Antioxidant Activity of Yogurt Fermented at Low Temperature and Its Anti-inflammatory Effect on DSS-induced Colitis in Mice

  • This study was performed to evaluate the antioxidant activity of yogurt fermented at low temperature and the anti-inflammatory effect it has on induced colitis with 2.5% dextran sodium sulfate (DSS) in Balb/c mice. Yogurt premix were fermented with a commercial starter culture containing Lactobacillus acidophilus, Bifidobacterium lactis, Streptococcus thermophilus, and Lactobacillus delbrueckii subsp. bulgaricus at different temperatures: $22^{\circ}C$ (low fermentation temperature) for 27 h and $37^{\circ}C$ (general fermentation temperature) for 12 h. To measure antioxidant activity of yogurt samples, DPPH, $ABTS^+$ and ferric reducing antioxidant potential (FRAP) assays were conducted. For animal experiments, inflammation was induced with 2.5% DSS in Balb/c mice. Yogurt fermented at low temperature showed higher antioxidant activity than that of the yogurt fermented at general temperature. In the inflammatory study, IL-6 (interleukin 6) was decreased and IL-4 and IL-10 increased significantly in DSS group with yogurt fermented at general temperature (DYG) and that with yogurt fermented at low temperature (DYL) compared to that in DSS-induced colitic mice (DC), especially DYL had higher concentration of cytokines IL-4, and IL-10 than DYG. MPO (myeloperoxidase) tended to decrease more in treatments with yogurt than DC. Additionally, yogurt fermented at low temperature had anti-inflammatory activity, although there was no significant difference with general temperature-fermented yogurt (p>0.05).

Effect of Kefir Extract on th Growth of Serum-Free Mouse Embyro (SFME) Cells

  • The antioxidative and protective activities of kefir, low-fat dry milk (NFDM) extract and fractions on SFME cells in serum-free medium were investigated. Kefir and low-fat kefir and NFDM extract were made by solubilizing the freeze dried powder forms in deionized water, filtering through glass prefilter, 12 ㎛ and 2 ㎛ membrane, and demineraling with chelating resin. Kefir, low-fat kefir and NFDM extract were fractioned into dialyzate and retentate by dialysis with membrane tube having the molecular cut-off of 3,500 Dalton. An antioxidative activity was analyzed by the in vitro model system using a linoleic acid. In the case of kefir an antioxidative activity was detected only in the retentate of kefir extract. On the other hand NFDM showed an antioxidative activity in extract, demineralized extract, dialyzate and retentate. The retentate of kefir extract had the higher antioxidative activity than that of NFDM extract. Kefir showed the protective effect of SFME cells in serum-free medium in extract, demineralized extract and retentate, but low-fat kefir didn't. NFDM had the similar protective effect on SFME cells as extract, demineralized extract and retentate of kefir.

Screening of Antioxidative Compounds toward Human Low Density Lipoprotein (LDL) from Useful Plants (유용식물로부터 Human Low Density Lipoprotein(LDL)에 대한 항산화제의 탐색)

  • This study was designed to investigate antioxidative activity of substances isolated from 25 kinds of useful plants resources toward free radical and human low density lipoprotein(LDL). Methanol extracts of Oenothers odorate had the highest antioxidative activity similar with ${\alpha}$-tocopherol. Methanol extracts of Oenothers odorate was extracted again by the ethylacetate. The ethylacetate soluble acidic fraction obtained from methanol extract of Oenothers odorate showed highest activity toward human LDL. Each fraction was purified through Sepadex LH-20 chromatography by elution of chloroform-methanol mixture (90:10 v/v). Fraction, F-2 obtained from Oenothers odorate showed at highest levels of electron donating activity. Fraction, F-2 was identified as 3,4-dihydroxybenzoic acid and 3-hydroxycinnamic acid.

Experimental Study of the Effect of Injinsaryungsan and Sosihotang on cholestatic liver injury induced by $ANIT({\alpha}-naphtylisothiocyanate)$ (인진사령산(茵陳四岺散)과 소시호탕(小柴胡湯)이 ANIT 로 유발(誘發)된 담즙울체성(膽汁鬱滯性) 간장애(肝障碍)에 미치는 영향(影響))

  • In an attempt to evaluate the effect of high and low concentration of Injinsaryungsan and high and low concentration of Sosihotang on cholestatic liver injery induced by $ANIT({\alpha}-naphthylisothiocyanate)$, biochemical changes in serum transaminase(GOT, GPT), alkaline phosphate, lactate dehydrogenase, total cholesterol, triglyceride, total-bilirubine were studied and the following results were obtained. 1. High concentration of Injinsaryungsan(2.2g/Kg) inhibited significantly the activity increases of GOT, GPT, ALP, LDH, TC, TG, T-Bilirubine induced by $ANIT({\alpha}-naphthylisothiocyanate)$. 2. Low concentration of Injinsaryungsan(1.1g/Kg) inhibited the activity increases of ALP, LDH, TC, TG with statistical significance, while inhibited the activity increase of GOT ,but with no statistical significance. 3. High concentration of Sosihotang(2.4g/Kg) inhibited the activity increases of LDH, TG, TC with statistical significance while inhibited the activity increases of GOT, GPT, ALP, T-bilirubine with no significance. 4. Low concentration of Sosihotang(1.2g/Kg) inhibited the activity increase of TG, while inhibited the activity increase of ALP, TC with no statistical sig-nificance, but didn't inhibite the activity increases of GOT, GPT, LDH, T-Bil. These results suggest that Injinsaryungsan has more significant effect on the liver injury induced by $ANIT({\alpha}-naphthylisothiocyanate)$ compared with Sosihotang and so can be applicable clinically to virus hepatitis and cholestatic liver injury. Further study will be required to evaluate the effect of Sosibotang on cholangitis and cholecystitis.

An Effect of Bromobenzene Treatment on the Liver Damage of Rats Previously Fed Low or High Protein Diet (단백식이 조건을 달리하여 성장시킨 흰쥐에 Bromobenzene 투여가 간손상에 미치는 영향)

  • To evaluate an effect of dietary protein on the liver damage, the bromobenzene was intraperitoneally injected to the rats fed a low or high protein diet and then the liver weight per body weight and serum levels of alanine aminotransferase (ALT) activities were determined to demonstrate the differences in liver damage between the groups fed low or high protein diet. Hepatic aniline hydroxylase (AH), glutthione (GSH) content and glutathione s-transferase(GST) activity were also determined to clarify causes of liver damage between the two groups. Increases of liver weight per body weight and serum ALT activities were higher in brombenzene treated rats fed low protein diet than those fed high protein diet. The increasing rate of hepatic AH activity was higher in bromobenzne-treated rats fed low protein diet than that in those fed high protein diet. Furthermore , hepatic glutathione contents and GST activities in bromobenzene-treated rats were higher in rats fed high protein diet than those fed low protein diet. In case of control group, the heaptic glutathione content and GST activity were also higher in rats fed high protein diet than those fed low protein diet.

  • This paper presents a register scheduling and allocation algorithm for high level synthesis. The proposed algorithm executes the low power scheduling to reduce the switching activity using shut down technique which was not unnecessary the calculation through the extraction DFG from VHDL description. Also, the register allocation algorithm determines the minimum register after the life time analysis of all variable. It is minimum the switching activity using graph coloring technique for low power consumption. The proposed algorithm proves the effect through various filter benchmark to adopt a new scheduling and allocation algorithm considering the low power.

SALIVARY REDUCTASE ACTIVITY AND DENTAL CARIES PREVALENCE IN PRESCHOOL CHILDREN (미취학아동의 타액환원효소활성과 치아우식증 이환실태에 관한 연구)

  • The purpose of this study was to do an epidemiological survey of the salivary reductase activity and dental caries prevalence of the large group of preschool children and analyze the validity of the salivary reductase activity test as a caries activity test. One thousand and forty-four preschool children were examined for caries experience and salivary reductase activity by dental survey and the Resazurin Disc Test. 39.8% of children showed low salivary reductase activity, 39.4% showed middle, and 20.8% showed high. Salivary reductase activity increased as age increased. All indexes of caries experience were high when salivary reductase activity was high. There was positive relation between salivary reductase activity and caries experience. The salivary reductase activity of children with no caries experience was lower than that of children with caries experience. The salivary reductase activity of children with low caries experience was lower than that of children with high caries experience. It seemed that the salivary reductase activity test had basic validity as a caries activity test. However, the test's ability to select the small risk group of high caries susceptibility should be enhanced.

  • To evaluate an effect of dietary protein on the intoxication of methanethiol in rats, the methanethiol was intraperitoneally injected to the rats fed a low or high protein diet and then the liver weight per body weight and seurm levels of alanine aminotransferase (ALT) activities were determined to investigate the differences in liver damage between the animal groups fed low protein diet and that fed high protein diet. On the other hand, the hepatic glutathione content and its conjugating enzyme, glutathione S-transferase (GST) activity were determined to clarify the cause of difference in liver function between the two groups. The increasing rate of liver weigh/body wt., serum levels of ALT to its control group were higher in methanethiol-treated rats fed low protein diet than those fed high protein diet. The hepatic content of glutathione and GST activity were higher in rats fed high protein diet than those fed low protein diet and the decreasing rate of hepatic glu-tathione content to its control group was higher in rats fed low protein diet than those fed high protein diet. Furthermore, the hepatic GST activity in methanethiol-treated rats was higher in rats fed high protein diet than those fed low protein diet. In case of control group, the GST activity was also higher in rats fed high protein diet than those fed low protein diet.

The Effect of Progressive Exercise on the Activities of the Elderly (점진적 근력 운동이 노인의 활동에 미치는 효과)

  • For the purpose of disclosing the effects of progressive low intensity exercise on the activity of the elderly, questionnaire survey and physical fitness examination were performed on the subjects who were 65 years old or older. The subjects were assigned into two groups using paired matching on gender and age: experimental group (33 persons) and control groups (35 persons). The subjects in experimental group was put on the low intensity exercise program (exercising upper and lower limbs for 12 weeks using dumbbell and lead-packed weight). The results showed that the difference of physical and social behavior scores among activities before and after exercise were significantly higher in all variables of general characteristics, health habits and status, perception, and physical fitness. The findings of this research suggested that the progressive low intensity exercise improves the activities of the elderly, physical activity, self assessed activity, activities of daily living, instrumental activities of daily living, social behavior of the elderly be affected by general characteristics, health habits and status, perception, and physical fitness.

Studies on the $K^+-dependent$ p-Nitrophenylphosphatase activity of the rat brain (백서 뇌 $K^+-dependent$ p-Nitrophenylphosphatase활성에 관한 연구)

  • In recent years much interesting information about the mechanism of the $Na^+-K^+$ activated ATPase has been obtained from investigation of the $K^+-activated$ phosphatase activity which appears to be catalysed by the same enzyme. Also several studies have indicated that a $K^+-activated p-nitrophenylphosphatase activity is intimately related to the ATPase activity. And then the exact relation of p-nitrophenylphosphatase activity to $Na^+-K^+$ ATPase activity is not known. The effects of some ions and drugs on the p-nitrophenylphosphatase activity of the rat brain were investigated and the results were summarized as follows. 1. The p-nitrophenylphosphatase was stimulated markedly by low concentrations of $K^+$, while the activity was activated slightly in the presence of $Na^+$ and oligomycin. 2. Addition of both ATP and $Na^+$ caused a remarkable increase in the activity of the $K^+-dependent$ phosphatase at low concentrations of $K^+$. 3. In the presence of $Na^+$ and low concentrations of $K^+$, oligomycin activated the p-nitrophenylphosphatase. 4. O1igomycin inhibited the stimulation of the enzyme activity caused by $Na^{+}+ATP$. 5. Ouabain inhibited the $K^+-dependent$ p-nitrophenylphosphatase activity more in the presence of ATP and $Na^+$ than in their absence. 6. Quinidine inhibited both $Na^+-K^+$ ATPase and p-nitrophenylphosphatase. These inhibitory effects of the drug were partially antagonized by increasing $K^+$ concentrations. The sensitivity of the $K^+-dependent$ p-nitrophenylphosphatase to quinidine was greater than the that of $Na^+-K^+$ ATPase.
