Minoxidil Regulates Aging-Like Phenotypes in Rat Cortical Astrocytes In Vitro

  • Mainly due to the slanted focus on the mechanism and regulation of neuronal aging, research on astrocyte aging and its modulation during brain aging is scarce. In this study, we established aged astrocyte culture model by long-term culturing. Cellular senescence was confirmed through SA-β-gal staining as well as through the examination of morphological, molecular, and functional markers. RNA sequencing and functional analysis of astrocytes were performed to further investigate the detailed characteristics of the aged astrocyte model. Along with aged phenotypes, decreased astrocytic proliferation, migration, mitochondrial energetic function and support for neuronal survival and differentiation has been observed in aged astrocytes. In addition, increased expression of cytokines and chemokine-related factors including plasminogen activator inhibitor -1 (PAI-1) was observed in aged astrocytes. Using the RNA sequencing results, we searched potential drugs that can normalize the dysregulated gene expression pattern observed in long-term cultured aged astrocytes. Among several candidates, minoxidil, a pyrimidine-derived anti-hypertensive and anti-pattern hair loss drug, normalized the increased number of SA-β-gal positive cells and nuclear size in aged astrocytes. In addition, minoxidil restored up-regulated activity of PAI-1 and increased mitochondrial superoxide production in aged astrocytes. We concluded that long term culture of astrocytes can be used as a reliable model for the study of astrocyte senescence and minoxidil can be a plausible candidate for the regulation of brain aging.

A Study on the Technology Measuring Partial Discharge for Long Term Aging Experiments of Insulation Materials (장시간 절연체 열화실험을 위한 부분방전측정기술 연구)

  • This paper described the measurement technology to analyze the partial discharge characteristics for long term aging of insulations. This system was consisted of high voltage generation and measurement part, PD detection part, digital conversion and signal processing part. We used the VXI system for digital conversion and signal processing part. In the digital conversion part, we studied the error of partial discharge magnitude and memory capacity for reading digital signal with the sampling rate. In the signal processing part, we showed the program algorithm to count pulses and read peak values of partial discharge. The allowable minimum sampling rate of digizer was decided to 250kS/s through analyzing test. We confirmed that this system was very useful in the study of $\phi-q-n$ characteristics of long term PD experiments with specimens being consisted of internal void defects and CIGRE II electrodes.

Multi-phase Accelerating Test Method of Thermal Aging Considering Heat Generation of Electric Equipment (전기기기의 발열을 고려한 다단계 가속열노화 방법)

  • Thermal aging test is performed to qualify the life time of equipment in thermally aged condition. Due to long life time more than 10 years like as in power plant, the equipment is subjected to the accelerated thermal aging condition which is able to shorten the long aging test period by increasing aging temperature. Normally, conservatism of thermal aging test causes to impose unbalanced and excessive thermal load on components of the equipment, and deformation and damage problems of the components. Additionally, temperature rise of each component through heat generation of the electric equipment leads to long-term problem of the test period. Multi-phase accelerating aging test is to perform thermal aging test in multiple aging conditions after dividing into groups with various components of equipment. The groups might be classified considering various factors such as activation energy, temperature rise, glass transition temperature and melting temperature. In this study, we verify that the multi-phase accelerating aging test method can reduce and equalize the thermal over load of the components and shorten aging test time.

  • A battery management system (BMS) provides some functions for ensuring safety and reliability that includes algorithms estimating battery states. Given the changes caused by various operating conditions, the state-of-health (SOH), which represents a figure of merit of the battery's ability to store and deliver energy, becomes challenging to estimate. Machine learning methods can be applied to perform accurate SOH estimation. In this study, we propose a Long-Term Recurrent Convolutional Network (LRCN) that combines the Convolutional Neural Network (CNN) and Long Short-term Memory (LSTM) to extract aging characteristics and learn temporal mechanisms. The dataset collected by the battery aging experiments of NASA PCoE is used to train models. The input dataset used part of the charging profile. The accuracy of the proposed model is compared with the CNN and LSTM models using the k-fold cross-validation technique. The proposed model achieves a low RMSE of 2.21%, which shows higher accuracy than others in SOH estimation.

  • The Wolsong Nuclear Power Plant (NPP) operates an on-site spent fuel dry storage facility using concrete silo and vertical module systems. This facility must be safely maintained until the spent nuclear fuel (SNF) is transferred to an external interim or final disposal facility, aligning with national policies on spent nuclear fuel management. The concrete silo system, operational since 1992, requires an aging management review for its long-term operation and potential license renewal. This involves comparing aging management programs of different dry storage systems against the U.S. NRC's guidelines for license renewal of spent nuclear fuel dry storage facilities and the U.S. DOE's program for long-term storage. Based on this comparison, a specific aging management program for the silo system was developed. Furthermore, the facility's current practices-periodic checks of surface dose rate, contamination, weld integrity, leakage, surface and groundwater, cumulative dose, and concrete structure-were evaluated for their suitability in managing the silo system's aging. Based on this review, several improvements were proposed.

  • Among the various statistical factors for South Korea, the population has been steadily decreased by lower birthrate. Nevertheless, the number of household is constantly increasing amid population aging and single life style. In general, residential electricity use is more the result of the number of household than the population. Therefore, residential electricity consumption is expected to be far higher for decades to come. The existing long-term load forecasting, however, do not necessarily reflect the growth of single and two-member households. In this respect, this paper proposes the long-term load forecasting for residential users considering the effect of changes of the housing type, and in the case study the changes of the residential load pattern is analyzed for accurate long-term load forecasting.

CREEP-FATIGUE CRACK GROWTH AND CREEP RUPTURE BEHAVIOR IN TYPE 316 STAINLESS STEELS- EFFECT OF HOLD TIME AND AGING TREATMENT

  • High temperature materials in service are subjected to mechanical damage due to operating load and metallurgical damage due to operating temperature. Therefore, when designing or assessing life of high temperature components, both factors must be considered. In this paper, the effect of tensile hold time on high temperature fatigue crack growth and long term prior thermal aging heat treatment on creep rupture behavior were investigated using STS 316L and STS 316 austenitic stainless steels, which are widely used for high temperature components like in automotive exhaust and piping systems. In high temperature fatigue crack growth tests using STS 316L, as tensile hold time increased, crack growth rate decreased in relatively short tensile hold time region. In long term aged specimens, cavity type microcracks have been observed at the interface of grain boundary and coarsened carbide.

Considerations on the Long-term Reliability of On-line Partial Discharge Ceramic Sensor for Thermal Power Generators and its Demonstration in the Field

  • The present study describes the considerations on the long-term reliability of the on-line partial discharge (PD) ceramic sensor for thermal power generators. Voltage acceleration aging tests were carried out under continuous and impulsive thermal aging at more than $100^{\circ}C$, considering the practical service environment. Experimental results show that the sensors have a life that could last for more than 100 years, excellent dielectric characteristics, and insulation strength. In addition, the ceramic on-line PD sensors were installed in a thermal power generator in Korea for demonstration. The results of the PD calibration and test voltage application prove that the on-line ceramic sensors have satisfactory performances for on-line PD measurement.

  • Recently polymer arresters are being used widely but we don't have appropriate long term characteristics test methods. Therefore we need to develop special test facility to evaluate long term reliability of polymer arresters. It's polymeric housing can be degraded by environmental stress and the interface between housing and inner module can be affected by moisture absorption. This moisture absorption can cause leakage current and tracking in the interface. We developed multi stress accelerated ageing test facility to simulate field conditions including UV, temperature, humidity, voltage, salt fog and rain. In addition, we carried out field exposure test at the outdoor test yard and characteristics analysis of field operated specimens to evaluate accelerating factor of this accelerated aging test.
