The Structure and Function of Locus Control Region (Locus Control Region의 구조와 기능)

  • Locus control region (LCR) is a cia-acting element which regulates the transcription of genes in developmental stage and/or tissue-specific pattern. Typically, LCR consists of several DNase I hypersensitive sites (HSs), where the binding motifs for transcriptional activators are present. The binding of activators to the HSs recruits chromatin modifying complexes to the LCR, opening chromatin structure and modifying histones covalently through the locus. LCR forms close physical contact with target gene located at a distance by looping away intervening region. In addition, non-coding RNA is transcribed from LCR toward target genes in continuously acetylated active domain. These structural and functional features of LCR suggest that the LCR plays many roles in chromatin activation and transcriptional regulation.

  • Locals of Control is considered one of the main imporment factors on responsible environmental behaviors(REB). It is defined as an individual's perception of his or ha ability to bring about change through his a her behavior. This psychological construct is divided in two, external and internal. Internal locus of control(ILOC) is imporment as predictor for responsible environmental behaviors. This study was conducted to investigate the Locus of Control and relations with REB in Undergraduate Students. The data were collected mainly through questionnaires and a total of 157 responded questionnaires was analyzed for the study. The statistical methods used in this study were frequency, percentile and correlation. The major findings of this study were (1) Most of undergraduate students had internal locus of control(75.2%). (2) According to region, rural students showed high levels in ILOC and REB.

3C (Chromatin Conformation Capture): A Technique to Study Chromatin Organization (3C (chromatin conformation capture): 크로마틴 입체 구조 연구를 위한 기법)

  • 3C (chromatin conformation capture) is a technique to analyze chromatin organization in nuclei of eukaryotic cells. The procedure of 3C includes the formaldehyde treatment of cells to fix interactions between proteins and between proteins and DNA in chromatin, the digestion of fixed chromatin with restriction enzyme, and the ligation of fragmented DNA. The efficiency of DNA ligation represents proximity between DNA fragments in chromatin organization. Studies in the ${\beta}$-globin locus using 3C showed that the locus control region is in close proximity to the transcriptionally-active globin genes, indicating that chromatin organization has a role in transcriptional regulation of the genes. 3C has been advanced by combining with ChIP and genome-wide sequencing. This review presents the principle and procedure of the 3C technique, the chromatin organization of the ${\beta}$-globin locus explained by 3C, and advanced techniques based on 3C.

Elderly's leisure participation by the type of leisure attitude and leisure constriants (노인의 여가태도 및 여가제약유형에 따른 여가활동 참여도)

  • the purpose of this study was to analyze the relationship between elderly's leissure participation and the type of leisure attitude and leisure constraints. The sample of this study consisted of 485 elderly over 55 years old living in Seoul Daejeon Jeonju and Daegu. Statistics employed for the analysis were frequencies means cross-tabs and multiple regression analysis. The results could be summarized as follows The types of leisure attitude and leisure constraints of elderly was affected by the age educational attaintment health status region total asset income pocket money family relationship locus of control and self-esteem. And educational attaintment health status region family relationship locus of control and self-esteem had effects on leisure participation level of elderly. These findings pointed that psychological factors such as family relation self-esteen and type of leisure attitude and leisure constraints were the variables explaining leisure activities of elderly.

  • An explorative and descriptive study in order to determine the effect of family structure and other socio-demographic variables on health behavior, belief, locus, and value and utilization of public health service was carried out. Data were collected from 1,653 subjects randomly sampled in three areas, Seoul, Kyunggi province, and Cheju province. From Seoul 849 subjects were selected, 397 subjects Kyunggi, and 407 subjects from Cheju, respectively. Self-reporting questionaires were administered during the period from March to June, 1992. The major findings were as follows: 1) The subjects visited herb-doctor's at irregular intervals mainly to have tonic medicine prepared. They preferred herb-doctor's rather than with doctors at clinics and hospitals. Statistically significant difference was found among the regions studied (p<0.05). 2) The reason for visiting hospitals was primarily for treatment of diseases. They preferred hospital because they felt that the hospitals offer much highly reliable treatment services as well as medical accessibility. For the purpose of hospital utilization, statistically significant differences existed among sex, educational level, family type and region. However, no significance was found among sex, educational level, and region (p<0.05). 3) The subjects utilized general hospitals mainly for diagnosis and treatment of diseases. They preferred general hospitals because of their much better facilities and reliability. Statistical significance was found among sex, educational level, and region (p<0.05). 4) The subjects visited dentist at irregular intervals basis. They visited once half a year or three to four months. their purpose of visit was mainly for diagnosis and treatment of diseases. Statistical significance differences were found among educational level, region and economic standard (p<0.05). 5) Whenever their illnesses were mild and the pharmacies was located in nearby they visited to pharmacies. They visited once a month and patient medicines. Statistically significant differences were found among sex, educational level and region (p<0.05). 6) The subjects believed that herb medicine was quite efficacious for treatment of some diseases, particularly by information handed down through time-honored tradition and experience. However, they recognized that the efficacy of folk medicine can vary with type and severity of diseases. Statistical significance was among sex, educational level, region and economic standard (p<0.05). 7) The reason why subjects believed that pray and superstition are effective for treatment of certain type of diseases, particularly in neuropathy, was the belief in God's almighty. Statistically significant differences were found among sex, educational level, regions and economic standard (p<0.05). 8) Most of subjects under same condition preferred western medicine because they believed that it is more scientific and prompts in showing therapeutic effect. Statistical significance was not found in the choice of type of public health service among, regions. But significant differences were found among sex, educational level and region (p<0.05). 9) The subjects looked for pharmacy if they thought the symptom was mild. However, they visited hospitals for chronic disease and general hospitals for emergency treatment. Statistical significances were found among educational level, region and economic standard (p<0.05). 10) Although most of students wanted to have a healthy life as for the component of health standard and value, they think that they are not healthy (p<0.05). As for the health behavior, significant difference was found in the proportion of smoking and drinking between educational level and region (p<0.05). The health locus was affected by educational level, and health behavior was influenced by region, sex and educational level. The utilization of type of public health service was influenced by family type and region, and health belief by region and educational level, and the health values by region and economic standard respectively, most of correlation showed statistical significance. Among them, the highest correlation was seen between locus of control and external/internal locus of control, which is quite obvious. The correlation between health belief and behavior was the next highest, but still low (0.343). All the other variables are low but significant except only a few of those. These findings indicate that health education should be incorporated into the curriculum so as to develop desirable health habit, and ability of self-control in accordance with their growth stages. A systematic and scientific understanding on the herb/folk medicine is needed, and greater reliability of the utilization of public health services are is still required. Health policy for equal distribution of health service throughout the country along the hierarchical health service system and complementary mutual assistance and cooperation among various health organizations are also required.

Anti-stress effects of Sihosogansan in the passive avoidance test and the forced swimming test (시호소간산(柴胡疏肝散)이 스트레스로 인한 기억저하와 우울행동에 미치는 영향)

  • Objective : Investigation of the anti-stress effects of Sihosogansan Methods : Passive avoidance test(PAT) was performed after applying immobilization stress in water to rats. Also, forced swimming test(FST) was performed to another rats and after FST, the degree of Tyrosine Hydroxylase(TH) expression was measured with immunohistochemical method in the regions of locus coeruleus(LC) and ventral tegmental area (VTA). Results : In the PAT after immobilization stress in water, response latency was significantly increased in the Sihosogansan(400mg/kg) group in comparison with the control group. In the FST, immobility was significantly decreased in the Sihosogansan groups (100mg/kg, 400mg/kg), comparing with the control group. Stress-induced TH increases were suppressed in the Sihosogansan groups (100mg/kg, 400mg/kg) at the LC and the VTA region respectively. Conclusion : Sihosogansan can improve memory ability of rats, reduce behavior of depression in rats, decrease TH-immunoreactive cells at the LC and VTA region in rat, and it may be concluded that Sihosogansan has significant effect in reducing stress.

Association between Histone Deacetylase 9 Gene Polymorphism and Stroke in Chinese Han Population

  • Objective : To explore the correlation between the polymorphism of histone deacetylase 9 gene (rs1060499865, rs723296, rs957960) and ischemic stroke (IS) in Chinese Han population in Dali region. Methods : This study included 155 IS patients and 128 healthy physical examinees. TaqMan-polymerase chain reaction technology and multivariate logistic regression were performed. Results : In the case group, there was no polymorphism of rs1060499865 observed in the two groups; whereas on the rs723296 locus the frequencies of C allele and TC genotype were significantly higher than that in the control group, alleles C and T were associated with a 2.158-fold increase in IS risk, and genotypes TC and TT were associated with a 2.269-fold increase in IS risk. The locus rs957960 exhibited no significant difference between the two groups. Conclusion : An association between rs723296 and the risk of IS was found in the Chinese Han population in Dali region. No significant association was found between rs1060499865, rs957960 and IS in the Chinese Han population in Dali region.

Genetic Diversity among Local Populations of the Gold-spotted Pond Frog, Rana plancyi chosenica (Amphibia: Ranidae), Assessed by Mitochondrial Cytochrome b Gene and Control Region Sequences

  • The Gold-spotted pond frog, Rana plancyi chosenica, designated as a vulnerable species by IUCN Red list. This species is a typical example facing local population threats and extinction due to human activities in South Korea. A strategic conservation plan for this endangered species is urgently needed. In order to provide information for future conservation planning, accurate information on the genetic diversity and taxonomic status is needed for the establishment of conservation units for this species. In this study, we used a molecular genetic approach using the mitochondrial cytochrome b gene and control region sequences to find the genetic diversity of gold-spotted pond frogs within South Korea. We sequenced the mitochondrial DNA cytochrome b gene and control region of 77 individuals from 11 populations in South Korea, and one from Chongqing, China. A total of 15 cytochrome b gene haplotypes and 34 control region haplotypes were identified from Korean gold-spotted pond frogs. Mean sequence diversity among Korean gold-spotted pond frogs was 0.31% (0.0-0.8%) and 0.51% (0.0-1.0%), respectively. Most Korean populations had at least one unique haplotype for each locus. The Taean, Ansan and Cheongwon populations had no haplotypes shared with other populations. There was a sequence divergence between Korean and Chinese gold-spotted pond frogs (1.3% for cyt b; 2.9% for control region). Analysis of genetic distances and phylogenetic trees based on both cytochrome b and control region sequences indicate that the Korean gold-spotted pond frog are genetically differentiated from those in China.

The Effect of Core Self-Evaluation of Female Salesperson on Sales Performance

  • Purpose: This study aimed to empirically demonstrate how self-esteem, self-efficacy, locus of control, and neuroticism, which is core self-evaluation factors of door-to-door salespersons within food and cosmetic retail industry, affected the sales performance of female salespersons via job satisfaction and organizational commitment. Research design, data and methodology: Using a 23-item questionnaire constructed based on previous studies, this study selected six domestic door-to-door sales companies and conducted a 1:1 face-to-face survey with female door-to-door salespersons belonging to branches and agencies in Seoul and the capital region. The data of 208 respondents were analyzed. The results of path analysis based on structural equation modeling and mediation analysis through the Sobel test were presented. Results: Self-efficacy and locus of control had the largest effect on job satisfaction and organizational commitment, respectively. Neuroticism, however, did not significantly influence job satisfaction and organizational commitment. Overall, core self-evaluation factors positively affected sales performance via job satisfaction but not via organizational commitment. Conclusions: This study found that factors which were oriented to female salespersons' ability and sales goals, such as self-efficacy and job satisfaction, were considered important in managing them in the door-to-door sales-distribution channels. In the future, policies should be developed for managing female door-to-door salespersons to help them achieve job satisfaction.

A Comparison of the Characteristics between Intake and Non-intake Respondents of Health Foods, and Analysis of Factors Affecting the Willingness to Consume Health foods in Busan and Gyeongnam Region (부산.경남 지역 소비자의 건강식품 섭취자와 비섭취자의 특성 비교 및 섭취 의도에 영향을 미치는 요인 분석)

  • This study compared the characteristics between intake and non-intake respondents of health foods, and analyzed the factors affecting the willingness to consume health foods, in order to provide the educational program for the desirable choice of health foods. The data were collected from 453 adults living in Busan and Gyeongnam through a self-administered questionnaire and were analyzed using SPSS Windows. Approximately 49% of the respondents consumed health foods at the time of survey. According to the results of chi-square and t tests, there were significant differences between the intake and non-intake respondents by variables, which included sex, age, educational level, marriage status, monthly household income, concerns about health, self-evaluation of health status, self-evaluation of health status compared to the same age, concerns about health foods, awareness of health foods, and internal health locus of control. In addition, the results of multinomial logistic regression analysis showed that the factors affecting willingness to consume health foods in the future were sex, educational level, marriage status, concerns about health and health foods, awareness of health foods, powerful others health locus of control, internal health locus of control, and current intake of health foods.
