Study of in Vivo Serum Lipid Regulation with Ulmus macrocarpa Hance Extract in Rats (왕느릅나무 추출물에 의한 설치류 혈중지질 개선 효과에 대한 연구)

  • A previous study reported that Ulmus macrocarpa Hance water extract (UME) can improve hyperlipid metabolism and the involvement of suppressed lipid synthesis through adenosine monophosphate-activated protein kinase (AMPK) pathway regulation was suggested. Further exploration of the lipid metabolism between liver and peripheral tissue was necessary to confirm that work, and so this study aimed to extend the possibility that UME can regulate serum lipids. After a 6-week in vivo trial of oral administration of UME to rats with induced hyperlipidemia, serum levels of triglyceride (TG) and total cholesterol (TC) were seen to decrease while HDL cholesterol increased. The UME administrations also decreased the TC and TG levels from the control in liver analysis. However, the suggestion that UME regulates the AMPK pathway to improve hyperlipid states through the suppression of hepatic lipogenesis seems to be only one part of the extract's effect. Indeed, serum concentrations of apolipoproteins A and B were returned to baseline levels of the control group in response to UME administration whilst the liver lipid content was much reduced; this cannot occur through the suggested suppression of hepatic lipogenesis alone. Therefore, a possible mechanism of UME could be that it improves blood circulation by modulating serum lipid levels through both the prior stimulation of lipid oxidation and the suppression of hepatic lipogenesis.

A Study on the Effects of Magnetic Field and BEP on Hyperlipidemia and Enzymatic Activities in Rats (자기장(磁氣場) 및 BEP 조사(照射)가 식이성(食餌性) 고지혈증(高脂血症)이 유발(誘發)된 흰쥐에 미치는 영향(影響)에 대(對)한 연구(硏究))

  • It has been known that Ki(氣) energy is very effective on many adult diseases. Oriental Medicine has acknowledged Ki as an existing reality and investigated its effects on the body. However, the existence of Ki has not been fully explained. In order to find a conclusive evidence on the existence of Ki, this experiment was done to study the mutual relationship of Ki with a magnetic field and BEP (biological energy projector). The BEP apparatus was irradiated under the magnetic field on rats in the hyperlipidemic induced state. Following criterias were measured in this experiment: weight change, weight of the visceral organs, serum, hepatic lipid peroxide, bleeding time, tissue factor, and etc. The following results were obtained in this study: 1. The weight of rat significantly decreased in the magnetic field treated group and radically reduced in the group treated with both magnetic field and BEP. 2. The weight of liver, heart, and kidney increased in both the magnetic field treated group and magnetic field+BEP group compared to the normal group, but decreased in comparison to the control group. No changes were witnessed in the weight of spleen. 3. Serum and hepatic total cholesterol, total lipid, and lipid peroxide level significantly decreased in both magnetic field treated group and magnetic field+BEP treated group, while lipase activity has increased noticeably. 4. Serum HDL showed a significant increase in both magnetic field treated group and magnetic field+BEP treated group compared to the control group, while LDL and VLDL level decreased significantly. 5. A bleeding time significantly increased in both magnetic field treated group and magnetic field+BEP treated group compared to the control group. A tissue factor value of the lung decreased in the magnetic field treated group and magnetic field+BEP treated groups while increased in the control group. 6. Serum and hepatic lipid peroxide and glutathione level were significantly decreased in the magnetic field treated group and magnetic field+BEP treated group, while hepatic glutathione level was significantly increased compared to the control group. 7. A significant increase was found in the serum hydroxyl radical and SOD activity in the dietary hyperlipidemic rats, and significant decrease was found in the serum lipid peroxide content and superoxidase activity. 8. Hepatic cytosolic enzyme xanthine oxidase and aldehyde oxidase showed a significant decrease in the magnetic field treated group and magnetic field+BEP treated group. Through the above experimental results, one can suggest that the magnetic field with BEP can suppress hyperlipidemia and boost lipid metabolism and restructuring a lipid in liver, which increases the function of liver. To conclude, BEP is considered to show more potent effects under the exposure of magnetic field because magnetic field seems to increase the flow of Ki in the body.

Effects of Freeze-Dried Garlic Powder on Lipid Improvement in Rats Fed a High Fat-Cholesterol Diet (고지방-콜레스테롤 식이 급이 흰쥐에 대한 동결건조 마늘분말의 지질개선 효과)

  • This study was carried out to investigate the effects of freeze-dried garlic powder (FDGP) on the lipid metabolism in rats fed a high fat-cholesterol (HFC) diet for 4 weeks to induce hyperlipidemia. The rats were divided into the following four experimental groups: normal diet group (NC), HFC diet group (HFC), HFC+FDGP 5 mg/100 g body weight (BW) group (FGT-1), and HFC+FDGP 15 mg/100 g BW group (FGT-2). Body weight gains and food efficiency ratios were significantly lower in the FGT groups compared to the HFC group, and the food intake was not significantly different in all experimental groups. Serum total lipid, triglyceride, and total cholesterol levels were significantly lower in the FGT groups compared to the HFC group. Total lipid content of the FGT-2 group was reduced by about 9% and 12% compared to the FGT-1 group and HFC group, respectively. The FGT groups also showed about 10% reductions low density lipoprotein (LDL)-cholesterol, atherogenic index (AI), and cardiac risk factor (CRF) compared to the HFC group. Liver function index (alkaline phosphatase, aspartate aminotransferase, and alanine aminotransferase) and lipid components were significantly lower in the FGT groups compared to the HFC group. Thiobarbituric acid reactive substances content of serum and liver tissue was reduced in the FGT groups compared to the HFC group, and was lower in the FGT-2 group compared to the FGT-1 group in serum. Antioxidant activities in serum and liver tissue were in the order of FGT-2> FGT-1> HFC group, significantly. In conclusion, FDGP effectively reduced the lipid components of serum and liver tissue. Feeding FDGP did not increase high density lipoprotein-cholesterol, but reduced LDL-cholesterol content, AI and CRF, especially. This shows that FDGP, will be effective in preventing and improving cardiovascular diseases such as arteriosclerosis and hyperlipidemia.

Protective Effect of Semisulcospira libertina Extract on Induced Hepatitis in Rats (다슬기 열수 추출물이 간독성이 유도된 흰쥐에 미치는 보호 효과)

  • This study aimed to investigate the protective effect of Semisulcospira libertina extract on liver injury induced by D-galactosamine in rats. After the administration of S. libertina extract, the local fat degeneration and infiltration of inflammatory cells in liver tissues were significantly decreased and peripheral hemorrhages around portal triads and central necrosis around central veins were found to be protective. The elevated levels of plasma ALT, AST, and LDH, the ALP activation lipid peroxidation, and the lipid contents of a damaged liver were recovered in experimental rats administrated with S. libertina extract, suggesting its role in blood enzyme activation and lipid content restoration within damaged rat liver tissues. Moreover, the expression rate of TNF-α, which accelerates inflammation and induces tissue damage and necrosis, was significantly decreased. In addition, the activities of antioxidant enzymes were more effectively upregulated compared to those of the control group induced hepatotoxicity. All data showed that S. libertina extract has a preventive role against liver damages, such as inflammation and tissue necrosis, as instigated with D-galactosamine by improving the activities of blood enzymes and antioxidant enzymes and modulating the expression of inflammation factor, suggest S. libertina extract is an effective medicinal resource for the restoration of hepatotoxicity.

Effects of the Extruded Pellets and Raw Fish-Based Moist Pellet on Growth and Body Composition of Flounder, Paralichthys olivaceus L. for 10 Months (상업용 부상사료와 어분기초생사료가 넙치의 성장과 체조성에 미치는 영향)

  • Effects of the commercially available or formulated extruded pellets (EP) and raw fish-based moist pellet (MP) on growth and body composition of flounder Paralichthys olivaceus L. were evaluated on commercial scale for 10 months. Survivals of flounder fed the MP, EP 1 and EP3 were not significantly different from those of fish fed the EP2 or EP5, but significantly (P<0.05) higher than that of fish fed the EP4. Weight gain of fish fed the MP was not significantly different from that of fish fed the EP3 and EP5, but significantly (P<0.05) higher than that of fish fed the EP1, EP2 or EP4. Improvement in weight gain of flounder fed the high lipid diets (EP1, EP2, EP3) compared to low lipid diet (EP4) in the $55\%$ protein level, and weight gain of fish fed the high lipid diet (MP) compared to low lipid diet (EP5) in the $61\%$ protein level indicated protein-sparing effect of lipid. FER for flounder fed the EP3. which was not significantly different from that for fish fed the EP1 or EP5 was significantly (P<0.05) higher thar. that for fish fed the EP2, EP4 and MP. FER significantly (P<0.05) changed over time. CF of flounder fed the MP was significantly (P<0.05) higher than that of fish fed EP1 or EP4. Moisture content of dorsal muscle in flounder fed the EP2 was significantly (P<0.05) higher than that in fish fed the EP1 or EP4. Crude protein content of dorsal. muscle in flounder fed the EP1, EP4 and EP5 was significantly (P<0.05) higher than that in fish fed the EP2. How- ever, crude lipid content of dorsal muscle, and moisture, crude protein and crude lipid content of liver in flounder was not significantly different among the diets. In considering performance of flounder, it can be concluded that the use of EP3 and EP5 is more recommendable than the MP, which commonly used in flounder farm.

A study on the comparison of antioxidant effects among wild ginseng, cultivated wild ginseng, and cultivated ginseng extracts (자연산 산삼, 산양삼 및 인삼의 항산화능 비교연구)

  • Objective: The objective of this study was to compare the antioxidant effects among wild ginseng, cultivated wild ginseng, and ginseng extracts. Methods: In vitro antioxidant activities were examined by total antioxidant capacity (TAC), oxygen radical scavenging capacity(ORAC), total phenolic content, 1, 1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, inhibition of induced lipid peroxidation using liver mitochondria, reactive oxygen species(ROS) scavenging effect using 2', 7'-dichlorofluorescein(DCF) fluorescence. Results: 1. TAC of 1.5 and 3.75 mg extracts was highest in cultivated wild ginseng, followed by wild ginseng and lowest in ginseng. 2. ORAC of 2, 10, and $20{\mu}g$ extracts was highest in cultivated wild ginseng, followed by wild ginseng and lowest in ginseng. 3. Total phenolic content of 0.375, 0.938, and 1.875 mg extracts was highest in cultivated wild ginseng, followed by wild ginseng and lowest in ginseng. 4. DPPH(1, 1 -Diphenyl-2-picrylhydrazyl) scavenging activity between wild ginseng and cultivated wild ginseng did not differ significantly (p>0.05). 5. Induced lipid peroxidation, measured by TBARS concentration in solution containing rat liver mitochondria incubated in the presence of $FeSO_4$/ascorbic acid was inhibited as amounts of wild ginseng, cultivated wild ginseng, and ginseng extracts increased. TBARS concentration of ginseng extracts were significantly (p<0.05) higher than wild ginseng or cultivated wild ginseng extracts. 6. DCF fluorescence intensity was decreased as concentrations of wild ginseng, cultivated wild ginseng, and ginseng extracts increased, demonstrating that ROS generation was inhibited in a concentrationdependent manner. Conclusions: In summary, the results of this study demonstrate that cultivated wild ginseng extracts had similar antioxidant activities to wild ginseng extracts and greater that of cultivated ginseng extracts.

Effects of Heated Oil and Vitamin E on Lipid Peroxidative Liver Damage in Rat (가열유와 Vitamin E가 흰쥐 간장내의 과산화적 손상에 미치는 영향)

  • In order to investigate the cellular peroxidative damage due to heated oil intake and the preventive effect of vitamin E on it rats were fed heated corn oil with acid value of 4.02 at the level of 10 Cal% and three different levels of vitamin E that were 0, 40 and 200 mg/kg diet. Control group was fed fresh corn oil and 40mg/kg diet of vitamin E. After ech feeding period of 0, 3 and 6 weeks, liver superoxide dismutase (SOD), glutathione peroxidase (GPX) activities and microsomal content of vitamin E and lipid peroxide (LPO) were measured as well as cellular morphology was examined. SOD activities and LPO contents were higher, while GPX activities and vitamin e contents were lower in heated oil groups than control group. Electromicroscopic observation revealed the loss of inner mitochondrial membrane and cristae and irregular arrangement of nuclear membrane and chromatin in heated oil groups. As dietary vitamin e level was increased, SOD activity and LPO content were decreased, but GPX activity and vitamin E content in the liver increased and cellular peroxidative damage reduced progressively. This phenomena was more remarkable in 6 weeks of feeding than 3 weeks.

The Screening of Hepatic Functional Improvement, Liver Protection and Antifibrotic Effect from Dried Extracts of Concha Cipangopaludinae in Rats (랫드에서 전라 추출물의 간기능 개선, 간보호 및 항섬유화 효과 검색)

  • Oxidative stress and its consequent lipid peroxidation exert harmful effects, which have been currently involved in the generation of carbon tetrachloride (CCl₄)-induced fibrosis(cirrhosis). In this study, it was investigated whether dried extract of 田螺(Concha Cipangopaludinae; CC) has liver functional improvement, antioxidative and antifibrotic effect in rats those were induced liver fibrosis by CCl₄ administration. The female Sprague-Dawley rats were divided into 3 groups(Normal, AC, AC-CC) and were observed in 6 weeks. Except for normal group, liver fibrosis(cirrhosis) in rats were developed by CCl₄ administration(0.8 ㎖/rat/week). And the rats were treated with prepared CC(p. o. 2 ㎖/day/rat). At the time of sacrifice, the liver, kidney and spleen were weighed and the ratio of organ weight/body weight was calculated. The MDA, hyp and biochemical parameters(AST, ALT, ALP, t-bili) were measured in sera and liver tissue of rats. The strong yellow color of urine was observed in all CCl₄-treated group compared with normal group, but jaundice didn't appear in CCl₄-treated group. The mortality of CCl₄-treated group is very low(<13%) during 6 weeks of observation time. The ratio of liver/body as well as the weight of liver in CCl₄-treated rats significantly increased compared with that in normal group(p<0.001). The level of clinical parameters in sera of all liver fibrosis(cirrhosis) developed rats were significantly higher than that of normal group(p<0.001-0.05). Especially the value of BUN, ALP, t-bilirubin in AC-CC group showed 20.9%, 19.6%, 47.9% lower than that in AC group. The content of hyp in CCl₄-treated rats was significantly higher than normal group(p<0.001~<0.05), and showed 12.2% lower value in the AC-CC group than AC group(p<0.05). The production of lipid peroxidation(MDA) in sera and liver tissue significantly increased under the fibrotic(cirrhotic) condition(p<0.001~<0.05). Especially the MDA value of AC-CC group in sera significantly 46.5% decreased compared with that of AC group(p<0.05), and the MDA value of AC-CC in liver tissue showed 21.4% lower than that of AC group. Concha Cipangopaludinae can be improved hepatic function, and maybe have effect of liver protection, antioxidation and antifibrosis.

Effects of Green Tea Catechin on Platelet Phospholipase $A_{2}$ Activity and the Liver Antioxidative Defense System in Streptozotocin-induced Diabetic Rats

  • The purpose of the study was to investigate the effects of dietary green tea catechin and vitamin E on the phospholipse {TEX}$A_{2}${/TEX} activity and th antioxidative defense system in streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley male rats weighing 100$\pm$10 gm were randomly assigned to one normal and five STZ-induced diabetic groups. The diabetic groups were assigned either a catechin-free diet (DM group), 0.5% catechin diet (DM-0.5C group), 1% catechin diet (DM-1C group), vitamin E-free diet (DM-0E group), and 400 mg vitamin E per kg diet (DM-400E group) according to the levels of dietary catechin or vitamin E supplementation. The vitamin E levels of the normal, DM, DM-0.5C, and DM-1C groups were 40 mg per kg diet. Diabetes was experimentally induced by an intravenous injection of streptozotocin after 4 weeks of feeding the five experimental diets. The animals were sacrificed on the 6th day of he diabetic state. The body weight gains were lower in all five diabetic groups after the STZ injection. The platelet phospholipase {TEX}$A_{2}${/TEX}({TEX}$PLA_{2}${/TEX}) activity in the diabetic groups was higher than that in the normal group. However, the enzyme activity in the DM-0.5C, DM-1C, and DM-400E groups was lower than that in the DM and DM-0E groups. The cytochrome {TEX}$P_{450}${/TEX} and cytochrome {TEX}$b_{5}${/TEX} content and NADPH-cytochrome {TEX}$P_{450}${/TEX} reductase activity were about 50~110% higher in the DM and DM-0E groups than in the normal group, yet significantly reduced by either catechin or vitamin E supplementation. The superoxide dismutase (SOD) content in the liver did not differ significantly in any of the groups. However, the glutathione peroxidase (GSHpx) activity was generally lower in the diabetic groups, compared with the normal group, whereas that of the DM-0.5C, DM-1C, and DM-400E groups was significantly higher compared with that of the DM and DM-0E groups. The levels of thiobarbituric acid reactive substances (TBARS) in the liver tissue were 148% and 201% higher in the DM and DM-0E groups, respectively, compared with the normal group, however, these levels were reduced by either catechin or vitamin E supplementation (DM-0.5, DM-1C and DM-400E). Accordingly, the present results indicate that STZ-induced diabetic rats exhibited an imbalance between free radical generation and scavenger systems in the liver which led to the acceleration of lipid peroxidation. However, these abnormalities were reduced and the antioxidative defense system was restored by either dietary catechin or vitamin E supplementation. In conclusion, the effects of dietary catechin or vitamin E in streptozotocin-induced diabetic rats would appear to inhibit lipid peroxidation as an anti-oxidant by regulating the activity of {TEX}$PLA_{2}${/TEX}.

Effects of Sipjeondaebotang on the white rat Hypothyroidism Induced by PTU(6-n-propyl-2-thiouracil) (PTU로 유발된 흰쥐의 갑상선 기능저하증에 미치는 십전대보탕(十全大補湯)의 효과)

  • Objectives : This study was to evaluate Sibjeondaebo-tang’s(Shiquan dabutang) effect to rat hypothyroidism which induced by 6-n-propyl-2-thiouracil(PTU)Methods : Aqueous extracts of Sibjeondaebo-tang(Shiquan dabutang)(21.29%) were administered by oral dose of 300 and 150 mg/kg(rat’s body weight) for 42 days from 2 weeks before start of PTU treatment. During this moment the body weight, thyroid gland weights, liver weight, serum thyroid hormone, lipid content and liver antioxidant defense system were measured with histopathological changes of thyroid glands, Liver and these result were compared with LevoT4 0.5mg/kg treated ratsResults : As a result of PTU treatment, marked decreases of body weight, liver weight, serum thyroid hormone levels of T3, T4, triglyceride content and liver CAT activation were observed with marked increases of thyroid gland weight, thyroid gland hormone TSH, activate liver H2O2 and SOD, serum Total cholesterol, HDL and AST amount were admitted. In addition, marked hyperplasia of follicular cells with reduce of follicular colloid contents and sizes were showed at histopathological inspections. These PTU-induces histopathological changes related to hypothyroidism were significantly reduced by treatment of both different dosages of Sibjeondaebo-tang(Shiquan dabutang).Conclusions: This study suggest that Sibjeondaebo-tang(Shiquan dabutang) extracts have favorable effect on the thyroid hormone productions with beneficial effect on hypothyroidism medicated by modulatory effects on antioxidant defense system.
