비파가루를 첨가한 설기떡의 품질 특성 (Quality Characteristics of Seolgidduk added with Loquat Fruit Powder)

  • This study investigated the optimal mix ratio of frozen-dried loquat fruit powder and the optimun conditions for making Seolgidduk by analyzing chemical properties, rating moisture contents, color, mechanical quality characteristics and sensory characteristics Seolgidduk was made with 0, 2, 4, 6 and 8% frozen-dried loquat fruit powder, which contains functional components. Chemical analysis showed that loquat fruit powder contained of 12.87% water, 2.23% crude protein, 0.55% crude fat, 3.57% crude ash and 3.89% crude fiber. Water content of Seolgidduk containing loquat fruit ranged from 36.58~39.18%. With more loquat fruit powder L value decreased, while the 'a' value and 'b' values increased. According to scanning electronic microscopy, Seolgidduk containing frozen-dried loquat fruit powder showed weak coherence. Hardness decreased with more frozen-dried loquat fruit powder. In the sensory test, Seolgidduk with 4% loquat fruit powder was the most preferred with less sourness, proper softness, moisture and chewiness. Based on the results of this experiment, Seolgidduk with 4% loquat fruit powder showed less hardness than the control group. These results show that quality and preference increased when 4% loquat fruit powder was added to Seolgidduk. Therefore, Seolgidduk with 4% of loquat fruit powder is expected to increase quality and preference of Seolgidduk.

Loquat (Eriobotrya japonica) extracts suppress the adhesion, migration and invasion of human breast cancer cell line

  • We examined the inhibitory effects of loquat methanol extract on the adhesion, migration, invasion and matrix metalloproteinase (MMP) activities of MDA-MB-231 human breast cancer cell line. Cells were cultured with DMSO or with 10, 25, or 50 ${\mu}g/ml$ of loquat methanol extract. Both leaf and seed extracts significantly inhibited growth of MDA-MB-231 cells in a dose-dependent manner, although leaf extract was more effective. Adhesion and migration were significantly inhibited by loquat extracts in a dose-dependent manner. Loquat extract also inhibited the invasion of breast cancer cells in a dose-dependent manner and leaf extract was more effective than seed extract. MMP-2 and MMP-9 activities were also inhibited by loquat extract. Our results indicate that methanol extracts of loquat inhibit the adhesion, migration and invasion of human breast cancer cells partially through the inhibition of MMP activity and leaf extract has more anti-metastatic effects in cell based assay than seed extract. Clinical application of loquat extract as a potent chemopreventive agent may be helpful in limiting breast cancer invasion and metastasis.

Loquat (Eriobotrya japonica) leaf extract inhibits the growth of MDA-MB-231 tumors in nude mouse xenografts and invasion of MDA-MB-231 cells

  • BACKGROUND/OBJECFTIVES: The present study was conducted to examine the inhibitory effect of loquat leaves on MDA-MB-231 cell proliferation and invasion. MATERIALS/METHODS: Female athymic nude mice were given a subcutaneous (s.c.) inoculation of MDA-MB-231 cells and randomly grouped to receive a s.c. injection of either 500 mg/kg ethanol, water extract or vehicle five times a week. Tumor growth, mitotic rate and necrosis were examined. MDA-MB-231 cells were cultured with DMSO or with various concentrations of loquat water or ethanol extract. Proliferation, adhesion, migration, invasion and matrix metalloproteinase (MMP) activity were examined. RESULTS: Tumor growth of xenograft nude mouse was significantly reduced by loquat extracts. The results of mitotic examination revealed that loquat extracts reduced tumor cell division. Both ethanol and water extracts significantly inhibited MDA-MB-231 cell proliferation. The protein expression of ErbB3 was significantly down-regulated by loquat leaf extracts. Loquat leaf extracts increased apoptosis of MDA-MB-231 cells following 24 hour incubation and the ethanol extract was more potent in inducing apoptosis than the water extract. Furthermore, loquat extracts inhibited adhesion, migration and invasion of MDA-MB-231 cells. MMP activity was significantly inhibited by loquat extracts. CONCLUSION: Our results show that extracts of loquat inhibit the growth of tumor in MDA-MB-231 xenograft nude mice and the invasion of human breast cancer cells, indicating the inhibition of tumor cell proliferation and invasion.

Differential Frost Tolerance and Enzymatic Activities in the Leaves and Immature Fruits of Loquat (Eriobotrya japonica Lindl.)

  • In this experiment, five commercial cultivars and one wild species of loquat were used to investigate frost tolerance and enzymatic activities in leaves and young fruits under cold stress at $-3^{\circ}C$. The frost injury, malondialdehyde (MDA) content, and oxygen-scavenging enzyme activities of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT) and ascorbate peroxidase (APX) were studied. This results showed that the wild species 'Wild Oak-leaf' loquat was the most frost tolerant among accessions tested, followed by the cultivar 'Golden Block'. Other cultivars, 'Wu Gong Bai', 'Taicheng 4', 'Xiangzhong 11' and 'Zaozhong 6', were relatively weak in frost tolerance. The enzymatic activities of SOD, POD and CAT increased initially and then decreased as the exposure time increased. However, the enzymatic peak occurred later in the frost-tolerant accession than in the frost-sensitive accession. The correlation coefficients of MDA contents between leaves and immature fruits were from 0.93 to 0.99 in the five commercial loquat cultivars. For the 'Wild Oak-leaf' loquat, the correlation coefficients of MDA and POD were 0.98 and 0.95, respectively, but the coefficients for SOD, CAT and APX were relatively low. In general, there were good correlations between loquat leaves and immature fruits in MDA content and enzyme activities. These results indicate that analysis of these physiological and biochemical activities in loquat leaves could potentially be used to predict the cold tolerance in loquat at immature fruit stage and to accelerate breeding programs for cold tolerance in loquat.

비파 부위별 추출물이 3T3-L1 세포와 돼지 지방전구세포의 분화에 미치는 효과 (Effects of Loquat(Eriobotrya japonica Lindl.) Extracts in Different Aerial Components on Differentiation of 3T3-L1 Cells and Pig Preadipocytes)

  • The current study was undertaken to determine the effects of the ethanol extracts of loquat (Eriobotrya japonica Lindl.) seeds, flesh or leaves on the differentiation of 3T3-L1 cells and male pig preadipocytes. The cell number was measured with the MTT assay after trypsin digestion. The cell differentiation was determined by measuring the glycerol-3-phosphate dehydrogenase (GPDH) activity and triglyceride(TG) content. No cytotoxicity was observed from the loquat flesh and leaf ethanol extracts at concentrations of 5, 10, 25, 50, 100 or $200{\mu}g/mL$ in 3T3-L1 cells and pig preadipocytes. However, the cell viability of neither cell line were affected by up $50{\mu}g/mL$ of loquat seed ethanol extract. Treatment with the loquat seed and leaf ethanol extracts significantly suppressed the terminal differentiation of both cell lines in a dose-dependent manner, as confirmed by the decrease in the glycerol-3-phosphate dehydrogenase(GPDH) activity and TG content. Treatment with the loquat seed and leaf ethanol extracts inhibited the GPDH activity and reduced the TG content of both cell types more effectively than that with the loquat flesh ethanol extract. The most potent anti-adipogenic effect was obtained in the case of the ethanol extract of loquat seeds.

Changes in Cell Ca2+ Distribution in Loquat Leaves and Its Effects on Cold Tolerance

  • Calcium has been associated with improved cold tolerance in many crops. The aim of this study was to investigate the changes in leaf cell $Ca^{2+}$ distribution and cell organelle ultrastructure of loquat (Eriobotrya japonica Lindl.) plants in response to cold stress at $-3^{\circ}C$, using transmission electron microscopy (TEM). Two loquat accessions, Zaozhong 6 (a commercial cultivar) and oakleaf loquat (a wild relative) were used. Cold tolerance, as measured by leaf browning rate, was higher in oakleaf plants, and calcium treatment improved cold tolerance in both species. Cold stress first induced inward transport of $Ca^{2+}$ from the intracellular space. Then, the imported $Ca^{2+}$ was aggregated around the chloroplast membrane, finally entering the chloroplast. This pattern of $Ca^{2+}$ distribution in leaf cells occurred earlier in Zaozhong 6 than in the wild loquat. With increasing time of cold exposure, the chloroplast membranes of Zaozhong 6 leaves were damaged, blurred and even disappeared, while those of wild oakleaf loquat leaves maintained their structure longer. In Zaozhong 6, cold stress induced a clear cavity between poorly structured granal thylakoids and vesicles appearing inside the chloroplast, while in oakleaf leaves cold stress had little effect on the ultrastructure of chloroplasts (although chloroplast membranes looked blurred). Loquat leaves accumulated free calcium ions around chloroplasts in response to cold stress, with earlier calcium accumulation occurring in the cold-sensitive cultivar Zaozhong 6 than in wild oakleaf loquat. These results demonstrate that these two loquat species have differences in both cold tolerance and calcium accumulation dynamics.

비파 부위별 에탄올 추출물의 항산화 활성 및 암세포 증식 억제효과 (Effects of Loquat (Eriobotrya japonica Lindl.) Ethanol Extracts of Different Aerial Parts on Antioxidant Activity and Antiproliferation of Human Cancer Cells)

  • 본 연구는 비파 씨, 과육 및 잎의 부위별 에탄올 추출물이 항산화효과 및 암세포 증식 억제효과를 알아보고자 실시하였다. 총 폴리페놀과 총 플라보노이드 함량은 비파 잎 에탄올 추출물에서 가장 높게 나타났으며, 다음으로는 비파 씨, 과육 추출물 순으로 나타났다. 비파 씨, 과육 및 잎 추출물의 DPPH radical 소거능의 $IC_{50}$값은 0.049, 0.063, 0.042 mg/mL로 비파 씨와 잎이 과육에 비하여 항산화 활성이 좋은 것으로 나타났다. Rancimat에 의한 항산화능 측정 결과도 비파 씨와 잎 추출물이 높게 나타났다. 비파 부위별 추출물의 암세포 성장억제 효과를 측정한 결과, 비파 씨 에탄올 추출물은 위암과 폐암세포는 농도 의존적으로 저하시켰으나, 간암세포의 경우는 $400{\mu}g/mL$부터 유의하게 저하시켰다. 비파 잎과 과육 에탄올 추출물은 고농도에서 암세포 증식 억제효과가 나타났다. 이상의 결과 비파 부위별 에탄올 추출물의 항산화 및 암세포 증식 억제효과는 모든 부위에서 나타났으며, 특히 비파 씨 추출물이 가장 좋은 결과를 보였다.

국내산 비파(Eriobotrya japonica Lindley)를 첨가한 Stirred Yoghurt의 제조 (Preparation of Stirred Yoghurt from Milk Added with Korean Loquat (Eriobotrya japonica Lindley))

  • 비파를 첨가한 stirred yoghurt를 제조하기 위하여 비파 착즙액 $15\%$를 Skim milk에 첨가하여 Streptococcus thermophilus 와 Lactobacillus acidophilus 혼합균주로 발효시킨 발효유에 비파 과육 첨가량 $(5\%,\;10\%,\;15\%,\;w/v)$과 올리고당 첨가량 $(10\%,\;15\%,\;20\%,\;25\%,\;w/v)$을 달리하여 제조한 stirred yoghurt의 품질 특성을 조사하였다. 관능검사 결과에서는 비파 과육 $15\%$와 올리고당 $20\%$를 첨가하였을 때 단맛, 신맛과 전체적인 기호도에서 가장 좋게 평가되었다. 색도 측정 결과 비파 과육 $15\%$ stirred yoghurt에 올리고당의 첨가량이 증가할수록 명도(L값)와 적색도(a값)는 감소하였고 황색도(b값)는 증가하였다. Texture 특성은 $15\%$ 비파 과육 첨가 stirred yoghurt에 올리고당 $20\%$ 첨가시 견고성(hardness), 탄성(springiness), 응집성(cohesiveness), 껌성(gumminess)과 탄력성(resilience)등에서 가장 높게 나타났다. 따라서 $15\%$비파 착즙액 첨가 발효유에 $15\%$비파 과육과 $20\%$ 올리고당의 첨가가 비파 첨가 stirred yoghurt 제조를 위해 가장 적합한 것으로 사료되었다

비파(Eriobotrya japonica) 열매 추출물의 산화방지능과 미백 효과 (Antioxidant and whitening effects of loquat (Eriobotrya japonica) fruit extracts)

  • 비파 열매 추출물의 총페놀 함량, 총플라보노드 함량, 산화방지능을 확인한 결과 전중 열매에서 가장 높은 총페놀 함량과 산화방지능(ABTS법)을 보였다. 진왕 열매는 전중 열매와 비교하여 총페놀 함량은 유의적으로 낮았지만, 가장 높은 총플라보노이드 함량과 산화방지능(DPPH법)을 보여주었다. 세 품종의 비파 열매추출물은 B16F1 흑색종 세포 내 산화스트레스를 감소시켰으며, 특히 전중과 진왕 열매 추출물은 멜라닌 합성에 관여하는 타이로시네이즈 활성을 효과적으로 감소시켰다. 하지만 전중 열매 추출물(125 ㎍/mL)을 제외한 모든 처리군에서 멜라닌 생성을 효과적으로 저해시키지는 못하였다. 본 연구 결과를 바탕으로 전중품종의 열매 추출물은 천연 산화방지제 및 피부미백 소재로의 활용 가치가 있는 것으로 판단된다.

비파엽차 제조 및 그 이화학적 특성 (Loquat (Eriobotrya japonica Lindl.) Leaf Tea Processing and Its Physicochemical Properties)

  • 한국산 비파엾을 이용하여 식품으로서의 활용도를 높이기 위한 일환으로 비파엽차를 제조하여 그 이화학적 특성을 분석한 결과는 다음과 같다. 비파엽차의 제조 중 일반성분은 2월 중순에 채취한 비파엽의 수분, 조지방 및 회분 함량은 10월에 비해 낮게 나타났으며, 무기성분은 2, 10월 중순에 채취한 비파엽의 경우 4파 볶음에서 각각 칼륨이 39,916.8, 23,950.0 ppm으로 높게 나타났다. ascorbic acid 유리당, 탄닌 및 카페인 함량은 각각 2.33%, 1.01%, 1.63% 및 113mg%로 2월 중순에 비해 10월에 채취한 비파엽 4차 볶음에서 가장 높게 나타났다. 비파엽차 및 그 침출액에 대한 색도는 10월 중순에 채취한 2차 볶음에서는 증가하였으나 4차 볶음의 경우 2월에 채취한 비파엽에 비해 낮게 나타났다. 또한 10월 중순 4차 볶음의 향기성분과 그 침출액에 대한 유리아미노산 함량은 각각 nerolidol과 DL-allohydroxylysine 성분이 가장 높게 나타났다.
