Management of Radiation Injuries by Panax ginseng Extract

  • Chemical radiation protection is an important strategy to protect living beings against the deleterious effects of radiation. In the present study, the radioprotective effect of hydro-alcoholic extract of Panax ginseng extract (PGR-HAE) was studied on radiation-induced deleterious alterations in Swiss albino mice. Oral administration of such extract (25 mg/kg b wt/day/animal) for 5 consecutive days, half an h. before whole-body exposure to 6 Gy gamma radiation, enhanced the 30 days survival and also inhibited the radiogenic sickness, weight loss and life shortening. PGR-HAE ameliorated radiation induced depletion in blood constituents at different necropsy intervals between 12 h to 30 d, and significantly increased the number of femoral spleen colony forming units that survived after irradiation. Furthermore, it checked depletion of glutathione and antioxidant enzymes (superoxide dismutase, catalase, and glutathione S-transferase) as well as elevation of lipid peroxidation (LPO) level in blood and liver. The significant reduction in the yield of LPO demonstrates that PGR-HAE protects the membranes against radiation-induced oxidative damage. These findings conclude that such plant extract provides significant radioprotection, and it may be potentially valuable in the prevention of injuries caused during planned and unplanned radiation exposure.

Effects of Korean Citrus junos and Medicinal Herbs on Liver Protection and Lipid Metabolism of Alcohol Fed Rats

  • In order to investigate whether or not the alcohol-treated rat liver cells can be protected by Korean Citrus junos and medicinal herbs, We compared the serum biochemistry of rats administered both alcohol and the complex of Korean Citrus junos and medicinal herbs to control rats treated with alcohol alone. The activities of Aspartate aminotransferase (AST) and Alanine aminotransferase (ALT) in the citron 3 (Citron 3, less mellowed citron which was ripened for three months)+Phellinus linteus, Alnus japonica, Dendropanax morbifera and citron 4 (Citron 4, completely mellowed citron which was ripened fer four months)+Phaseolus radiatus, Cordyceps militaris group were significantly low when compared with the negative control group (p<0.05). The levels of triglyceride (TG) in all experimental groups were significantly lower than the negative control group (p<0.05). The concentrations of total cholesterol in the citron 3+Phellinus linteus, Atnus japonica, Dendropanax morbifera and citron 4+Phaseolus radiatus, Cordyceps militaris, Phellinus linteus were lower than the negative control group (p<0.05). The activities of alcohol dehydyogenase (ADH) in all experimental groups were significantly high when compared with the normal control group (p<0.05). These results suggest that the complex of Korean Citrus junos and medecinal herbs could be an excellent candidate for protecting vat liver cell damage induced by alcohol.

Citrus junos Fractions Decrease Alcohol-induced Liver Damage and Influence Lipid Metabolism in Alcohol-fed Rats

  • The effect of treatment with Citrus junos fractions (citron 3W, citron 3H, citron 4W and citron 4H) upon rat hepatocytes exposed to alcohol was investigated. We compared the serum biochemistry of rats administered both alcohol and Citrus junos fractions to control rats treated with alcohol alone. The effects of Alanine amino transferase (ALT) were significantly lower in the citron 3H extract group compared with the negative control group (p<0.05) and other experimental groups were not significantly low but a little low compared to negative control group. The levels of triglyceride (TG) were significantly low in all experimental groups compared with negative control group. Especially triglyceride level of citron 3H was lowest near to normal control group. The concentration of total cholesterol was significantly high in negative groups compared with normal control group but in all experimental groups, the concentration of total cholesterol was similar to that of negative control group. Total cholesterol of the citron 4W group was somewhat low compared with negative control group. In contrast, activities of alcohol dehydrogenase (ADH) were significantly higher in all experimental groups compared with the negative control (p<0.05) group. These data suggest that Citrus junos fractions may represents an excellent candidate for protection of rat hepatocytes from alcohol-mediated damage.

Light- and Relative Humidity-Regulated Hypersensitive Cell Death and Plant Immunity in Chinese Cabbage Leaves by a Non-adapted Bacteria Xanthomonas campestris pv. vesicatoria

  • Inoculation of Chinese cabbage leaves with high titer (107 cfu/ml) of the non-adapted bacteria Xanthomonas campestris pv. vesicatoria (Xcv) strain Bv5-4a.1 triggered rapid leaf tissue collapses and hypersensitive cell death (HCD) at 24 h. Electrolyte leakage and lipid peroxidation markedly increased in the Xcv-inoculated leaves. Defence-related gene expressions (BrPR1, BrPR4, BrChi1, BrGST1 and BrAPX1) were preferentially activated in the Xcv-inoculated leaves. The Xcv-triggered HCD was attenuated by continuous light but accelerated by a dark environment, and the prolonged high relative humidity also alleviated the HCD. Constant dark and increased relative humidity provided favorable conditions for the Xcv bacterial growth in the leaves. Pretreated fluridone (biosynthetic inhibitor of endogenous abscisic acid [ABA]) increased the HCD in the Xcv-inoculated leaves, but exogenous ABA attenuated the HCD. The pretreated ABA also reduced the Xcv bacterial growth in the leaves. These results highlight that the onset of HCD in Chinese cabbage leaves initiated by non-adapted pathogen Xcv Bv5-4a.1 and in planta bacterial growth was differently modulated by internal and external conditional changes.

황칠나무 추출물의 고지혈증 완화 효과 스크리닝 (Screening test for Dendropanax morbifera Leveille extracts: in vitro comparison to ox-LDL-induced lipid accumulation, ethanol-induced fatty liver and HMG-CoA reductase inhibition)

  • 이 연구는 고지혈증에 대한 생리 반응의 조절과 관련하여 Dendropanax morbifera 잎의 효과를 비교하기 위한 것이다. 추출물은 표본 연령, 수확시기, 추출 방법에 따라 달랐다. 이 추출물이 처리된 RAW 264.7 세포는 산화 저밀도 지단백질에 의해 자극되었고, 세포 지질 축적은 Oil Red O (ORO) 염색법을 사용하여 정량화되었다. HepG2 세포에서 에탄올을 사용하여 독성을 유도한 다음, 상기와 같이 지질 축적을 정량화하였다. 추출물의 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase 억제 활성을 측정하였다. RAW 264.7 세포에서 가장 지질 축적을 억제한 시료는 8년생 11월 채취 주정 60% 추출물로 처리된 세포에서 관찰되었다. 에탄올로 유도한 HepG2 세포에서 가장 큰 보호효과를 나타낸 시료는 5년생 5월 채취 주정 30% 추출물이었다. 가장 강력한 HMG-CoA reductase 억제 작용을 한 시료는 8년생 5월 채취 열수 100% 추출물이다. 전체적으로 봤을 때 지질 축적 억제는 주정 추출물에서, 콜레스테롤 합성 효소 저해 효과는 열수추출물에서 좋은 효과를 보여 이들 원료를 기반으로 지질 대사 개선 기능성 원료를 개발할 수 있을 것이라 기대한다.

녹차 첨가 서리태 청국장의 3-morpholinosydnonimine에 의한 산화적 스트레스 개선 효과 (Protective effects of Seoritae Chungkukjang added with green tea powder against 3-morpholinosydnonimine-induced oxidative stress)

  • 본 실험에서는 청국장의 항산화 기능성 증진을 위해 녹차를 첨가한 서리태 청국장을 제조한 후 LLC-$PK_1$ 세포를 이용하여 SIN-1유도 세포 독성에 대한 보호효과를 검토하였다. 항산화능이 대두보다 우수한 서리태를 주재료로, 부재료로 녹차를 농도별(0.5%, 2.0%, 5.0%)로 달리하여 청국장을 각각 제조한 후 $ONOO^-$에 의해 유도된 산화적 스트레스에 대해 서리태 청국장과 녹차첨가 서리태 청국장의 산화스트레스에 대한 개선효과를 비교한 결과, 녹차첨가 서리태청국장 추출물은 SOD, GSH-Px와 같은 항산화효소의 활성을 증가시키고, 지질과산화를 억제하여 세포생존율을 증가시킴으로써 산화적 스트레스에 대한 보호효과를 나타냈었으며, 특히 녹차를 5.0% 첨가했을 때의 효과가 가장 높은 것으로 나타났다. 따라서, 녹차 첨가 서리태 청국장은$ONOO^-$에 의해 유발된 산화적 스트레스를 개선함으로써 이로 인한 질병을 예방하는 항산화제로서 역할을 할 것으로 생각된다

스피루리나 첨가 샐러드 드레싱 보충 식이가 마우스 혈장 항산화 지표 및 DNA보호에 미치는 영양 (Effects of Spirulina Added Salad Dressing on the Antioxidant Index and DNA Protection in Mice)

  • The effects of spirulina-added salad dressing on lipid profiles and antioxidant biomarkers such as total glutathionine, TBARS value, carbonyl value, GPx, GR, SOD and paraoxonase activity in plasma or liver of mice were evaluated Sixteen male ICR mice weighing 20$\pm$2 g were divided into two groups and fed low fat ($5\%$ fat) diet (low fat control: LFC) and low fat control plus dressing diet (LFD) for eight weeks. Body weight, tissue weights of liver, heart and kidney, and the distribution of body fat deposition were not significantly different between two groups. Also, the profile of TG, TC, LDL and HDL cholesterol were similar between two groups. The DNA damage was determined using the comet assay (single cell gel assay) with alkaline electrophoresis and quantified by measuring tail length (TL). Spirulina salad dressing consumption resulted in significant decrease in lymphocyte DNA damage expressed by TL (LFC: $28.8{\mu}m$, LFD: $20.3{\mu}m$). Additionally, salad dressing consumption for 8 wks decreased the lipid peroxidation assayed by TBARS to $12.6\%$ compared with the control. The levels of antioxidant vitamins such as $\beta$-carotene were significantly higher in plasma of LFD group than those in LFC group based on HPLC method This study shows that spirulina-added salad dressing exerts degenerative disease-protective effects on oxidative DNA damage and lipid peroxidation possibly via a free radical levels.

고려인삼 추출물이 고온환경에 노출된 흰쥐에 미치는 영향 (Effects of Korean Ginseng(Panax Ginseng, C. A. Meyer) Extracts on Rat Exposed to Heat Environment)

  • 주기적인 고온환경 부가와 백삼 및 홍삼 추출물의 투여가 흰쥐의 체중, 식이섭취량, 혈액성분, 장기중량 및 간의 지질과 산화에 미치는 영향을 살펴보았다. 흰쥐를 이용한 동물모델 실험결과, 매일 $38^{\circ}C$에서 5시간씩 7일간 고온환경을 부가한 경우 주당 평균체중 증가률이 감소하는 현상이 관찰되었으나 뚜렷한 식이섭취량 변화는 없었다. 반면 고온환경 부가전후에 백삼과 홍삼 추출물을 투여한 경우에는 고온환경 부가에 따른 평균 체중증가률 감소가 억제되었다. 고온환경 부가에 의한 혈액성분 변화를 살펴본 결과에서는 적혈구수와 혈색소량은 큰 차이를 나타내지 않았으나 백혈구수는 다소 증가하는 경향을 나타내었으며 인삼 추출물 투여는 백혈구수 증가도 억제하는 효과를 나타내었다. 고온환경 부가에 따라 장기중 간장의 중량비가 감소하였고 간의 MDA 함량이 크게 증가하였다. 인삼 추출물 투여시에 고온환경부가에 따른 간의 중량감소 및 MDA 함량 증가 등의 변화도 억제되는 효과가 있었다. 특히 간의 중량변화 및 간의 지질과산화 억제효과에 있어서는 백삼보다 홍삼 추출물의 효과가 다소 우수하였으며 통계학적으로 유의적인 수준은 아니었다. 향후 인삼중의 유효성분 구명이나 관련 물질들의 활성 변화에 대한 연구가 필요할 것으로 판단되지만 고온 등 급격한 주위환경가 변화에 대한 인삼의 생체항상성 유지효과를 일부 증명할 수 있는 결과인 것으로 판단되었다.

  • In manufacturing emulsion lipstick in the past, heat and emulsifyoil phase made with oil, wax and lipophilic emulsifier with water phase together; and then mold above-mentioned materials, so-called lipophilic emulsified lipstick production method has been used. However it is hard to manufacture products using the above said method and furthermore, weak in moisturizing effect, lip care and stability. In this paper, I will discuss about multi-emulsion lipstick complementing already existing emulsion lipstick in terms of stability, giving moisturing effect of lip, and outstanding protection effectiveness of skin by safely gelatinating the state between amphiphilic lipid and water in stable.

Protective effect of green tea extract on doxorubicin induced cardiotoxicity in rats

  • Doxorubicin induces oxidative stress leading to cardiotoxicity causing electrocardiogram abnormalities and increases in biomarkers associated with toxicity. Green tea extract (GTE) is reported to possess antioxidant activity mainly via its polyphenolic constituent, catechins. This study was intended to determine the effect of various doses of GTE (25, 50 and 100 mg/kg/day p.o. for 30 days) on doxorubicin-induced electrocardiographic and biochemical changes in rat heart. The latter included lactate dehydrogenase, creatine kinase, and glutamic oxaloacetate transaminase in serum and superoxide dismutase, catalase, and reduced glutathione, as well as membrane bound enzymes like $Na^+K^+ATPase,\;Ca^{2+}ATPase,\;Mg^{2+}ATPase$ and decreased lipid peroxidation in heart tissue Results demonstrated that rats which received GTE were less susceptible to such changes indicating protection afforded by GTE.
