A portable RF HTS SQUID-based susceptometer was applied to the measurement of fatigue behavior for type 316L(N) stainless steel containing 0.04% to 0.15% nitrogen content. Strain-controlled low cycle fatigue (LCF) tests were conducted at RT and $600^{\circ}C$ in air an atmosphere, and the magnetic moments were measured after the fatigue test using HTS SQUID. The magnetic moment of an as-received sample is higher than that of a fatigued sample in all the temperature ranges irrespective of the nitrogen content. The fatigue life decreased with an increasing test temperature up to $500^{\circ}C$, but increased at $600^{\circ}C$. The change of the magnetic moments by LCF test is attributed to the stress induced micro defects.

Genetic improvement in potato can be carried out through several approaches, as sexual crosses, somatic hybridization, mutation and genetic engineering. Although the approach is different, but the goal is the same, to get a superior cultivar. Mutation and genetic engineering are very interesting methods for genetic improvement of potato plants. Mutation by gamma-ray irradiation have been performed to get some new potato cultivars which are more resistant to disease and have higher productivity. We have carried out a mutation of some potato cultivars and obtained some excellent clones to be potentially released as new superior cultivars. By the mutation method, we have released one potato cultivar for the French fries industry, and we registered one cultivar of potato for chips, and two cultivar for vegetable potatoes. Actually we are doing multi-location trial for three clones to be released as new cultivars. Through genetic engineering, several genes have been introduced into the potato plant, and we obtained several clones of transgenic potato plants. Transgenic potato plants containing FBPase gene encoding for fructose bisphosphatase, have a higher rate of photosynthesis and higher tuber productivity than non-transgenic plants. This result suggests that FBPase plays an important role in increasing the rate of photosynthesis and potato tuber productivity. Some transgenic potatoes containing the Hd3a gene are currently being evaluated for their productivity. Over expression of the Hd3a gene is expected to increase tuber productivity and induce flowering in potatoes. Transgenic potato plants containing MmPMA gene encoding for plasma membrane ATPse are more tolerant to low pH than non-transgenic plants, indicating that plasma membrane ATPase plays an important role in the potato plant tolerance to low pH stress. Transgenic potato plants containing c-lysozyme genes, are highly tolerant of bacterial wilt diseases caused by Ralstonia solanacearum and bacterial soft rot disease caused by Pectobacterium carotovorum. Expression of c-lyzozyme gene plays an important role in increasing the resistance of potato plants to bacterial diseases.
This study was designed to determine the effect of season and split-sex feeding on performance, stress and carcass quality of finishing pigs. Two hundred and seventy cross-bred finishing pigs $(82.1{\pm}0.82kg)$ were randomly divided into six groups in 2 seasons (summer and winter) ${\times}$ 3 split-sex feeding (female only, castrated male and mixed) factorial arrangement of treatments. In winter, feed intake and weight gain of finishing pigs were better than summer (p<0.05). Split-sex feeding was affected the feed conversion, feed conversion of finishing pigs were superior in winter and mixed groups. Plasma concentrations of cortisol as a stress indicators was greater in the mixed groups vs female only and castrated male groups. There were no interactions between the season and split-sex feeding in their effects on these blood variables. The season was affected the carcass characteristics and PSE incidence of the finishing pigs, PSE incidence was low in winter and mixed groups. Meat quality did not show any significant difference due to the difference of season and split-sex feeding of finishing pigs. There was no significant difference in the pH, drip loss and meat color of pork loin between the season and split-sex feeding of finishing pigs. In conclusion, the results of this study suggest that we need more precise on-farm management practice in summer and application of split-sex feeding skills to reduce the stress and improve welfare status of finishing pigs.

This paper dealt with melting characteristics of low-voltage miniature cartridge fuse used for 220 V electronic equipment. The experimental sample is low-voltage miniature cartridge fuse with rating of 250 V(3A) and size of $5{\times}20$ mm. In order to evaluate melting and scattering characteristics of the fuse, we applied to 8/20 ${\mu}s$ surge current, overload current and external thermal stress such as flame of fire. From the experimental results, the fuse element was melted and scattered by applied surge current(above 0.79 kA) and overload current(above 4.5 A). It was also attached to the inner surface of the fuse tube. The fuse element was attached as a thin film on inner surface of fuse tube when large surge current was applied. It was confirmed, however, the fuse element was not changed by external thermal stress such as flame and hot-air.
Schizosaccharomyces pombe gene encoding redox enzymes, such as thioltransferase (TTase) and thioredoxin (TRX), were previously cloned and induced by oxidative stress. In this investigation, their expressions were examined using $\beta$-galactosidase fusion plasmids. The expression of the two cloned genes appeared to be growth-dependent. The synthesis of $\beta$-galactosidase from the TTase-lacZ fusion was increased in the medium with the low glucose level, whereas it was significantly decreased in the medium without glucose or with galactose. It was also decreased in the nitrogen-limited medium. The synthesis of galactosidase from the TRX-lacZ fusion was unaffected by galactose or low glucose. However, it was lowered the absence of glucose. The synthesis of $\beta$-galactosidase from the TTase-lacZ fusion was shown to be enhanced in a higher medium pH. Our findings indicate that S. pombe TTase and TRX genes may be regulated by carbon and nitrogen sources, as well as medium pH.

Transformation plasticity is that when a phase transformation of ferrous or non-ferrous alloys progresses even under an extremely small applied stress compared with a yield stress of the material, a permanent deformation occurs. One of widely accepted description for the transformation was proposed by Greenwood and Johnson [1]. Their description is based on an assumption that a weaker phase of an ideal plastic material could deform plastically to accommodate the externally applied stress and the internal stress caused by the volumetric change accompanying the phase transformation. In this study, an implicit finite element model was developed to simulate the deformation behavior of a low carbon steel during phase transformation. The finite element model was coupled with a phase field model, which could simulate the kinetics for ferrite to austenite transformation of the steel. The thermo-elasto-plastic constitutive equation for each phase was adopted to confirm the weaker phase yielding, which was proposed by Greenwood and Johnson [1]. From the simulation, the origin of the transformation plasticity was quantitatively discussed comparing with the other descriptions of it.
BACKGROUND: The aim of this study was to evaluate the combined effect of low-level laser treatment (LLLT) and recombinant human bone morphological protein-2 (rhBMP-2) applied to hypoxic-cultured MC3T3-E1 osteoblastic cells and to determine possible signaling pathways underlying differentiation and mineralization of osteoblasts under hypoxia. METHODS: MC3T3-E1 cells were cultured under 1% oxygen tension for 72 h. Cell cultures were divided into four groups: normoxia control, low-level laser (LLL) alone, rhBMP-2 combined with LLLT, and rhBMP-2 under hypoxia. Laser irradiation was applied at 0, 24, and 48 h. Cells were treated with rhBMP-2 at 50 ng/mL. Alkaline phosphatase activity was measured at 3, 7, and 14 days to evaluate osteoblastic differentiation. Cell mineralization was determined with Alizarin red S staining at 7 and 14 days. Western blot assays were performed to evaluate whether p38/protein kinase D (PKD) signaling was involved. RESULTS: The results indicate that LLLT and rhBMP-2 synergistically increased alkaline phosphatase (ALP) activity and mineralization. Western blot analyses showed that expression of type I collagen, runt-related transcription factor 2 (RUNX2), and Osterix (Osx), increased and expression of hypoxia-inducible factor 1-alpha ($HIF-1{\alpha}$), decreased more in the LLLT and rhBMP-2 combined group than in the rhBMP-2 or LLL alone groups. Moreover, LLLT and rhBMP-2 stimulated p38 phosphorylation and rhBMP-2 and LLLT increased Prkd1 phosphorylation. CONCLUSION: Combined treatment with rhBMP-2 and LLL induced differentiation and mineralization of hypoxic-cultured MC3T3-E1 osteoblasts by activating p38/PKD signaling in vitro.
Animal welfare during transport became an largely issue because of increasing demand for improved animal welfare standards. Most studies on the animal welfare during transportation have concentrated on the atmosphere and the temperature of the truck compartments. Thus, the objective of study was to collect and quantify three axis acceleration and determine the effect of bedding for transporting pigs from farm to slaughterhouse. A total of 2,840 crossbred fattening pigs with a live weight of approximately 115 kg were used. They were raised in the same commercial farms and transported to the same commercial slaughterhouse. A 3×2×2 completely randomized factorial design was used to investigate effects of rubber type bedding (bedding or non-bedding) and two levels of driving style (aggressive or normal) in three different time periods with different outside temperatures. Air temperature treatments were as follow: high temperature ([HT] higher than 24℃); low temperature ([LT] lower than 10℃); normal temperature ([NT] 10℃ to 24℃). In our experiment, pigs transported under aggressive driving style showed lower (p < 0.05) pH and water holding capacity (WHC) than those transported under normal driving style. Pigs transported under normal driving style showed a lower percentage of drip loss (DL) (p < 0.05) than those transported with an aggressive driving style. Also, transported with bedding showed higher (p < 0.05) lying behavior but lower (p < 0.05) sitting behavior than those transported without bedding. Pigs transported under normal driving style showed lower (p < 0.05) cortisol level than those transported under aggressive driving style. In conclusion, aggressive driving style cause acute stress in pigs, while bedding helps alleviate acute stress in pigs during transportation in LT.
This study was conducted to determine the effects of exogenous zinc-metallothionein (Zn-MT) on anti-oxidative function and pork quality. After feeding a corn-soybean meal-based diet for two weeks, 48 pigs ($Duroc{\times}Landrace{\times}Chinese\;Black Pig$) were assigned randomly to four groups. Pigs in Group 1 were maintained under non-stress conditions, whereas pigs in Groups 2, 3 and 4 were aggressively handled for 25 min to produce stress. Pigs in Groups 1, 2, 3, and 4 received intramuscular administration of saline (control group; CON), 0 (negative control group; NCON), 0.8 (low dose group; LOW), and 1.6 (high dose group; HIGH) mg rabbit liver Zn-MT per kg body weight, respectively. Pigs were slaughtered at 3 and 6 h post-injection. Zn-MT treatment increased (p<0.05) the activities of superoxide dismutase (SOD) and glutathione-peroxidase (GSH-PX) while decreasing the concentration of malondialdehyde (MDA) in liver. These responses were greater (p<0.05) at 6 h than at 3 h post Zn-MT injection. Zn-MT treatment increased (p<0.05) hepatic SOD mRNA levels in a time and dose-dependent manner and decreased (p<0.05) serum glutamate-pyruvate transaminase and lactate dehydrogenase activities (indicators of tissue integrity). Zn-MT administration decreased (p<0.05) lactate concentration and increased (p<0.05) pH and water-holding capacity in the longissimus thorasis meat. Collectively, our results indicate that intramuscular administration of Zn-MT to pre-slaughter stressed pigs improved tissue anti-oxidative ability and meat quality.
The electrical stability of ZPCCY-based varistors composing of ZnO-Pr$_{6}$O$_{11}$-CoO-C $r_2$$O_3$- $Y_2$$O_3$ ceramics were investigated in various DC accelerated aging stress with sintering times. Sintering time greatly affected electrical properties and stability. Sintering time decreased nonlinear exponent in the range of 51.2∼23.8 and increased leakage current in the range of 1.3∼5.6 ${\mu}$A. The varistor sintered for 1 h exhibited high nonlinearity, whereas relatively low stability. On the contrary, the varistor sintered for 3 h exhibited low nonlinearity, whereas relatively high stability. But the varistor sintered for 2 h exhibited not only good nonlinearity, with nonlinear exponent of 38.6 and leakage current of 3.6 ${\mu}$A but also high stability, in which the variation rates of varistor voltage, nonlinear exponent, leakage current, and dissipation factor are -0.80%, -1.81 %, +74.4%, and +0.88%, respectively.
