The Efficacy of Ephedra sinica, Panax ginseng, and Alisma orientale Extract on Insulin resistance induced by Non-alcoholic fatty liver disease (NAFLD) (마황, 인삼, 택사 복합추출물의 endocannabinoid system 억제를 통한 비알콜성 지방간 유도 인슐린저항성 개선 효과)

  • Objectives This study aimed to investigate the efficacy of Ephedra sinica (E. sinica), Panax ginseng (P. ginseng), and Alisma orientale (A. orientale) Extract (MIT) on insulin resistance induced by Non-alcoholic fatty liver disease (NAFLD). Methods C57BL /6 male mice (8-week-old, 20 g) were divided into four groups: control group (Ctrl), high-fat diet group (HFDF), high fat diet with metformin administration group (METT), and high fat diet with MIT administration group (MITT). Each 10 mice were allocated to each group (a total of 40 mice). All mice were allowed to eat fat-rich diet freely throughout the experiment. To examine the effect of MIT, we observed Cannabinoid receptor type 1 (CB1), Cannabinoid receptor type 2 (CB2), G protein-coupled receptor 55 (GPR55), and Transforming growth factor-β (TGF-β). Results In the MITT group, positive reactions of the CB1, CB2, and GPR55 were significantly was significantly suppressed compared to the HFDF group. The positive reactions of the CD36 and TGF-β in the liver tissue were significantly suppressed in MITT. Conclusions MIT has the effect of improving NAFLD induced insulin resistance through the regulation of the lipid metabolism.

  • Toll-like receptors (TLRs) are the mammalian ortholog of Drosophila melanogaster protein Toll, originally identified for its involvement in embryonic development. In mammals, TLRs are mainly known for their ability to recognize pathogen- or damage-associated molecular patterns and, consequently, to initiate the immune response. However, it is becoming clear that TLRs can play a role also in mammal embryo development. We have previously described TLR4 and TLR7 expression in developing mouse peripheral nervous system and gastrointestinal tract. In the present study, we extended the investigation of TLR4 and TLR7 to the respiratory system and to the two main accessory organs of the digestive system, the liver and pancreas. TLR4 and TLR7 immunostaining was performed on mouse conceptuses collected at different stages, from E12 to E18. TLR4 and TLR7 immunoreactivity was evident in the embryo pancreas and liver at E12, while, in the respiratory apparatus, appeared at E14 and E17, respectively. Although further studies are required to elucidate the specific role of these TLRs in embryo development, the differential spatiotemporal TLR4 and TLR7 appearance may suggest that TLR expression in developing embryos is highly regulated for a possible their direct involvement in the formation of the organs and in the acquisition of immune-related features in preparation for the birth.

Rotations between Erythroblasts and Kupffer Cells in Human Fetal Hepatic Erythropoiesis - Trasmission and Scanning Electron Microscopic Observation (태아 간 적혈구형성에서 별큰포식세포의 적혈구모세포섬형성 - 투과 및 주사전자현미경적 관찰)

  • The relationship between intravascular erythroblasts and Kupffer cells in the human fetal liver from 11 to 20 week gestation was studied ultrastructurally. The walls of the developing sinusoids consisted of two cell types devoid of basal lamina, the nonfenestrasted endothelial cells and Kupffer cells. Kupffer cells examined were easily identified by their content of phagosmes and their morphological features, and partially proliferated by mitotic division which was different way of proliferation from adult. Some extruded nuclei of acidophilic erythroblasts were trapped within Kupffer cells which exhibited various stages of intracellular digestion of the nuclei. During high activity of human fetal hepatic erythropoiesis, Kupffer cells were found in association with developing erythrob-lasts, which was similar with erythroblastic islands. The developing erythroblasts were partially surrounded by multilaminated membrane system of the Kupffer cell consisting erythroblastic island, or in contact with Kupffer cell via cytoptasmic processes in the sinu-soidal lumen. The presence of these islands was confirmed by transmission and scanning electron microscopic study. The results demonstrate that Kupffer cells in fetal heaptic erythropoiesis phagocytized expelled nuclei and contributed to erythropoiesis mechanically and physiologically by the hypertrophy and the formation of erythroblastic islands.

Effect of Diazinon on the Cytoplasmic Organelles of Hepatocytes in Albino Mice (Diazinon이 Mouse의 간세포내 미세구조에 미치는 영향)

  • The organic phosphorus compounds have been widely used as an insecticide, since toxicity of these compounds is especially drastic to the insects than to men and other mammals. The organic phosphates are rapidly hydrolized and hence have little cumulative and ecologic effects. However, due to their acute toxic effects organophosphate have recorded rather high fatalities in men and domestic animals. The organic phosphorus compounds are powerful inhibitors to the carboxylic esterase enzymes such as acetylcholinesterase and pseudocholinesterase. As a result of firm binding characteristics of phosphate radicals to the active sites of enzyme, the activities of these enzymes are inhibited by the organophosphates. The organophosphates such as diazinon is easily observed from skin, gastrointestinal tract, conjunctivas and respiratory tract, and it is converted to more toxic form during metabolism in the liver The present study was carried out in order to investigate the hepatotoxicity of diazinon by observing the changes in the ultrastructure of cytoplasmic organelles of hepatic cells in albino mice. The animals were killed at 6, 12 and 24 hours after administration of 25mg/kg diazinon. The piece of hepatic tissue obtained from each animal was ultrathinly sectioned. The specimens stained by uranyl acetate and lead citrate double contrast methods were observed with JEM model 100B electron microscope. The results obtained were as follows: 1) A prominent dilatation and sacculation of the cisternae of rough endoplasmic reticulum associated with detachment of membrane bound-ribosomes, and disaggregation of the free ribosomes were recognized. 2) The hypertrophy of the smooth endoplasmic reticulum associated with depletion of the glycogen particles was observed. 3) The atrophy of cisternae of Golgi complex was observed. 4) A large number of secondary lysosomes (autophagic vacuoles and residual bodies) were formed. Consequently it is suggested that diazinon would induce disorganization of the cytoplasmic organelles of hepatocytes in albino mice.

  • After Triton WR-1339 (TX; 600mg/kg) intraperitoneal injection, hepatic tissues of ICR mice were intragastric injected with Hyulboochucketang extract(HCE; 3.3ml/kg/day) were observed to investigate the suppressive effect of lipid accumulation that evoke by the antihyperlipidemic effect of HCE. These hepatic tissues were fixed in fromol-calcium solution and were cryocut. These tissues stained by H&E for general morphology, sudan black B for lipid and perchloric acid-naphthoquinone(PAN) method for cholesterol. After TX treatment, the increase of hepatocyte having meshlike cytoplasm(HHMC) were shown in all hepatic lobules and the hepatic plates were disappeared in the aggregative region of HHMC. The number of blue black colored lipid drop and dark green colored asterisk shaped cholesterol particle in hepatic cytoplasm were increased and the size of lipid drop and cholesterol particle were enlarged. But, in HCE-treated mice, the HHCM were disappeared and hapatic plate were rearranged. The number of lipid drop and cholesterol particle were decreased than TX-treated mice and the size of lipid drop and cholesterol particle were diminished. As results indicated that the HCE work on the suppression of lipid accumulation in hepatic tissue of hyperlipidemic mice caused by disturbance of lipid metabolism.

Hepatic Steatosis Alleviated in Diabetic Mice upon Dietary Exposure to Fibroin via Transgenic Rice: Potential STAMP2 Involvement in Hepatocytes

  • Many benefits of silk protein fibroin (SPF) have been suggested in biomedical applications; and notably, significant SPF effects have been observed for metabolic syndromes that are directly linked to insulin resistance, such as type 2 diabetes mellitus (T2DM). Based on our previous findings, we believe that SPF from spiders exhibits outstanding glucose-lowering effects in diabetic BKS.Cg-m+/+Leprdb mice. In order to evaluate the dietary effects of SPF in diabetic animals, we generated several lines of transgenic rice (TR) that expresses SPF, and the feeding of TR-SPF to diabetic animals decreased blood glucose levels, but did not change insulin levels. Western blot analyses of hepatic proteins showed that AMP-activated protein kinase (AMPK) expression and phosphorylation both decreased in TR-SPF-fed groups, compared with controls. This finding suggests that the glucose-lowering effects in this diabetic animal model might be AMPK-independent. In contrast, six-transmembrane protein of prostate 2 (STAMP2) was upregulated after TR-SPF exposure. Together with STAMP2, the Akt protein phosphorylation increased after TR-SPF exposure, which indicates that STAMP2 leads to Akt phosphorylation and thus increases insulin sensitivity in hepatocytes. Importantly, the hepatic steatosis that was seen in the liver of diabetic mice was remarkably alleviated in TR-SPF-fed mice. Hepatocytes that were immunopositive for STAMP2 were overwhelmingly observed in hepatic tissues from TR-SPF-fed mice compared to the control. Taken together, these results suggest that feeding diabetic mice with TR-SPF upregulates STAMP2 expression and increases Akt phosphorylation in hepatic tissues and thus potentially alleviates insulin resistance and hepatic steatosis.

Isolation and Identification of an Antioxidant Enzyme Catalase Stimulatory Compound from Garnoderma lucidum

  • Antioxidant enzymes are scavenger reactive-oxygen intermediates and are involved in many cellular defense systems. We previously reported that a crude extract of Garnoderma lucidum, a medicinally potent mushroom, profoundly increased the catalase gene expression and enzyme activities in mouse livers (Park et al., J. Biochem. Mol. Biol. 34. 144-149, 2001). In this study, we elucidated the detailed mechanism whereby G. lucidum stimulates the catalase activity and expression. The major active fraction was isolated from G. lucidum and methyl linoleate was considered the most major component of the fraction. In order to determine whether methyl linoleate increases mRNA and protein synthesis of catalase, Northern and Western blot analyses were performed in vivo with methyl linoleate-treated mouse liver homogenate after feeding methyl linoleate to the mice. Northern and Western blot analyses of the crude liver homogenates in the mice that were administered methyl linoleate revealed that the expression catalase was significantly increased when compared to the untreated controls. In addition, the catalase protein levels and enzymatic activities increased in the mouse liver homogenates. These results suggest that methyl linoleate that is produced by G. lucidum stimulates the catalase expression at the transcription level.

A study of infectious duck diseases in western Gyeongnam province, Korea (경남 서부지역 사육오리의 질병 감염 조사)

  • We investigated the prevalence of infectious duck diseases using 156 ducks reared in 18 farms of western Gyeong-nam province. As a result, duck viral hepatitis (12.8%), colibacillosis (7.1%), and fungal disease (9.0%) were detected. However, avian influenza and riemerellosis were not detected. During autopsies, we could grossly observed red swollen liver (12.8%), petechial or ecchymotic hemorrhage on liver (11.5%), and fibrinous perihepatitis (9.0%). Gray-white necrotic spot (23.1%), swollen spleen (22.8%), swollen kidney (20.5%), hyperemia or hemorrhage on tracheal mucous membrane (8.3%), and nodule in long or air sac (9.0%) were also found.

Endoscopic ultrasound-guided intervention for inaccessible papilla in advanced malignant hilar biliary obstruction

  • Advanced malignant hilar biliary obstruction (MHBO) with inaccessible papilla poses a significant challenge to endoscopists, as drainage of multiple liver segments may be warranted. Transpapillary drainage may not be feasible in patients with surgically altered anatomy, duodenal stenosis, prior duodenal self-expanding metal stent, and after initial transpapillary drainage, but require re-intervention for draining separated liver segments. Endoscopic ultrasound-guided biliary drainage (EUS-BD) and percutaneous trans-hepatic biliary drainage are the feasible options in this scenario. The major advantages of EUS-BD over percutaneous trans-hepatic biliary drainage include a reduction in patient discomfort and internal drainage away from the tumor, thus reducing the possibility of tissue or tumor ingrowth. With innovations, EUS-BD is helpful not only for bilateral communicating MHBO but also for non-communicating systems with bridging hilar stents or isolated right intra-hepatic duct drainage by hepatico-duodenostomy. EUS-guided multi-stent drainage with specially designed cannulas and guidewires has become a reality. A combined approach with endoscopic retrograde cholangiopancreatography for re-intervention, interventional radiology, and intraductal tumor ablative therapies has been reported. Stent migration and bile leakage can be minimized with proper stent selection and technique, and stent blocks can be managed with EUS-guided interventions in a majority of cases. Future comparative studies are required to establish the role of EUS-guided interventions in MHBO as rescue or primary therapy.

Effects of Dietary Polyunsaturated Fat and Se on Levels of Serum Vitamin E and Creatine Phosphokinase and Morphology of Rat Hepatocytes (Polyunsaturated Fat 및 Se 식이가 흰쥐의 혈청 비타민 E 및 Creatine Phosphokinase 와 간세포의 형태에 미치는 영향)

  • Vitamin E status affected by dietary high PUFA and Se was examined by biochemical and morphological means. Rats were fed four different diets(I : 15% p/s=1 control diet, II : 15% perilla oil diet, III : 15% perilla oil, vitamin mix -vitamin E, IV : 15% perilla oil, vitamin mix-vitamin E and salt mix -Se ) for $4\frac{1}{2}$ weeks. Various dietary treatments had no significant effects on body weight gains of rats. Activities of serum creatine phosphokinase known as an indicator of vitamin E deficiency were significantly higher( P < 0.001) in groups fed diets high in PUFA, regardless of the addition or omission of vitamin E from the vitamin mixture than those in control group. Vitamin E concentrations of serum and liver were affected by experimental diets and serum levels were more affected than those in liver. Electron microscopic observations of the liver revealed 1) the presence of swollen and degenerated mitochondria and lysosome-like body(II), and 2) markedly swollen and degenerated mitochondria, numerous lysosomes and decreased in size and number of microvilli along the bile canaliculus ( III, and 3) a remarkable accumulation of lipid droplets, nuclear pyknosis, degenerated mitochondria and increased number of lysosomes scattered along the cell junction in the hepatocytes (IV).
