웅담과 우황약침이 지방세포 대사에 미치는 영향 (The Effect of Fel Ursi and Bovis Calculus Pharmacopuncture on Preadipocyte Proliferation and Lipolysis of Adipocyte, Localized Fat Accumulation)

  • calculus pharmacopuncture. Porcine skin including fat tissue after treated Fel ursi and Bovis calculus pharmacopuncture by means of the dosage dependent variation are investigated the histologic changes after injection of these pharmacopuncture. Results: Following results were obtained from the preadipocyte proliferation and lipolysis of adipocyte and histologic investigation of fat tissue. 1. Fel ursi and Bovis calculus pharmacopuncture showed the effect of decreased preadipocyte proliferation on the high dosage($1mg/m{\ell}$). 2. Fel ursi pharmacopuncture showed the effect of decreased the activity of glycerol-3-phosphate dehydrogenase (GPDH) on the high dosage($1mg/m{\ell}$) and Bovis calculus pharmacopuncture significantly showed from $0.1mg/m{\ell}$ concentration. 3. Fel ursi pharmacopuncture was not showed the effect of lipolysis, but Bovis calculus pharmacopuncture was increased the effect of lipolysis in all concentration significantly. 4. Investigated the histological changes in porcine fat tissue after treated Fel ursi and Bovis calculus pharmacopuncture, we knew that these pharmacopuncture was showed significant activity to the lysis of cell membranes in all concentration. Conclusion : These results suggest that Fel ursi and Bovis calculus pharmacopuncture efficiently induces diminish proliferation of preadipocyte and lipolysis in adipose tissue.

전기자극이 체성분에 미치는 효과 (The Effect of Electrical Stimulation of Body Composition in Obese Person)

  • The purpose of this study is to reveal the effect of electrical stimulation of body composition in obese person. Subjects were 30s to 40s aged healthy workers(2004. 3. 8~4. 17) in the S general Hospital in Suwon and they were brought to manage obesity. Subjects were divided into control group(Female<0.85, Male<0.90) and study group(Female>0.85, Male>0.90) by WHR(waist-hip ratio) that is measured by Automatic body composition analyzer(InBody 3.0). And we divided the study group with randomized methods into group A(n=8) and group B(n=8). Then we compared and analyzed the change of muscle mass, body fat, abdominal girth, WHR, BMI(body mass index) after application of electrical stimulation, three times a week, for 30 minutes in each session with 50 Hz of pulse frequency, $20\;{\mu}s$ or $250\;{\mu}s$ of pulse duration. There was statistically meaningful decrement of body fat(p<0.05) and abdominal girth(p<0.05) but not of body weight, muscle mass, WHR and BMI in the control group after application of electrical stimulation with 50 Hz, $20\;{\mu}s$. There were meaningful change of abdominal girth(p<0.05), WHR(p<0.05) and BMI(p<0.05), but not of body weight, muscle mass and body fat after application of electrical stimulation with 50 Hz, $20\;{\mu}s$ in group A. We applied electrical stimulation with 50 Hz, $250\;{\mu}s$ in group B, then there were meaningful change of body weight(p<0.05), body fat(p<0.01), abdominal girth(p<0.01), WHR(p<0.05) and BMI(p<0.01) but not of muscle mass only. Consequently, the pulse duration is the main parameter of electrical stimulation that affect the body composition of obese person in this study and if we combined the diet control to reduce blood components we could have better result. So it would be more effective to manage localized obesity(in abdomen, thigh, upper arm, etc.) if you apply electrical stimulation considering the pulse duration.

슬관절의 국소적 색소 융모 결절성 활액막염에 대한 관절경적 치료 (Arthroscopic Treatment for Localized Pigmented Villonodular Synovitis of the Knee)

  • 색소 융모 결절성 활액막염은 드물게 발생하는 단관절성 병변으로 비특이적 증상 발현으로 인해 진단이 어려운 것으로 알려져 있다. 본 연구는 관절경적 시술로 진단 및 제거술을 시행 받았던 11명의 환자를 대상으로 임상 양상, 진단 및 치료 결과를 조사하였다. 모든 예에서 조직학 소견상 색소 융모 결절성 활액막염에 합당한 소견을 나타내었다. 6명은 남자였고 5명은 여자였으며, 연령층은 15세에서 59세(평균 34.6세) 였다. 증상발현 기간은 2개월에서 3년으로 관찰 되었고 (평균 29.9개월), 11례 모두에서 수술전 슬관절 동통을 호소하였다. 4례에서는 외상의 과거력이, 9례에서는 1개의 종물이 발견되었으나 나머지 2례에서 각각 2개 3개의 종물이 관찰되었다. 11례 중 5례가 내측 반월판 전각 부위의 활액막에서 발견되었으며, 2례는 슬개하 지방 그리고 나머지는 각각 슬개상부, 후내측 구획, 외측 반월 연골의 전방, 내측 구에서 1례씩 관찰되었다. 수술 후 평균 29.9개월의 추시 관찰상 11례 모두 재발의 소견은 관찰되지 않았다. 색소 융모 결절성 활액막염의 치료에 있어서 관절경 수술은 최소한의 이환율을 가지며 진단과 치료가 동시에 가능한 좋은 치료 방법 중 하나로 사료된다.

침 요법을 통한 국내 비만치료의 최근 경향 (Acupuncture Therapy of the Obesity - Current Status in Korea and Recent Developments)

  • In this study, after we analyzed 44 recent research paper written about obesity treatement, we tried to understand the acupuncture on the obesity treatment of the recent trend. We searched the recent papers with the key word of obesity, obese, and Acupuncture and are limited since 2004 in the search site KISS, KSI. Recently, acupuncture application for obesity treatment were 6 type. Traditional Acupuncture, Auricular Acupuncture, Electro Acupuncture, Herbal Acupuncture, Chi Acupuncture, and Acupoint Catgut Embedding Therapy. Although acupuncture application were effective method to decrease weight and localized fat, they were critical to evaluate to their effectiveness through strict and scientific clinical trials.

추출방식에 따른 방풍통성산(防風通聖散)이 지방세포 대사에 미치는 영향 (The Effect of Bangpungtongsung-san Extracts on Adipocyte Metabolism)

  • Objective : The purpose of this study is to investigate the effects of Bangpungtongsung-san extracts on the preadipocytes proliferation, of 3T3-L1 cell line. lipolysis of adipocytes in rat's epididymis and localized fat accumulation of porcine by extraction methods(alcohol and water). Methods : Diminish 3T3-L1 proliferation and lipogenesis do primary role to reduce obesity. So, 3T3-L1 preadipocyte and adipocytes were performed on cell cultures, and using Sprague-Dawley rats for the lipogenesis, and treated with 0.01-$1mg/m{\ell}$ Bangpungtongsung-san Extracts depend on concentrations. Porcine skin including fat tissue after treated Bangpungtongsung-san Extracts by means of the dosage dependent variation are investigated the histologic changes after injection of these extracts. Results : Following results were obtained from the 3T3-L1 preadipocyte proliferation and lipolysis of adipocyte in rats and histologic investigation of fat tissue. 1. Bangpungtongsung-san extracts were showed the effect of decreased the activity of glycerol-3-phosphate dehydrogenase(GPDH) on the high dosage($1.0mg/m{\ell}$) and Specially, alcohol extract of Bangpungtongsung-san was clear as time goes by high concentration. 3. Bangpungtongsung-san extracts were showed tries to compare the effect of lipolysis, alcohol extract of Bangpungtongsung-san on the high dosage($1.0mg/m{\ell}$) was observed the effect is higher than water extract. 4. Investigated the histological changes in porcine fat tissue after treated Bangpungtongsung-san extracts, we knew that water extract of Bangpungtongsung-san was showed the effect of lipolysis on the high dosage($10.0mg/m{\ell}$) and alcohol extract of Bangpungtongsung-san was showed significant activity to the lysis of cell membranes in all concentration. Conclusion : These results suggest that Bangpungtongsung-san efficiently induces diminish proliferation of preadipocyte and lipolysis in adipose tissue.

Genetic Variation of Growth Hormone Gene and Its Relationship with Milk Production Traits in China Holstein Cows

  • Associations were analysed between polymorphisms localized in intron 3 of the growth hormone gene (GH-MspI) and milk production traits of 543 China Holstein cows. A PCR-RFLP method was used for identification of genotypes. The following frequencies of genotypes and alleles were found: 0.77, 0.21 and 0.02 for +/+, +/- and -/-, respectively, and 0.87 and 0.13 for $GH^+$ and $GH^-$, respectively. Significant differences between herds were observed in the frequencies of both genotypes and alleles. The results of least squares analysis showed that in all three lactation phases the GH +/+ cows yielded most milk (p<0.01 for lactation I and p<0.05 for lactations II and III), whereas +/- cows showed higher milk fat content than +/+ individuals (p<0.05 for lactation I and II, and p<0.01 for lactation III). The +/+ cows yielded more fat than +/- individuals (p<0.01 only in lactation I). The +/+ cows yielded more milk protein than +/- individuals (p<0.01 for lactation I, II, and III). The +/+ cows produced milk of higher protein content than that of +/- individuals (p<0.05 only in lactation II). Based on these results, we conclude that the +/+ of GH locus should be the favored genotype in China Holstein cow breeds for use in marker-assisted selection programmes.

  • The study was designed to observe the effect of different dietary fats on the incidence of colorectal tumor and in vivo cell proliferation in colon carcinogenesis. Male Sprague Dawley rats were intrarectally infused with chemical carcinogen(methylnitrosourea, MNU) and fed 16%(w/w) fat diet containing one of dietary fats(beef tallow, corn oil, perilla oil) for 30 weeks. To measure in vivo cell proliferation, the incorporation of 5-bromo-2-deoxyuridine(BrdU) into DNA was localized using the monoclonal anti-BrdU antibody. Large number of tumors were found in the distal colon and tumor incidence was increased in the order of perilla oil(57.7%)$\alpha$-linolenic acid rich in perilla oil could have a protective effect against colon cancer compared to saturated fatty acid or n-6 linoleic acid.

Identification of New Microsatellite DNAs in the Chromosomal DNA of the Korean Cattle (Hanwoo)

  • To isolate the microsatellites from the chromosomal DNA of the Korean cattle (Hanwoo) and to use those for the genetic selection, four bacteriophage genomic libraries containing the chromosomal DNA of six Hanwoo steers showing the differences in meat quality and quantity were used. Screening of the genomic libraries using $^{32}P-radiolabeled 5'-({CA})_{12}-3$nucleotide as a probe, resulted in isolation of about 3,000 positive candidate bacteriophage clones that contain $(CA)_n$-type dinucleotide microsatellites. After confirming the presence of microsatellite in each positive candidate clone by Southern blot analysis, the DNA fragments that include microsatellite and flanking sequences possessing less than 2 kb in size, were subcloned into plasmid vector. Results from the analysis of microsatellite length polymorphism, using twenty-two PCR primers designed from flanking region of each microsatellite DNA, demonstrated that 208 and 210 alleles of HW-YU-MS#3 were closely related to the economic traits such as marbling score, daily gain, backfat thickness and M. longissimus dorsi area in Hanwoo. Interestingly, HW-YU-MS#3 microsatellite was localized in bovine chromosome 17 on which QTLs related to regulation of the body fat content and muscle ypertrophy locus are previously known to exist. Taken together, the results from the present study suggest the possible use of the two alleles as a DNA marker related to economic trait to select the Hanwoo in the future.

Characterization and comparison of the pathogenicity of viscerotropic velogenic Newcastle disease virus isolates in Korea

  • A total of 18 Newcastle disease virus (NDV) isolates that were recovered from 1949 through 1997 were characterized and pathotyped. All viruses were highly virulent as determined by intracerebral pathogenicity indices ${\geq}1.81$ in day-old. These pathotypes are typical for viscerotropic velogenic NDV (VVNDV) pathotype viruses. Some differences were observed for the chicken red blood cell elution rate and thermostability of the hemagglutinin at $56^{\circ}C$. Three antigenic groups were identified by a hemagglutination-inhibition assay using NDV monoclonal antibodies. And the predominant gross lesions were as follows: discharge from the nasal cavity, tracheal mucus, petechial hemorrhage in the heart fat, kidney urates and hemorrhage with or without necrosis in the gastrointestinal tract. Severe hemorrhagic or necrotic lesions were also noted in the lymphoid organs and were localized primarily in the spleen and cecal tonsil. However, differences in the occurrence and frequency of the gross lesions were observed between the virus strains. Among them, NDV strains that induced neurological symptoms belonged only to genotype VI. This strain had spread throughout Korea during the late 1980s to the 1990s, which suggests that specific VVNDVs genotypes might result in neurological symptoms.
