감압처리(減壓處理)에 따른 토마토과실(果實)의 주요성분(主要成分) 및 함유지질(含有脂質)의 변화(變化) (Changes in Chemical Component and Lipid in Tomato Fruits Under the Treatment of Sub-Atmospheric Pressure)

  • 토마토 상종(商種) 중(中) "강력미수(强力米壽)"를 노지(露地) 재배(栽培)한 것을 mature green stage에서 수확하여 상온(常溫) 감압하(減壓下)에서 처리(處理)하였을 때의 저장 생리(生理)와 관련(關聯)하는 주요성분(主要成分) 및 호흡량(呼吸量), 숙도(熟度), chlorophyll, 지질(脂質)등의 변화(變化)를 조사(調査)한 바, 1. 과실(果實)의 climacteric rise의 발현(發現)은 감압구(減壓區) 보다 10 일(日)이상 지연되었으며, 2, 처리기간(處理期間) 중(中) 숙도(熟度)의 변화(變化)는 감압정도(減壓程度)에 따라 차(差)가 있어, 대체(大體)로 감압도(減壓度)가 높을수록 지연되었다. 이 관계(關係)는 climacteric rise와도 밀접(密接)한 관계가 있었으며 처리구(處理區)에 관계(關係)없이 dark pink stage와 climacteric maximum는 일치(一致)하였다. 3. 과실(果實)의 색상(色相)과 관계(關係)가 깊은 chlorophyll의 감소(減少)도 상압구(常壓區)가 감압구(減壓區)보다 현저하였다. 4. 과실(果實)의 함유지질(含有脂質)은 중성지질(中性脂質), 인지질(燐脂質). 당지질(糖脂質)이었으며 그중(中) 중성지질(中性脂質)이 46%, 인지질(燐脂質)이 32%, 당지질(糖脂質)이 13%이였다. 중성지질(中性脂質)의 구성(構成)은 sterol ester, triglyc ericle, free fatty acid, 1.3 diglyceride, 1.2 diglyceride, sterol, monoglyceride 의 7종(種)이었으며, 이중(中) triglyceride가 49.3%로서 가장 함량(含量)이 많았다. 인지질(燐脂質)은 phosphatidic acid, cardiolipin, phosphatidyl choline, phosphatidyl ethanolamine, phos phatidyl glycerol, phosphatidyl inositol phosphatidyl serine의 7종(種)으로 구성(構成)되며 이중(中) phosphatidyl choline 및 phosphatidyl ethanol amine이 가장 많이 함유되고 있었다. 5. 저장중(貯藏中) 총지질(總脂質)은 시일(時日)이 경과함에 따라 점차(漸次) 감소(減少)하는 경향(傾向)이었는데 상압구(常壓區)에 비하여 감압구(減壓區)의 변화(變化)가 적었다. 중성지질(中性脂質)은 climacteric rise 및 숙도(熟度)와 관련하여 변화(變化)하였으며 climacteric onset기(期)에. 가장 많이 감소(減少)하다가 다시 상승(上昇)하는 경향(傾向)이었다. 중성지질(中性脂質) 중(中) climacteric rise 및 숙도(熟度)와 관련하여 변화(變化)한 것은 diglyceride 및 sterol ester의 2종(種)이었으며 glyceride가 중성지질(中性脂質) 전량(全量)의 변화(變化)와 동일(同一)한 경향(傾向)인데 반(反)하여 sterol ester은 climactric onset까지 증가(增加)하다가 기후(其後) 감소(減少)하였다. 인지질(燐脂質)도 저장기간(貯藏期間) 중(中) 처리구(處理區)에 관계(關係)없이 다같이 감소(減少)되었는데, 그 정도(程度)는 상압구(常壓區)에 비(比)하여 감압구(減壓區)의 변화(變化)가 적었다. 인지질(燐脂質) 중(中) ph. choline과 ph. ethanlamine은 climacteric rise및 숙도(熟度)와 관련하여 변화(變化)하였다. 중성지질(中性脂質) diglyceride 및 sterol ester와 인지질(燐脂質)의 ph. choline 과 ph. ethanol amine은 climacteric rise와 관련하여 변화(變化)하였다.

Bioconversion of Citrus unshiu peel extracts with cytolase suppresses adipogenic activity in 3T3-L1 cells

  • BACKGROUND/OBJECTIVES: Citrus flavonoids have a variety of physiological properties such as anti-oxidant, anti-inflammation, anti-cancer, and anti-obesity. We investigated whether bioconversion of Citrus unshiu with cytolase (CU-C) ameliorates the anti-adipogenic effects by modulation of adipocyte differentiation and lipid metabolism in 3T3-L1 cells. MATERIALS/METHODS: Glycoside forms of Citrus unshiu (CU) were converted into aglycoside forms with cytolase treatment. Cell viability of CU and CU-C was measured at various concentrations in 3T3L-1 cells. The anti-adipogenic and lipolytic effects were examined using Oil red O staining and free glycerol assay, respectively. We performed real time-polymerase chain reaction and western immunoblotting assay to detect mRNA and protein expression of adipogenic transcription factors, respectively. RESULTS: Treatment with cytolase decreased flavanone rutinoside forms (narirutin and hesperidin) and instead, increased flavanone aglycoside forms (naringenin and hesperetin). During adipocyte differentiation, 3T3-L1 cells were treated with CU or CU-C at a dose of 0.5 mg/ml. Adipocyte differentiation was inhibited in CU-C group, but not in CU group. CU-C markedly suppressed the insulin-induced protein expression of CCAAT/enhancer-binding protein ${\alpha}$ ($C/EBP{\alpha}$) and peroxisome proliferator-activated receptor gamma ($PPAR{\gamma}$) as well as the mRNA levels of $CEBP{\alpha}$, $PPAR{\gamma}$, and sterol regulatory element binding protein 1c (SREBP1c). Both CU and CU-C groups significantly increased the adipolytic activity with the higher release of free glycerol than those of control group in differentiated 3T3-L1 adipocytes. CU-C is particularly superior in suppression of adipogenesis, whereas CU-C has similar effect to CU on stimulation of lipolysis. CONCLUSIONS: These results suggest that bioconversion of Citrus unshiu peel extracts with cytolase enhances aglycoside flavonoids and improves the anti-adipogenic metabolism via both inhibition of key adipogenic transcription factors and induction of adipolytic activity.

3T3-L1 지방세포에서 Bacillus subtilis KC-3 발효두유의 항비만 효과 (Antiobesity Effect of the Bacillus subtilis KC-3 Fermented Soymilk in 3T3-L1 Adipocytes)

  • 3T3-L1 지방세포에서 상업용 Bacillus subtilis 균주와 순창 민속마을 전통 청국장에서 분리한 Bacillus subtilis KC-3(KCCM 42923) 균주를 이용해 두유를 발효시키고 이를 발효하지 않는 두유와 지방 생성 억제 효과를 비교하였다. 렙틴의 분비량은 B. subtilis MYCO 10001 발효두유(F-MYCO)와 B. subtilis KC-3 발효두유(F-KC)에서 유의적으로 감소하였다. 이러한 지방 생성 억제 효과가 지방의 축적과도 관련이 있는지 알아보기 위하여 지방구를 관찰한 결과 두유와 발효두유 모두에서 축적된 지방의 양이 감소했고 그중 F-KC는 유의적으로 감소하여(p<0.05) 지방의 생성과 축적이 억제된 것을 알 수 있었다. 지방축적의 감소가 지방 분해와도 관련이 있는지 조사하기 위하여 글리세롤의 분비량을 측정하였는데 발효되지 않은 두유의 글리세롤 분비 정도는 control과 비슷하였으나 발효두유의 모든 군에서는 글리세롤의 분비량이 증가하였고 특히 F-KC에서 글리세롤 분비량이 유의적으로 증가하였다. 또한 F-KC의 지방 축적 감소가 지방 생성 억 제로부터 기인된 것인지 조사하기 위하여 지방생성에 중추적 역할을 맡고 있는 전사인자인 $PPAR{\gamma}$와 SREBP-1c의 mRNA 발현을 확인한 결과 두유나 다른 발효두유에 비하여 F-KC에서 이들 유전자 발현이 감소한 것으로 나타났다. 따라서 B. subtilis KC-3에 의해 발효된 두유의 항비만 효과는 지방 생성의 중요한 전사인자인 $PPAR{\gamma}$와 SREBP-1c의 발현 억제에 기인한 것으로서 그 결과 지방의 생성을 억제하고 지방 축적을 효과적으로 감소 시키는 것으로 보인다.

Veronica incana 추출물의 생물학적 활성 평가 (Evaluation of Biological Activity of Veronica incana Extracts)

  • Objectives : The aim of this study is to evaluate the potential biological activity of Veronica incana extracts (VIE) through in vitro, ex vivo, and in vivo experiments. Methods : In vitro, we conducted analyses on the total polyphenol (TP) and total flavonoid (TF) levels, alongside DPPHand ABTS radical scavenging activities. Ex vivo evaluations on adipose tissue measured glycerol release as a marker of lipolysis. In LPS-induced RAW 264.7 cells, we quantified nitric oxide (NO) production. Following H2O2 induction in U2OS cells, we performed mitochondrial assays such as MitoSox and MitoTracker. Moreover, Bodipy assays were conducted in 3T3-L1 cells. In vivo, we performed anti-osteoarthritis effect of VIE against monosodium iodoacetate (MIA)-induced osteoarthritis in rats. Results : The results presented encompass a myriad of models, from cell culture to animal experiments as well as ex vivo studies. VIE demonstrated high TP and TF contents, potent DPPH and ABTS scavenging activities, and regulated glycerol release. Moreover, the inhibition of NO production in LPS-induced inflammation was notably confirmed and the reduction of lipid droplets was distinctly shown. Furthermore, in H2O2-induced U2OS cells, MitoSox was effectively reduced while MitoTracker noticeably increased. In vivo assays confirmed a significant increase in hindpaw weight distribution (HWD) decreased by MIA after VIE treatment. Additionally, VIE inhibited serum inflammatory cytokines (TNF-𝛼, IL-6, and IL-1𝛽) and MDA levels in joint tissue. Conclusion : In conclusion, Veronica incana exhibited various pharmacological effects including antioxidant, anti-obesity, and anti-inflammatory properties.

유청단백질로 만들어진 식품포장재에 관한 연구

  • Edible films such as wax coatings, sugar and chocolate covers, and sausage casings, have been used in food applications for years$^{(1)}$ However, interest in edible films and biodegradable polymers has been renewed due to concerns about the environment, a need to reduce the quantity of disposable packaging, and demand by the consumer for higher quality food products. Edible films can function as secondary packaging materials to enhance food quality and reduce the amount of traditional packaging needed. For example, edible films can serve to enhance food quality by acting as moisture and gas barriers, thus, providing protection to a food product after the primary packaging is opened. Edible films are not meant to replace synthetic packaging materials; instead, they provide the potential as food packagings where traditional synthetic or biodegradable plastics cannot function. For instance, edible films can be used as convenient soluble pouches containing single-servings for products such as instant noodles and soup/seasoning combination. In the food industry, they can be used as ingredient delivery systems for delivering pre-measured ingredients during processing. Edible films also can provide the food processors with a variety of new opportunities for product development and processing. Depends on materials of edible films, they also can be sources of nutritional supplements. Especially, whey proteins have excellent amino acid balance while some edible films resources lack adequate amount of certain amino acids, for example, soy protein is low in methionine and wheat flour is low in lysine$^{(2)}$. Whey proteins have a surplus of the essential amino acid lysine, threonine, methionine and isoleucine. Thus, the idea of using whey protein-based films to individually pack cereal products, which often deficient in these amino acids, become very attractive$^{(3)}$. Whey is a by-product of cheese manufacturing and much of annual production is not utilized$^{(4)}$. Development of edible films from whey protein is one of the ways to recover whey from dairy industry waste. Whey proteins as raw materials of film production can be obtained at inexpensive cost. I hypothesize that it is possible to make whey protein-based edible films with improved moisture barrier properties without significantly altering other properties by producing whey protein/lipid emulsion films and these films will be suitable far food applications. The fellowing are the specific otjectives of this research: 1. Develop whey protein/lipid emulsion edible films and determine their microstructures, barrier (moisture and oxygen) and mechanical (tensile strength and elongation) properties. 2. Study the nature of interactions involved in the formation and stability of the films. 3. Investigate thermal properties, heat sealability, and sealing properties of the films. 4. Demonstrate suitability of their application in foods as packaging materials. Methodologies were developed to produce edible films from whey protein isolate (WPI) and concentrate (WPC), and film-forming procedure was optimized. Lipids, butter fat (BF) and candelilla wax (CW), were added into film-forming solutions to produce whey protein/lipid emulsion edible films. Significant reduction in water vapor and oxygen permeabilities of the films could be achieved upon addition of BF and CW. Mechanical properties were also influenced by the lipid type. Microstructures of the films accounted for the differences in their barrier and mechanical properties. Studies with bond-dissociating agents indicated that disulfide and hydrogen bonds, cooperatively, were the primary forces involved in the formation and stability of whey protein/lipid emulsion films. Contribution of hydrophobic interactions was secondary. Thermal properties of the films were studied using differential scanning calorimetry, and the results were used to optimize heat-sealing conditions for the films. Electron spectroscopy for chemical analysis (ESCA) was used to study the nature of the interfacial interaction of sealed films. All films were heat sealable and showed good seal strengths while the plasticizer type influenced optimum heat-sealing temperatures of the films, 130$^{\circ}$C for sorbitol-plasticized WPI films and 110$^{\circ}$C for glycerol-plasticized WPI films. ESCA spectra showed that the main interactions responsible for the heat-sealed joint of whey protein-based edible films were hydrogen bonds and covalent bonds involving C-0-H and N-C components. Finally, solubility in water, moisture contents, moisture sorption isotherms and sensory attributes (using a trained sensory panel) of the films were determined. Solubility was influenced primarily by the plasticizer in the films, and the higher the plasticizer content, the greater was the solubility of the films in water. Moisture contents of the films showed a strong relationship with moisture sorption isotherm properties of the films. Lower moisture content of the films resulted in lower equilibrium moisture contents at all aw levels. Sensory evaluation of the films revealed that no distinctive odor existed in WPI films. All films tested showed slight sweetness and adhesiveness. Films with lipids were scored as being opaque while films without lipids were scored to be clear. Whey protein/lipid emulsion edible films may be suitable for packaging of powder mix and should be suitable for packaging of non-hygroscopic foods$^{(5,6,7,8,)}$.

인삼(人蔘)의 지방질(脂肪質) 성분(成分)에 관한 연구 (Studies on the Lipid Components of Panax ginseng)

  • 한국산(韓國産) 수삼(水蔘)과 건삼(乾蔘)을 클로로포름-메탄올(2:1, v/v) 및 에틸 에테르로 각각 추출(抽出)한 지방질(脂肪質)은 관, 얇은 막 및 기체-액체 크로마토그래피로 분리(分離), 정량(定量)하여 다음과 같은 결과(結果)를 얻었다. 1. 수삼(水蔘)은 약 0.62%의 조(粗) 지방질(脂肪質)을 함유하고 있었고, 그 중 중성 지질은 45.28%, 당 지질은 18.12%, 인지질은 33.60%였다. 그러나 건삼(乾蔘)은 약 0.82%의 조지방질을 함유하고 있었고, 그중 중성 지질은 86.48%, 당 지질은 9.20%, 인 지질은 4.32%였다. 2. 수삼(水蔘)과 건삼(乾蔘)의 중성(中性) 지질(脂質) 중에는 다같이 triglyceride(중성 지질의 $37.6{\sim}42.5%$)와 스테롤 에스테르($16.5{\sim}19.6%$)가 주된 성분이었고, 그외에 monoglyceride, diglyceride, 유리(遊離) 지방산(脂肪酸), 유리(遊離) 스테롤이 부성분(副成分)으로 존재 하고 있었다. 3. 당(糖) 지질(脂質) 중에서는 digalactosyl diglyceride (당지질의 23.5%)가 수삼(水蔘)중에, steryl glycoside(28.9%)가 건삼(乾蔘)중에 각각 그 함량이 가장 많았다. 그리고 esterified steryl glycoside와 monogalactosyl diglyceride가 수삼(水蔘)과 건삼(乾蔘)에 다같이 존재하고 있었으며, 그외에 수삼(水蔘)에는 4가지, 건삼(乾蔘)에는 2가지의 미확인 당지질이 존재하고 있었다. 4. 수삼(水蔘)과 건삼(乾蔘)의 인(燐) 지질(脂質) 중에는 다같이 prosphatidyl choline(인 지질의 $31.3{\sim}31.9%$과 phosphatidyl glycerol $34.8{\sim}36.7%$이 주된 성분이었고 그 외에 phosphatidyl ethanolamine과 phosphatidy1 inositol이 존재하고 있었다. 5. 수삼(水蔘)과 건삼(乾蔘)의 주요(主要) 지방산은 다같이 linoleic $62.29{\sim}64.32%$, palmitic $15.63{\sim}13.16%$, oleic $5.73{\sim}7.23%$ 및 linolenic acid$3.52{\sim}6.11%$였다. 중성 지질 획분의 지방산 조성은 총지방질의 지방산조성과 그 패턴이 비슷하였다. 그리나 당 지질과 인 지질 획분의 지방산 조성은 중성 지질 획분에 비하여 linoleic acid의 함량이 적고 Palmitic acid의 함량은 많은 편이었다.

한국산 들깻잎의 지방질 및 지방산조성에 관한 연구 (Studies on Lipid and Fatty acid Composition of Korean Perilla Leaves(Penilla frutescens var. japonica HARA))

  • 근래에 와서 널리 식용되고 있는 2종류의 들깻잎 즉 잎 뒷면이 녹색을 띄는 들깻잎(GPL)과 자색을 띄는 들깻잎(VPL)을 시료로 하여 이들의 지방질 및 지방산 조성을 비교 측정하였다. 총지방질 함량은 건중당 GPL에서는 5.24%, VPL에서는 5.02%로 두 시료간에 차이가 거의 없었다. 총지방질중 중성지방질은 $34.67{\sim}36.36%$, 당지방질은 $49.06{\sim}50.94%$, 인지방질은 $14.39{\sim}14.58%$로 함유되어 VPL에서는 중성지방질과 인지방질이 GPL에서는 당지방질이 약간 높게 함유되었다. 중성지방질의 주요성분은 sterol ester-hychocarbons $(58.07{\sim}58.54%)$과 triglyceride(14.9%)였으며 당지방질에서는 monogalactosyl diglyceride$(42.4{\sim}44.9%)$ digalactosyl diglyceride$(13.4{\sim}15.4%)$였고, 그리고 인지방질에서는 phosphatidyl ethanolamine$(40.1{\sim}45.0%)$이었으며 이들 함량은 GPL과 VPL간에 유의적인 차이가 없었다. GPL과 VPL간에 구성분의 유사도는 중성지방질(r=1.00), 당지방질(r=0.997) 그리고 인지방질(r=0.968)의 순으로 감소되었다. 총지방질 지방질획분의 구성 주요지방산은 총지방질 중에는 linolenic acid$(62{\sim}64%)$, palmitic$(10{\sim}12%)$ linoleic acid$(9{\sim}10%)$였고, 중성지방질에는 linolenic acid$(42.2{\sim}43.2%)$, palmitic acid(15.2%), myristic acid$(14.4{\sim}14.9%)$, 당지방질에는 linolenic acid$(74.6{\sim}79.4%)$, oleic acid$(8.0{\sim}10.8%,$, palmitic acid$(5.2{\sim}7.9%)$, 인지방질 중에는 linolenic acid$(36.4{\sim}39.1%)$, linoleic acid$(22.6{\sim}25.4%)$, palmitic acid$(22.4{\sim}22.5%)$였다. 불포화지방산의 비율은 VPL의 당지방질에서 가장 높았다. GPL과 VPL간에 지방산 구성유사도는 인지방질에서 가장 작았고, 인지방질과 다른 지방질 획분간에도 그러하였다.

태음인(太陰人) 열다한소탕(熱多寒少湯)이 3T3-L1 지방전구세포(脂肪前驅細胞) 분화(分化)에 미치는 영향(影響) (Effects of Yeoldahanso-tang, a Sasang Constitutional Herbal Prescription for Taeeumin, on the Adipogenesis in 3T3-L1 Cells)

  • Objectives : Although Yeoldahanso-tang (YDHST) has been widely used for treatment of obesity and its related diseases such as hyperinsulinemia and hypertension for Taeeumin, no scientific evidence has reported yet to support its ability to work against these metabolic disorders. Our study was aimed to investigate the anti-obesity effect of YDHST extract on the cellular differentiation of 3T3-L1 preadipocytes into adipocytes. Methods : 3T3-L1 preadipocytes were differentiated into adipocytes by adding insulin, dexamethasone and 3-isobutyl-1-methylxanthine (IBMX) for 8 days in the absence or presence of YDHST extract. Anti-obesity effects of YDHST extract were evaluated by Oil Red O staining, glycerol-3-phosphate dehydrogenase (GPDH) activity, triglyceride contents, and leptin production. Results : YDHST extract remarkably prevented lipid accumulation with no cytotoxicity in the differentiated 3T3-L1 cells. In addition, YDHST extract decreased contents of triglyceride 3T3-L1 adipocytes. Consistently, YDHST extract caused a significant inhibition of GPDH activity and leptin production in a dose-dependent manner. Conclusions : Our findings suggest that Sasang constitutional herbal formula YDHST for Tae-eumin has anti-obesity activity by regulation of the adipogenesis process in vitro. Additional study will be required to further confirm the inhibitory effect on adipocyte differentiation by using in vivo animal model.

Effects of Conjugated Linoleic Acid on Adipocyte Secreted Proteins in vitro

  • Conjugated linoleic acid (CLA) is a class of positional, geometric conjugated dienoic isomers of linoleic acid (LA). CLA activates the immune system, protects against tumorigenesis, and reduces the incidence of atherosclerosis. Trans-10, cis-12 CLA has specific effects on lipid metabolism, it has been shown to reduce body fat gain and regulates some adipocyte secreted proteins in vivo and in vitro. Here we report that a CLA mixture affects cytokine secretion from rat primary adipocytes. Rat primary adipocytes were treated with 1 mM, 100 $\mu$M, 1 $\mu$M or 100 nM CLA mixture doses; and leptin, tumor necrosis factor alpha (TNF a ), interleukin-6 (IL-6) and glycerol levels in the medium were measured. Leptin secretion was lower, TNF $\alpha$ secretion higher and IL-6 secretion did not change in response to the CLA mixture. Leptin and TNF $\alpha$ secretions did not change with CLA mixture treatment in a dose-dependent manner. In addition, the CLA mixture did not appear to enhance lipolysis in rat primary adipocytes. In conclusion, our study demonstrates that the decrease in leptin and increase in TNF $\alpha$ secretion in adipocytes treated with CLA mixture may be due to the apoptotic effect and to a reduction in peroxisome proliferator-activated receptor gamma (PPAR ${\gamma}$ ) ligands.

Enhancing liquid-chilled storage and cryopreservation capacities of ram spermatozoa by supplementing the diluent with different additives

  • Objective: In the present study, we determined efficiency of incorporating caffeine, melatonin or omega-3 polyunsaturated fatty acid in the diluent on mitigating consequences of (a) liquid chilled- and (b) cryo-storage of ram spermatozoa. Methods: In the first experiment, ejaculates (n = 30) were collected from 5 adult rams and were pooled, diluted (1:10) with Tris-citric acid (base diluent) and were split into 4 aliquots assigned for: control (untreated), caffeine (0.1 mM), melatonin (0.3 mM) or omega-3 fatty acids (0.3 mM) (T0). The diluted specimens were stored at 4℃ for 48 h, during which sperm physical and cytological properties were evaluated along with oxidative stress indices (T24, T48). In the second experiment, 15 ejaculates (3 per male) were pooled, diluted with glycerolized base diluent (4% glycerol, v/v) and were split corresponding to the same previous treatment groups before being processed for cryopreservation. Post-thaw physical and kinematic sperm properties were assessed by a computer-assisted sperm analysis system. Results: The results clarified superiority of both melatonin and omega-3 supplementation on maintaining (p<0.05) sperm properties, while reducing (p<0.05) lipid peroxidase reaction and enzymatic activities of alanine aminotransferase, aspartate aminotransferase, and alkaline phosphatase in preservation medium, compared to caffeine either during liquid-chilled storage or cryopreservation of spermatozoa. Conclusion: Melatonin and omega-3 are regarded efficient alternatives to caffeine when processing ram spermatozoa for application of artificial insemination or in vitro fertilization.
