Betula Platyphylla var. Japonica Extract Prevent Ultraviolet C Light-induced Cell Damage in Chinese Hamster Fibroblast (V79-4) Cells

  • The present study reports the protective properties of a total methanol extract of B. platyphylla var. japonica against ultraviolet (UV)-C irradiation. Pretreatment of Chinese hamster fibroblast (V79-4) cells with a total methanol extract significantly increased cell survival following $300\;J/m^2$ of UV-C irradiation. The total methanol extract was further fractionated into 5 fractions: n-hexane, dichloromethane, ethylacetate, n-butanol and water fractions. Among these fractions, B. platyphylla var. japonica ethylacetate, butanol and water fractions showed significant protective effects against the cellular damage induced by UV-C irradiation. In order to elucidate the mechanism underlying this protective effect, DPPH (Editor note: abbreviations should be spelled out at first use.) radical scavenging and lipid peroxidation inhibitory activity were measured. Significant radical scavenging and lipid peroxidation inhibitory activities were observed for the ethylacetate fraction. In summary, the present data demonstrate that an extract of B. platyphylla var. japonica has a significant protective effect against UV-C irradiation. The underlying mechanism of this protective effect may involve radical scavenging and inhibition of lipid peroxidation by the B. platyphylla var. japonica extract.

  • The ethanol extracts of the mixed vegetables (Bioactive Vegetables, BV) and the mixed fruits (Bioactive Fruits, BF) were evaluated for their in vivo antioxidant activities. Four weeks treatment of oral administration was performed to mice. A $KBrO_3$ as a potent oxidant was used to induce the oxidative stress for in vivo experiment. BV and BF were shown to possess the significant inhibitory effect of lipid peroxidation as measured by the level of malondialdehyde (MDA) formation although the potencies were not higher than those of well-known antioxidants such as vitamin C, trolox and quercetin. Furthermore, BV and BF inhibited DNA damage assessed by single cell gel electrophoresis (comet assay) and reduced the micronucleated reticulocyte (MNRET) formation of peripheral blood. Antioxidants tested also revealed potent inhibitory activities higher than BV and BF. These antigenotoxic activity profiles were similar to that of abovementioned inhibition of lipid peroxidation. Therefore, BV and BF having mild antioxidant activity as functional food candidates may be useful natural antioxidants by the inhibiting of lipid peroxidation and the protecting oxidative DNA and chromosomal damage.

  • Inhibitory effects of Hyeulbuchuckeutang(H.B.T) On the peroxidations of lecithin-liposomes by active oxygens, hydroxyl radical and superoxide anion, derived from hydrogen $peroxidase-Fe^{2+}$ system and xanthine- xanthine oxidase system. These effects were similar to and stronger than those of catalase, mannitol, superoxide dismutase or $dl-{\alpha}-tocopherol$ as a scavenger or an antioxidant. H.B.T. inhibited the peroxidation of lecithin-liposome and active oxygens in concentration-dependent manner. H.B.T. also dose-dependently protected the cell death mduced by tert-butvlhydroperoxide(tBHP) and significantly increased cell viability in the rat normal liver cell (Ac2F). These results suggested that H.B.T might playa protective role in lipid peroxidation by free radicals and tBHP.

  • The effects of aqueous extracts from Korean commercial teas on excessive free radicals were examined utilizing spin trapping, 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical and lipid peroxidation. A potent scavenging effect of green tea and oriental senna tea was dound using sipin trapping. The most effective teasagainst the DPPH radical was green tea, followed inorder by pine leak tea, Chinese gutta percha tea and orietnal senna tea. Similar to the effects of DPPH radical , green tea, pine leaf tea, Chinese gutta percha tea and oriential senna tea had an inhibitory effect on lipid peroxidation. These findings predict that Korean tea is a promising material for scavenging free radicals, and for curing diseases related to free-radical reactions.

Ginseng Extract Protects Unsaturated Fatty acid from Decomposition Caused by Iron-Mediated Lipid Peroxidation

  • We hypothesized the primary effect of ginseng was to protect cell membrane fatty acids from decomposition caused by free radicals. To confirm the antioxidant effect of ginseng, we measured the inhibitory effect on the formation of thiobarbituric acid-reactive substances, an indicator of lipid peroxidation, and evaluated the free radical scavenging effect of ginseng by electron spin resonance spectrometer, and gas chromatography. The results showed that thiobarbituric acid-reactive substances formed and the loss of arachidonic acid during lipid peroxidation, and that hydroxyl (-like) radical peak formed by the iron complex (ferric nitrilotriacetate, an known free radical generator in vitro) were completely inhibited by ginseng extract. This antioxidant effect of ginseng may be responsible for its wide pharmacological actions in clinical practice. As the free radical reactions in general are rapid and non-specific, ginseng seems to act as a normalizer, rather than a general tonic, at the stages of acute or chronic active phase of the various diseases.

  • The MeOH extract of the twig of Morus alba L. (Moraceae) inhibited strong lipid peroxidation activity. In order to find out active principle from the plant, acivity-guided fractionation was performed and five antioxidant compounds were isolated. Their chemical structures were identified as 6-geranylapigenin (1), 6-geranylnorartocarpetin (2), resveratrol (3), oxyresveratrol (4) and quercetin (5) by physicochemical and spectrometric methods. Compounds 1-5 significantly inhibited lipid peroxidation in rat brain homogenate $(IC_{50}\;values\;of\;3.37,\;3.74,\;0.23,\;0.29\;and\;0.06\; {\mu}M,\;respectively)$.

  • Four diarylheptanoids oregonin, hirsutanonol, 1,7-bis- (3,4-dihydroxyphenyl)-heptane-5-O-$\beta$-D-xylopyanoside and 1,7-bis-(3,4-dihydroxyphenyl)-heptane-5-0-$\beta$-D-glucopyranoside isolated from Alnus hirsuta were evaluated for their antioxidative effects with 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and lipid peroxidation generation system mediated by addition of $H_2O$$_2$and/or Fe$^{2+}$ to rat liver homogenate. All these diarylheptanoids showed significant inhibitory activities against the DPPH radical and demonstrated strong activities in preventing the lipid peroxidation induced by $H_2O$$_2$.>.

The Inhibitory Effects of the Methanolic Fraction of Pueraria Radix on Hydrogen peroxide-induced Lipid peroxidation and Cadmium-induced cytotoxicity

  • The effects of the methanol subfraction of Pueraria radix on hydrogen peroxide-induced lipid peroxidation and Cd-induced cytotoxicity were investigated in NIH3T3 fibroblasts. After the methanol subfraction treatment, the content of MDA induced by 600${\mu}$M H2O2 significantly decreased in proportion to the subfraction concentrations as well as 50${\mu}$M CdCl2-induced cytotoxicity. Especially, 200$\mu\textrm{g}$/mg concentration of methanol subfraction was strongly shown inhibition of lipid peroxidation and detoxification of Cd. These results suggest that the methanol subfraction of Pueraria radix retains a potential antioxidant and protective effect against cadmium.

Antioxidative and Antimutagenic Effects of $Arctium$ $lappa$ Ethanol Extract (우엉 에탄올 추출물의 항산화활성과 항돌연변이 효과)

  • The antioxidant activities of the ethanol extract of Arctium lappa were assessed by measuring the 1,1-diphenyl-2-picrylhydrazyl( DPPH) radical scavenging effect, inhibition of $Fe^{2+}$-induced lipid peroxidation, inhibition of malondialdehyde(MDA)-bovine serum albumin(BSA) conjugation reaction and antimutagenic capacities using the Ames test. The DPPH radical scavenging activity and inhibition of $Fe^{2+}$-induced lipid peroxidation of the $Arctium$ $lappa$ ethanol extract significantly increased in a dose-dependent manner. In the radical scavenging assay using DPPH, the $IC_{50}$ of the Arctium lappa extract was 296 ${\mu}g$/assay(1.29 mg of dry sample). In addition, the $IC_{50}$ in the inhibition of $Fe^{2+}$-induced lipid peroxidation was 1,759 ${\mu}g$/assay(7.65 mg of dry sample). This extract also significantly inhibited the MDA-BSA conjugation reaction with an $IC_{50}$ of 57.58 mg/assay(250 mg of dry sample). However, no inhibitory effects against the direct and indirect mutagenicities in $Salmonella$ Typhimurium TA98 and TA100 were observed. Based on these results, the ethanol extract of $Arctium$ $lappa$ was shown to display considerable antioxidative activities.

A Nucleoside with Lipid Peroxidation Inhibitory Activity from Agrocybe cylindracea. (버들송이로부터 분리한 Nucleoside계 화합물의 지질과산화 저해활성)

  • In the screening for lipid peroxidation inhibitors from edible mushroom, Agrocybe cylindracea, a bioactive compound AG 8 was isolated. The AG 8 was purified from methanol extract of its fruit body by Diaion HP-20 column chromatography, ethyl acetate extraction, and silica gel column chromatography, consecutively. Based on various NMR studies including $^1$H irradiation and HMBC experiments, the AG 8 was identified as MTA, 5'-deoxy-5'-methylthioadenosine. This compound inhibited lipid peroxidation with an $IC_{50}$/ value of 3.2 $\mu\textrm{g}$/$m\ell$. The MTA was isolated for the first time from basidiomycetes.
