Effect of Feeding Ammoniated Wheat Straw Treated with and without Hydrochloric Acid on Meat Quality and Various Sensory Attributes of Growing Male Buffalo (Bubalus bubalis) Calves

  • An experiment was conducted to study the effect of feeding ammoniated wheat straw treated with and without HCl on meat quality and various sensory attributes of growing male buffalo (Bubalus bubalis) calves. Due to urea-ammoniation, the CP content of wheat straw increased from 2.90 to 6.96%. The addition of HCl along with urea during urea-ammoniation further increased the CP content to 10.09%. The proximate composition (% fresh basis) of psoas major, longissimus dorsi and semitendinosus were comparable among the groups. However, comparatively higher cumulative muscle mean protein and ash percentage and lower moisture percentage in groups II and III in comparison to group I indicated the desirable effect of feeding AWS and HCl-AWS. The cumulative muscle mean pH in group III (5.54) was comparatively lower than group I (5.65) and II (5.62). The cumulative group mean pH and water holding capacity (% water retained) in semitendinosus was comparatively lower than psoas major and longissimus dorsi. The cumulative muscle mean SSP (g%) was significantly (p<0.05) lower in group III (41.78) than group I (42.45). The cumulative group mean cooking loss (%) was significantly (p<0.01) lower in psoas major (31.61) and longissimus dorsi (29.78) than semitendinosus (35.10). The cumulative group mean SFV (kg/$cm^{2}$) of psoas major, longissimus dorsi and semitendinosus were 6.38, 6.32 and 6.56, respectively, being lowest in psoas major and highest in semitendinosus. However, the cumulative muscle mean SFV (kg/$cm^{2}$) in group I, II and III were 6.53, 6.56 and 6.17, respectively, being lowest in group III and highest in group II. The scores of the cooked (2% common salt) buffen for various sensory attributes viz. appearance, flavour, juiciness, texture, mouth coating and overall palatability were comparable among the groups. Results suggested that feeding of ammoniated wheat straw treated with and without HCl to growing male buffalo calves for 180 days had no adverse effect on the meat quality and various sensory attributes.

한우 성장단계 특이발현 유전자의 발현양상 분석 (Expression Patterns of the Differentially Expressed Genes During Growth Stages of Hanwoo(Korean Cattle))

  • 한우 성장단계 특이발현 유전자 탐색에 관한 연구(장 등, 2002)에서 선정된 후보유전자 중, EPV 20, TCTP 및 aldolase A를 비롯하여 24개월령 cDNA probe에 대하여 강한 signal을 나타낸 ADRP 유전자의 발현양상을 분석하기 위하여 성장단계별 한우 등심조직 시료를 사용하여 semiquantitative RT-PCR 및 northern blot 분석을 실시하였다. EPV 20 유전자는 한우 등심조직에서 성장단계에 따른 발현량 차이가 거의 없는 결과를 근거로, 근육조직에서 항상 발현되는 유전자로 판단하였다. 또한 발달단계에 따라 발현량 차이가 있는 것으로 보고된 aldolase A 유전자 역시 뚜렷한 발현량 차이는 없었으며, aldolase A의 muscle specific promoter와 근육분화 관련 transcription factor에 관한 연구를 통하여 근육분화 및 근육수축에 있어 aldolase A의 역할 및 조절작용을 밝힐 수 있을 것으로 사료된다. 성장관련 단백질로 알려져 있는 TCTP의 발현양상을 분석한 결과, 한우 등심조직에서 성장함에 따라 발현량이 약간 증가하는 양상을 나타내었다. 이와 같은 발현양상 분석결과로 TCTP 유전자는 성장과 관련된 기능이 있지만 동물의 성장에 있어 직접적으로 관여하지는 않을 것으로 판단된다. 지방세포 분화의 초기단계에서 발현량이 급증하고 lipid droplet 형성에 관여하며 지방축적과도 관련이 있는 것으로 보고된 ADRP 유전자의 transcript의 크기는 약 1.82 kb 이었고, 24개월령 한우 등심조직에서 발현량이 급격히 증가하였으며, 30개월령 조직에서는 감소하는 양상을 나타내었다. 이와 같은 결과로부터 ADRP 유전자를 한우 성장단계 특이발현 유전자로 선정하였으며, ADRP 유전자의 발현양상은 근육내 지방축적과 관련이 있을 것으로 추정하였다. 이후에는 발현조절 기작의 해명 및 근육내 지방축적과의 관련성 분석을 위하여 ADRP 유전자의 전체적인 구조 및 전사조절 인자 분석을 비롯하여 다른 지방대사 및 지방축적 관련 유전자와의 상호작용 및 다형성 탐색분석에 관한 연구가 필요 할 것으로 판단하였다.

돼지 저지방 부의 근육들의 육질 특성 (Meat Quality of Pork Muscles from Low-fat Cuts)

  • 돼지고기 저지방 부위 근육들의 특성에 맞는 이용법을 구명하는데 필요한 기초 자료를 제시하고자 삼원교잡종 돼지도체 10두(거세돼지 5두, 암퇘지 5두)에서 분리한 21개 근육의 육질 특성을 조사하였다. 돼지 저지방 부위를 구성하고 있는 주요 21개 근육의 육질특성을 조사한 결과, subscpularis 근육의 pH가 가장 높았고, gluteus superrificialis 근육이 가장 낮았다(p<0.05). 보수력은 subscpularis 근육이 가장 높았으며(p<0.05), 가열감량은 biceps femoris 근육이 가장 많았고, subscpularis 근육이 적었다(p<0.05). 포장감량은 bluteus superrificialis 근육이 가장 많았고(p<0.05) 콜라겐 함량은 infraspinatus와 gastrocnemius 근육이 가장 많았던 반면에 adductor 근육이 가장 적었다(p<0.05). 전단력은 biceps femoris 근육이 가장 높았고, subscpularis 근육이 가장 낮았다(p<0.05). 단백질 용해성은 pectoralis profundi(fan) 근육이 가장 높았고(p<0.05), 겔 강도는 semitendinosus 근육이 가장 높았다(p<0.05). 육색특성에서 명도 값은 tensor fasciae latae 근육이 가장 높았고(p<0.05), 적색도는 superaspinatus, brachiocephalicus, infraspinatus 근육들이 높았으며, 미오글로빈 함량은 vastus intermedius 근육이 가장 높았고, longissimus dorsi 근육이 가장 낮았다(p<0.05). 관능적 특성 중 연도는 infraspinatus 근육이 가장 연하였던 반면에 biceps femoris 근육이 가장 질겼다(p<0.05). 풍미에서는 pectoralis profundi(fan) 근육이 가장 높은 평가를 받았고, vastus intermedius 근육이 가장 낮은 평가를 받았다(p<0.05). 다즙성에서는 supraspinatus, infraspinatus, semitendinosus, vastus intermedius 근육들이 가장 다즙했던 반면에 longissimus dorsi 근육이 가장 건조했다(p<0.05). 전체기호도는 semitendino년와 infraspinatus 근육이 가장 높은 평가를 받았고, biceps femoris와 longissimus dorsi 근육이 가장 낯은 평가를 받았다(p<0.05). 이상의 결과를 종합하면 돼지 저지방 부위를 구성하는 21개 근육들은 육질적인 측면에서 서로 다른 특성들을 나타내는 것으로 조사되었으며, 이러한 결과는 각 근육들의 특성에 적합한 이용방법 개발에 필요한 기초 자료로 유용할 것으로 사료된다.

  • 통계모형의 상호작용 효과를 분석하기 위해 비모수적인 방법인 다중인자 차원 축소(MDR) 방법을 사용해왔다. MDR 방법은 사례-대조 데이터에만 적용 할 수 있다. 본 논문에서는 연속형 데이터에도 적용 할 수 있는 더미(dummy) 변수를 활용한 MDR방법을 소개한다. 아울러 이를 통해 한우의 주요 경제형질인 등심단면적 (longissimus muscle dorsi area: LMA), 도체중(carcass cold weight: CWT), 일당증체량(average daily gain: ADC)에 영향을 주는 우수 유전자 단일염기다형성(SNP)을 규명한다.

  • 소의 질을 평가하기 위해서는 중요한 인자인 근육내 지방(또는 마블링)을 조절하는 분자를 연구해야 한다. cDNA microarray를 사용하여 등지방 조직과 최장근의 유전자발현 차이를 비교하였다. 이 연구를 통해, 우리는 한우의 지방조직에 1211개, 근육조직에서 1346개의 특이 유전자를 확인하였다. bovine chip은 지방조직의 920개 유전자와 근육조직의 760개 유전자로 이루어진 1680개의 특이 유전자로 구성되어있다. 이 실험에서 Microarray 분석은 등지방조직(Cy3)과 최장근(Cy5)의 유전자 발현에 있어서 큰 차이를 보여준다. 차이를 보이는 많은 특이유전자 중에서, 12-리폭시게나아제 유전자와 프로스타글란딘 D 합성효소는 근육내 지방의 축적을 조절하는 중요한 효소이다. 본 연구에서, 일반적으로 발현되지만 한우의 근육과 지방 조직에서 차이를 보이는 많은 유전자를 hybridization 분석을 통해 발견하였다. 선택된 유전자의 발현 수준은 반정량적 RT-PCR을 통해 확인하였고, 그 결과는 cDNA microarray와 유사하였다.

Effects of Maternal Nutrition during Pregnancy on the Body Weight, Muscle Fiber Number, Carcass Traits, and Pork Quality Traits of Offspring

  • The purpose of the current study was to examine the influence of different maternal nutrition treatments during pregnancy on body weight, muscle fiber number, carcass traits, and pork quality traits of offspring. A total of 18 crossbred sows (Landrace${\times}$Yorkshire${\times}$Duroc) were randomly assigned to one of three nutritional treatment groups; control, high energy, and high protein. The control group was fed a standard diet, the high energy group was fed a diet that contained 30% increased metabolizable energy, and the high protein group was fed a diet that contained 30% increased limiting amino acids compared to the control. The sows in each group were fed equal quantities of each diet (1.9 kg/d) for the entire gestational period. A total of 36 piglets from each sow were used to evaluate changes in body weight, muscle fiber number in the longissimus dorsi muscle at birth, carcass traits, and pork quality traits. Birth weight of offspring born to sows in the high energy diet group was significantly higher compared to the high protein diet group (p<0.05). However, body weight of offspring after birth was not significantly different between the groups. Muscle fiber number for the longissimus dorsi muscle at birth was not significantly different between the groups. In addition, there were no significant differences in carcass traits or pork quality traits between offspring born to sows in the control group and those born to sows that received high energy or high protein diets during pregnancy. Based on these results, it appears that maternal nutrition treatment during pregnancy, regardless of whether it is with high energy or high protein diets, does not have a significant effect on body weight, muscle fiber number at birth, carcass traits, or pork quality traits.

Fatty Acid Profiles and Stearoyl-CoA Desaturase Gene Expression in Longissimus dorsi Muscle of Growing Lambs Influenced by Addition of Tea Saponins and Soybean Oil

  • This study was conducted to determine the effects of dietary addition of tea saponins (TS) and soybean oil (SO) on fatty acid profile and gene expression of stearoyl-CoA desaturase (SCD) in longissimus dorsi (LD) muscle of growing lambs. Thirty-two Huzhou lambs were assigned to four dietary treatments in a $2{\times}2$ factorial arrangement with main effects of TS (0 or 3 g/d) and SO (0 or 30 g/kg of diet DM). The diet without additives was considered as NTNS (no TS or SO). After a feeding trial for 60 d, four lambs of each treatment were slaughtered to collect the samples of LD muscle. Percentage of trans-11 vaccenic acid was enhanced (p<0.05) in muscle of lambs fed TS and SO. The proportion of total conjugated linoleic acid (CLA) was increased (p<0.05) by SO, but decreased (p<0.05) by TS in LD muscle. The percentage of total saturated fatty acids in muscle was decreased (p<0.05) by addition of TS and SO, while addition of SO increased (p<0.05) the percentage of total polyunsaturated fatty acids. The ratio of cis-9, trans-11 CLA to tran-11 vaccenic acid was decreased (p<0.05) by TS, but increased (p<0.05) by SO. The same effects were observed in SCD mRNA expression. From these results it is indicated that including TS and SO in the diet of growing lambs affect the fatty acid profiles of LD muscle and that the proportion of cis-9, trans-11 CLA in the muscle influenced by TS and SO may be related to the SCD gene expression.

Association of DNA Methylation Levels with Tissue-specific Expression of Adipogenic and Lipogenic Genes in Longissimus dorsi Muscle of Korean Cattle

  • Epigenetic factors, such as DNA methylation status, may regulate adipogenesis and lipogenesis, thus affecting intramuscular fat (IMF) deposition in longissimus dorsi muscle (LM) of beef cattle. In Korean cattle steers, the LM consists mainly of muscle tissue. However, the LM tissue also contains IMF. We compared the gene expression levels between the IMF and muscle portions of the LM after tissue separation. Real-time polymerase chain reaction analysis showed that the mRNA levels of both adipogenic peroxisome proliferator-activated receptor gamma isoform 1 (PPARG1) and lipogenic fatty acid binding protein 4 (FABP4) were higher (p<0.01) in the IMF than in the muscle portion of the LM. We determined DNA methylation levels of regulatory regions of the PPARG1 and FABP4 genes by pyrosequencing of genomic DNA. DNA methylation levels of two of three CpG sites in the PPARG1 gene promoter region were lower (p<0.05) in the IMF than in the muscle portion of the LM. DNA methylation levels of all five CpG sites from the FABP4 gene promoter region were also lower (p<0.001) in the IMF than in the muscle portion. Thus, mRNA levels of both PPARG1 and FABP4 genes were inversely correlated with DNA methylation levels in regulatory regions of CpG sites of the corresponding gene. Our findings suggest that DNA methylation status regulates tissue-specific expression of adipogenic and lipogenic genes in the IMF and muscle portions of LM tissue in Korean cattle.

Influence of freeze-thawed cycles on pork quality

  • Objective: This study was conducted to determine the effect of freeze-thawed cycles (Fresh meat, F-T 1 cycle and F-T 2 cycles) on the quality characteristics of porcine longissimus dorsi muscle. Methods: A total of 20 three-crossbred pigs (Duroc×[Large White×Landrace]) were randomly obtained from a commercial slaughterhouse in Thailand. Muscle samples were immediately taken from 10 to 11th of the longissimus dorsi for histochemical analysis. The muscles were cut into 2.54 cm-thick chops. A minimum of 20 chops were used for each treatment (fresh meat, freeze-thawed 1 and 2 cycles). Individually chops were packaged in polyethylene bags and frozen at -20℃ for 6 months followed by thawing in refrigerator at 4℃ for 24 h (the 1st freeze-thawed cycle). The freeze-thawed procedure was repeated for two cycles (the 2nd freeze-thawed cycle). Thawing loss, shear force value, citrate synthase activity and muscle fiber characteristics were determined on the muscles. Results: Results showed that increasing of freeze-thawed cycle increased the thawing loss (p<0.01) and citrate synthase activity (p<0.001). Shear force value of fresh meat was higher than freeze-thawed 1 and 2 cycles (F-T 1 cycle and F-T 2 cycles). Freeze-thawed cycles affected muscle characteristics. Muscle fiber area and muscle fiber diameter decreased with an increasing number of freeze-thawed cycles (p<0.001), while the thickness of endomysium and perimysium were increased (p<0.001). Conclusion: Repeated freeze-thawed cycles degraded muscle fiber structure and deteriorated pork quality.

Identification of Superior Single Nucleotide Polymorphisms (SNP) Combinations Related to Economic Traits by Genotype Matrix Mapping (GMM) in Hanwoo (Korean Cattle)

  • It is important to identify genetic interactions related to human diseases or animal traits. Many linear statistical models have been reported but they did not consider genetic interactions. Genotype matrix mapping (GMM) has been developed to identify genetic interactions. This study uses the GMM method to detect superior SNP combinations of the CCDC158 gene that influences average daily gain, marbling score, cold carcass weight and longissimus muscle dorsi area traits in Hanwoo. We evaluated the statistical significance of the major SNP combinations selected by implementing the permutation test of the F-measure. The effect of g.34425+102 A>T (AA), g.8778G>A (GG) and g.4102+36T>G (GT) SNP combinations produced higher performance of average daily gain, marbling score, cold carcass weight and the longissimus muscle dorsi area traits than the effect of a single SNP. GMM is a fast and reliable method for multiple SNP analysis with potential application in marker-assisted selection. GMM may prospectively be used for genetic assessment of quantitative traits after further development.
