악안면 골신장술의 치료계획을 위한 3차원 시뮬레이션 프로토콜의 개발 (Development of Computer Assisted 3-D Simulation and Prediction Surgery in Craniofacial Distraction Osteogenesis)

  • There are significant limitations in the precision of mandibular distraction in setting a desired occlusal and facial esthetic outcome. The purpose of this study is to present the simulation method for the distraction osteogenesis treatment planning. 3-D surgery simulation software programs V-works and V-Surgery(Cybermed, Seoul, Korea) were used from the 3D CT data in addition to the conventional data facial photography, panorama and cephalogram, dental cast model. We have utilized already for the various surgical procedures to get information preoperatively for the maxillofacial surgery like cancer localization and reconstructive surgery, orthognathic surgery and implant surgery in the department of Oral and Maxillofacial surgery, Seoul National University Hospital. On the software, bone cutting can be done at any place and any direction. Separated bone segment can be mobilized in all 3 dimensional direction. After the 3D simulation on the software program, mock surgery on the RP model can be performed. This planning method was applied to two hemifacial microsomia patients. With this protocol, we could simulate the movement of bony segment after maxillofacial distraction osteogenesis

추적 레이다용 Dual-band 모노펄스 수신기 (Dual-band Monopulse Receiver for Tracking Radar)

  • 레이다 기술의 국산화가 이슈가 되고 연구 개발에 박차를 가하고 있는 가운데 본 논문의 Dual-band 모노 펄스 수신기는 추적 레이다를 기반으로 한 실험적 수행 결과를 바탕으로 하고 있다. Dual-band 레이다를 Single-band 레이다와 비교해 보면 높은 비용과 소비 전력 등의 단점을 갖고 있지만 재밍이나 검파 거리, 이미지 신호 제거, 강우 감쇄 특성 면에서 이점을 가질 수 있다. 수신기는 X-band RF head 모듈, Ka-band RF head 모듈, 그리고 공통 IF모듈로 구성되며 X-band와 Ka-band의 각 신호는 IF 모듈의 스위칭에 의해 선택되어진다. 또한 IF모듈의 국부 신호단에서는 위상 제어기가 장착되어 합 채널을 기준으로 고각과 방위각 채널의 위상을 제어할 수 있도록 하였다. 측정 결과 전체 이득은 $40{\pm}3 dB$였으며, 송신/수신간 격리도는 39 dBc, 동적 영역 110 dB, 그리고 잡음지수는 X/Ka-band 각각 4.5dB/6.9dB로 나타났다.

Translation elongation factor-1A1 (eEF1A1) localizes to the spine by domain III

  • In vertebrates, there are two variants of eukaryotic peptide elongation factor 1A (eEF1A; formerly eEF-$1{\alpha}$), eEF1A1 and eEF1A2, which have three well-conserved domains ($D_I$, $D_{II}$, and $D_{III}$). In neurons, eEF1A1 is the embryonic type, which is expressed during embryonic development as well as the first two postnatal weeks. In the present study, EGFP-tagged eEF1A1 truncates were expressed in cortical neurons isolated from rat embryo (E18-19). Live cell images of transfected neurons showed that $D_{III}$-containing EGFP-fusion proteins (EGFP-$D_{III}$, -$D_{II-III}$, -$D_{I-III}$) formed clusters that were confined within somatodendritic domains, while $D_{III}$-missing ones (EGFP-$D_I$, -$D_{II}$, -$D_{I-II}$) and control EGFP were homogeneously dispersed throughout the neuron including axons. In dendrites, EGFP-$D_{III}$ was targeted to the heads of spine- and filopodia-like protrusions, where it was colocalized with $SynGAP{\alpha}$, a postsynaptic marker. Our data indicate that $D_{III}$ of eEF1A1 mediates formation of clusters and localization to spines.

Molecular and biochemical characterization of hemoglobinase, a cysteine proteinase, in Paragonimus westermani

  • The mammalian trematode Paragonimus westermani is a typical digenetic parasite, which can cause paragonimiasis in humans. Host tissues and blood cells are important sources of nutrients for development, growth and reproduction of P. westermani. In this study, a cDNA clone encoding a 47 kDa hemoglobinase of P. westermani was characterized by sequencing analysis, and its localization was investigated immunohistochemically. The phylogenetic tree prepared based on the hemoglobinase gene showed high homology with hemoglobinases of Fasciola hepatica and Schistosoma spp. Moreover, recombinant P. westermani hemoglobinase degradaded human hemoglobin at acidic pH (from 3.0 to 5.5) and its activity was almost completely inhibited by E-64, a cysteine proteinase inhibitor. Immunohistochemical studies showed that P. westermani hemoglobinase was localized in the epithelium of the adult worm intestine implying that the protein has a specific function. These observations suggest that hemoglobinase may act as a digestive enzyme for acquisition of nutrients from host hemoglobin. Further investigations may provide insights into hemoglobin catabolism in P. westermani.

  • B. rapa로부터 분리한 BrMT3 유전자를 도입시킨 효모와 애기장대가 카드뮴을 비롯한 중금속에 저항성을 보이는 것이 확인되었고 이 결과를 토대로 이 유전자가 중금속 흡착을 통한 환경 정화 및 스트레스에 내성을 갖는 형질전환 식물체를 개발하는데 유용하게 사용될 것으로 기대된다.

  • 최근 두개부 종양의 방사성물질의 자입과 방사선입체조사에 의한 뇌수술이 개발되어 의료계 에 많은 관심을 끌고 있다. 또한 방사선수술등은 비관혈적인 체외조사이므로 뇌정위수술용 좌표계의 전산화단층촬영을 이용한 표적중심결정이 매우 중요하다. 현재 알려진 방법은 뇌정위수술용좌표계의 전산화단층찰영에 대한 직교성하에서 비교적 정확하게 표적의 위치를 결정하게 되나, 임상현장에서 직교성유지는 실제 어려운 실정이다. 이에 필자들은 임의의 비직교성 스켄하에서 정확한 표적좌표를 얻기위한 알고리즘을 사용하였으며, 표적오차는 평균 0.02$\pm$0.3mm를 보였다.

Development and Evaluation of Maximum-Likelihood Position Estimation with Poisson and Gaussian Noise Models in a Small Gamma Camera

  • It has been reported that maximum-likelihood position-estimation (MLPE) algorithms offer advantages of improved spatial resolution and linearity over conventional Anger algorithm in gamma cameras. The purpose of this study is to evaluate the performances of the noise models, Poisson and Gaussian, in MLPE for the localization of photons in a small gamma camera (SGC) using NaI(Tl) plate and PSPMT. The SGC consists of a single NaI(Tl) crystal, 10 cm diameter and 6 mm thick, optically coupled to a PSPMT (Hamamatsu R3292-07). The PSPMT was read out using a resistive charge divider, which multiplexes 28(X) by 28(Y) cross wire anodes into four channels. Poisson and Gaussian based MLPE methods have been implemented using experimentally measured light response functions. The system resolutions estimated by Poisson and Gaussian based MLPE were 4.3 mm and 4.0 mm, respectively. Integral uniformities were 29.7% and 30.6%, linearities were 1.5 mm and 1.0 mm and count rates were 1463 cps and 1388 cps in Poisson and Gaussian based MLPE, respectively. The results indicate that Gaussian based MLPE, which is convenient to implement, has better performances and is more robust to statistical noise than Poisson based MLPE.

Expression Characterization, Polymorphism and Chromosomal Location of the Porcine Calsarcin-3 Gene

  • Calcineurin is a calmodulin dependent protein that functions as a regulator of muscle cell growth and function. Agents capable of interacting with calcineurin could have important applications in muscle disease treatment as well as in the improvement of livestock production. Calsarcins comprise a family of muscle-specific calcineurin binding proteins which play an important role in modulating the function of calcineurin in muscle cells. Recently, we described the first two members of the calsarcin family (calsarcin-1 and calsarcin-2) in the pig. Here, we characterized the third member of the calsarcin family, calsarcin-3, which is also expressed specifically in skeletal muscle. However, unlike calsarcin-1 and calsarcin-2, the calsarcin-3 mRNA expression in skeletal muscle kept rising throughout the prenatal and postnatal development periods. In addition, radiation hybrid mapping indicated that porcine calsarcin-3 mapped to the distal end of the q arm of pig chromosome 2 (SSC2). A C/T single nucleotide polymorphism site in exon 5 was genotyped using the denaturing high performance liquid chromatography (DHPLC) method and the allele frequencies at this locus were significantly different among breeds.

Overexpression of RUNX3 Inhibits Malignant Behaviour of Eca109 Cells in Vitro and Vivo

  • Runt-related transcription factor 3 (RUNX3) is a tumor suppressor gene whose reduced expression may play an important role in the development and progression of esophageal squamous cell cancer (ESCC). The aim of this study was to investigate the clinical relevance of RUNX3 in ESCC patients and effects of overexpression on biological behaviour of Eca109 cells in vitro and in vivo. Immunohistochemistry was performed to detect the clinical relevance of RUNX3 and lymph node metastasis in 80 ESCC tissues and 40 non-cancerous tissues using the SP method. RT-PCR and Western blotting were applied to assess the RUNX3 level and verify the Eca109 cell line with stable overexpression. Localization of RUNX3 proteins was performed by cell immunofluorescence. CCK-8 and Scrape motility assays were used to determine proliferation and migration and the TUNEL assay to analyze cell apoptosis. Invasive potential was assessed in cell transwell invasion experiments. In nude mice, tumorigenesis in vivo was determined. Results showed decreased expression of RUNX3 in esophageal tissue to be significantly related to lymph node metastasis (LNM) (P<0.01). In addition, construction of a recombinant lentiviral vector and transfection into the human ESCC cell line Eca109 demonstrated that overexpression could inhibit cell proliferation, migration and invasion, and induce apoptosis. The in vivo experiments in mice showed tumorigenicity and invasiveness to be significantly reduced. Taken together, our studies indicate that underexpression of RUNX3 in human ESCC tissue is significantly correlated with progression. Restoration of RUNX3 expression significantly inhibits ESCC cells proliferation, migration, invasion and tumorigenesis.

Cytogenetic Mapping of Carthamus tinctorius L. with Tandemly Repeated DNA Sequences by Fluorescence in situ Hybridization

  • Dual-color fluorescence in situ hybridization karyotype analysis was created using repetitive sequences including two types of rDNA repeats (45S and 5S rDNAs) and Arabidopsis-type telomere sequence repeats. The somatic metaphase cells of Carthamus tinctorius were observed as diploids (2n=2x=24). A symmetrical or slightly asymmetrical karyotype with seven pairs of metacentric and five pairs of submetacentric chromosomes was observed. The lengths of the somatic metaphase chromosomes ranged from 4.18 to $6.53{\mu}m$, with a total length of $60.71{\mu}m$. One locus of 45S rDNA was located on the pericentromeric regions of three pairs of chromosomes and the other pair was situated on the terminal regions of the short arms of a single pair of chromosomes. One locus of 5S rDNA was detected on the interstitial regions of the short arms of two pairs of chromosomes. Arabidopsis-type telomeric repeats were detected on the terminal regions of all pairs of chromosomes. Co-localization of loci between telomeric repeats and 45S rDNA was observed in a single pair of chromosomes. The results provide additional information for the existing physical mapping project of C. tinctorius and will also serve as a benchmark to a more intricate cytogenetic investigation of C. tinctorius and its related species.
