INITIATION OF LIPID PEROXIDATION AS A RESULT OF THE COMBINED ACTION OF FERRIC IRON AND LIGHT ON MEMBRANES

  • The synergic effect of iron plus blue light on the peroxidation of membrane lipid was investigated, using liposomes made of phospholipid. While strong irradiation did not affect Fe$^{+2}$-promoted lipid peroxidation that turned out to be O$_2$-dependent, ferric iron in bright light exerted a pronounced effect on the initiation of lipid peroxidation: this combined action of light and Fe$^{+3}$ on liposomal membranes was apparently independent of O$_2$. When liposomal samples containing Fe$^{+3}$ were subjected to irradiation, some portions of Fe$^{+3}$ were converted into Fe$^{+2}$. The extent of the Fe$^{+3}$-Fe$^{+2}$ conversion increased with increasing time of irradiation, which resembled the dependence of Fe$^{+3}$-promoted lipid peroxidation on irradiation. Further, it was observed that the effect of irradiation in liposomal samples containing Fe$^{+2}$ was strikingly mimicked by that of Fe$^{+2}$ addition to the same samples. The obligatory requirement of a suitable Fe$^{+3}$/Fe$^{+2}$ ratio for the genesis of iron-dependent lipid peroxidation, a controversial proposition, was also confirmed by the observation that lipid peroxidation was substantially enhanced by the addition of a mixture of Fe$^{+3}$ and Fe$^{+2}$, as compared to the addition of Fe$^{+3}$ or Fe$^{+2}$ alone. The results obtained in this study not only suggest that light acts as an effector for initiating lipid peroxidation, when Fe$^{+3}$ is present in membrane systems, but also imply that any chemical or physical factor that influences the redox states of iron in membranes can play a role in lipid peroxidation reactions.

  • The effects of membrane lipid peroxidation and retinyl palmitate on rat liver microsomal functions were investigated in vitro. Rat liver homogenates exposed to oxygen tension for 0, 3, 6, 9 or12 hours and lipid peroxidation levels were evaluated by the measurements of fluorescence intensity, malondialdehyde (MDA) and retinyl palmitate. The fluorescence intensity of homogenates and microsomes were elevated and retinyl palmitate concentrations were decreased. But the concentration of MDA was not affected to exposure time. Therefore, fluorescence intensity and retinyl palmitate concentration were used to analyze the correlation between lipid peroxidation and microsomal functions. To investigate the liver microsomal functions, the microsome was isolated from rat liver homogenates exposed to oxygen. The concentration of cytochrome P450 and the activity of NADPH-cytochrome P450 reductase in liver microsomes were gradually decreased with increasing the exposure time. The correlation between fluorescence intensity of microsomes showed a very high inverse correlation of -0.97 and -0.93, respectively. The decrease of cytochrome P450 concentration was due to the regeneration of cytochrome P450 to cytochrome P420. Also, the activities of cytochrome P450-dependent aminopyrine demethylase and benzpyrene hydroxylase of liver microsomes were gradually decreased with increasing the exposure time. The correlation with fluorescence intensity of microsome showed a high inverse correlation of -0.97 and -0.91, respectively. The retinyl palmitate concentrations of rat liver homogenates were decreased with increasing the exposure time. The decrease of retinyl palmitate concentration was followed by a low concentration of cytochrome P450 and activity of NADPH-cytochrome P450 reductase. The correlation indicated high direct correlation of 0.92 and 0.93, respectively. The decrease of retinyl palmitate concentration was also accompanied by the reduction of aminopyrine demethylase and benzpyrene hydroxylase activities. The correlation was analyzed a high direct correlation of 0.90 and 0.85, respectively. In conclusion, these studies have shown that the membrane lipid peroxidation of rat liver microsome proportionally decreased microsomal enzyme activities in vitro experiments.

New Family of Monoglucosylglyceride Diacyl Glycerol Lipids Containing Very Long Chain bifunctional Acyl Chains in Sarcina ventriculi

  • Recent research on the fatty acyl chains in the membrane lipids in Sarcina ventriculi has shown that unusually long chain bifunctional fatty acyl components are the major components of the total lipid. However, these studies did not yield any information on the complete structures of the lipid species containing these fatty acids. In this study, the structures of a new family of glucolipids containing bifunctional acyl chains are described. These structures were determined using NMR(Nuclear Magnetic Resonance) Spectroscopy, GC (Gas Chromatography)/MS (Mass Spectrometry), FTIR (Fourier Transform Infrared) spectroscopy, and FAB (Fast Atom Bombardment) mass spectrometric studies. One of the major bifunctional acyl components of the $\alpha$-glucolipids was an $\omega$-formylmethyl ester indicating the presence of plasmalogen. The general structure of the lipid components was one in which the two head groups were separated by a membrane-spanning acyl species. One head group component is a glycerol moiety of each head group, and the other is a glyceryl clucoside. Two regular chain fatty acids, one on the glycerol moiety of each head group, are also present and meet in the middle of the membrane, roughly equidistant from each head group.

대두 사포닌의 활성에 관한 기전 연구

  • Saponins are glycosidic compounds present in many plant foods. They are characterized by their ability to lyse cell membranes due to their surface-active properties. Saponins are believed to interact primarily with cholesterol in the cell membrane. In this study, the interaction of soybean(SS) with cell membrane was investigated using erythrocytes as a model. Mechanisms of interaction was also investigated by measuring their binding capacity with different membrane lipid fractions. Throughout the study, gypsophilla saponin(GS) and quillaja saponin(QS) were used to evaluate the membranolytic activity of soybean saponins. All saponins released hemoglobin in a concentration-dependent manner. SS induced 40% hemolysis at the concentration of 400 ppm, however there was no increase in hemoglobin release above 400ppm concentration. 5ppm of GS and 8 ppm of QS hemolyzed 100% of erythrocytes. Isolation of SS fractions by thin layer chromatography revealed that only one non-polar saponin possesses strong hemolytic activity. When saponins were incubated decreased the release of cholesterol. When the hemolytic activity of saponins was measured in the presence of other major membrane lipid components, sphingomyelin significantly reduced the hemolytic activity of SS, while cholesterol reduced the activity of QS. GS showed high affinity to other component(s) in the incubation media as well as lipids. These results suggest that the membranolytic activity of saponins are related to their specific chemical structure, which determines the interaction behavior between saponins and different membrane components, and thereby influence the biological activity.

Diffusion-based determination of protein homodimerization on reconstituted membrane surfaces

  • The transient interactions between cellular components, particularly on membrane surfaces, are critical in the proper function of many biochemical reactions. For example, many signaling pathways involve dimerization, oligomerization, or other types of clustering of signaling proteins as a key step in the signaling cascade. However, it is often experimentally challenging to directly observe and characterize the molecular mechanisms such interactions-the greatest difficulty lies in the fact that living cells have an unknown number of background processes that may or may not participate in the molecular process of interest, and as a consequence, it is usually impossible to definitively correlate an observation to a well-defined cellular mechanism. One of the experimental methods that can quantitatively capture these interactions is through membrane reconstitution, whereby a lipid bilayer is fabricated to mimic the membrane environment, and the biological components of interest are systematically introduced, without unknown background processes. This configuration allows the extensive use of fluorescence techniques, particularly fluorescence fluctuation spectroscopy and single-molecule fluorescence microscopy. In this review, we describe how the equilibrium diffusion of two proteins, K-Ras4B and the PH domain of Bruton's tyrosine kinase (Btk), on fluid lipid membranes can be used to determine the kinetics of homodimerization reactions.

  • We fabricated the sample of u1tra thin lipid membrane(L-${\alpha}$-DLPC ) by LB methode. The $\pi$-A isotherm of the DLPC was measured at the air-water interface varying with the compressing speed and amounts of solutions for spreading. For good property of lipid monolayer film, it was necessary for the lower speed of compressing, and 40${\mu}\ell$ of solutions for spreading. The molecular arrangement of deposited films were evaluated by measuring the absorption, transmitance and intensity with the UV spectrophotometer. The Y-type multilayers prepared at 50mN/m showed weaker than Z-type. So we found building-up of structurally high quality LB films is essential to study properties of the films and to get reproducible data.

  • In this study, deposited lipid membranes on the electrode and detected thermally stimulated displacement current generated from it. The researchers examined displacement current of electric conduction organic monolayer generated due to orient change of monolayers alkylchain and changed of dipole moment vertical component due to thermally stimulated. We paid attention to the phase transition temperature obtained by the thermally stimulated displacement current of lipid membrane layers this time. We detected the thermally stimulated displacement current peak of layers. From above results the transition temperature dilauroylphosphatidylcholine layers is about 43.deg. C. This study also compared above results with those obtained by differential thermal analysis method.

  • Change of membrane property can affect the activity of membrane proteins. In this work, we investigated the single channel properties of large conductance $Ca^{2+}$-activated $K^{+}$(BK) channels in planar lipid bilayers of different thickness. First, we recorded the activity of single BK channels from rat skeletal muscle incorporated into the control bilayer, then increased the bilayer thickness by perfusing the recording solution with the one saturated with n-pentane, or reduced the thickness by adding diheptanoylphosphatidylcholine (di$C_{7:0}$PC) to the recording soluton.(omitted)

Preparation and Characterization of a Polar Milk Lipid-enriched Component from Whey Powder

  • Milk fat globule membrane (MFGM) is a lipid carrier in mammals including humans that consists mainly of polar lipids, like phospholipids and glycolipids. In this study, a process to enrich polar lipids in commercial butter and whey powder, including polar lipids of MFGM, was developed. WPC (whey protein concentrate) 60 was selected as the most suitable raw material based on the yield, phospholipid, protein, and lactose content of the polar lipid fraction obtained by ethanol extraction of two WPC (WPC60 and WPC70) and two buttermilk (A and B). After fractionation under optimum conditions, the polar-lipid enriched fraction from WPC60 contained 38.56% phospholipids. The content of glycolipids, cerebroside, lactosylceramide, ganglioside GM3, ganglioside GD3, was 0.97%, 0.55%, 0.09%, and 0.14%, respectively. Rancimat results showed that the oxidation stability of fish oil increased with an increase in the polar-lipid fraction by more than 30 times. In addition, the secretion of IL-6 and TNF-α decreased in a concentration-dependent manner after treatment of RAW 264.7 cells with 0.1 to 100 ppm of the polar lipid fraction. In this study, polar lipid concentrates with antioxidant and anti-inflammatory activity, were prepared from milk processing by-products. The MFGM polar lipid concentrates made from by-products are not only additives for infants, but are also likely to be used as antioxidants in cooking oils and as active ingredients for functional foods.
