Wavelength selection by loading vector analysis in determining total protein in human serum using near-infrared spectroscopy and Partial Least Squares Regression

  • In multivariate analysis, absorbance spectrum is measured over a band of wavelengths. One does not often pay attention to the size of this wavelength band. However, it is desirable that spectrum is measured at only necessary wavelengths as long as the acceptable accuracy of prediction can be met. In this paper, the method of selecting an optimal band of wavelengths based on the loading vector analysis was proposed and applied for determining total protein in human serum using near-infrared transmission spectroscopy and PLSR. Loading vectors in the full spectrum PLSR were used as reference in selecting wavelengths, but only the first loading vector was used since it explains the spectrum best. Absorbance spectra of sera from 97 outpatients were measured at 1530∼1850 nm with an interval of 2 nm. Total protein concentrations of sera were ranged from 5.1 to 7.7 g/㎗. Spectra were measured by Cary 5E spectrophotometer (Varian, Australia). Serum in the 5 mm-pathlength cuvette was put in the sample beam and air in the reference beam. Full spectrum PLSR was applied to determine total protein from sera. Next, the wavelength region of 1672∼1754 nm was selected based on the first loading vector analysis. Standard Error of Cross Validation (SECV) of full spectrum (1530∼l850 nm) PLSR and selected wavelength PLSR (1672∼1754 nm) was respectively 0.28 and 0.27 g/㎗. The prediction accuracy between the two bands was equal. Wavelength selection based on loading vector in PLSR seemed to be simple and robust in comparison to other methods based on correlation plot, regression vector and genetic algorithm. As a reference of wavelength selection for PLSR, the loading vector has the advantage over the correlation plot since the former is based on multivariate model whereas the latter, on univariate model. Wavelength selection by the first loading vector analysis requires shorter computation time than that by genetic algorithm and needs not smoothing.

Fractographic Analysis Method of Fatigue Fracture Surface under Program and Random Loading for Aluminum Alloy (알루미늄 합금의 랜덤하중 하에서 발생한 피로파면 해석 방법)

  • Striation is a typical pattern observed on the fatigue fracture surface and the spacing is known to correspond to a macroscopic fatigue crack growth rate, and many models for the predict in the formation of such striation have been proposed. However, these theories and methods can't be applied under random loading spectrum. In this study, the fatigue tests were carried out on aluminum alloy under random loading spectrum. The fatigue fracture surfaces were observed in the scanning electron microscope (SEM) and great quantities of SEM micrographs were synthesized and saved in computer system. The space and morphology of several large-scale striations, which are expected to from at the relatively greater load range in loading block, were observed. The crack length for each loading blocks was decided in consideration of regularity and repetition of those striations. It is shown that the applicability of fractographic methods on the fatigue fracture surface under random loading spectrum.

A Study on Fluctuating Pressure Load on High Speed Train Passing through Tunnels

  • The carbody structure of a high speed train passing through a tunnel is subjected to pressure fluctuation. Fatigue strength of the carbody structure against the fluctuating pressure loading should be proved in the design stage for safety. In this study, to get the useful information on the pressure fluctuation in the tunnel, measurement has been conducted during test running of KHST on the high speed line for two years. The measured results were analyzed and arranged to be used for carbody design. A prediction method for the magnitude and frequency of pressure change was proposed and the propagating characteristics of pressure wave was investigated. By statistical analysis for the measured results, a pressure loading spectrum for the high speed train was given. The proposed method can also be used to estimate the pressure loading spectrum for new high speed line at design stage combined with the results of train performance simulation.

  • In this study, the counting method for multi-axial and multi-point load states was proposed. Using this counting method, the load spectrum is generated from the service load history which is measured for boom structure of excavator. Loading state for loading points of boom structure is described as a multi-dimensional state space. From this load spectrum, the stress spectrum was generated by FEM analysis using the superposition of the unit load. The cumulated damage at the severe damage point of In nm structure by the failure example is calculated by Palmgren-Miner's rule. As a result of this study, the fatigue life estimation using the multi-axial and multi-point load counting method is useful.

Evaluation of typhoon induced fatigue damage using health monitoring data for the Tsing Ma Bridge

  • This paper aims to evaluate the effect of typhoons on fatigue damage accumulation in steel decks of long-span suspension bridges. The strain-time histories at critical locations of deck sections of long-span bridges during different typhoons passing the bridge area are investigated by using on-line strain data acquired from the structural health monitoring system installed on the bridge. The fatigue damage models based on Miner's Law and Continuum Damage Mechanics (CDM) are applied to calculate the increment of fatigue damage due to the action of a typhoon. Accumulated fatigue damage during the typhoon is also calculated and compared between Miner's Law and the CDM method. It is found that for the Tsing Ma Bridge case, the stress spectrum generated by a typhoon is significantly different than that generated by normal traffic and its histogram shapes can be described approximately as a Rayleigh distribution. The influence of typhoon loading on accumulative fatigue damage is more significant than that due to normal traffic loading. The increment of fatigue damage generated by hourly stress spectrum for the maximum typhoon loading may be much greater than those for normal traffic loading. It is, therefore, concluded that it is necessary to evaluate typhoon induced fatigue damage for the purpose of accurately evaluating accumulative fatigue damage for long-span bridges located within typhoon prone regions.

Noise Source Identification of a Car A/V System (차량탑재용 A/Y 시스템의 소음원 규명)

  • This paper presents the noise source identification of a car A/V system. There are two different kinds of noise sources noise generated by loading mechanism and rattle noise by externally forced vibration. A dynamometer has been made to produce stationary inertia to the loading mechanism of A/V system. Sound pressure spectra and sound intensity were measured by operating the dynamometer setup as various motor speeds, and the results were analyzed. A dominant rattle noise source about A/V system's components has been found by multi-dimensional spectral analysis. Residual spectrum method was applied for eliminating coherence between the vibration sources. In result, the dominant rattle noise source was identified by partial coherent output spectrum of individual vibration component.

  • Crack closure and growth behavior of physically short fatigue cracks under random loading are Investigated by performing narrow- and wide-band random loading tests for various stress ratios. Artificially prepared two-dimensional, short through-thickness cracks are used. The closure behavior of short cracks under random loading is discussed, comparing with that of short cracks under constant-amplitude loading and also that of long cracks under random loading. Irrespective of random loading spectrum or block length, the crack opening load of short cracks is much lower under random loading than under constant-amplitude loading corresponding to the largest load cycle in a random load history, contrary to the behavior of long cracks that the crack opening load under random loading is nearly the same as or slightly higher than constant-amplitude results. This result indicates that the largest load cycle in a random load history has an effect to enhance crack opening of short cracks.

CCQC modal combination rule using load-dependent Ritz vectors

  • Response spectrum method is still an effective approach for the design of buildings with supplemental dampers. In practice, complex complete quadratic combination (CCQC) rule is always used in the response spectrum method to consider the effect of non-classical damping. The conventional CCQC rule is based on exact complex mode vectors. Sometimes the calculated complex mode vectors may be not excited by the external loading and errors in the structural responses always arise due to the mode truncation. Load-dependent Ritz (LDR) vectors are associated with the external loading and LDR vectors not excited can be automatically excluded. Also, contributions of higher modes are implicitly contained in the LDR vectors in terms of static responses. To improve the calculation efficiency and accuracy, LDR vectors are introduced in the CCQC rule in the present study. Firstly, the generation procedure of LDR vectors suitable for non-classical damping system is presented. Compared to the conventional LDR vectors, the LDR vectors herein are complex-valued and named as complex LDR (CLDR) vectors. Based on the CLDR vectors, the CCQC rule is then rederived and an improved response spectrum method is developed. Finally, the effectiveness of the proposed method in this paper is verified through three typical non-classical damping buildings. Numerical results show that the CLDR vector is superior to the complex mode with the same number in the calculation. Since the generation of CLDR vectors requires less computational cost and storage space, the method proposed in this paper offers an attractive alternative, especially for structures with a large number of degrees of freedom.

  • Grinding System is very difficult to examine closely wear phenomenon or dynamic characterastic because it is very complex and different from a general cutting system, Considering automatization and precision it is very important to examine closely grinding system. In this study grinding wheel surface is acquired by using computer vision system in order to explain wear and loading phenomenon. We investigate the relationship between wear and Fourier spectrum of acquired image and observe the entropy variation in the process of manufacturing.

  • In order to evaluate the earthquake damage level of small and medium - sized bridges without earthquake monitoring system, we proposed an algorithm for estimating the seismic force at the target bridge location using the ground acceleration data from the earthquake observatories near the structure. In general, response spectrum analysis, which is the most widely used dynamic analysis method to design and evaluate the structural system numerically is required a response spectrum to determine the dynamic loading. In this study, selection methods of the three closest observatories from the target structure and estimation method of ground response spectrum at arbitrary locations are developed. The proposed method can consider the distance and phase between the target bridge and the seismic station and from the relationship between the acceleration amplitudes and the location of the selected seismic station, the earthquake loading of the target bridge can be determined. The proposed algorithm is estimated to be more conservative than the response spectrum evaluated by actual earthquake data.
