꾸지뽕나무 열매 추출물의 항산화 활성 (Antioxidative Activity of Extrcts from Fruit of Curdrania tricuspidata)

  • The comparative activities of acetone, ethanol, and aqueous fractions extracted from fruit powder of Cudrania tricuspidata by different temperature were tested by in vitro experimental models; peroxidation of linoleic acid and autooxidation of rat hepatic and renal microsomes by using thiobarbituric acid (TBA) for assay of free malondialdehyde production, and scavenging activities of free radicals by DPPH (α, α'-diphenyl-β-picrylhydrazyl). In DPPH method, acetone fraction extracted at 30℃ showed the highest free radical scavenging activities and acetone fractions extracted at 30℃ and 60℃ and ethanol fraction extracted at 30℃ showed stronger than BHT (butylated hydroxitoluene) although used ten-fold lower concentrations. In thiocyanate method used linoleic acid an inhibitory effects of all fractions showed higher than control treatment. TBA method used linoleic acid showed the highest antioxidative activity in acetone fraction extracted at 30℃ and 60℃. an inhibition activity against lipid peroxidation in hepatic microsomes of rats showed the highest at acetone faction extracted at acetone fraction among extracted fractions was shown to be the most potent antioxidative properties and this action was more potent in fractions extracted at 30℃ than those extracted at 60℃.

Structure-Activity Relationships of Gagaminine and Its Derivatives on the Inhibition of Hepatic Aldehyde Oxidase Activity and Lipid Peroxidation

  • In order to determine the structure-activity relationships for antioxidative effects of gagaminine, a steroidal alkaloid isolated from the roots of Cynanchum wilfordi (Asclepiadaceae), two derivatives identified as sarcostin and penupogenin were prepared from gagaminine by hydrolysis and reduction. These compounds were evaluated for the inhibitory effects on the aidehyde oxidase activity and on lipid perbxidation in vitro. Furthermore, their effects were compared with those of gagaminine and the related compounds, cinnamic acid and nicotinic acid. The results of this study prove that the cinnamoyl group in the structure of gagaminine is critical in inhibition of the aldehyde oxidase activity while the nicotinoyl group may be necessary for anti-lipid peroxidation of the compound.

무 유식물에서의 니켈내성과 히스티딘의 작용 (Nickel Tolerance and the Complexing Role of Histidine in Raphanus sativus)

  • The effect of nickel (Ni) on growth and some tolerance strategies with regard to heavy metal tolerance mechanism was investigated in radish (Raphanus sativus) seedlings. The protective effect of histidine on nickel stress conditions was also monitored. The seedling growth decreased with an increase in metal concentrations. The inhibitory effect was more pronounced in the root elongation than in the shoot elongation. Increasing Ni supply showed a progressive increase of Ni concentrations in the roots and shoots. Ni content was higher in the shoots than in the roots. In the presence of nickel, radish exhibited an antioxidative defense mechanism, as evidenced by the elevated malondialdehyde(MDA), showing that nickel is an efficient inducer of lipid peroxidation. Exposure of radish to elevated concentrations of nickel was accompanied by an increase in the proline content. Supplemental histidine in the presence of Ni ameliorated metal-induced growth inhibition and lipid peroxidation. Combinations of Ni and histidine resulted in a significant decline in proline content compared with Ni stress alone, indicating that histidine may provide protection against the adverse effect of Ni stress. From the results it is suggested that histidine is an efficient chelator by complexing metal ion within the plant and may playa role in nickel tolerance implicated in metal detoxification.

  • The ethanol extract of peony root (Paeonia Lactiflora Pall, Paeoniaceae) and its major active components including gallic acid and methyl gallate were evaluated for their protective effects against free radical generation and lipid peroxidation. And protective effects against hydrogen peroxide-induced oxidative DNA damage in a mammalian cell line were performed. The ethanol extract of peony root (PRE), gallic acid and methyl gallate were shown to possess the significant free radical scavenging effect against 1,1-diphenyl-2-picryl hydrazine (DPPH) radical generation and were revealed the inhibitory effect of lipid peroxidation as expressed by malondialdehyde (MDA) formation. They were also found to strongly inhibit hydrogen peroxide-induced DNA damage from NIH/3T3 fibroblasts, assessed by single cell gel electrophoresis. Furthermore, oral administration of 50% PRE (50% ethanol extract), gallic acid and methyl gallate potently inhibited micronucleated reticulocyte (MNRET) formation of mouse peripheral blood induced by KBrO3 treatment in vivo. Therefore, PRE containing gallic acid and methyl gallate may be a useful natural antioxidant by scavenging free radicals, inhibition of lipid peroxidation and protecting oxidative DNA damage.

Total Phenolic Contents, Radical Scavenging Capacities and Inhibitory Effects on Lipid Peroxidation and LDL Oxidation of Prunus persica Branch

  • This study was undertaken to elucidate the antioxidant activity of the ethanol (EEPB) and water (WEPB) extracts of Prunus persica branches. The extracts contained a high phenolic content and revealed a potent hydrogen donating activity in DPPH scavenging assay. Compared to $\alpha$-tocopherol, EEPB (p < 0.001) and WEPB (p < 0.05) significantly inhibited $FeCl_2$-ascorbic acid-induced lipid peroxidation, and also exhibited potent antiradical activities against hydroxyl radical, superoxide anion, nitric oxide and peroxynitrite. In copper- and AAPH-mediated human low-density lipoprotein (LDL) oxidation systems, the extracts demonstrated a strong antioxidant function by metal chelating, rather than direct scavenging, action. Furthermore, EEPB at 5 ${\mu}g/mL$ concentration showed 80.77% inhibition of the electrophoretic mobility of LDL, compared to 77.69% for ascorbic acid and 76.92% for BHT. These results suggest that PB branch extracts may protect against oxidative stress-induced diseases.

Antioxidative Constituents from Paeonia lactiflora

The ethanol extracts of the peony root (PREs) and its active constituents, gallic acid and methyl gallate, exhibited a significant free radical scavenging effect against 1,1-diphenyl-2-picryl hydrazine (DPPH) radical generation and had an inhibitory effect on lipid peroxidation, as measured by the level of malondialdehyde (MDA) formation. The PREs did not have any pro-oxidant effect. They strongly inhibited the hydrogen peroxide-induced DNA damage from NIH/3T3 fibroblasts, as assessed by single cell gel electrophoresis. Furthermore, the oral administration of 50% PRE (50% ethanol extract of peony root), gallic acid and methyl gallate potently inhibited the formation of micronucleated reticulocytes (MNRET) in the mouse peripheral blood induced by a $KBrO_3$ treatment in vivo. Therefore, PREs containing gallic acid and methyl gallate may be a useful antigenotoxic antioxidant by scavenging free radicals, inhibiting lipid peroxidation and protecting against oxidative DNA damage without exhibiting any pro-oxidant effect.

칠기탕(七氣湯) 약침액(藥鍼液)이 활성산소종, 활성질소종, 지질과산화 및 LDL의 산화에 미치는 영향 (Effect of Chilgitang Extract for Herb-Acupuncture on ROS, RNS, Lipid Peroxidation and LDL oxidation)

  • Objectives : This study was undertaken to determine the in vitro antioxidant activity of the extract of Chilgitang herb-acupuncture solution (CHAS). Methods : The radical scavenging capacity was tested by 2,2-diphenyl-1-picryl-hydrazyl (DPPH), hypoxanthine-xanthine oxidase system, DCFH-DA assay, nitric oxide and peroxynitrite generating system. In addition, antioxidant activity on copper and AAPH mediated human low-density lipoprotein (LDL) oxidation was measured by using TBARS assay and relative electrophoretic mobility assay. The amount of total phenolic compounds was assayed by the Folin-Ciocalteu method. Results : CHAS revealed a potent scavenging activity on DPPH radical(82%), superoxide anions(73%), hydroxyl radical(63%), nitric oxide (99%) and peroxynitrite (99%). Moreover, CHAS showed a strong inhibitory effect (59%) on $FeCl_2$-ascorbic acid induced lipid peroxidation of rat liver homogenate. CHAS also markedly inhibited copper(81%) and AAPH(56%)-mediated LDL oxidation, and effectively suppressed the electrophoretic mobility during exposure of human LDL to copper ions. CHAS (82 mg/g) contained higher concentration of total phenolic compounds than that of water extract (45 mg/g) obtained from Chilgitang. Conclusions : These results indicate that CHAS may protect against ROS- or RNS involved diseases, including cardiovascular diseases.

울금(Curcuma longa L.)의 생리활성 및 지질과산화 저해능에 미치는 영향 (Effects of Turmeric (Curcuma longa L.) Bioactivity Compounds and Lipid Peroxidation Inhibitory Action)

  • 울금(Curcuma longa L.)의 생리활성 및 지질과산화 저해능에 미치는 영향을 확인하고 기능성 식품 소재로 활용 가치를 검토하기 위하여 연구를 수행하였다. 총 카로티노이드(total carotenoid) 함량은 $1.581{\pm}0.005mg$ ${\beta}$-carotene equivalents (BCE)/g dry weight으로 관찰되었다. 70% 메탄올, chloroform:methanol (CM, 2:1, v/v), 에틸 아세테이트(ethyl acetate, EA) 3가지 용매를 사용한 추출 수율은 70% 메탄올(16.54%), CM (5.64%), EA (4.14%) 순으로 나타났다. 총 페놀 함량은 EA, CM 및 70% 메탄올에서 각각 106.287, 90.614 및 18.527 mg gallic acid equivalents (GAE)/g의 함량으로 나타났으며, 추출 용매 별 항산화능은 0.2, 0.4, 0.6, 0.8 mg/mL의 농도로 측정하였고, 양성대조구로 사용된 BHA (butylated hydroxyanisole) 및 trolox 보다는 낮은 항산화능을 보였다. Nitric oxide (NO) 라디칼 소거능은 70% 메탄올에서 28.65~48.43%, CM 18.86~55.10%, EA에서 15.68~56.25%로 관찰되었다. Nitrite ($NO_2$) 소거능은 70% 메탄올, CM 및 EA 순으로 나타나 EA 추출물에서 유의적인 차이를 보이며 강한 $NO_2$ 소거능을 나타내었다(p<0.05). ${\beta}$-carotene 탈색 저해능은 70% 메탄올, CM 및 EA에서 각각 1.64~23.79%, 6.99~41.16% 및 10.20~48.52%로 관찰되었다. 한편, 지질과산화 저해능은 70% 메탄올, CM 및 EA 추출물에서 높게 측정되었다.

적미 추출물과 분획물의 항산화 활성 및 암세포 성장억제효과 (Effect of Methanol Extracts of Red Colored Rices on Antioxidant Activity and Growth Inhibitory Activities of Cancer Cells)

  • 자광도와 홍향미, 동진현미를 물과 methanol로 추출하여 과산화지질생성 억제능 및 4종류의 인체유래의 암세포 즉, 간암세포 HepG2, 위 암세포 SNU-1, 유방암세포 MCF-7 및 대장암세포 SNU-C4에 대한 성장억제효과를 검토하였다. 물 추출물보다는 methanol 추출물에서, 동진현미보다는 자광도와 홍향미에서 높은 과산화지질생성 억제능과 암세포의 성장억제효과가 나타났다. 과산화지질생성 억제능은 홍향미 methanol 추출물(1 mg/assay)에서 80%, 자광도 methanol 추출물(1 mg/assay)에서 68%였다. 암세포 성장억제효능은 MCF-7에 자광도 methanol 추출물 첨가시 57%로 가장 낮았다. 또한 methanol 추출물을 극성이 서로 다른 4개의 유기용매로 분획하여 얻은 획분으로 과산화지질생성 억제능과 인체유래 암세포의 성장억제효과를 확인하였다. 자광도와 홍향미 methanol 추출물 분획물의 암세포에 대한 성장억제효과는 세포주에 따라 다소 차이는 있으나 chloroform, hexane 및 ethyl acetate 획분에서 성장억제효과가 큰 것으로 나타났다.

뜰보리수 에탄올 추출물의 산화적 스트레스 억제 효과와 암세포 증식 억제 효과 (Antioxidative and Cytotoxic Effects of Ethanol Extracts from Elaeagnus multiflora)

  • Elaeagnus multiflora, generally referred to as the cherry silverberry, is a plant. Elaeagnus multiflora fruit, leaves, and roots have been traditionally utilized in China as a treatment for cough, diarrhea, itch, and foul sores, and even cancer. More importantly, it is being investigated as a food that is capable of reducing the incidence of cancer, and also as a means of halting or reversing the growth of cancers. Considering the dearth of information regarding the medicinal properties of Elaeagnus multiflora, we assessed the antioxidative and cytotoxic effects of Elaeagnus multiflora by examining its scavenging effects on the 1,1-diphenyl-2-picryl-hydrazyl(DPPH) radical, its inhibitory effects on lipid peroxidation, and its inhibitory effects on cancer cell proliferation in HeLa cells, MCF-7 cells, and SNU-638 cells via MTT assay. Ethanol extracts of Elaeagnus multiflora flesh and seed inhibited DPPH radical production($36.91{\pm}1.00$, $94.17{\pm}0.37$) and lipid peroxidation ($28.18{\pm}5.02$, $40.30{\pm}1.45$, respectively). The Elaeagnus multiflora seed is believed to exert a possible antioxidative effects against the DPPH radical. The ethanol extracts of Elaeagnus multiflora flesh and seed exerted the cytotoxic effects on Hela cells ($6.93{\pm}1.92$, $84.97{\pm}0.47$), MCF-7 cells($5.45{\pm}0.41$, $84.97{\pm}0.47$), and SNU-638 cells($19.39{\pm}0.43$, $76.84{\pm}0.63$) used in this study. This result suggests that Elaeagnus multiflora seeds in contrast to its flesh, is believed to exert a possible anticancer effect. Elaeagnus multiflora seeds are considered to be a the candidate for preventative and dietetic treatment as an anticancer functional food.
