Coptis rhizoma was extracted with methanol or hot water. Methanol extracts had higher berberine content than water extracts, whereas the contents of Ca, Mg, Zn and K were higher in the water extracts. Rats were fed on the purified diets rich in 0.12% cholesterol only or supplemented with 0.5% of each extracts for four weeks. The concentration of total serum cholesterol was significantly lower in the rats given the extracts, irrespective of the extractants. Although the concentration of serum HDL-cholesterol was comparable among the groups, the ratio of HDL-/total-cholesterol was higher in rats given the extracts. The concentration of serum free fatty acids and ketone body were significantly elevated in rats given the extracts. On the other hand, the extracts supplements significantly decreased the contents of liver cholesterol and triacylglycerol. The amount of fecal bile acids was markedly higher in rats given the extracts, especially in those given water extracts. The results show an effective hypocholesterolemic action of Coptis rhizoma and it seems that the lower concentration of serum and liver cholesterol is resulted from the increased fecal bile acids in rats.
Overnutrition is one of the major causes of non-alcoholic fatty liver disease (NAFLD). NAFLD is characterized by an accumulation of lipids (triglycerides) in hepatocytes and is often accompanied by high plasma levels of free fatty acids (FFA). In this study, we compared the energy metabolism in acute steatotic and non-steatotic primary mouse hepatocytes. Acute steatosis was induced by pre-incubation with high concentrations of oleate and palmitate. Labeling experiments were conducted using [$U-^{13}C_5$,$U-^{15}N_2$] glutamine. Metabolite concentrations and mass isotopomer distributions of intracellular metabolites were measured and applied for metabolic flux estimation using transient $^{13}C$ metabolic flux analysis. FFAs were efficiently taken up and almost completely incorporated into triglycerides (TAGs). In spite of high FFA uptake rates and the high synthesis rate of TAGs, central energy metabolism was not significantly changed in acute steatotic cells. Fatty acid ${\beta}$-oxidation does not significantly contribute to the detoxification of FFAs under the applied conditions.

This study was designed to examine the effect of ${\omega}-3$ fatty acid, linlenic acid, EPA, DHA on serum lipid and cytokines of male rats(Sprague-Dawley). Animals of 3 groups were administrated perilla oil, salmoon oil, and tuna oil of 0.4 $m{\ell}/day$ for 8 weeks respectively. These oils were used for a source of linolenic acid, EPA and DHA. ${\omega}-3$ polyunsaturated fatty acid decreases significantly body weight, serum $PGE_2$ content and serum cytokines content of the rat, and increases internal organs weight, specially liver weight and serum HDL-cholesterol level of the rat. In the results, authors propose to use perilla oil for source of effective ${\omega}-3$ poly-unsaturated fatty acid(linolenic acid) to Prevent cardiovascular and immune diseases.
Objective: This study was to evaluate effects of mixed plant oils (identified as mixed oil 1 [MO1] and mixed oil 2 [MO2]) on performance, serum composition, viscera percentages, meat quality, and fatty acid deposition of broilers. Methods: A total of 126 one-day-old Arbor Acres male broiler chicks (weighing $44.91{\pm}0.92g$) were randomly allocated to 1 of 3 treatments with 7 replicate pens per treatment (6 broilers per pen). Dietary treatments included a corn-soybean basal diet supplemented with 3% soybean oil (CTR), basal diet with 3% MO1 (a mixture of 15% corn oil, 10% coconut oil, 15% linseed oil, 20% palm oil, 15% peanut oil and 25% soybean oil; MO1), or basal diet with 3% MO2 (a combination of 50% MO1 and 50% extruded corn; MO2). The trial consisted of phase 1 (d 1 to 21) and phase 2 (d 22 to 42). Results: Compared to CTR, broilers fed MO (MO1 or MO2) had greater (p<0.05) average daily gain in phase 1, 2, and overall (d 1 to 42), redness in thigh muscle, concentrations of serum glucose, serum albumin, saturated fatty acids (SFA) and n-6/n-3 polyunsaturated fatty acids (PUFA) ratio in breast muscle, while these broilers also showed lower ($p{\leq}0.05$) drip loss and concentrations of C18:3n-3 and PUFA/SFA ratio in breast muscle. Broilers fed MO2 had higher (p<0.05) liver percentage, while broilers fed MO1 had lower ($p{\leq}0.05$) feed conversion ratio in phase 1 and increased (p<0.05) contents of C18:2n-6, C20:5n-3, C22:6n-3, and n-3 PUFA in breast muscle compared to CTR. Conclusion: Mixed plant oils had positive effects on performance, serum parameters, meat quality, liver percentage and fatty acid deposition in broilers, which indicates they can be used as better dietary energy feedstocks than soybean oil alone.
Fatty acid binging proteins(FABP) are distinct but related gene productes which are found in many mamalian cell types. FABP bind long chain fatty acids in vitro. However, their functions and mechanisms of action, in vivo, remain unknown . Also not known is whether all FABP function similaryly in their respective cell types. or whether different FABP have unique functions. The puropose of the present study was to assess whether different members of the FABP family exhibit different structural and function properties. A comparison was made between heart(H-FABP) and liver (L-FABP). The results show that the binding sites of both FABP are hydrophobic in nature, although the L-FABP site is more nonpolar than the H-FABP site. Additionally, the bound ligand experiences less motional constraint within the H-FABP binding site than within the L-FABP binding site. In accordance with these differences in structural properties, it was found that anthroyloxy-fatty acid transfer from H-FABP to membranes is markedly faster than from L-FABP. moreover, the mechanism of fatty acid transfer to phospholipid membranes appears to occur via transient collisional interactions between H-FABP and membranes. In contrast , transfer of fatty acid from L-FABP occurs via an aqueous diffusion mechanism.
To evaluate the effects of n-3 and n-9 fatty acid combination on broiler chicks, diets containing the combinations of five different fat sources including flaxseed oil, fish oil, EPA, DHA and olive oil were provided, and all chicks were processed at 4 weeks of growth. Liver, breast and thigh samples were collected and fatty acid composition and/or CIE $L^*$, $a^*$ and $b^*$ measurement were measured. Also, live chick and liver weights were weighed and the ratio was provided as an evidence of fat accumulation in liver. No significant difference was determined in both live and liver weight ratio and liver color. EPA was low in FHO as compared to livers from others. In contrast, DHA was significantly high in FHO. In broiler breasts derived from FDO, AA and n-3 fatty acid content was high, but only numerical differences of EPA and DHA were determined in breasts from FDO. The thighs from FHO showed high in EPA, DHA and n-3 fatty acid content but had low in AA and n-6 to n-3 ratio. Therefore, the results indicate that broiler chicken diets containing either FDO or FHO may be possible combination diets increasing n-3 polyunsaturated fatty acids in broiler chicks.

This study was carried out to evaluate the nutritional properties of spreadable liver product. The composition of fresh hog liver used as raw material was 70.5% moisture, 5.2% fat, 20.3% protein and 1.2% ash. The composition of the spreadable liver product was 55.3% moisture, 19.4% protein, 21.6% fat and 3.7% ash. The total amino acid content of fresh liver was 18.69 g/100 g with glutamic acid at the highest level of 2.57 g/100 g. The total amino acid content of liver product was 18.03 g/100 g with glutamic acid at 2.28 g/100 g. The essential amino acid content was found to be 42% of total amino acids. The fatty acid analysis of fresh liver revealed oleic acid to be present at the highest level of 28% in the unsaturated fatty acid portion, and palmitic acid to be highest at 20.7% in the saturated fatty acid portion. The oleic acid content of the liver product was 40%, and the palmitic acid content was 20.4%. The level of cholesterol in fresh liver was 178.0 mg/100 g compared to 118.0 mg/100 g in the liver product. Regarding mineral analysis, the K and P contents of fresh liver were 362.2 mg/100 g and 339.1 mg/100 g, respectively, and 336.1 mg/100 g and 213.3 mg/100 g in the liver product, respectively. Many other minerals including Na, Mg, Ca and Fe were present in the product. Based on these results, the spreadable liver product made with hog liver was found to be a quality food with nutritional benefits and that is easy to consume.
The objective of this experiment was to determine the effects of n-3 polyunsaturated fatty acids (n-3 PUFAs)-enriched diet supplemented with different levels of $\alpha$-tocopherol on the activities of hepatic lipogenic-related enzymes and the contents of liver and plasma lipid fractions in laying Tsaiya ducks. A total of 180 30-wk-old laying Tsaiya ducks, at the beginning of peak production, were allotted into 6 treatments with 3 replicates each. Ducks were fed one of the 6 experimental diets, containing 4% tallow (control), and 4% fish oil supplemented with graded levels of $\alpha$-tocopheryl acetate ($\alpha$-tocopherol) at 0, 100, 200, 300 and 400 mg/kg, respectively, for 6 wks. Feed and water were supplied ad libitum throughout the experimental period. The results indicated that the n-3 PUFAsenriched diet supplemented with different levels of $\alpha$-tocopherol did not affect (p>0.05) egg weight, feed intake, body weight change or liver and abdominal fat weights. Egg production, egg mass and feed efficiency significantly (p<0.05) improved as dietary $\alpha$-tocopherol levels increased. The activities of hepatic lipogenic-related enzymes including acetyl-CoA carboxylase (EC 6. 2. 1. 3; ACC), glucose-6-phosphate dehydrogenase (EC 1. 1. 1. 49; G-6-PDH), ATP-citrate cleavage enzyme (EC 4. 1. 3. 8; CCE), NADP-malate dehydrogenase (EC 1.1.1.40; NADP-MDH) and fatty acid synthetase (FAS) were higher (p<0.05) in birds fed with the tallow diet than in those fed with fish oil diets and increased with increasing dietary $\alpha$-tocopherol levels. None of the dietary treatments significantly affected the contents of triglyceride and total cholesterol in the liver, or total cholesterol, phospholipid and total lipid in the plasma. However, the contents of phospholipid and total lipid in the liver, and triglyceride in the plasma increased as dietary $\alpha$-tocopherol levels increased. Increasing dietary $\alpha$-tocopherol levels decreased the non-esterified fatty acid (NEFA) content in the plasma and trended to decrease the cholesterol contents in the egg yolk. The lipid metabolism of laying Tsaiya ducks was influenced not only by the dietary fat but also by the supplementation levels of $\alpha$-tocopherol.
The biogenesis and biological roles of extracellular vesicles (EVs) in the progression of liver diseases have attracted considerable attention in recent years. EVs are membrane-bound nanosized vesicles found in different types of body fluids and contain various bioactive materials, including proteins, lipids, nucleic acids, and mitochondrial DNA. Based on their origin and biogenesis, EVs can be classified as apoptotic bodies, microvesicles, and exosomes. Among these, exosomes are the smallest EVs (30-150 nm in diameter), which play a significant role in cell-to-cell communication and epigenetic regulation. Moreover, exosomal content analysis can reveal the functional state of the parental cell. Therefore, exosomes can be applied to various purposes, including disease diagnosis and treatment, drug delivery, cell-free vaccines, and regenerative medicine. However, exosome-related research faces two major limitations: isolation of exosomes with high yield and purity and distinction of exosomes from other EVs (especially microvesicles). No standardized exosome isolation method has been established to date; however, various exosome isolation strategies have been proposed to investigate their biological roles. Exosome-mediated intercellular communications are known to be involved in alcoholic liver disease and nonalcoholic fatty liver disease development. Damaged hepatocytes or nonparenchymal cells release large numbers of exosomes that promote the progression of inflammation and fibrogenesis through interactions with neighboring cells. Exosomes are expected to provide insight on the progression of liver disease. Here, we review the biogenesis of exosomes, exosome isolation techniques, and biological roles of exosomes in alcoholic liver disease and nonalcoholic fatty liver disease.
This study evaluates the effect of Dioscorea japonica Thunb subfractions on hyperglycemia and the composition of energy metabolites in diabetic rats. Diabetes emllitus was induced in male Sprague-Dawley rats by an injection of streptozotocin(STZ) dissolved in a citrate buffer into the tail vein at a dose of 45㎎/㎏ of body weight. Diabetic rats were assigned to 6 groups; STZ-control, subfraction A, B , C, D and E groups. All groups were fed an AIN-76 diet. The second butanol fraction of Dioscorea administered orally with carboxymethyl cellucose for 10 days after the STZ injection Body weight gain, diet intake and organ weights were monitored Levels of hematocrit, blood glucose, liver and muscle glycogen were measured. Levels of cholesterol, triglycerides and free fatty acids were also assayed. Body weight losses were observed by subfraction A group. Liver and kidney weights were not affected in any of the subgractioned groups. The decrease of blood glucose in daibetic rats which were fed Dioscorea japonica Thunb was significantly greater than the dicrease of blood glucose in the STZ-control group. cholesterol plasma level was not influenced in any subfraction of Dioscorea japonica Thunb. Liver triglyceride levels were significantly lowered in subfraction A compared with the STZ-control group. This study's results suggest that oral administration of subfraction C of Dioscorea japonica Thunb frction is capabl of reducing blood glucose, plasma triglyceride and free fatty acid levels, and therefore Dioscorea japonica Thunb may contain antihyperglycemic compounds.
