Inhibition of Tumor Formation and Changes in Hepatic Enzyme Activities by Kimchi Extracts in Sarcoma-180 Cell Transplanted Mice

  • Inhibitory effects of the methanol extract, hexane extract, methanol soluble fraction (MSF) and juice from 3 weeks fermented Kimchi on the tumor formation in sarcoma-180 cell transplanted mice were studied. Effects of the solvent extracts and juice of the Kimchi on the levels of lipid peroxide, glutathione, and the enzyme activities of the liver were also investigated in normal and sarcoma-180 cell transplanted mice. At 32 days following trans-plantation, MSF reduced the tumor formation by 54% compared with the control group, resulting in the smallest tumor weight. Lipid peroxided content in liver increased by the transplantation of sarcoma-180 cells. However, it decreased when MSF of Kimchi was treated to the mice. MSF also suppressed xanthine oxidase activity in cytosol of the liver cells in mice transplanted by sarcoma-180 cells. Kimchi extracts had no inhibitory effect on hepatic aminopyrine-N-demethylase activity in sarcoma-180 cell transplanted or normal mice. Methanol extract and hexane extract of Kimchi slightly increased hepatic glutathione contents in sarcoma-180 treated mice. The injection of MSF from Kimchi markedly increased glutathione levels in the liver of sarcoma-180 treated mice. The injection of MSF from Kimchi markedly increased glutathione levels in the liver of sarcoma-180 treated mice compared to the controls. The MSF recovered the activities of hepatic glutathione reductase and glutathione S-transferase that decreased by the injection of sarcoma-180 cells. These results showed that MSF of Kimchi could suppress the growth of tumors, inhibiting lipid peroxide production and xanthine oxidase activity, in mice. We also suggested that Kimchi extract might play an important role in the prevention of cancer by enhancement of the glutathione level itself as well as via glutathione reductase and glutathione S-transferase.

식이내 Se수준과 알콜 섭취가 흰쥐의 지질과산화와 지방대사에 미치는 영향 (The Effect of Dietry Se Levels and Alcohol Administration on the Lipid Hyperoxidation and the Lipid Metabolism in the Rats)

  • The purposes of this studies were to investigate the effect of dietry Se levels and alcohol administration on the lipid hyperoxidation and the lipid metabolism in the rat. Seventy two male rats of Sprague-Dawley Strain weighting about 58~629 were divided into 12groups. The dietary Se levels were 10, 0.4 and Omg, and the dietary a-tocopherol levels were 150 and 0mg per kg diet, respectively. Alcohol-administrated groups received drinking water solution containing 10% of ethanol from the 3-weeks of experimental periods. The obtained experimental results are summarized as follows. 1. Food intake, body weight gain and food efficiency ratio were significantly lower in H-, L-and alcohol administrated groups(-A) by administrated Se and alcohol in diet. The weight of liver and spleen tended to be greater in H- and alcohol administrated groups. 2. The glutathione values in liver tend to be lower in alcohol or Vit. E administrated groups than nonadministrated groups. Also there were higher in H- and L- than C-groups, but the increasing range decreased due to administrated alcohol. The lipid peroxide values In liver were significantly higher in alcohol groups, and L- and tocopherol groups were higher values. Specially the increasing of lipid peroxide values were significantly effected by alcohol in low Se and Vit. E groups. 3. The contents of total glyceride in plasma were higher in alcohol groups, there were significantly higher values in alcohol administrated groups under low Se and Vit. E groups. The contents of total cholesterol and HDL-cholesterol In plasma were significantly higher in alcohol groups. 4. The contents of total lipid in liver were higher alcohol groups, and slightly higher values in low Se groups(L-groups) than other groups, also higher values in low Vit. E groups. Those of total glyceride in liver were significantly higher in alcohol groups, appeared highest values when alcohol was administrated in low Se and Vit. E groups. The increasing of total glyceride content was significantly effected by alcohol in low Se groups than that in C-groups.

β-Carotene의 보충 급여가 당뇨쥐의 지질과산화물과 항산화비타민 함량에 미치는 영향 (Effect of Dietary Supplementation of β-Carotene on Lipid Peroxide Level and Antioxidative Vitamins of Diabetic Rats)

  • 당뇨병 상태에서 항산화영양소인 carotenoids의 공급이 체내 항산화비타민의 함량에 어떠한 영향을 미치는지에 대하여 조사하고자 Sprague-Dawley종 흰쥐를 이용하여 당뇨를 유도하고 비타민 A와 E의 함량 변화를 측정하였다. 당뇨를 유도하기 전에는 체중증가량에 있어서 별다른 차이를 나타내지 않았으나 당뇨가 유도된 후에는 체중증가량이 감소되었으나 $\beta$-carotene의 섭취에 의한 영향은 나타나지 않았다. 식이섭취량은 당뇨쥐에서 대조군에 비해 별다른 차이를 나타내지 않았으나 식이효율은 대조군에 비해 당뇨군에서 유의적으로 감소되었다. 당뇨 유도로 흰쥐의 혈당치는 대조군에 비하여 2배 이상 증가하였으나 당뇨군 사이에서 $\beta$-carotene의 급여에 의한 혈당강하 효과는 관찰되지 않았다. 혈장의 retinol 함량은 당뇨군에서 유의적으로 감소하였으나 $\beta$-carotene의 급여에 의한 영향은 관찰되지 않았다. 당뇨군에서 retinol함량이 유의적으로 감소되었고 혈장의 $\alpha$-to-copherol의 함량은 혈장 지질 함량을 이용하여 보정하였을 때 당뇨군에서 현저한 감소를 보였으나 $\beta$-carotene의 급여에 의한 영향은 나타나지 않았다. 간조직 중의 retinol과 retinyl palmitate 함량은 당뇨 유발군에서 유의적인 감소를 보였으며, $\beta$-carotene 급여에 의해 증가하는 경향을 나타내었다. 그러나 $\alpha$-tocopherol 함량은 당뇨 유도와 $\beta$-carotene급여에 의해 영향을 받지 않았다. 이상의 결과를 통해 볼 때 당뇨병 환자의 혈관계 합병증을 예방하기 위해 한국인의 식생활에서 $\beta$-carotene 섭취를 효과적으로 할 수 있는 실제적인 식사관리 방안을 강구한다면 당뇨병환자의 혈관계 합병증을 완화시킬 수 있으리라고 여겨진다.

사물탕(四物湯)이 혈관내피세포(血管內皮細胞)에 미치는 영향(影響) (Effects of Samul-Tang Extract on Vascular Endothelial Cells from Hydrogen Peroxide-induced Injury)

  • This study is designed to investigate the effects of Samul-Tang extract on the response of lactic dehydrogenase(LDH) release, cellular activity, lipid peroxidation, DNA synthesis and the changes of total protein of bovine pulmonary artery endothelial cells(PAEC) from hydrogen peroxide$(H_2O_2)$-induced injury. The results are as follows : 1. Samul-Tang significantly decreased $H_2O_2$-induced release of LDH from injured bovine PAEC. 2. Samul-Tang significantly repressed $H_2O_2$-induced cellular activity from injured bovine PAEC. 3. Samul-Tang significantly repressed $H_2O_2$-induced lipid peroxidation from injured bovine PAEC. 4. Samul-Tang significantly stimulated DNA synthesis in bovine PAEC. 5. Samul-Tang significantly repressed $H_2O_2$-induced changes of total protein volume from injured bovine PAEC. Above results suggest that Samul-Tang can protect bovine PAEC from $H_2O_2$-induced injury. These results can be effectively applied to the prevention and cure of cardiovascular and cerebrovascular diseases.

실크 피브로인 분말이 혈청중의 활성산소 및 제거효소에 미치는 영향 (Effects of Silk Fibroin Powder on Oxygen Radicals and Their Scavenger Enzymes in Serum of Rats)

  • This study was designed to investigate the effects of silk fibroin powder on oxygen radicals and their scavenger enzymes in serum of rats. Sprague-Dawley(SD) male rats (160${\pm}$10 g) were fed experimental diets (SFP-2.5 and SFP-5.0 groups)added 2.5 and 5.0 g/kg BW/day for 6 weeks. Triglyceride levels were remarkably inhibited (15∼25%) in SFP-2.5 and SFP-5.0 groups compared with control group. Hydroxyl radical ($.$OH) formations resulted in a marked decrease (15%∼25%) compared with control group, while superoxide radical and hydrogen peroxide formations resulted in a significant decrease (6∼12% and 10%). Lipid peroxide and oxidized protein (>C=O group) productions resulted in a significant decrease (6∼12% and 6%) compared with control group. Superoxide dismutase (SOD) and catalase (CAT) activities were remarkably increased (35∼55% and 40∼50%), but glutathione peroxidase (GSHPX) activities were significantly increased (13%) compared with control group. These results suggest that anti-aging effect of silk fibroin powder (SFP) may play a pivotal role in attenuating a various age-related changes.

누에분말이 혈청중의 활성산소 및 제거효소에 미치는 영향 (Effects of Silkworm Powder on Oxyen Radicals and Their Scavenger Enzymes in Serum of Rats)

  • This study was designed to investigate the effects of silkworm powder (SWP) on oxygen radicals and their scavenger enzymes in serum of rats. Sprague Dawley (SD) male rats (160${\pm}$10g) were fed experimental diets (SWP-200and SWP-400 groups)added 200 and 400mg/kg BW/day for 6 weeks. Triglyceride (TG)levels were significantly inhibited (10% and 25%) in SWP-200 and SWP-400 groups, but there were no singificant differences in total, LDL-and HDL-cholesterol levels in both SWP-200 and SWP-400 groups. Hydroxyl radical ($.$OH) formations resulted in a marked decreases (about 20%) in both SWP-200 and SWP-400 groups compared with control group. Superoxide radical (O2.-)and hydrogen peroxide formations resulted in a significant decreases in SWP-400 group compared with control group. Lipid peroxide (LPO)and oxidized protein(>C=O group) productions resulted in a significant decreases (SOD) and catalase (CAT) activiteies were remarkably increased (10∼25% and 40∼50%)in SWP-200 and SWP-400groups. Glutathione peroxidase(GSHPX) activities were significantly increased (about 10%) in SWP-400 groups compared with control group. These results suggest that anti-aging effect of silkworm powder (SWP) may play a pivotal role in attenuation a various chronic degenerative diseases age-related changes.

어유섭취가 출생후 발달과정의 흰 쥐의 간과 뇌조직의 지질과산화와 그 관련기능에 미치는 영향 (Effect of Dietary Fish Oil on Lipid Peroxidation in Rats Liver and Brain During Postnatal Development)

  • 어유를 섭취하였을 때, 체내 과산화물 생성과 항산화 기능을 조사하며, 그 결과를 다른 식이지방과 비교하기 위하여, 고등어유, 대두유, 쇠기름, 들기름, 채종유의 5 종의 지방을 먹이의 10% 되게 식이를 조제하여 70g 내외의 암, 수컷의 흰쥐에게 섭취시켰다. 출생후 초기 성장시에 적응도를 관찰하기 위하여, 위의 쥐들을 34일간 조제식이로 사육한후, 교배시켜 출생한 2 대째 쥐들을 수유시기(17일, 26일)과 이유시기(39일)에 희생시키고, 출산, 수유를 마친 어미쥐(나이 123일, 총 식이일수 81일)를 희생하여 간과 뇌조직의 지질과산화물, $\alpha$-tocopherol, glutathione 양을 정량하고, glutathione peroxidase, superoxide dismutase의 활성을 측정하였다. 간조직의 지질과산화물 값은 고등어유를 섭취한 어미쥐와 수유기(17, 26일)의 새끼쥐들에게서 타군에 비해 높았으나 39일의 새끼쥐에서는 채종유보다는 높았으나 대두유, 들기름군과 같은 수준이었다. 뇌조직에서는 간에 차이가 거의없었다. 간조직의 $\alpha$-tocopherol 농도와 환원형의 glutathione(GSH) 의 농도가 어미쥐에서 타군에 비해 현저히 낮았고, 새끼쥐에서도 $\alpha$-tocopherol 경우는 같은경향이나 그 정도가 덜 현저하며, GSH의 경우는 타군들과 차이가 없었다. 산화형 glutathioe (GSSG)의 농도는 어미쥐, 새끼쥐모두에서 식이지방에 따른 차이가 일관성 있게 나타나지 않았다. Glutathione peroxidase 활성은 간장과 뇌조직에서 성장과정중의 새끼쥐들에서 모두 약간씩 증가하였고, 채종유군에서 타군에 비해 증가정도가 낮아 활성이 39일에 현저히 낮았다. Superoxide dismutase 의 활성은 성장기간에 따른 변화는 적었고, 고등어유군이 타군에 비해 비교적 낮은값을 유지하였다. 어미쥐와 17일째 새끼쥐에서는 달리 26일과 39일째 새끼쥐의 간조직 glutathioe peroxidase 의활성이 지질과산화물 농도와 정의 상관관계가 보여져, 출생후 초기 성장과정에서 체내 적응성이 존재함을 나타내주었다.

오배자 추출물이 고지방식이를 급여한 흰쥐의 간기능, 혈청지질구성 및 항산화계에 미치는 영향 (Effect of Rhus chinensis Gall Extract on Liver Function, Plasma Lipid Composition and Antioxidant System in Rats with High Fat Diet)

  • The purposes of this study is to investigate the effect of Rhus chinensis gall extract on liver function, plasma lipid composition and antioxidant system in the obese rats with high fat diet for seven weeks. Thirty two male rats of Sprague Dawely strain were divided into four groups. they are normal group (basal diet), control group(high fat diet), III group(high fat diet with Rhus chinensis gall extract 25mg/kg body weight per day) and IV group(high fat diet with Rhus chinensis gall extract 250mg/kg body weight per day). Rats in III group and IV group were administered with Rhus chinensis gall extract accordingly. Weight gains showed a tendency to decrease in rat with Rhus chinensis gall extract group but showed no differences among treatment groups. Food intake and Food efficiency ratio were not significantly different among treatment groups. Plasma total cholesterol showed a tendency to decrease in Rhus chinensis gall extract group. HDL cholesterol showed a tendency to decrease in 25mg/kg of Rhus chinensis gall extract group. However, in the 250mg/kg of Rhus chinensis gall extract group, these values showed no significant difference compared to the control group (p<0.05). GPT activities showed no significant difference among treatment groups. GOT activities showed a tendency to decrease in the groups of Rhus chinensis gall extract groups. Lipid peroxide level was significantly higher in control group than those of normal group. In Rhus chinensis gall extract groups, lipid peroxide level showed a tendency to decrease, but glutathione peroxidase activity was progressively increased. Results of our research in this paper show that Rhus chinensis gall extract might improve liver function, antioxidant system and plasma lipid composition in rats with high fat diet.

  • 결착제의 조성을 달리하여 제조한 견과-깨 엿강정의 저장 중 지방질 산화 정도와 산화방지제 함량을 평가하였다. 엿강정 제조시 결착제를 물엿 대신 꿀, 조청, 그리고 설탕 대신 트레할로스로 전부 또는 일부 대체하였을 때 엿강정의 저장 중 지방질 산화가 증가, 가속되었으며 트레할로스에 의한 산소 퍼짐도 증가가 일부 기여했을 가능성이 있다. 또한 엿강정을 저장하였을때 산화방지제 중 리그난의 안정성이 가장 높았으며, 토코페롤 분해속도는 설탕을 트레할로스로 대체한 엿강정에서 낮아 트레할로스가 토코페롤의 지방질 산화방지작용을 억제한 가능성을 제시하였다.

홍삼 산성다당체의 생리활성 연구(111)-아세트아미노펜 처리 흰쥐의 대사기능에 미치는 영향 (Biological Activities of Acidic Polysaccharide of Korean Red Ginseng.111.-Effects on Metabolizing Activities in Acetaminophen- treated Rats)

  • Pretreatment of acidic polysaccharide of Korean red ginseng (AcPS) for two weeks remarkably lowered the elevated content of lipid peroxide and levels of aminotransferases, sorbitol dehydrogenase, ${\gamma}$-glutamyltransferase, alkaline phosphatase and lactate dehydrogenase in liver intoxicated by acetaminophen (AA) . Pretreatments of AcPS also strengthen the liver function of glutathione related detoxication system indicated by glutathione contents and activities of glutathione S-transferase and glutathione reeducates which were affected by AA treatments. Activity of ${\gamma}$-glutamylcysteine syntheses was not changed by AcPS pretreatment whereas the activity of flu tathione reeducates was increased significantly. These results collectively indicate that the treatments of AcPS can promote the metabolism of lipid and reduce the production of peroxide in acetaminophen-intoxicated animals.
