This study aimed to find out the effect of freeze-drying leek against cadmium poisoning on the cholesterol and enzyme activities in serum and superoxide radical, SOD and catalase in liver and kidney of the male rats during the administrered period. In this experiment, male rats of Sprague-Dawley strain were used. The rats were divided into 3 groups which were fed differently either for 5 weeks or for 10 weeks:basal diet, basal diet and cadmium in water and 3 % leek added diet and cadmium in water. Cadmium was administered ad libiturn 100 ppm CdCl$_{2}$ in water. The followings are the results of this experiment. 1. Leek reduced the cholesterol and the activities of GPT increase resulted from cadmium treatment. 2. Leek reduced the rate of cadmium in liver and kidney. 3. Leek reduced the activities of SOR and catalase in liver and kidney, while it enhanced the activities of SOD. 4. Leek reduced the necrosis and swelling in liver and kidney casused by cadmium treatment. This experiment showed that leek-addition group had protective effect against cadmium poisoning and increased ALPase activities in serum. Leek alleviated GPT activities in serum and cadmium concentration, necrosis, and swelling in liver and kidney. Therefore, this experiment concluded that leek has defensive power against cadmium poisoning.
This study investigated the anti-obesity effect of a pear (Fructus Pyri Pyrifoliae) extract fermented by lactic-acid bacteria on obesity induced by a high-fat diet in rats. Body-weight measurement, blood analysis, and light microscope observation of adipose tissue in liver and epididymis were conducted after 8 weeks. Gene expression of leptin, adiponectin, and tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) in liver cells were also investigated. Compared to CON, PFA and PFB showed 8% weight reduction along with weight reduction of adipose tissue in liver and epididymis. Observing the microstructure of liver cells showed that lipid droplets were smaller in PFA and PFB than in CON. We confirmed that Fructus Pyri Pyrifoliae extract fermented by lactic-acid bacteria can affect gene expression of leptin, adiponectin, and $TNF-{\alpha}$ in liver cells, showing an obesity treatment effect. From the results above, it was observed that weight gain from increased gene expression in adipose cells as well as from the increased proportion of adipose cells caused by a high-fat diet can be statistically significantly reduced by taking Fructus Pyri Pyrifoliae extract fermented by lactic-acid bacteria. Therefore, Fructus Pyri Pyrifoliae extract fermented by lactic-acid bacteria can be effective for preventing and treating obesity by reducing weight and adipose cells.
Objectives : The purpose of this study is to investigate the effect of electro-acupuncture(EA) at ST36 on $CCl_4{^-}$ induced chronic hepatic damage in rats. Methods : The authors performed several experimental items, including measurements of liver index, hematological analysis for WBC, lymphocytes, neutrophils, monocytes, hemoglobin, and biochemical assays for ALT, AST, ALP in serum, and histological analysis of liver tissue. Results : Liver index(liver weight/body weight), the levels of WBC, Neutrophils and Monocytes in blood, the levels of ALT, AST and ALP in serum which were increased by $CCl_4{^-}$ intoxication were reduced significantly by EA at ST36. The levels of lymphocyte and hemoglobin in blood which were decreased by $CCl_4{^-}$ intoxication were significantly increased by EA at ST36. Conclusions : EA at ST36 has a hepatoprotective effect on chronic hepatic damage in $CCl_4{^-}$ intoxicated rats.
In the present study, to determine whether the antioxidative components of Korean red ginseng protect against radiation damage and the possible relationship among the radioprotective effects and antioxidant actions, the effects of total saponin (200 mg/kg, ip) and lipophilic fraction (200 mg/kg, oral) preferment of mice on the survival ratio, major antioxidant enzymes (SOD, catalase and glutathione peroxidase) activities, glutathione levels and lipid peroxidation in the liver were exiled for 2 weeks after whole ${\gamma}$-body ${\gamma}$-irradiation (6.5 Gy). The 30-day survival ratio increased from 10% to 57% and 40% for mice treated with total saponin and lipophilic fraction, respectively. On day 14 after ${\gamma}$-irradiation, the ginseng total saponin pretreatment produced a slight increase of antioxidant enzymes activities and significantly Increased reduced glutathione (GSH) contents (p<0.05) in the liver compared with non-treated group. Pretreatment with ginseng total saponin significantly deceased GSSG/total GSH ratio (p<0.05) without change of GSSG in the liver and inhibited the radiation-induced incense in the hepatic malondialdehyde levels. (p<0.05) In these results, GSH plays an important role in the liver in several detoxifications and the reduction of lipid peroxides. Thus, it appears that total saponin of red ginseng exerts its radioprotective effect by accelerating the production of endogenous antioxidants, such as glutathione from radiation induced damages and thereby oxygen free radicals.
The aim of the present study was to optimize in vitro method for the prediction of drug-induced liver injury using human liver microsomes (HLM). Cytotoxicity test of cyclophosphamide and acetaminophen in HepG2 cells cultured with HLM showed that the newly established condition using 0.375 mg/ml HLM for 24 hr incubation was comparable or more sensitive than the previously established condition using 0.75 mg/ml HLM for 12 hr incubation. Although the cytotoxic effect of troglitazone was completely attenuated by 0.75 mg/ml HLM, it was augmented by 0.375 mg/ml HLM in the presence of the NADPH-generating system. The cytotoxic effect of chlormezanone, a withdrawn drug due to hepatotoxicity in human, was increased by HLM in the presence of the NADPH-generating system. In contrast, the cytotoxic effect of methapyrilene, a withdrawn drug due to hepatotoxicity in rats, was decreased by HLM in the presence of the NADPH-generating system. The present study suggests that the optimized in vitro method using HLM can be useful for the prediction of drug-induced hepatotoxicity.

Conclusion : Use of Western medicine with herbal medicine can improved liver function test of drug-induced liver injury effectively and herbal medicine do not disturb the liver function.
In order to evalute the effects of radiation on mammalian neuronal system, we have examined the effect of gamma-ray radiation on the monoamine oxidase (MAO) activity in monoaminergic neurons. Following the whole body irradiation, MAO activity in the rat brain was measured as well as in the liver for the comparative studies between the neuronal and nonneuronal system. The effects of some radiation protectors and sensitizers were also examined in addition to the $O_2$ effect. The results can be summarized as follows. 1) The MAO activity of rat brain was minimally affected by the radiation dose up to 1,700 cGy Radiation dose above 2,500 cGy inhibited the brain MAO activity by no less than $l0\%.$ MAO-A form was found to be particularly sensitive to radiation. The liver MAO was somewhat inhibited (by about $5\%$) but hardly dependent on the dose of radiation. 2) The inhibitory effect on the brain was initiated immediately by the radiation dose of 2,500 cGy. On the contrary, for the liver, the inhibitory effect became apparent only 2 days after irradiation. 3) Two days after a dose of 2,500 cGy, Vmax and Km of the brain mitochondrial MAO decreased. For liver, Vmax decreased while Km increased, which indicates the kinetic patterns for the neuronal and nonneruronal systems are not affected similarly by radiation. 4) The effect of several known radiation protectors and sensitizers on MAO activity was tested ut no definite results were obtained. The level of -SH group increased in some degree upon radiation but not by the compounds. 5) MAO activity was not affected by $O_2$ concentration, while an elevated level of lipid peroxidase was found under the same condition. The results described here indicate that characteristics of MAO, one of the most important central nervous system enzymes, are liable to radiation, which is partially differentiated from the liver MAO. Also indicated are that the -SH groups are hardly related to the effect of radiation but the production of the lipid peroxide seems to be somewhat correlated to the effect of radiation.
To investigate the effect of 'Saengkankunbi-Tang' on the liver tissue recovery in about 70% cutted rat liver, the observation on the photographing by the Gamma-camera and the electromicroscopic section against the rabbit liver induced by $CCl_4$, these studies were conducted. The results were summarized as follows; Observed on the liver tissue recovery, function regeneration and body weight increase in about 70% cutted rat liver, the recovery of liver tissue and body weight were increased with the passage of the time and also they were showed to the high increase rate in proportion to administration amount. The glucose content in the serum was found in increasing tide as compared with the control group. The alkaline phosphate activity in the serum showed the significant difference in the 10th day and 13th day as compared with the control group. The ammonia content in the blood showed the significant difference only in the 10th day. Observed on the photographing by the Gamma-camera against the rabbit liver induced by $CCl_4$, sample groups were shown to be significant recovery as compared with the control group. Observed on the electromicroscopic section, sample groups showed the control action of the nuclear destruction against the rabbit liver induced by $CCl_4$ and also they showed the control action of the expansion of glycogen and granular endothelial reticulum in cytoplasm. According to the above finding, it is presume that 'Saengkankunbi-Tang' can be applicable to the extensive treatment of liver disease.
To evaluate the effect of cyclohexane(CH) treatment on the serum levels of glutathion S-transferase(GST) activity in liver damaged animals, damaged liver was induced with pretreatment of 50% $CCl_4$ dissolved in olive oil (0.1 m1/100g body weight) intraperitoneally 17 times every other day. To $CCl_4$-treated rats, CH (1.56 g/kg body weight, i.p) was injected once and then the animals were sacrificed at 4 hours after injection of CH. The $CCl_4$-treated animals were identified as severe liver damage on the basis of liver functional findings, 1,e, increased serum levels of alanine aminotransferase(ALT), alkaline phosphate(ALP) and xanthine oxidase(XO) activities. On the other hand, $CCl_4$-treated animals injected with CH once($CCl_4$-pretreated animals) showed more decreased serum levels of ALT and XO, and more increased those of ALP rather than $CCl_4$-treated animals. In case of comparing the GST with ALT activity in liver, both $CCl_4$-treated and pretreated animals showed similar changing pattern of enzyme actvity. Especially $CCl_4$-pretreated animals showed significantly increased serum level of GST actvity compared with the $CCl_4$-treated those, whereas those of ALT showed reversed tendency. In aspects of GST enzyme kinetics, $CCl_4$-pretreated animals showed higher Vmax of liver GST enzyme than $CCl_4$-treated animals. In conclusion, injection of CH to the liver damaged rats led to enhanced liver damage and more increased activity of serum GST which may be chiefly caused by the enzyme induction.
Objectives : The present study attempted to reveal the effects of the combination of Fel Ursi and Bezoar on the improvement of the function of the liver, through examining the effects of Fel Ursi, Bezoar, and Woong-Whang Tang composed of Fel Ursi and Bezoar. Method : Acute hepatitis was induced by galactosamine to rats, and then Fel Ursi, Bezoar Bovis, and Woong-Whang Tang were introjected to measure the influence of serums such as AST, ALT, ALP, ${\gamma}GT$, and lipid peroxide of liver tissues which are considered as the index of the function of the liver. Results : 1. Fel Ursi decreased the lipid peroxide of ALT, ${\gamma}FT$, and liver tissue and that of mitochondria in liver tissues of the rats with galactosamine-induced acute hepatitis, but it did not have any significant effect on AST and ALP. 2. Bezoar Bovis decreased the lipid peroxide of AST, ALT, ALP, ${\gamma}GT$, and the liver tissues among serum of the rats suffering from acute hapatitis induced by galactosamine, but it did not have any significant effect on that of mitochondria of the liver tissues. 3. Woong-Whang Tang had a significant effects on the lipid peroxide of AST, ALT, ALP, ${\gamma}GT$, and the liver tissues, and on the decrease of lipid peroxide of mitochondria, among serum of the rats suffering from acute hapatitis induced by galactosamine. Conclusion : Fel Ursi and Bezoar Bovis were judged to be effective on the acute hepatitis of the liver by galactosamine. In particular, Woong-Whang Tang which was composed of the combination of Fel Ursi and Bezoar Bovis was more efficient in the improvement of the function of the liver and the amount of lipid peroxide than the respective use of Fel Ursi or Bezoar Bovis.
