D}$ and $B_{max}$) to control rat ventricular strips and Langendorff preparations which were not previously exposed to ouabain were compared with those to both preparations that had been first exposed to a complete ouabain dose range of dose-response curve (10$^{-8}$ to 10$^{4}$M). In rat ventricular strips and Langendorff perfused heart preparations, cumulative dose-response curves of ouabain revealed biphasic positive inotropic effects, a "low-dose" effect and a "high-dose" effect with E $d_{50}$ values of 0.5 $\mu$M and 35 $\mu$M ouabain, respectively. The "low-dose" effect in ventricular strip disappeared or was diminished significantly when the ouabain dose-response curve was repeated after the washout of the effects of the first dose-response curve, whereas there were no significant differences in the maximal "high-dose"effect in both exposures to oubain. However, both of the control and ouabain-preexposed Langendorff perfused hearts revealed the same low-dose effects. The $K_{D}$ value for [$^3$H] ouabain binding and the ouabain binding site concentration ( $B_{max}$) estimated by [$^3$H]ouabain displacement assay in control preparations were 230 nM and 2 pmol/mg protein, respectively. [$^3$H]Ouabain binding parameters were not changed by repeated exposure to high concentrations of ouabain. These results suggest that digitalis receptor desensitization in the rat ventricular strip may due to the change of post-receptor events induced by ouabain binding to a high affinity site ($\alpha$$_2$isoform).).).).).

  • A waveform for shock response spectrum test on a shaker is synthesized using wavelets such that a specified shock response spectrum of a test profile is achieved. The parameters of a wavelet are center frequency, amplitude, number of half cycles, delay and polarity. The amplitude of each wavelet component is iteratively adjusted so a specified shock response spectrum is matched. The waveform so synthesized is regarded as a reference acceleration waveform for a shaker shock response spectrum test. The author proposes the use of a long duration and low peak waveform. The usefulness of this approach is illustrated with some examples.

The Relationship between Viscosity of Soluble Dietary Fiber and their Hypoglycemic Effects (수용성 식이섬유소의 점도가 혈당강하에 미치는 영향)

  • Experiments were carried out in humans to assess the relationship between viscosity and post-prandial glucose response of soluble fibers. Eight(3 male and 5 female) healthy individuals were tested for their glucose response after taking control meals or 3 test fiber meals of different viscosities. Meal viscosity of the test food was adjusted to be between 200 and 70, 000 cps. There was a significant increase in post-prandial glucose response(p=0.01) in control meals by solely increasing the volume of water. Fiber with the highest viscosity konjac mannan demonstrated the lowest relative glucose response(70.1$\pm$6.6), followed by medium viscosity xanthan (79.3$\pm$8.7)and low viscosity psyllium (86.3$\pm$10.5). The difference is significant at the level of p<0.05. However, no significant difference in relative glucose response of the same fiber was found when the 4 levels of water were added to make different meal viscosity of each fiber (p=0.476). This result suggested that hypoglycemic action of soluble dietary fibers is related to fiber viscosity rather than meal viscosity.

  • Fast response characteristics of twisted nematic liquid crystal display (TN-LCD) cell with different nematic liquid crystals (NLCs) and cell gap on a rubbed polyimide (PI) surface were studied. High transmittance and fast response time of the TN-LCD on the rubbed PI surface were achieved by using high birefringence ($\Delta$ n) and low cell gap. It is considered that the transmittance and response time of the TN-LCD on the rubbed PI surface decreased as $\Delta$ nd decrease.
