Although intravenous fat emulsions are well accepted as a consituent of a total parenteral nutrition regimen it is still controversial how much it can be used and who to use it in consideration of physical situation of useres. In this study the effect of two marketed lipid formulas on serum lipids change was investigated. Each lipid formula was injected to twelve normal adult volunteers and a set of blood samples was drawn at 5 minute interval during the experiment. Changes of triglycerides. free fatty acids free glycerol total cholesterol and phospolipids in the serum wre determined. To calculate serum lipids clearance the phar-macokinetics of serum triglyceride fractional removal rate(k) and half-life time(t/2) were calculated using intravenous fat tolerance-test(IVFTT) None of the parameters determined in this experiment was statistically different between two marketed formulas. In summary the bolus injection of the lipid formulas did not produce any adverse effects and their elimination kinetics from the blood stream were similar.
This study compared the lipid metabolism of white Leghorn layers between the growing and the laying periods. The study was conducted on thirty layers in the growing period (14 weeks old) and in the initial laying period (36 weeks old; 14 weeks from the onset of laying). Results indicated that all plasma lipid traits were significantly different (p<0.05) between the two periods. The estrogen concentration in the laying period was about 3 fold that in the growing period. Triacylglycerol (TG) concentration in the laying period was about 12 fold that in the growing period. The phospholipids, cholesterol, glycerol and non esterified fatty acid (NEFA) in the laying period were also higher than those in the growing period (p<0.05). Lipogenic enzyme activities in the laying period were higher than in the growing period (p<0.05). High-density lipoprotein (HDL) was the largest lipoprotein portion in growing layers. In laying hens, the major lipoprotein portion was very low-density lipoprotein (VLDL). It was also shown that protein is the primary component of HDL and TG is the largest portion of VLDL in both periods. In laying hens, VLDL contained more TG than that in growing layers (p<0.05). The VLDL particle size in laying hens was larger than that in growing layers (p<0.05). This study indicates that the lipid metabolism traits of a layer in the laying period were different from those in the growing period. The lipogenesis related traits in the laying period were markedly greater than in the growing period.
The composition of lipid class and fatty acid of free lipids(FL) as non-starch lipid and bound lipids(BL) as starch-lipid extracted from starch In naked barley(Hordeum vulgare L.) was investigated with the chromatographic procedures. FL were extracted from barley starch by petroleum ether(PE) and then BL were reextracted from PE extracted starch by the solvent systems of water-saturated butanol (WSB) at $25^{\circ}C$ and at $95^{\circ}C$ respectively. The contents of neutral lipid(NL), glycolipids(GL) and phospholipids(PL) in FL were 69.9%, 27.3%, and 2.8%, on the other hand those of BL were 34.9-54.6%, 30-45.5% and 15.4-19.6%, respectively. The identified components of NL in starch-lipid were triglycerides (70.4-82.4%), free fatty acid (8.4-26.2%), esterified sterols and free sterols, and also the major GL in starch-lipid was monogalactos-yldiglycerides(87.2-91.1%). Of the PL in FL and BL, diphosphatidyl glycerols, lysophosphatidyl choline, phosphatidyl choline & phosphatidyl serine were the major components. The predominent fatty acids found in NL, GL and PL of barley starch were palmitic acid and linoleic acid, and also myristic, stearic, oleic, linolenic acids were determined.

Silkworm (Bombyx mori) pupae have been widely worked in wound dressing, hepatoprotective activity, antigenotoxicity, control of glucose level and anti-cancer activity. To investigate the anti-obesity activity of ethanol extract of silkworm pupae powder fermented with Cordyceps militaris (ESfC), the free glycerol release and cAMP concentration as well as fat accumulation were measured in the primary adipocytes of SD (Sprague Dawley) rats and high fat diet (HFD)-treated C57BL/6 mice treated with 12 weeks. Firstly, the presence of the cordycepin with lipid lowering effect was confirmed in ESfC using HPLC analysis. The level of free glycerol and cAMP concentration was significantly increased in the primary adipocytes treated with high dose of ESfC ($400{\mu}g/ml$) although these levels were consistently maintained in other dose ESfC treated groups. In HFD-induced obesity model, the increased fat weight and size of adipocytes in HFD+Vehicle treated group was recovered in HFD+ESfC treated group. Also, the liver weight and the number of lipid droplets were higher in HFD+Vehicle treated group than No treated group. But, this level was significantly decreased in HFD+ESfC treated group compared with HFD+Vehicle treated group. Furthermore, a similar recovery was detected on the phosphorylation of periliphin and HSL, and ATGL expression. Overall, the results of the present study provide some scientific evidences that ESfC can stimulate lipolysis in primary adipocytes and prevent fat accumulation in HFD-treated obesity model, and therefore have the potential for use as anti-obesity agents to treat obese patient.
Conjugated linoleic acid (CLA) reduces fat deposition in several mammalian species. The proposed mechanisms for this effect are reduced preadipocyte proliferation and differentiation. The objective of this study was to investigate the inhibitory effects of diglyceride (DG), CLA, DG-CLA of proliferation and differentiation of 3T3-L1 preadipocytes. Cell viability was determined using WST-8 analysis and cell differentiation was determined by glycerol-3-phosphate dehydrogenase (GPDH) activity. Lipid accumulation in differentiating 3T3-L1 cells was measured by Oil red O staining. The proliferation of preconfluent 3T3-L1 cells by treatments of DG, CLA, and DG-CLA was reduced in a dose-dependent manner. CLA among them was the most effective in reduction of viable cells with increasing concentrations. Treatments of the DG, CLA, and DG-CLA at the concentration of $100{\cdot}\ddot{I}g/ml$ for 48h significantly inhibited differentiation of 3T3-L1 cells (p<0.05). In addition. cytoplasmic lipid accumulation during differentiation of the 3T3-L1 preadipocytes was also inhibited by treatments of the test solutions. DG-CLA was the most effective in the inhibition of differentiation and lipid accumulation in 3T3-L1 cells. These results indicate that the DG including CLA as fatty acids is more effective for anti-obesity than DG or CLA alone and that consumption of DG-CLA as a dietary oil may give a benefit for controlling overweight in humans.

Lipid accumulation in differentiating 3T3-L1 cells was determined by Oil red O staining. There were four experimental groups including vehicle control (DMSO), CLA, triglyceride (TG)-CLA, and DG-CLA. Treatments of CLA, TG-CLA, and DG-CLA at the concentrations of $10{\sim}1000{\mu}g/ml$ reduced proliferation of preconfluent 3T3-L1 cells in a dose-dependent manner. Among them CLA was the most effective in the proliferation inhibition of preconfluent 3T3-L1 cells with increasing concentrations. Treatments of CLA and DG-CLA at the concentration of $100{\mu}g/ml$ significantly inhibited differentiation of postconfluent 3T3-L1 cells as measured by GPOH activity (p<0.05). In addition, treatments of CLA, TG-CLA, and DG-CLA effectively inhibited lipid accumulation during differentiation of 3T3-L 1 cells. OG-CLA had the most inhibitory effect on the differentiation and lipid accumulation. These results suggest that the compounds including CLA have a respectable anti-obesity effect and that consumption of DG-CLA as a dietary oil may give a benefit for controlling overweight in humans.
The purpose of this study was to evaluate anti-obesity activity of Cirsium setidens Nakai test material (CNTM) in 3T3-L1 adipocytes and obese C57BL/6J mice fed with a high-fat diet using various obesity-related in vitro experiments. During adipocyte differentiation, CNTM significantly inhibited lipid accumulation and ROS production compared to controls. To evaluate whether CNTM could exert glycerol release effects on mature 3T3-L1 adipocytes, we treated cells with various concentrations of CNTM for 1 h. Treatment of mature adipocytes with $160-320{\mu}g/mL$ of CNTM increased the release of glycerol, but not in a significant dose-dependent manner. Anti-adipogenic and anti-lipogenic effects of CNTM seemed to be mediated by the inhibition of $PPAR{\gamma}$ and $C/EBP{\alpha}$. Moreover, CNTM stimulated fatty acid oxidation in an AMPK-dependent manner. CNTM-treated groups of C57BL/6J mice showed reduced body weights and adipose tissue weight with improving serum lipid profiles and adiponectin protein expression in obese C57BL/6J mice fed with a high-fat diet. These results suggest that CNTM might have anti-obesity effect on adipogenesis and lipid metabolism in vitro and in vivo. This presents the possibility of developing a treatment for obesity using nontoxic natural resources.

Anti-lipogenic effects of tannic acid on 3T3-L1 adipocytes as well as on rats fed high fat diet (HFD) were investigated. Tannic acid stimulated lipolysis through suppression of the leptin secretion and an increase of glycerol release in a dose dependent manner in 3T3-L1 adipocytes. For animal study, the rats were fed either HFD or HFD supplemented with 1%(w/w) tannic acid (HFDT) for 12 weeks, respectively. Body weight gain, liver weight, and visceral fat mass in rats fed HFDT were significantly decreased compared to those of rats fed HFD. The lipid profiles of HFDT group were significantly decreased compared with HFD group in the serum and liver, whereas fecal total cholesterol excretion was increased in HFDT group. These results suggest that anti-lipogenic effect of tannic acid in 3T3-L1 adipocytes and in rats fed HFD may be due to the stimulation of lipolysis and the reduction of lipid levels.
Resveratrol (3,4,5-trihydroxy-trans-stilbene), a phytoalexin found in grape skin, grape products, and peanuts as well as red wine, has been reported to have various biological and pharmacological properties. The purpose of this study was to investigate the anti-obesity effect of resveratrol-amplified grape skin extracts on adipocytes. The anti-obesity effects of grape skin extracts were investigated by measuring proliferation and differentiation in 3T3-L1 cells. The effect of grape skin ethanol extracts on cell proliferation was detected by the MTS assay. The morphological changes and degree of adipogenesis of preadipocyte 3T3-L1 cells were measured by Oil Red-O staining assay. Treatment with extracts of resveratrol-amplified grape skin decreased lipid accumulation and glycerol-3-phosphate dehydrogenase activity without affecting 3T3-L1 cell viability. Grape skin extract treatment resulted in significantly attenuated expression of key adipogenic transcription factors, including peroxisome proliferator-activated receptor, CCAAT/enhancer-binding proteins, and their target genes (FAS, aP2, SCD-1, and LPL). These results indicate that resveratrol-amplified grape skin extracts may be useful for preventing obesity by regulating lipid metabolism.

Hydrolysate silk fibroin (HSF) is a fibrous protein composed of parallel $\beta$-structures and is made from pure silk elements including 18 amino acids, with glycine, alanine, and serine comprising of over 80% of the amino acids. Numerous studies have documented a range of effects of HSF, including moisturizing, antioxidant activity, nervous system disorders, and many more. We investigated whether HSF has anti-obesity effects in vitro. The effects of HSF inhibition on lipid accumulation and acceleration of lipid degradation in 3T3-L1 cells were studied. Treatment of 3T3-L1 cells with HSF caused significant inhibition of cell viability, an increase in glycerol release, and a decreased in adipocyte differentiation. Moreover HSF stimulated downregulated of adipogenic enzyme expressions (PPAR${\gamma}$ and C/EBP${\alpha}$) and up-regulated of fatty oxidation enzyme expressions (CPT-1 and UCP-2). Based on these results, hydrolysate silk fibroin can be suggested as a potential therapeutic substance as part of a prevention or treatment strategy for obesity.
