The Effect of Micro Nano Multi-Scale Structures on the Surface Wettability (초소수성 표면 개질에 미치는 마이크로 나노 복합구조의 영향)

  • Surface wettability in terms of the size of the micro nano structures has been examined. To evaluate the influence of the nano structures on the contact angles, we fabricated two different kinds of structures: squarepillar-type microstructure with nano-protrusions and without nano-protrusions. Microstructure and nanostructure arrays were fabricated by deep reactive ion etching (DRIE) and reactive ion etching (RIE) processes, respectively. And plasma polymerized fluorocarbon (PPFC) was finally deposited onto the fabricated structures. Average value of the measured contact angles from microstructures with nanoprotrusions was $6.37^{\circ}$ higher than that from microstructures without nano-protrusions. This result indicates that the nano-protrusions give a crucial effect to increase the contact angle.

Anti-oxidant and α-Glucosidase Inhibition Activity of Extracts or Fractions from Diospyros lotus L. Leaves and Quantitative Analysis of Their Flavonoid Compounds (고욤잎 추출물과 분획물의 항산화 및 alpha-glucosdiase 저해 활성 및 플라보노이드 화합물의 정량)

  • Persimmon leaves were commonly consumed as beverages, but were also used as popular folk medicine in Asia. The purpose of this work was to assess the biological activities of Diospyros Lotus L. extracts (DLLE). Various solvent extracts, including n-Hexnae, $CHCl_3$, EtOAc, and n-BuOH fractions, were obtained from the methanol extract of Diospyros Lotus L. leaves. The increasing interest in the powerful biological activity of plant phenolics and flavonoids outlined the necessity for determining their content in medicinal herbs. In this study, the total polyphenol and flavonoid contents (TPC and TFC) in the EA fraction were higher than those of other fractions. The biological activities of DLLE were tested using the 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging activity assay, as well as superoxide dismutase (SOD) activity as an anti-oxidant effect and ${\alpha}$-glucosidase inhibitory activity as an anti-diabetic effect. The EA fraction with high TPC and TFC values showed the highest anti-oxidant effect and high ${\alpha}$-glucosidase inhibition. The EA fractions were further purified into eight fractions using open column chromatography. Higher anti-oxidant and anti-${\alpha}$-glucosidase activity were observed in polar fractions. The content of the flavonoids, including quercein-3-O-rutinoside, kaempferol-3-O-glucoside, myricetin, luteolin, and kaempferol, were analyzed in effective fractions using high-performance liquid chromatography (HPLC). The results suggest that DLLE have anti-oxidative and anti-diabetic effects and thus, have the potential as anti-diabetic materials and as a source for natural health products.

Allelopathy of Tagetes minuta L. Aqueous Extracts on Seed Germination and Root Hair Growth

  • Present paper showed allelopahtic effects of Tagetes minuta aqueous extracts on seed germination and root hair development. Allelopathy of aqueous extracts derived from T. minuta examined using two test plant species (Lotus comiculatus var. japonicus and Lactuca sativa). The seeds of test species were inoculated in petri dishes containing 0, 10,50 and 100% aqueous extracts from T. minuta. At day 5, the relative seed germination ratio to control was evaluated, and the development of seedling root hairs was observed through light microscopy. Seed germination of L. comiculatus var. japonicus was significantly inhibited proportional to the concentrations of aqueous extract, but that of L. sativa wasn't inhibited. The inhibitory allelopathic effect of T. minuta was found in the development and growth of seedling root hairs. It was concluded that the inhibitory allelophatic effects have been to be investigated using various bioassay, for the allelopathy of plant species shows species-specific and organ-specific.

  • Hangover is associated with alcohol metabolism in body after the ingestion of an alcoholic beverage. It has been known that hangover is caused by increasing blood acetaldehyde concentration. This study was carried out to evaluate effect against blood ethanol(EtOH) and acetaidehyde(AcH) on the seven natural products samples(Lotus seed, Sweet chest nut rose, Kohki, Gurume-K, Gurume-J, Phytic acid and Chlorophyll). Also, samples which were selected as good products were mixed. Then, effect against blood EtOH and AcH formulas were tested in vivo. (omitted)

Adding Effect of Lotus Leaf Extract on the Quality of Beef Jerky (우육포의 품질에 영향을 미치는 연잎추출물 첨가효과)

  • This study was carried out in order to investigate the effects of lotus leaf extracts on the quality characteristics of beef jerky, such as chemical composition, free amino acid, Hunter's color, physical properties, water activity, pH, TBARS value, VBN content, total bacterial count and sensory score. According to the results, there were no significant differences in moisture, protein, fat, ash, L-value, b-value, hardness, springiness, cohesiveness, gumminess, chewiness, water activity, pH, flavor, taste, texture, juiciness and overall acceptability. Total free amino acid content was highest in T0 and, lowest in T3 (p<0.05). The a-value was highest in T3 (p<0.05). The TBARS value and total bacterial count of T2 and T3 were significantly lower than those of T0 and T1 (p<0.05). The VBN content was lowest in T3 (p<0.05) and the sensory color was highest in T3 (p<0.05).

Quality of Soybean Paste (Doenjang) Prepared with Lotus Root Powder (연근 분말을 첨가한 된장의 품질 특성)

  • Soybean paste (Doenjang) was prepared by adding Lotus root powder (LRP) at $5\~15\%$ (w/w) to improve quality of the Doenjang and to give some functional properties. Moisture content was ranged about $50.05\~54.04\%$ and amino nitrogen content was $635\~648\;mg\%$ following the LRP contents. Crude protein amounts were $11.55\~12.56\%$ that was no difference between test samples. Carbohydrates contents increased $1.5\~2$ times in the test samples than the control depending on the LRP contents. However, contents of crude lipid $(6.99\~8.55\%)$ and ash $(13.99\~15.17\%)$ were decreased as increased the amounts of LRP. The pH of the product was decreased until 45 days during aging period and then slightly increased without significantly differences among test samples. The values of acidity were $1.85\~2.18\%$ at the early stage of aging but slightly increased with further aging. The total phenolic content in the Doenjang adding $15\%$ LRP was $461.8\;mg\%$ which is higher than $368.6\;mg\%$ in the control. Doenjang prepared with LRP showed an anti-browning effect of the product itself.

Antioxidant Activity and Nitrite Scavenging Ability of Olive Leaf (Olea europaea L.) Fractions (올리브 잎 분획물의 항산화기능과 아질산염 소거능력 평가)

  • In this study, the antioxidant activities and nitrite scavenging abilities of olive leaf fractions acquired from plants cultivated in Australia (Olea europaea L. var. Picual) and Spain (Olea europaea L. var. Hojiblanca) were evaluated. Oleuropein was found to be the major phenolic compound in the leaves, with the butanol fractions presenting the highest contents. Antioxidant activity was evaluated in terms of superoxide dismutase (SOD)-like activity, 1,1-diphenyl-2-picryl hydroxyl (DPPH) radical scavenging activity, and the inhibitory effect on the auto-oxidation rate of linoleic acid. The SOD-like activities of the olive leaf extracts ranged from 0 to 36.8%. DPPH radical scavenging activity was highest in the ethanol extract of the Australian cultivated olive leaves. Finally, the chloroform fractions of the extracts showed inhibitory effects on the auto-oxidation rate of linoleic acid as well as nitrite scavenging ability.

Antioxidant and Cytoprotective Activity of the Olive Leaf (Olea europaea L. var. Kalamata) Extracts on the Mouse Embryonic Fibroblast Cell

  • Oleuropein content of olive leaf extracts (OLE; ethanol extract) was evaluated by high performance liquid chromatography analysis. Oleuropein contents were $4.21{\pm}0.57$, $3.92{\pm}0.43$, $0.32{\pm}0.03$, $5.76{\pm}0.32$, and $32.47{\pm}0.25$ mg/100 g for ethanol extract, and hexane, chloroform, ethyl acetate, and butanol fraction, respectively. The removal of DPPH free radical increased in OLE and all 5 fractions of OLE in a concentration dependent manner. In order to investigate the antioxidant effect of OLE in vitro, 80%(v/v) ethanol OLE, $H_2O_2$, or combined treatment of 80%(v/v) ethanol OLE and $H_2O_2$ were applied on mouse embryonic fibroblast (MEF) cells. Cells were damaged by oxidative stress decreased their viability followed by increasing concentration of $H_2O_2$, but co-treatment of OLE and $H_2O_2$ showed an increase in cell growth about 20% compare to the cells treated with $H_2O_2$. OLE suppresses cytotoxicity induced by $H_2O_2$ in dose dependent manner. OLE treatment on MEF cells was also examined by analyzing cell cycle and apoptotic rate using flow cytometry. Apoptotic and necrotic cell accumulation was decreased in addition of OLE to $H_2O_2$ compare to the oxidative damaged cells. Taken together, these results demonstrated that OLE suppresses cytotoxicity induced by $H_2O_2$ and protect cells against oxidative stress on MEF cells.

Anti-inflammtory effects of the MeOH extract of Petiolus Nelumbinis (LPS로 활성화된 RAW 264.7 cell에서 하경(荷梗)의 염증매개물질 억제효과)

  • Petiolus Nelumbinis, branches of lotus leaf or lotus flower is a traditional oriental herbal medicine widely used for treating a superheat or disorder of qi flow. Although there are many clinical results and literature study, it has been rarely conducted to evaluate the immuno-biological activity. The present study was conducted to examine the anti-inflammatory effects of PNM (Petiolus Nelumbinis MeOH extract) in vitro. To determine cytotoxic concentration of PNM, the cells were treated with PNM for 24 h after LPS addition, and the cell viability was tested by MTT assay. Both of dosages (30 and 100 ${\mu}g/ml$) of PNM had no cytotoxicity. In these concentrations, PNM significantly reduced the elevated levels of NO and $PGE_2$ by LPS. These inhibitory effects of PNM were due to the reduced expressions of iNOS and COX-2 protein. TNF-$\alpha$, IL-1$\beta$ and IL-6 are frequently encountered pro-inflammatory cytokines, and LPS plays a key role in inducing to the massive production of these cytokines. Thus, we next determined the levels of these cytokines. Although PNM had no significant inhibitory effect on the production of TNF-$\alpha$, the elevated levels of IL-1$\beta$ and IL-6 by LPS were dose-dependently reduced in PNM-treated groups. These results demonstrate that PNM has anti-inflammatory effects by inhibiting the production of proinflammatory cytokines, NO and $PGE_2$ in LPS-activated macrophage. Moreover, the reduction of NO and $PGE_2$ levels was due to the inhibition of iNOS and COX-2 protein expression by PNM.

Effect of Treatment with Ozonated Water on Shelf Life of Refrigerated Meat (오존수 처리가 냉장 쇠고기의 화학적 품질에 미치는 영향)

  • Ozone is a strong oxidant and potent disinfecting agent. In this study, volatile basic nitrogen, thiobarbituric acid reactive substances, acid value and pH all of which are quality indicators in meat products, were evaluated. The meat was treated with the ozonated water (0.2 ppm) for 0, 5, 10, 30, and 60 min and then stored at $5^{\circ}C$ for 24 d. The volatile basic nitrogen content of meat was 22.40 mg% after 9 d of storage and 23.24 mg% at 15 d of storage with ozonated water (0.2 ppm) treatment. During 24 d of storage, the pH, thiobarbituric acid reactive substances, and acid value were decreased when subjected to ozonated water treatment. These results suggested that the ozonated water treatment effectively improved the chemical properties and food safety.
