Effect of Transportation at High Ambient Temperatures on Physiological Responses, Carcass and Meat Quality Characteristics in Two Age Groups of Omani Sheep

  • The aim of this study was to determine the effects of short road transportation in an open truck during hot season on live weight shrink, physiological responses, and carcass and meat quality of Omani sheep at 6 and 12 months of age. Thirty-six male sheep, 18 of each age group, were used. Age groups were assigned randomly to transported and not-transported groups. The transported group was transported to the slaughterhouse the day of slaughter in an open truck covering a distance of approximately 100 km. The average temperature during transportation was $37^{\circ}C$. The not-transported group was kept in a lairage of a commercial slaughterhouse with ad libitum feed and water for 48 h prior to slaughter. Blood samples were collected from sheep before loading and prior to slaughter via jugular venipuncture to assess their physiological response to transport in relation to hormonal levels. Animals were weighed just before loading onto a truck and after transport to assess shrinkage. Muscle ultimate pH, expressed juice, cooking loss percentage, WB-shear force value, sarcomere length and colour L*, a*, b* were measured on samples from longissimus dorsi, biceps femoris and semitendinosus muscles collected at 24 h postmortem at $1-3^{\circ}C$. Live weight shrinkage losses were 1.09 and 1.52 kg for 6 and 12 month transported sheep, respectively. The transported sheep had significantly (p<0.05) higher cortisol, adrenaline, noradrenaline, and dopamine concentration levels prior to slaughter at both ages than the not-transported sheep. Transportation significantly influenced meat quality characteristics of three muscles. Muscle ultimate pH and shear force values were significantly higher, while CIE L*, a*, b*, expressed juice and cooking loss were lower in transported than not-transported sheep. Age had a significant effect on meat quality characteristics of Omani sheep. These results indicated that short-term pre-slaughter transport at high ambient temperatures can cause noticeable changes in physiological and muscle metabolism responses in sheep.

  • 꺽지의 식성 분석을 위해 금강에서 2016년 3월부터 12월까지 채집을 하였고 크기는 전장이 38~70 mm, 71~109 mm, 110 mm 이상 등 3개 집단으로 구분하였다. 먹이생물은 수서곤충에 속하는 하루살이목(Ephemeroptera), 잠자리목(Odonata), 강도래목(Plecoptera), 딱정벌레목(Coleoptera), 파리목(Diptera), 날도래목(Trichoptera) 등과 어류 이었다. 꺽지는 수서곤충을 주로 섭식하였고 개체수에 있어 98.8%, 생체량은 55.8%, 상대중요성 지수는 97.1%로 대부분을 차지하였다. 식성은 육식성이었으며 먹이 섭취 특성과 형태는 Keenleyside의 분류를 적용하면 육식포식자(stalker)에 속하였다. 꺽지의 주요 먹이원은 하루살이류, 파리류, 날도래류 등이었고 잠자리류, 강도래류, 딱정벌레류, 어류 등은 희소하게 섭식하고 있었다. 꺽지는 체장이 증가함에 따라 섭식한 먹이의 생체량에 있어 하루살이류가 점진적으로 감소하였고 날도래류와 어류가 증가하는 먹이 전환이 발생 하였다. 딱정벌레류와 파리류는 큰 변동이 없이 일정하였다. 전반적으로 꺽지는 먹이생물의 구성이 계절에 따라 큰 변화는 없었다. 하천에 있어 꺽지의 자연증식과 서식지를 복원할 경우 주요 먹이원인 하루살이류, 파리류, 날도래류 등이 다량 서식할 수 있도록 조성하여야 한다.

Ruminal pH pattern, fermentation characteristics and related bacteria in response to dietary live yeast (Saccharomyces cerevisiae) supplementation in beef cattle

  • Objective: In this study we aimed to evaluate the effect of dietary live yeast supplementation on ruminal pH pattern, fermentation characteristics and associated bacteria in beef cattle. Methods: This work comprised of in vitro and in vivo experiments. In vitro fermentation was conducted by incubating 0%, 0.05%, 0.075%, 0.1%, 0.125%, and 0.15% active dried yeast (Saccharomyces cerevisiae, ADY) with total mixed ration substrate to determine its dose effect. According to in vitro results, 0.1% ADY inclusion level was assigned in in vivo study for continuously monitoring ruminal fermentation characteristics and microbes. Six ruminally cannulated steers were randomly assigned to 2 treatments (Control and ADY supplementation) as two-period crossover design (30-day). Blood samples were harvested before-feeding and rumen fluid was sampled at 0, 3, 6, 9, and 12 h post-feeding on 30 d. Results: After 24 h in vitro fermentation, pH and gas production were increased at 0.1% ADY where ammonia nitrogen and microbial crude protein also displayed lowest and peak values, respectively. Acetate, butyrate and total volatile fatty acids concentrations heightened with increasing ADY doses and plateaued at high levels, while acetate to propionate ratio was decreased accordingly. In in vivo study, ruminal pH was increased with ADY supplementation that also elevated acetate and propionate. Conversely, ADY reduced lactate level by dampening Streptococcus bovis and inducing greater Selenomonas ruminantium and Megasphaera elsdenii populations involved in lactate utilization. The serum urea nitrogen decreased, whereas glucose, albumin and total protein concentrations were increased with ADY supplementation. Conclusion: The results demonstrated dietary ADY improved ruminal fermentation dose-dependently. The ruminal lactate reduction through modification of lactate metabolic bacteria could be an important reason for rumen pH stabilization induced by ADY. ADY supplementation offered a complementary probiotics strategy in improving gluconeogenesis and nitrogen metabolism of beef cattle, potentially resulted from optimized rumen pH and fermentation.

Effects of Particle Size of Barley on Intestinal Morphology, Growth Performance and Nutrient Digestibility in Pigs

  • A growth trial and a digestibility trial were conducted to examine the effect of feed particle size on the performance, nutrient digestibility, gastric ulceration and intestinal morphology in pigs fed barley-based diets. Barley was processed through a hammer mill to achieve four diets varying in particle size (average particle $size{\pm}standard $deviation): coarse ($1,100{\pm}2.19\;{\mu}m$), medium ($785{\pm}2.23\;{\mu}m$), fine ($434{\pm}1.70\;{\mu}m$) and mixed (1/3 of coarse, medium and fine) ($789{\pm}2.45\;{\mu}m$). Sixty-four entire male pigs were used in the growth trial and the diets were fed ad libitum between 31 kg and 87 kg live weight. Following slaughter, stomach and ileal tissues were scored for integrity (ulceration or damage) and histological measurements taken. Twenty-four entire male pigs were used in the digestibility trial, which involved total faecal collection. Over the entire growth phase, there were no differences (p>0.05) in average daily gain and feed conversion ratio between pigs fed diets of different particle size. Pigs fed the coarse and medium diets had lower (p<0.05) stomach ulceration scores (0.20 and 0.25, respectively, on a scale from 0 to 3) than those fed the mixed (0.69) or the fine diets (1.87). The stomachs of all animals fed the fine diet had lesions and stomach ulcerations were present only in this group. Pigs fed the fine diet had thicker (p<0.001) ileal epithelial cell layer with no differences (p>0.05) being observed for villous height or crypt depth. Faecal digestibility coefficients of neutral and acid detergent fibre were the highest (p<0.05) for the mixed diet, intermediate for the fine and coarse diets and the lowest for the medium diet. A similar numerical trend (p = 0.103) was observed for the apparent faecal energy digestibility coefficient. It is concluded that, with barley based diets, a variation in average particle size between $400{\mu}m$ and $1,100{\mu}m$ had no effect on pig performance but the fine dietary particle size affected the integrity of the stomach, as well as the structure of the small intestine, thus compromising overall gut health. Our data also demonstrate that changes in particle size distribution during the digestion process, rather than average particle size or particle size variation, are related to apparent faecal digestibility.

Colostrum Protein Isolate Increases Gut and Whole Body Growth and Plasma IGF-I in Neonatal Pigs

  • The growth rate of the young pig is generally much less than it's potential and may be constrained by endocrine status as well as nutrient intake. Growth factors are present in relatively high quantities in colostrum and play an important part in gut development. It is possible that supplementation of colostrum protein isolate may stimulate gut and whole body growth in the pig. Eight male and 8 female (Large Whitex${\times}$Landrace) piglets were weaned at 1 d of age after each pig had obtained colostrum from their dam, and were trained to consume one of two liquid diets. The two diets were based on either a colostrum protein isolate (n = 4 males and 4 females) or whey protein concentrate (n = 4 males and 4 females) and were formulated to contain equal levels of crude protein and amino acids. Pigs were fed their diets ad libitum for 28 days after which time 12 pigs were euthanised and various tissues and organs weighed. Pigs were bled for IGF-I analyses at 21 and 28 days of age. Daily gain was higher in pigs consuming the colostrum isolate (171 vs. 216 g/d, p = 0.010), particularly between 2 and 4 weeks of age (212 vs. 298 g/d, p = 0.010). Pigs tended to consume more of the liquid feed containing colostrum isolate (25.5 vs. 29.1 kg, p = 0.074) and gained more live weight per unit of liquid feed (0.203 vs. 0.223 g/g, p = 0.056). There were no effects of sex on growth performance. Pigs consuming the diet supplemented with colostrum isolate had higher (p<0.05) full gut weight (445 vs. 554 g, p = 0.026), empty gut weight (356 vs. 463 g, p = 0.008), stomach weight (42.2 vs. 54.4 g, p = 0.001), small intestine weight (222 vs. 275 g, p = 0.025) and large intestine weight (63.7 vs. 98.0 g, p = 0.005). Plasma IGF-I (99 vs. 150 ng/ml, p<0.001) and IGF-II (265 vs. 406 ng/ml, p<0.001) were higher in pigs fed colostrum isolate. Pigs consuming colostrum protein isolate ate more, grew faster and had higher plasma IGF-I concentrations than pigs consuming a diet with similar macronutrient content but devoid of growth factors.

Voluntary Intake of Insoluble Granite-grit Offered in Free Choice by Broilers: Its Effect on Their Digestive Tract Traits and Performances

  • In this study, the effects of granite-grit offered free-choice on voluntary intakes of grit and subsequently on some morphologic traits of the digestive tract and performance of broilers were investigated. A total of 240 7d-old male broiler chicks (Ross 308) were allotted randomly to 10 floor pens supplied with wood shavings. The experiment lasted for 42 days. There were 2 dietary treatments, each consisting of 5 replicates. Each replicate consisted of 24 birds kept in an experimental unit with a floor size of $2{\times}2m$. Dietary treatments consisted of control (C) in which broilers were fed standard broiler rations, and acid insoluble granite-grit choice (AIGG) in which broilers were fed standard broiler rations and grit in separate troughs. Mean amounts consumed varied quite widely from week to week, but on average broilers ate 3.41 g per d per bird during the experimental period. Birds had a higher voluntary intake of granite-grit at an early age (7 to 21 d of age) than later (22 to 42 d of age). The voluntary intake of granite-grit of AIGG broilers increased (p<0.05) from 2.7 g/day at 7-14 d to 4.4 g at 15-21 d of age, and then it decreased to 3.4, 3.2 and 3.4 g/day between 22-28, 29-35 and 36-42 d of age, respectively. This level of grit intake increased (p<0.05) weights of empty gizzard (0.97 vs. 1.30 g), edible inner organs (3.51 vs. 3.69 g), and length of gut (8.86 vs. 9.01 cm) as a proportion of body weight and the content of insoluble ash (8.4 vs. 42.2 g/kg) in the faeces compared to the control group. Feeding free-choice grit had little or no effect on final live weights (2,542 vs. 2,543 g), daily gains (69 vs. 69 g), carcass weights (1,924 vs. 1,911), dressing percentages (75.6% vs. 75.1%) and feed efficiencies (1.69 vs. 1.66). Birds given grit did not gain more weight than those not given grit but they tended to have (p<0.07) lower feed intake (116.7 vs. 114.5 g), and consequently lower protein and energy intake. In conclusion, the granite-grit consumed voluntarily by broilers increased gut length and empty gizzard weight without affecting growth performance of broilers. Thus, it can be assumed that the voluntary consumption of granite-grit was too low to affect performance.

Studies on the induction of pregnancy and the number of fetuses during pregnancy in rats

  • This study used adult wistar-based rats to observe the sexual cycle as a morphological characteristic of vaginal epithelial cells by vaginal smearing, and investigated the fetal number through mating with male rats of the same strain. The target animal was a 12 to 13-week-old Wistar-based mature unlighted rat (weight 220 g to 240 g), room temperature 23 ± 2℃, 14 hours artificial lighting (05:00 to 19:00 hours), 10 hours Adapted individuals were used for rearing for at least 2 weeks under the conditions of the darkroom (19:00 to 05:00). The feed was managed for free feeding of pellet feed for animals and water. The vaginal smearing method was used for the experiments by observing the sexual cycle every morning and confirming that the normal sexual cycle of 4 or 5 days was repeated at least 2 cycles or more. As a result, the proestrus was found to have few red blood cells, the cells and nuclei were rather large and round, and many nucleated cells were identified. In the case of the estrus, the cells were large and the nuclei were not stained, and most of the keratinocytes were found. In addition, in the metestrus and diestrus, there were many white blood cells, and it was confirmed that nucleated epithelial cells and keratinocytes were significantly reduced. The pregnancy period was 21 ± 1.8 days, and the number of live births per delivery was 11.9 on average. The number of fetuses on the 8th and 10th days of pregnancy were 15.2 ± 0.4 and 15.4 ± 0.3, respectively. On the contrary, the number of fetuses on the 12th day of pregnancy was 12.9 ± 0.6, which was significantly (p < 0.05) decreased compared to the 10th day of pregnancy, and the number of fetuses was similar until delivery. As a result of investigating the change of body weight according to the birth weight and growth stage after delivery, the birth weight of female and male was 9.2 ± 2.0 g and 9.8 ± 2.5 g, respectively. After that, until the 16th day, the female and the male showed similarly moderate weight gain, and then showed a rapid weight gain until the 21st day of lactation. With reference to the results of this study, it is expected to be used as basic data for determining the mating time of rodents and controlling pregnancy and fetal number.

Effects of Dietary Alpha-lipoic Acid on Anti-oxidative Ability and Meat Quality in Arbor Acres Broilers

  • An experiment was conducted to evaluate the effects of dietary alpha-lipoic acid (LA) on growth performance, carcass characteristics and meat quality in Arbor Acres broilers. A total of 240 1-d-old male Arbor Acres broilers were randomly allocated to 4 dietary treatments (0, 300 ppm, 600 ppm, and 900 ppm dietary LA supplementation, respectively). Birds were slaughtered at 42 days old. Live body weight (BW), average daily gain (ADG), average feed intake (AFI), feed conversion ratio (FCR), dressing percentage, breast muscle percentage, thigh muscle percentage, abdominal fat percentage, muscle color (L*, a*, b*), pH values at 24 h postmortem, meat shear force value (SFV) and anti-oxidative ability were measured. Results showed that addition of 600 ppm or 900 ppm LA decreased BW (p<0.01), ADG (p<0.01) and AFI (p<0.05) compared with other diets. FCR was not affected by dietary LA content. LA had no marked effect on dressing percentage, breast muscle percentage or thigh muscle percentage. Abdominal fat percentage was lower (p<0.05) in the 900 ppm LA supplementation group than the control group. Dietary 900 ppm LA increased (p<0.05) breast and thigh muscle pH value at 24 h postmortem compared with the control treatment. Dietary LA increased thigh muscle a* value, though no significant difference was found in thigh muscle a* value among the treatments. Dietary LA significantly decreased breast muscle L* value (p<0.05), breast muscle b* value (p<0.01) and thigh muscle b* value (p<0.05). Broilers fed LA had higher breast muscle a* value (p<0.05) and thigh muscle L* value (p<0.05). All test groups had lower (p<0.05) breast muscle SFV than the control group. Dietary 600 ppm or 900 ppm LA both decreased (p<0.01) thigh muscle SFV compared with the control treatment. Dietary 900 ppm LA significantly increased (p<0.05) TAOC, SOD and GSHPx compared with no LA treatment. Broilers fed LA had lower (p<0.01) MDA compared with the control treatment. These results suggested that dietary LA enhanced the anti-oxidative ability and oxidative stability, and contributed to the improvement of meat quality in broilers.

보리함유 고단백질 사료의 급여가 비육돈의 생산성 및 도체특성에 미치는 영향 (Effects of High Protein Diet Containing Barley on Growth Performance and Carcass Characteristics in Finishing Pigs)

  • 본 연구는 보리함유 고단백질 사료의 급여가 비육돈의 생산성 및 도체특성에 미치는 영향에 대하여 알아보기 위해 시험을 실시하였다. 3원 교잡종[(Landrace${\times}$Yorkshire)${\times}$Duroc]의 비육돈 56두를 공시하였으며, 시험 개시 시 체중이 85.08 kg이었고, 6주간 사양시험온 실시하였다. 시험설계는 1) CON(기초사료) 및 2) HB5(5% 보리함유 고단백질 사료)로 2개 처리를 하여 처리 당 7반복, 반복 당 4두씩 완전임의 배치하였다. 전체 사양시험 기간 동안의 체중, 일당증체량, 일당사료섭취량 및 사료효율은 처리구간에 유의적인 차이가 없었으며(p>0.05), 육색에 있어서 황색도를 나타내는 b값은 대조구가 HB5처리구와 비교하여 유의적으로 높았다(p<0.05). 보수력은 HB5처리구가 대조구와 비교하여 유의적으로 높았고(p<0.05), 육즙감량은 3일, 5일 및 7일에서 HB5처리구가 대조구와 비교하여 유의적으로 낮은 감량을 나타내었다(p<0.05). 등지방두께, 생시체중, 도체중, 지육율 및 도체등급은 처리구간에 유의적인 차이가 없었다(p>0.05). 결론적으로, 본 시험의 결과 비육돈에 보리함유 고단백질 사료의 급여는 육색, 보수력 및 육즙감량에 영향을 미치는 것으로 사료된다.

Effects of Feeding Urea and Soybean Meal-Treated Rice Straw on Digestibility of Feed Nutrients and Growth Performance of Bull Calves

  • The experiment was conducted for a period of 56 days with twelve Bangladeshi bull calves of average body weight of $127.20{\pm}11.34$ kg. The calves were divided into 3 groups having 4 animals in each. The animals were fed urea-treated rice straw designated as A) 4% urea-treated rice straw, B) 4% urea+4% soybean-treated rice straw and C) 4% urea+6% soybean-treated rice straw. In addition, all the animals were supplied 2 kg green grass, 350 g Til-oil-cake and 100 g common salt per 100 kg body weight of animals. Straw was treated with 4% urea solution and soybean meal at 4 and 6% were added to treated straw and kept for 48 h in double layer polythene bags under anaerobic condition. Urea treatment improved crude protein (CP) content of rice straw from 2.68 to 8.70% and it was further increased by 10.74 and 12.12% with the addition of 4 and 6% soybean meal. Dry matter (DM) intake (kg) was higher (p<0.05) in C (4.2) followed by B (4.1) and A (4.0). Crude protein intake was significantly higher (p<0.05) in group B and C than group A. Total live weight gains were 20.2, 24.8 and 25.6 kg for calves of group A, B and C respectively (p<0.01). The addition of soybean meal to treated rice straw did not affect the coefficients of digestibility of DM, OM, EE and NFE. However, CP and CF digestibility were significantly higher in group B and C (p<0.05). The values for digestible crude protein (DCP), digestible ether extract (DEE), digestible nitrogen free extract (DNFE) and total digestible nutrients (TDN) were significantly (p<0.05) higher in diet C and B in comparison to diet A, but there were no significant difference in digestible organic matter (DOM) and digestible crude fibre (DCF) value among the groups. It may be concluded that 4% urea treated rice straw can be fed to growing bull calves with 2 kg green grass and a small quantity of concentrate without any adverse effect on feed intake and growth. Moreover, soybean meal at 4 and 6% can be added to urea treated rice straw at the time of treatment for rapid hydrolyzing of urea, which resulted an improvement in nutrient digestibility and better utilization of rice straw for growth of growing bull calves.
