Isolation and Characterization of a Gene Encoding Hexokinase from Loquat (Eriobotrya japonica Lindl.)

  • Hexokinase is the first enzyme in the hexose assimilation pathway; it acts as a sensor for plant sugar responses, and it is also important in determining the fruit sugar levels. The full-length cDNA of a hexokinase gene was isolated from loquat through reverse transcription polymerase chain reaction (RT-PCR) and rapid amplification of cDNA ends, which was designated as EjHXK1. EjHXK1 is 1,839 bp long and contains an entire open reading frame encoding 497 amino acids. The predicted protein of EjHXK1 shares 72%-81% similarity with other plant hexokinases. Phylogeny analysis indicated that EjHXK1 is closely related to maize and rice hexokinases. Transient expression of the 35S: EjHXK1-GFP fusion protein was observed on the cell membrane and cytoplasm. Real-time RT-PCR indicated that EjHXK1 is expressed in loquat leaves, stems, flowers, and fruits. EjHXK1 transcripts were higher during early fruit development, but decreases before maturation, which is consistent with hexokinase enzyme activity during fruit development and conducive for hexose accumulation in mature fruits. These results imply that EjHXK1 may play important roles in the regulation of sugar flux during fruit ripening.

Protective Effects of Loquat (Eriobotrya japonica Lindl.) Leaf Extract on Ethanol-Induced Liver Damage in Rats (비파 잎 추출물이 에탄올을 투여한 흰쥐의 간 손상 지표의 개선효과)

  • This study was performed to determine the hepatoprotective effects of ethanol extract of loquat leaf (LL) on alcohol-induced liver damage in rats. Sprague-Dawley rats (n=32) were divided into the following four groups: normal group (NOR), ethanol administrated group (ET), ethanol plus LL 200 mg/kg BW/day administrated group (ET-LLL), and ethanol plus LL 400 mg/kg Bw/day administrated group (ET-LLH). Body weight gain and food intake of the ET group were significantly reduced compared to those of the ET-LLL and ET-LLH groups. Serum aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), and lactate dehydrogenase (LDH) activities elevated by ethanol administration were significantly reduced by LL administration. Serum triglyceride (TG) and total cholesterol (TC) contents and hepatic TG and TC contents of the ET group were significantly elevated compared to those of the NOR group. However, TG and TC contents in the serum and liver were significantly reduced in the ET-LLH group. Hepatic glutathione (GSH) contents of the ET-LLL and ET-LLH groups were significantly elevated, and hepatic thiobarbituric acid reactive substances (TBARS) contents were reduced compared to that of the ET group. Taken together, these results suggest that LL may have a possible protective effect on the improvement of hepatic injury by ethanol administration.

Quality Characteristics of Cookies Prepared with Loquat (Eriobotrya japonica Lindl.) Leaf Powder (비파잎 분말을 첨가한 쿠키의 품질특성)

  • The purpose of this study is to investigate the quality characteristics of cookies prepared with Loquat (Eriobotrya japonica Lindl.) leaf powder (0, 1, 2, 3, and 4%) for flour. The pH of cookie dough decreased significantly in response to the addition of all levels of Loquat leaf powder (LLP). The spread factor of the cookies also increased significantly with more LLP content added to the cookies recipe. In addition, Hunter's color L and a values significantly decreased with increasing LLP content, whereas the b value was increased. According to hardness measurements, the substitution of 1~4% for LLP resulted in increased hardness when samples were compared to the control. Moreover, the addition of LLP resulted in increased DPPH free radical scavenging activity when compared to the control. The results of sensory evaluation showed that the cookies containing 2% had the highest scores.

The Optimal Addition Level of Loquat Leaf Extract in the Rice Beverages (반응 표면 분석을 통한 비파엽 첨가 쌀음료의 비파엽 첨가량의 결정)

  • The optimal amount of loquat leaf extract (LLE, $18^{\circ}$ Brix), fructose, and cream were determined to develop a rice beverage supplemented with LLE. The loquat leaves were extracted by water at $80^{\circ}C$ for 6 hrs and then concentrated to 18% of solid for 1 hr at the same temperature to extraction. To investigate the optimal concentration of the additives, the response surface methodology (RSM) was employed for the experimental design. The addition amounts of the additives were coded. Sweetness, bitterness, off-taste, color, taste preference, and total preference were affected significantly (P<0.05) by the addition of LLE, fructose, and cream. Cream masked the bitterness and off-taste. The total preference of the LLEsupplemented rice beverage showed a lower score than the commercialiy available rice beverages. To achieve the preference level of the commercial rice beverages, the regression between fructose and LLE concentration was calculated as: fructose= $0.618{\times}(LLE)^2+0.406{\times}(LLE)$. Conclusively, cream had an effect of masking the bitterness of rice milk, and the preference was highly related to the addition amount of fructose.

Nutritional Components and Antioxidant Activities of Commercial Loquat (Eryobotrya japonica) Leaf Tea (시판 비파나무잎차의 영양성분 및 항산화 활성)

  • Nutritional components and antioxidant activities of commercial loquat leaf tea (CLLT) were evaluated. Proximate compositions were as follows; moisture 5.34%, crude protein 8.38%, crude fat 13.26%, nitrogen free extract 46.21%, crude fiber 19.24% and crude ash 7.57%, respectively. Ascorbic acid and total phenolics content of CLLT was 0.47 mg/100 g and 42.55 mg/GAE g, respectively. Mineral elements were Ca 1,624.01 mg/100 g, K 1,099.66 mg/100 g, and Mg 192.70 mg/100 g, respectively. Amino acid contents of CLLT were very rich in glutamic acid 565.98 mg/100 g and deficient in cystine 12.97 mg/100 g. The 2,2'-azino-bis (3-ethylbenzthiazoline-6- sulfonic acid) diammonium salt (ABTS), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities of hot water extract from CLLT were 72.51% and 90.11%, respectively at a concentration of $1,000{\mu}g/mL$. Reducing power and ferric reducing antioxidant power (FRAP) of hot water extract from CLLT were increased in a dose dependent manner. Therefore, these results suggest that the hot water extract of CLLT possess antioxidant activities and thus it has great potential as a source for functional food such as natural antioxidant.

Nitrite-scavenging and Antimutagenic Effects of Various Solvent Extract from Different Parts of Loquat (Eriobotrya japonica, Lindl.) (비파 부위별 용매추출물의 아질산염 소거 및 항돌연변이 효과)

  • The nitrite-scavenging and antimutagenic effects of various solvent extracts from different parts of the loquat. were investigated to develope as source of functional food. The nitrite-scavenging and antimutagenic effects of fractions obtained from methanol extract were most highest in ethyl acetate fraction. Ethyl acetate fraction showed better results in the nitrite scavenging activity of 80% in leaf, 81% in seedless fruit, 73% in peel and 63% in seed. The antimutagenic effect showed strong ethyl acetate fraction of 53.1%, 48.3% in leaf and seedless fruit. The antimutagenic effect increased with increasing concentration of ethyl acetate fraction against mutagenicity induced by direct mutagen of IQ in Salmonella typhimurium TA 100. Addition of 2.5 mg and 5 mg of ethyl acetate fraction showed inhibitory effect of 65.5%, 71.2% in leaf and 63.4%, 69.2% in seedless fruit, respectively.

  • This study is the investigation of the tree type of six waterlogged archaeological wood excavated from the remains related to Yun Sundo in Bogildo, Wando Island; the remains had been excavated and surveyed by Jeonnam Cultural property Research Center. The identification result was four Pinus sp. (hard pine), one loquat and one Symplocaceae. All the identified tree types are subtropical ones and they are common in Bogildo, Wando Island.

Antioxidant Activities of Extract Fractions of Leaves from Loquat (Eriobotrya japonica Lindl.) by Cultivars (비파의 품종별 항산화 활성 평가)

  • In this study, we compared the antioxidant activities of three loquat (Eriobotrya japonica Lindl.) by cultivars (Daebang, Bubang and native cultivar). The leaves were extracted by 80% ethanol and then fractionated with various solvents (n-hexane, ethyl acetate, n-butanol, and water) for biological activities and bioactive compounds analysis. Total polyphenol content of extracts and fractions was in the range of 84.93~478.50 mg/g gallic acid equivalent (GAE). From highest to lowest GAE, the fractions were n-butanol> ethyl acetate> n-hexane> 80% ethanol> water. Among the three cultivars, the highest polyphenol content was found from native cultivar. As for DPPH radical scavenging activity, the n-butanol fraction showed the highest activity, and native cultivar was the highest on the $IC_{50}$ values (0.18 mg/mL). In the nitrite scavenging activity, the ethyl acetate fraction (54.99~60.86%) showed the most effective activity on the Bubang cultivar was higher than others. The ursolic acid content of the ethyl acetate fraction showed the highest (51.41 mg/g) in the Daebang cultivar. Based on all these results;the Bubang cultivar showed relatively higher antioxidant and nitrite scavenging activities, but the ursolic acid content was higher in the Daebang cultivar. These results suggest that extracts from loquats (Eriobotrya japonica Lindl.) can be used as bioactive and functional materials that could be important information for industrial use in the future.

Anticancer Effect of Eriobotrya japonica Lindl by Specificity Test with Several Cancer Cell Lines (비파(枇杷)의 항암효과에 대한 암세포특이성 검정)

  • Antitumouric effect of loquat was investigated treating the extract from leaves, stems, fruits or seeds to normal and cancer cell lines to check by MTT method wheather the cancer cells are spec­ifially attacked. The results are summarized as follows. Water of MeOH extract from each organ were applied to human normal and cancer cell lines, SNU-1 and SNU-C4. The water extract from fruit flesh gave no dffect to normal cell lines by killed all the cancer cell lines. The water extract from fruit flesh was purified by Sephadex LH-20 and separated into 9 fractions which were than applied to 8 cancer cell lines. The eighth fraction out of the 9 fractions gave no effect to normal cells but exerted specific cytotoxicity to breast cancer, stomach and liver cancer cells. The eighth fraction was orally administed and injected to 10 mice each suffering from the abdominal cancer induced by myeloma cells, SP2/0-Ag14. In the groups received the treatment, only one mouse each died in 2 months but the rest survived until the end of the experimental period, which those in the control plot died in 10 to 13 days. The present results confirmed that loquat contained some substance that had specific cytotoxicity to human cancer cells.
