형개련교탕(荊芥連翹湯) 추출물의 경구투여(經口投與)가 rat의 수태능(受胎能) 및 초기(初期) 배발생(胚發生)에 미치는 영향(影響) (The Effect of Hyeonggaeyeongyotang Extract on the Fertility, Early Embryonic Development in Wistar rats by Oral Gavage Administration)

  • Objectives : this study was to access the effect of Hyeonggaeyeongyotang water extracts, a polyherbal formula has been used as folk medicine, on the fertility and early embryonic development of male and female Wistar rats when administered by oral gavage. Methods : In male rats, Hyeonggaeyeongyotang extract were dosed 4 weeks before pairing and 2 weeks after mating including the mating periods up to termination after necropsy of the majority of the females. In female rats, they were dosed 2 weeks before pairing, and from Day 0 to Day 7 of gestation. This study was conducted in accordance with the recommendations of the KFDA Guideline [2005-60] for Detection of Toxicity to Reproduction for Medicinal Products. Results: 1. No Hyeonggaeyeongyotang extract treatment-related changes on the clinical signs and mortalities, the Food consumptions, the Body weights and gains were demonstrated in all dosed levels tested in this study except for 500ml/kg-dosing male group in which a significant(p<0.05) increase of body gains was detected during day 0-7 after dosing. 2. No Hyeonggaeyeongyotang extract treatment-related changes on the pre-coital intervals, the estrus cycles, the mating index, conception rate and fertility index were demonstrated in all dosed levels tested in this study. 3. No Hyeonggaeyeongyotang extract treatment-related gross findings on reproductive organs, the weights of reproductive organs, histopathological findings on reproductive organs, the corpora lutea number, implantation site number, live fetus number, number of resorpted embryo and pre-and post-implatation loss were demonstrated in all dosed levels tested in this study. Conclusions : Base on the results, it is considered that the NOAEL (No-Observed-Adverse-Effect Level) for fertility and early embryonic development toxicity of Hyeonggaeyeongyotang extract was under 2000ml/kg/day in Wistar male and female rats because there no treatment-related changes on the fertility and early embryonic developmental index were demonstrated in all dosed levels tested.

고속충격하중을 받는 강섬유보강콘크리트 패널의 내충격성능 (Impact Resistance of Steel Fiber-Reinforced Concrete Panels Under High Velocity Impact-Load)

  • 본 실험적 연구는 고속 비상체에 의한 강섬유보강콘크리트의 내충격성을 파악하는데 그 목적이 있다. 이 연구에서는 패널두께 대 탄환지름 비가 3.5 이하인 패널 실험체에 강섬유 혼입률, 패널 두께, 충격 속도, 골재 크기를 변수로 조절하면서 고속충격을 가하여서 실험체의 성능을 비교하였다. 강섬유 혼입률이 증가할수록 중량손실량 및 표면 탈락률은 감소하지만, 관입깊이는 증가하는 추세를 보였다. 그리고 충격하중을 받을 때의 거동은 골재 20 mm를 사용하였을 경우 더욱 불리하게 나타났다. 실험결과는 기존 모델에 의한 예측값과 비교하였고, 이를 통해 패널두께 대 탄환지름 비가 3.5 이하일 때 보수적인 예측을 하는것을 확인하였다. 이 중 수정 NDRC 제안식과 ACE 제안식이 Hughes 제안식보다 안정되게 예측하는 것으로 나타났다. 관입깊이와 배면박리한계두께에 있어서는 강섬유 혼입률에 따라서 예측식과 오차가 크게 나타나기도 하지만, 관통깊이는 수정 NDRC 제안식 및 Hughes 제안식에 의해 비교적 정확하게 예측되었다.

  • This thesis is the final report which has long been studied by the author to obtain the design basis for various hydrological constructions with the specific system suitable to the natural environmental conditions in Korea. This report is divided into two parts: one is to estimate runoff volume from watersheds and the other to estimate the peak discharge for a single storm. According to the result of observed runoff record from watersheds, it is known that Kajiyama formula is useful instrument in estimating runoff volume from watersheds in this country. But it has been found that this formula shows us 20-30% less than the actual flow. Therefore, when wihed to bring a better result, the watershed characteristics coefficient in this formula, that is, f-value, should be corrected to 0.5-0.8. As for the method to estimate peak discharge from drainage basin, the author proposes to classify it in two ways; one is small size watershed and the other large size watershed. The maximum -flood discharge rate $Q_p$ and time to peak Pt obtained from the observed record on the small size watershed are compared by various methods and formulas which are based upon the modern hydrological knowledge. But it was fou.d that it. was not a satisfied result. Therefore, the author proposes. tocomputate $Q_p$, to present 4.0-5.0% for the total runoff volume ${\Sigma}Q$.${\Sigma}Q$ is computed under the assumption of 30mm 103s in watershed per day and to change the theoritical total flow volume to one hour dura tion total flow rate when design daily storm is given. Time to peak Pt is derived from three parameters which are u,w,k. These are computed by relationship between total runoff volume (ha-m unit)and $Q_p$. (C.M.S. unit). Finally, the author checked out these results obtained from 51 hydrographs and got a satisfied result. Therefore the author suggested the model of design dimensionless unit-hydrograph. And the author believes that this model will be much available at none runoff record river site. In the large size watersheds in Korea when the maximum discharge occurs, the effective rainfall is two consequtive stormy days. So the loss in watershed was assutned as 6Omm/2days,and the author proposed 3-hour-daration hydrograph flow distribution percentage. This distribution percentage will be sure to form the hydrograph coordinate.

HEVC의 재귀적 CU 구조에 대한 조건부 확률 기반 고속 탐색 알고리즘 (Conditional Probability Based Early Termination of Recursive Coding Unit Structures in HEVC)

  • MPEG과 ITU-T에서 최근 표준화가 진행되고 있는 HEVC는 H.264/AVC에 비해, CU(coding unit), PU(prediction unit), TU(transform unit)의 다양한 형태 분할 단위를 갖는 것을 큰 특징으로 한다. 이 중, CU와 TU는 쿼드트리 형태의 재귀적 분할 구조를 가지도록 구성되는데, 압축 효율은 향상시키지만 높은 부호화 복잡도를 갖는 단점이 있다. 본 논문에서는 이러한 재귀적 분할 구조에서의 rate-distortion cost를 조건부 확률을 이용한 통계적 분석 방법을 사용하여, 분할이 일어나는 경우와 그렇지 않은 경우로 분류하는 방법을 제안한다. 제안한 방법을 HEVC의 재귀적 CU 부호화에 적용한 결과, 부호화 복잡도를 32% 가량 감소시키면서 압축 효율하락은 0.4-0.5%로 억제할 수 있었다. 또한, HM4.0에 구현되어 있는 고속 탐색 알고리즘과 함께 사용하는 경우, 압축 효율 하락을 0.9%로 억제하면서 부호화 복잡도를 1/2로 감소시킬 수 있었다.

  • 공명기의 공명주파수를 계산할 수 있는 전통적인 공식의 정확도를 높이기 위한 추가 관계식을 수치적인 방법으로 얻는다. 그 전통적인 공식은 단순함 때문에 산업계에서 널리 사용되고 있지만, 덕트 소음 저감용 공명기의 공동과 목에 대한 정보를 충분히 제공하지 못한다. 그러므로, 산업 현장에서는 그 공식이 갖는 형상의 불확실성 때문에 실제 공명기 설계에 많은 어려움을 겪고 있다. 이 단점을 극복하기 위해서, 본 연구에서는 수치 계산 결과를 이용하여 추가 근사식을 유도하고자 한다. 이를 위해, 목과 공동의 형상 계수를 정의하고, 공명기의 투과 손실 곡선 상의 피크 주파수와 계산식의 공명 주파수가 일치해야 한다는 조건을 사용하여 형상 계수들 사이의 관계식을 유도하였다. 유도된 관계식의 유효성을 수치적인 방법과 실험적인 방법으로 검증하였다.

Protective Effects of Dodam Water Extract (Dodam) Against Rotenone-Induced Neurotoxicity in Neuro-2A Cells

  • Dodam formula (Dodam) has been used for neurodegenerative disease in Oriental medicine. Dodam is capable of protecting diverse kinds of cells from damage caused by a variety of toxic stimuli. In the present study, we investigated the underlying protective mechanism of Dodam on rotenone-induced cytotoxicity in rat neuroblastoma Neuro-2A cells. Treatment with Neuro-2A cells with rotenone caused the loss of cell viability, and condensation and fragmentation of nuclei, which was associated with the elevation of ROS level, and lipid peroxidation, the increase in Bax/Bcl-2 ratio. Rotenone induced mitochondrial dysfunction characterized by mitochondrial membrane potential loss and cytochrome-c release. These phenotypes induced by rotenone were reversed by pretreatment with Dodam. Our results suggested that major features of rotenone-induced neurotoxicity are partially mediated by mitochondrial dysfunction and oxidative stress, and that Dodam markedly protects Neuro-2A cells from oxidative injury. These data indicated that Dodam might provide a useful therapeutic strategy in treatment of the neurodegenerative diseases caused by oxidative injuries.

Metabolite profiles of ginsenosides Rk1 and Rg5 in zebrafish using ultraperformance liquid chromatography/quadrupole-time-of-flight MS

  • Background: In the present study, metabolite profiles of ginsenosides Rk1 and Rg5 from red ginseng or red notoginseng in zebrafish were qualitatively analyzed with ultraperformance liquid chromatography/quadrupole-time-of-flight MS, and the possible metabolic were pathways proposed. Methods: After exposing to zebrafish for 24 h, we determined the metabolites of ginsenosides Rk1 and Rg5. The chromatography was accomplished on UPLC BEH C18 column using a binary gradient elution of 0.1% formic acetonitrile-0.1% formic acid water. The quasimolecular ions of compounds were analyzed in the negative mode. With reference to quasimolecular ions and MS2 spectra, by comparing with reference standards and matching the empirical molecular formula with that of known published compounds, and then the potential structures of metabolites of ginsenosides Rk1 and Rg5 were acquired. Results: Four and seven metabolites of ginsenoside Rk1 and ginsenoside Rg5, respectively, were identified in zebrafish. The mechanisms involved were further deduced to be desugarization, glucuronidation, sulfation, and dehydroxymethylation pathways. Dehydroxylation and loss of C-17 residue were also metabolic pathways of ginsenoside Rg5 in zebrafish. Conclusion: Loss of glucose at position C-3 and glucuronidation at position C-12 in zebrafish were regarded as the primary physiological processes of ginsenosides Rk1 and Rg5.

Lactobacillus fermentum으로 발효한 쌍화탕의 파골 세포 분화와 난소 적출한 랫트의 골다공증에 미치는 영향 (Effect of Ssangwha-tang Fermented by Lactobacillus fermentum on Osteoclast Differentiation and Osteoporosis of Ovariectomized Rats)

  • Objective : Ssangwha-tang is a traditional medicine formula widely prescribed for a decrease of fatigue after an illness in Korea. The aim of this study is to investigate the effect of Ssangwha-tang fermented by Lactobacillus fermentum (SF) on osteoclast differentiation in vitro and on bone metabolism of an ovariectomized rat in vivo. Methods : Tartrate-resistant acid phosphatase activity and staining were applied to evaluate the formation of osteoclasts. Ovariectomized rats were orally administrated with SF (30 ml/kg/day) for 12 weeks. Serum aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, total cholesterol, high-density lipoprotein-cholesterol, low-density lipoprotein-cholesterol, triglyceride, phosphate, calcium levels were determined. Effect of SF on bone loss were studied by histological analysis and the measurement of bone mineral density. Results : SF significantly inhibited tartrate-resistant acid phosphatase activity and formation of osteoclasts in RAW264.7 cells stimulated by receptor activator for nuclear factor ${\kappa}B$ (NF-${\kappa}B$) ligand (RANKL). In addition, SF significantly decreased the level of triglyceride and increased the level of low-density lipoprotein. Moreover, the decrease of trabeculae of the femur was partially prevented in ovariectomized rats administrated with SF. Conclusions : SF treatment could prevent ovariectomy induced bone loss and its effects could be medicated by the inhibition of osteoclastogenesis.

고지방식이 비만마우스 모델에서 파키스탄산 및 중국산 마황으로 조성된 강지환(降脂丸)의 체중감량 효과 비교 (Comparison of Pakistani Ephedra Herba and Chinese Ephedra Herba Containing Gangjihwan in the Improvement Effects of Weight Loss in a High Fat Diet-Fed Obese Mice)

  • Objectives : This study investigated the improvement effects of Pakistani (DF-a) and Chinese Ephedra herba-containing Gangjihwan (DF-b) on obesity in a high fat diet-fed obese mouse model. Methods : Eight-week-old C57BL/6N mice were divided into four groups: a normal lean group given a standard diet, an obese control group given a high fat diet, and DF-a and DF-b groups given a high fat diet with DF-a (80 mg/kg), and DF-b (80 mg/kg), respectively. After 8 weeks of treatment, body weight gain, feeding efficiency ratio, blood lipid markers, fat weight and histology were examined. Results : 1. Body weight gain and fat mass were significantly decreased in DF-a and DF-b groups compared with control. The extent of decreases was eminent in DF-a group. 2. Feeding efficiency ratio and circulating leptin concentration were significantly decreased in DF-a and DF-b groups compared with control, whereas circulating adiponectin concentration was increased in DF-a and DF-b groups compared with control. 3. Consistent with their effects on body weight gain and fat mass, circulating concentrations of triglyceride, glucose and insulin were decreased in DF-a and DF-b groups compared with control. 4. The size of adipocytes were decreased by DF-a and DF-b compared with control, whereas the adipocyte number per unit area was increased by them, suggesting that DF-a and DF-b decreased the number of large adipocytes. 5. Consistent with their effects on body weight gain, liver fibrosis was reduced in DF-a and DF-b groups compared with control. Conclusions : In conclusion, these results suggest that DF-a and DF-b not only decrease feeding efficiency ratio, plasma leptin concentration, and blood anti-obesity biomarkers, but also reduce fat mass, contributing to the improvement of obesity. DF-a and DF-b also inhibit liver fibrosis. In addition, these effects were similar between Pakistani Ephedra herba and Chinese Ephedra herba-containing Gangjihwan.

첨가물이 냉동반죽의 품질향상에 미치는 영향 (Effects of Additives on the Improvement of Frozen Dough Quality)

  • 본 연구에서는 냉동반죽의 제조시 발생하는 효모 및 글루텐 조직의 물리 화학적 손실로 인한 제품의 품질손실을 효과적으로 억제 예방 할 수 있는 첨가물로서 ascorbic acid, L-cysteine, SSL을 선택하고, 이 첨가물들의 최적 배합비율을 반응표면 분석법을 통하여 도출해 냈다. 그 결과로 ascorbic acid 160.4ppm, L-cysteine 63.1ppm, SSL 0.6%를 첨가하여 냉동반죽을 제조하였을 때 빵의 경도와 관능검사결과 대조구와 매우 유사한 물성을 보여주었다. 선정된 최적배합비율에 xanthan gum 0.3%(formula A)과 Ultra tex-3 5%(formula B)를 첨가한 반죽의 물성을 Farinograph를 통해 측정하였다. 두 처리구 모두 대조구에 비해 안정도가 떨어지는 경향이 나타났으나, development time이 대조구에 비해 짧고 수분흡수율이 높아 이를 이용하여 냉동반죽을 제조할 경우 빵의 부피와 조직에 유익한 효과를 줄것이라고 예상되었다. 냉동반죽을 $-20^{\circ}C$에서 냉동저장시 2주가 경과하면서 대조구의 pH와 총산도가 급격히 변화하기 시작하여 4주 경과 후부터 반죽의 품질특성과 빵의 품질특성모두 급격히 저하되기 시작하였다. 이와는 대조적으로 선정된 formula A와 B는 10주간의 냉동저장기간 동안 반죽 및 빵의 품질 변화가 적고 일정한 품질을 유지할 수 있는 것을 보여주었다. 이상과 같이 연구결과 선정된 ascorbic acid, L-cysteine, SSL, xanthan gum 및 Ultra tex-3 등의 최적배합 비율은 냉동반죽의 저장과정에서 발생할 수 있는 본질적인 문제점들을 효과적으로 극복함으로서 제빵제조시 우수한 품질특성을 갖는 냉동반죽을 제조할 수 있을 것이라 사료된다.이었다. 관능검사 결과 $10^{\circ}C$에서 30일 저장한 초고압처리구가 신맛과 쓴맛이 적고 단맛과 종합적인 기호도가 높아 가장 우수한 품질을 나타내었다.accase의 활성이 나타나지 않았다. 저해제의 영향을 조사한 결과, 일반적으로 구리를 포함하는 단백질의 저해제인 $NaN_{3}$, TGA, DDC를 일정농도로 처리한 실험구에서는 효소의 활성이 완전하게 억제되었으며, p-coumaric acid와 EDTA 처리구에서는 효소의 활성이 억제되지 않았다. 한국산 주걱송편버섯 SCH-3 균주로부터 생산되는 laccase의 N-말단의 아미노산의 서열은 P. coccineus의 laccase와 호주에서 분리 동정된 P.cinnabarinus PB의 laccase와 94%가 같았다.적 특성치 중에서 경도와 씹힘성은 염도와 양의 상관을, 관능적 특성치중에서 전반적인 수용도는 감칠맛과 전반적인 맛에 대한 수용도와 양의 상관을 나타내었다(p<0.05). 적었고 자당(蔗糖)은 칠복(七福)이 가장 많고 원기(元氣)가 가장 낮았으며 가용성전당(可溶性全糖)은 유심(琉心)이 가장 많고 천미(千美)가 가장 적었다. 3. 전단백질함량(全蛋白質含量)은 수원(水原) 147호(號)가 가장 높고 유심(琉心)이 가장 낮았으며 가용성단백질함량(可溶性蛋白質含量)은 칠복(七福)이 가장 높고 유심(琉心)이 가장 낮았으며 전단백질함량(全蛋白質含量)과 수용성단백질함량(水溶性蛋白質含量) 간(間)의 차이(差異)도 각각(各各) 상이(相異)하였다.& Inner part)의 순서등 5품종을 선정하였다.
