비접촉식 저온 대기압 면방전 플라즈마를 이용한 저장 '세토카' 감귤의 선도유지 효과 (Preservatory effect of stored 'Setoka'(Citrus sp.) using the noncontacted low temperature atmospheric pressure surface discharged plasma)

  • 국내 주요 재배되는 만감류 중의 하나인 '세토카'는 시설재배하여 3월 까지 수확이 이루어진다. 껍질이 얇은 '세토카'는 저장 중 풍미 변화와 부패율 정도가 다른 만감류에 비하여 상대적으로 높다. 비접촉식 대기압 면방전 저온 플라즈마 기술은 전기적 방전을 통하여 주변의 기체를 플라즈마 상태로 전환시키는 것으로 이를 통해 오존($O_3$)과 질소산화물(NOx) 등이 발생되고, 이들 활성종 기체들에 의해 살균효과를 볼 수 있다. 본 실험에서는 서귀포에서 수확한 '세토카'를 일반상온저장, 저온저장($10{\pm}1^{\circ}C$, $80{\pm}3%$), 저온저장에 플라즈마 처리군으로 나누어 80일 동안 저장하고 10일 간격으로 품질 및 부패율을 조사하였다. 총 당함량은 3 처리 모두 일정하게 유지되었으며, 산 함량은 저장기간 동안 꾸준히 감소하였고, 특히 상온저장 처리에서 저장 30일부터 급격하게 감소하여 0.5%이하의 산 함량을 나타내었다. 경도는 저장 40일까지 소폭으로 증가하는 경향이었으나 처리간 유의적인 차이는 보이지 않았다. 감모율은 상대적으로 저장 온도가 높은 상온저장에서의 중량감소가 컸다. 저장 80일 후의 부패율은 상온저장 50.5%, 저온저장 5.6%, 저온저장에 플라즈마 처리군에서는 1.9%로 조사되었다. 상온저장에서 발생된 저장병의 73%가 곰팡이병이었으나 플라즈마 처리군에서 발생된 부패병은 대부분 무름병 증상이며, Penicillium 균에 의한 부패병은 발생되지 않움으로써 '세토카' 저장 시 플라즈마의 살균효과를 확인하였다.

Effect of Caffeine, cAMP and Cattle Seminal Plasma on Freezability of Buffalo Bull Semen

  • An experiment was conducted to investigate the effect of caffeine, cAMP and cattle seminal plasma on preservation of semen at ultra low temperature ($-196{^{\circ}C}$). Each semen sample was divided into four parts equal in volume and sperm concentration; three were treated with caffeine, or cAMP, or cattle seminal plasma (CSP) and the fourth was kept as control. Sperm motility, abnormal spermatozoa, live-dead count and acrosomal damage were studied at different stages of freeze preservation viz.; just after dilution, at $5{^{\circ}C}$, at glycerolisation, before freezing, just after freezing, 24 hours of storage, and one week of storage. Sperm motility (58.39, 61.33, 52.00 and 50.39 per cent), non-eosinophilic spermatozoa (72.55, 69.98, 63.31 and 67.64 per cent), abnormal spermatozoa (5.71, 4.98, 8.04 and 5.66 per cent) and acrosomal damage (13.28, 13.33, 14.80 and 14.65 per cent) were observed in cAMP, caffeine, cattle seminal plasma and control, respectively, at every stage of freeze preservation. From this study it could be concluded that freezability of buffalo semen can be improved through the addition of caffeine followed by cAMP and cattle seminal plasma.

문화재 보존시설에서의 습도지시카드 적용성 연구 (The Application of Humidity Indicators for Environmental Measurement Apparatus of Preservation Facility)

  • Cultural heritages are affected by various environmental factors. Main factors are temperature, humidity, light and indoor air pollutants such as ozone, nitrogen oxides. Especially humidity condition is very important. Low humidity condition can cause contraction and high humidity condition can lead to growth of microorganism. This study is conducted to appraise an applicability of the Humidity Indicator which is one of the simple method for humidity control. The Humidity Indicator shows the humidity conditions through color change. Therefore, photometer was used to correctly measure the color change of Humidity Indicator. As a result of lab test, Humidity Indicators was stabilized after one hour from en exposure in a certain humidity condition and the indicators indicated clear correlations between color changes of Humidity Indicators and humidity conditions. Some Humidity Indicators which had been selected through the lab tests were applied to preservation facilities of cultural heritages and the indicators indicated closed correlations with humidity conditions in indoor of facilities.

마늘 퓨레의 변색에 관여하는 저장온도와 pH의 영향 (The Effects of Storage Temperature and pH on Color Change in Garlic Puree)

  • 마늘퓨레를 제조하였을 때 원료로 사용된 마늘의 저장온도와 퓨레의 pH 및 온도가 변색에 미치는 영향을 알아보고 그 원인을 구명하기 위하여 본 실험이 수행되었다. 마늘퓨레의 변색은 저온저장했던 마늘로 퓨레제조시 심하게 나타났으며 pH 4.0, 2$^{\circ}C$$25^{\circ}C$ 조건에서 청변이 가장 심하였다. 변색의 양상은 녹변, 청변, 그리고 갈변의 세가지 형태로 나타났으며 산화적 갈변을 제외한 녹변과 청변은 변색정도의 차이일 뿐 같은 기작에 의한 것으로 생각되었다. 변색색소의 파장흡수는 486nm, 580nm, 그리고 656nm에서 특징적으로 나타났다. 알리네이즈 효소활성을 억제하였을 경우 청변은 전혀 일어나지 않아 변색의 원인이 효소적 반응에 의해 생성된 색소전구물질들에 의한 것으로 사료된다. 그러나 원료마늘의 저장온도 중 저온이 특히 마늘 퓨레 변색에 영향을 미친 원인을 설명하기 위해서는 보다 깊은 연구가 더욱 필요하다.

한강수계 유량곡선식 개발 및 수질오염물질 항목간 상관성 분석에 관한 연구- 벽계천 중심으로 - (Study on the Analysis of Development of Stage-Discharge Curve for Han River and Correlation between Items of Water Pollutants- Focused on Byeokgyeo Stream -)

  • We drew the stage-discharge relations of Byeokgye Stream, located in Yangpyeong-gun, Gyeonggi-do, and analysed the correlation between items of water pollutants by measuring the flow rate and water pollutants thirty-four times from April 2010 to December 2010. The results showed that it tended to be low water season from April to June and from October and December, while tending to be water season from July to September. The average flow rate was $2.137\;m^3/sec:\;0.464\;m^3/sec$ in low water season and $13.970\;m^3/sec$ in high water season. The stage-discharge curve thereon was $Q=40.107{\times}(h-1.200)^{2.877}$. As to the correlations, the correlation between the water temperature and COD was 0.58, and the correlations of SS with BOD and COD were 0.46 and 0.40 respectively. The correlation between SS and T-P was 0.73, showing higher than other items.

저장량과 환기구크기가 움저장 생강의 저장성에 미치는 영향 (The Effects of Storage Amount and Ventilator Size on the Quality of Ginger During Cellar Storage)

  • An experiment was conducted to develop the simple methods of ginger storage which decrease the weight reduction and maintain good quality of ginger during the cellar storage. The stored boxes with volume of 0.03㎥ and ventilator diameter of 3, 4, 5cm was hurried under the ground(60, 80, 100cm) in the green house. During the cellar storage at the 100cm depth the average temperature and relative humidity in the stored box were remained in 11.7~16.3$^{\circ}C$ and 73%, respect. The higher storage amount and smaller size of ventilator size increased the CO2 concentration in the stored box, and the concentration in the stored box with 50% storage quantity rate and 3cm ventilator diameter size was more than 10% for about 2 months from early Feburary. The decay rate of ginger during the cellar storage increased with higher amount of storage quantity and smaller size of ventilator. The CO2 concentration was low and remained relatively constant with the deeper location of stored box under the ground, and the decay rate was lower in the deeper stored box. Germination rate increased with the deeper location of stored box, and with the lower storage quantity and larger ventilator size. The germination rate was low in the higher decay rate box. Weight loss, total sugar and moisture contents of ginger were decreased, while crude fiber and ash were increased during the storage.

약주 제조를 위한 저온 적응성 효모의 특성 (Characteristics of yeast with low temperature adaptation for Yakju brewed)

  • 발효식품에서 분리한 효모 482균주의 저온적응성 및 알코올 생성능을 확인하고 ${\beta}$-glucosidase와 cerulenin, TFL저항성을 실험하여 저온적응성이 우수하고 향기성분 활성이 좋은 Y297 균주를 선발하였다. 26s rRNA 염기서열을 분석한 결과, Y297은 Saccharomyces cerevisiae로 동정되었다. 선발된 균주는 $15^{\circ}C$에서는 $25^{\circ}C$에 비해 정지상으로에 도달하는 시간은 오래 걸리지만 보다 많은 균수를 유지하였다. YPD 액체배지에 알코올 농도 10%와 glucose 60% 그리고 NaCl 8%가 포함된 배지에서 내성을 확인할 수 있었다. YPD(glucose 25%)배지에서 배양한 효모의 세포질 단백질을 추출하여 ${\beta}$-glucosidase와 esterase의 활성을 측정한 결과 시판효모보다 우수한 활성을 나타냈다. 이러한 결과를 통해 선발된 Y297는 약주제조에 적합한 종균효모로써의 가능성을 확인할 수 있었다.

열풍건조 온도에 따른 고구마 잎 메탄올 추출물의 항산화 효과 (Effect of hot-air drying temperature on antioxidative activity of sweetpotato leaves)

  • 고구마 잎을 건조 채소화 하기 위한 기초자료를 제공하기 위해서 열풍건조한 고구마 잎 메탄올 추출물에 함유된 총 탄닌, 총 플라보노이드, 총 폴리페놀 함량을 분석하고, 항산화 효과로 DPPH 라디칼 소거활성, ABTS 라디칼 소거능, 아질산염 소거능 등을 분석 비교 검토하였다. 총 탄닌 함량은 신미 $40^{\circ}C$에서 10.87 mg/g, $70^{\circ}C$에서 7.28 mg/g으로 감소되었고, 총 플라보노이드는 하얀미 $40^{\circ}C$에서 55.37 mg/g, $70^{\circ}C$에서 39.63 mg/g으로 감소되었다. 즉 저온건조가 고온건조보다 이들 물질의 함량이 많았다. DPPH 라디칼소거 활성은 신미와 하얀미 $40^{\circ}C$에서 84.33%와 85.25%로 가장 높았으며, ABTS 라디칼 소거능도 $40^{\circ}C$ 처리구에서 80%가 넘는 높은 값이었다. 아질산염 소거능은 신미와 하얀미가 $40^{\circ}C$에서 76.15%와 73.74%로 가장 높았고 $70^{\circ}C$에서는 낮았다. 품종 간에는 하얀미가 신미보다 페놀성 물질과 항산화 물질이 더 많은 것으로 나타났다. 즉 건조한 고구마 잎의 항산화 효과는 품종간의 차이는 있었으나, $40^{\circ}C$ 시료에서 높고, $70^{\circ}C$ 시료에서 낮았다. 이는 건조온도에 의해 영향을 받는 것으로서 낮은 온도에서 건조하는 것이 유효성분의 감소가 적어 항산화 능력이 높은 결과이었다.

Effect of Low Temperature Preservation and Cell Density on Metabolic Function in a Bioartificial Live

  • Difficulties associated with bioartificial liver (BAL) preservation limit not only the commercialization of BAL, but also its clinical trials. In this study, the possibility of cold preservation of BAL cartridges containing porcine hepatocytes was examined at 4$^{\circ}C$. In an in vitro perfusion culture System, BAL cartridges maintained cytochrome P450 metabolic function for at least 50 days. However, all BAL cartridges completely lost their ammonia eliminating ability when stored at 4$^{\circ}C$. We a1so studied the effect of cell density on the maintenance of BAL liver function In a highly differentiated and healthy state. As expected, BALs containing a larger number of hepatocytes demonstrated higher metabolic functions. When metabolic functions were compared per gram of hepatotytes, no large differences were observed between devices containing different densities of hepatocytes. Decreased cell density did not Successfully prolong BAL function. The viability and function of isolated hepatotytes highly depend on the culture conditions, such as cell density, substrata, culture media, and additives to the culture media. Perfusion culture of BAL cartridges at 4$^{\circ}C$ gave a promosing result with respect to the maintenance of P450 activity. However, as indicated by the rapid loss of ammonia metabolic activity, many factors still remain to be optimized for preservation of BAL keeping high metabolic functions for a longer time.

전통 한국 식이의 비교 영양학 (Comparative Nutrition of Traditional Korean Diet)

  • Rice is the primary main dish of Traditional Korean diet. Although there have been changes in food consumption and nutrient intake among Koreans, traditional dietary pattern is stil dominant among Koreans. Traditional Korean diet has emphasized breakfast, which is the most frequently missed meals in Korea today but important for daily work performance and health. Compared to diets of the U.S. and Greece, Korean diet is high in carbohydrate and low in fat and cholesterol due to low intake of meat. Koreans also consume large amount of plant food, which makes fiber content of diet to be high. However fruit and milk consumption tends to be low in Korea. Koreans use fermented food, including kimchi, very frequently as well as foods cooked and consumed at high temperature and over direct fire. Traditional cooking methods are time consuming which limits the usage among modern city dwellers with working women. Despite the strengths of traditional Korean diets in reducing risk factors of chronic diseases, preservation of the tradition in modern Korean society requires special attention and efforts to make them more adaptable to contemporary life styles.
