Free Radical Involvement in the DNA Damaging Activity of Fumonisin Bl

  • Fumonisin B1, a mycotoxin, is thought to induce esophageal cancer in humans and apoptosis in animal cells by inhibiting ceramide synthase. Dumonisin Bl may also generate reactive oxygen species directly or indirectly, leading to DNA damage and lipid peroxidation. In this study, a DNA fragmentation assay, dichlorofluorescein (DCF) analysis, and single cell gel electrophoresis (SCGE) were used to investigate the involvement of cellular free radicals, specifically hydrogen peroxide, in the DNA damaging activity of fumonisin B1. From an in vitro DNA fragmentation assay, E. coli DNA, damage by fumonisin Bl was increased by the addition of superxide dismutase (SOD) and decreased by catalase. SCGE and DCF analysis in vivo showed that the nuclear DNA damage and intracellular free radicals in cultured rat hepatocytes treated with fumonisin B1 were increased with the concentration of fumonisin Bl . DNA damage and free radical generation were inhibited by the addition of catalase. Fumonisin Bl , in the presence of SOD, produces hydrogen peroxide causing oxidative DNA damage and protein malfunction, leading to genotoxicity and cytotoxicity of the toxin.

  • Anti-lipid peroxidative activity and phytochemical study on the leaves of Glycine max Meer. were investigated. The methanol extract of the leaves of G. max reduced the level of lipid peroxides induced by bromobenzene in vitro. From the leaves of this plant, apigenin, genistein $7-O-{\beta}-D-glucopyranoside$, kaempferol $3-O-{\beta}-D-glucopyranoside$, and kaempferol 3-O-sophoroside were isolated and characterized by spectral data.

Effects of Antioxidant Vitamins Supplementation on Antioxidative Status and Plasma Lipid Profile in Korean NIDDM Patients (항산화비타민 보충이 인슐린비의 의존형 당뇨병 환자에서 항산화상태에 혈액지방성분에 미치는 영향)

  • We investigated the effects of antioxidant vitamins supplementation on antioxidative status and plasma lipid profile in female Korean non-insulin diabetes mellitus(NIDDM) patients. Forty-five patients were groups by types of vitamin to take into three groups-Vitam in E group(400IU/day, n=15), Vitamin C group (1,000mg/day, n=15) and Vitamin E plus C group (400 IU plus 1,000mg/day). Supplementation period was 4 weeks. After vitamins supplementation, plasma vitamin E concentration significantly increased in vitamin E and vitamin E + C group, but plasma retinol concentration were not affected by vitamin E or vitamin C supplementation. And plasma levels of lipid peroxide measured as thiobarbituric acid reactive substances(TBARS), indicator of lipid peroxidation and increased susceptibility of LDL towards lipid peroxidation, were significantly decreased in all three groups after vitamins supplementation. Also catalase activities in erythrocytes were significantly decreased after antioxidant vitamin supplementation in all subjects. And after vitamins supplementation, post prandial 2 hour glucose and total cholesterol levels was decreased in all subjects. And after vitamins supplementation, post prandial 2 hour glucose and total cholesterol levels was decreased in all patients, especially there was a significant difference in vitamin C, Vitamin E + C group. In this study, antioxidant vitamins supplementation might have a protective function against the free radical-mediated lipid peroxidation and decrease the plasma total cholesterol levels in Korean female NIDDM patients.

Effects of Selected Oligosaccharides on Fecal Microflora and Lipid Constitution in Rats (다양한 종류의 올리고당이 흰쥐의 분변내 균총과 지질 성상에 미치는 영향)

  • Certain indigestible oligosaccharides may benefit gastrointestinal tract via fermentation and proliferation of desirable bacterial species. The purose of this study was to elucidate the effect of selected oligosaccharides, such as fructooligosaccharides(FOS), soybean oliosaccharides(SOE), and highly concentrated branched oligosaccharides(HiBOS), on fecal micorflora proliferation, lipid concentration, lipid peroxide formation and antioxidant enzymes acitivies in plasma and liver of the rats. Thirty two male Sprague-Dawley rats were randomly assigned to one of four treatments ; 1) control diet(AIN-93G diet); 2) control diet +5% FOS ; 3) control diet + 5% SOE ; 4) control diet + 5% HiBOS. The duration of the study was 4 weeks. Fecal bifidobacteria concentration were significantly higher (p<0.05) in the HiBOS group compared with the control after 4 weeks of dietary treatment. FOS and SOE groups also had higher fecal bifidobacteria levels than control, but statistical significance was not found. The concentration of plasma total lipid was decreased by oligosaccharide consumption, especially in HiBOS group(p<0.05). The concentration of plasma total triglyceride was significantly lower in all of the oligosaccharide containing groups compared with the control(p<0.05). The plasma total cholesterol concentration tended to be lower in the oligosaccharide consuming groups than control. The concentrations of hepatic total lipid, triglyceride and total cholesterol were not affected by consumption of oligosaccharides. Thiobarbituric acid reactive substance(TBARS) concentrations and antioxidant enzyme activities in plasma and liver were not affected much by experimental diets. There results suggest that dietary oligosaccharides may be beneficial for increasing intestinal bifidobacteria and lowering plasma lipid levels.

Effect of Polyacetylene Compounds from Korean Ginseng on Lipid Peroxidation (고려인삼의 폴리아세틸렌 성분이 과산화 지질 형성에 미치는 영향)

  • The effect of three polyacetylene compounds. panaxydol. panaxynol and panaxytriol isolated from Korean ginseng on $CCI_4-induced$ lipid peroxidation in vitro and in vivo hepatic microsomal lipid peroxidation were investigated. Lipid peroxide levels both in serum and liver and serum enzyme (GOT. GPT. LDH) activities of normal or $CCI_4-treated$ mice and rats were also determined after administration of polyacetylenes. Hepatic microsomal cytochrome P-450 content and activities of aniline hydroxylase and aminopyrine demethylase were measured after treatment of polyacetylenes with or without carbon tetrachloride. As results. treatment with polyacetylenes to control mice did not influence the levels of lipid peroxides and serum enzyme activities while panaxynol did. Panaxynol itself inhibited liver lipid peroxidation in normal mice. Polyacetylene compounds protected hepatic lipid peroxidation and lowered serum lipid peroxide levels induced by $CCI_4$ Polyacetylenes prevented leakage of LDH to serum but elevated GOT and GPT levels caused by $CCI_4$ were not changed by polyacetylene pretreatment. $CCI_4$ caused losses in the content of cytochrome P-450 and activities of aniline hydroxylase and aminopyrine demethylase. When polyacetylenes were treated without $CCI_4$ panaxydol and panaxynol induced aniline hydroxylase and all three polyacetylenes induced aminopyrine demethylase. Cytochrome P-450 contents were not affected by polyacetylenes. In vitro hepatic microsomal lipid peroxidation was inhibited by polyacetylenes and $DL-{\alpha}-tocopherol$ in a concentration-dependent manner.

Effect of Hovenia dulcis THUNBER var. koreana Nakai Fruits Extracts on Glucose, Lipid Metabolism and Antioxidant Activities in Streptozotocin Induced Diabetic Rat

  • This investigation was performed to study the antioxidant activities of Hovenia dulcis THUNBER var. koreana Nakai fruits extracts and the effect of Hovenia dulcis fruits extracts on glucose, lipid metabolism in diabetic rats. DPPH free radical scavanging activitiy and superoxide anion radical Scavenging of Hovenia dulcis fruits $80\%$ methanol extracts were $0.06\pm0.002mg$ polyphrnol/ml and $0.l2\pm0.00lmg$ polyphmol/ml, respectively. Hovenia dulcis fruits $80\%$ methanol extracts were partitioned into hexan, dichloro methane, ethyl acetate and butanol, successively. Ethyl acetate fraction were good antioxidant activity. Streptozotocin (45 mg/kg body weight, i.p.) induced diabetic rats showed a significant increases of plasma glucose, triglyceride and total cholesterol. Concomitantly significant decrease of plasma high density lipoprotein. Glutathione level were decrease in cytosol of liver. Lipid peroxide were increase in microsome of liver. Group 1 and 2 were treated with Hovenia dulcis fruits ethyl acetate extracts 50 mg/kg body weight and 20mg/kg body weight for 24 days, individually. Group land 2 rats showed decreased plasma glucose, triglyceride, total cholesterol and lipide peroxide in microsome of liver tissue of rats, and increased plasma high density lipoprotein and glutathione in cytosol of liver tissue rats. The result suggest that Hovenia dulcis THUNBER var. koreana Nakai fruits extracts may effectively normalize the impaired antioxiants status in streptozotocin induced diabetic rats. Hovenia dulcis fruits ethyl acetate extracts were used to improve the imbalance between free radicals and antioxidant system due to the diabetes.

Evaluation of Antioxidative Activity of Agrimonia pilosa-Ledeb Leaves on Non-lipid Oxidative Damage

  • Present study was conducted to evaluate the anti oxidative activity of the Agrimonia pilosa-Ledeb leaves on non-lipid oxidative damage. The antioxidative activity of methanolic (MeOH) extract of the Agrimonia pilosa-Ledeb leaves on non-lipid oxidation, including liposome oxidation, deoxyribose oxidation, protein oxidation, chelating activity against metal ions, scavenging activity against hydrogen peroxide, scavenging activity against hydroxyl radical and 2'-deoxyguanosine (2'-dG) oxidation were investigated. The MeOH extract of the Agrimonia pilosa-Ledeb leaves exhibited high anti oxidative activity in the liposome model system. Deoxyribose peroxidation was inhibited by the MeOH extract of the Agrimonia pilosa-Ledeb leaves and MeOH extract of the Agrimonia pilosa-Ledeb leaves provided remarkable protection against damage to deoxyribose. Protective effect of MeOH extracts of the Agrimonia pilosa-Ledeb leaves on protein damage was observed at $600{\mu}g$ level (82.05%). The MeOH extracts of the Agrimonia pilosa-Ledeb leaves at $300{\mu}g$ revealed metal binding ability (32.64%) for hydrogen peroxide. Furthermore, the oxidation of 2'-deoxyguanosine (2'-dG) to 8-hydroxy-2-deoxyguanosine (8-OH-2'dG) was inhibited by MeOH extracts of the Agrimonia pilosa-Ledeb leaves and scavenging activity for hydroxyl radical exhibited a remarkable effect. From the results in the present study on biological model systems, we concluded that MeOH extract of the Agrimonia pilosa-Ledeb leaves was effective in the protection of non-lipids against various oxidative model systems.

Lipid Oxidative Stability of Fried Products Added with Egg Yolk Powder During Storage

  • In this study, we examined the effects of egg yolk powder added to flour dough on the lipid oxidation of fried products during storage. The flour dough containing the egg yolk powder (0, 5, and 10%) was fried in sunflower oil at $180^{\circ}C$ for 90 sec. The fried products were then stored at $60^{\circ}C$ for 9 days in the dark. The lipid oxidation of the fried products was evaluated by fatty acid composition, peroxide values (POV), conjugated dienoic acid (CDA) contents, and thiobarbituric acid (TBA) values. The color and phospholipids (PL) contents of the fried products were also determined by colorimetry and high performance liquid chromatography, respectively. The addition of egg yolk powder to the dough decreased the POV, CDA contents, and TBA values of the fried products during storage. Although POV, CDA contents, and TBA values significantly increased in the products without egg yolk powder during storage, little change was observed in the products with egg yolk powder. The PL contents remained relatively constant in the flied products added with egg yolk powder during storage. The lightness and greenness of the fried products decreased, and the yellowness increased, as the storage time increased. The results clearly indicate that the addition of egg yolk powder to the dough improved the lipid oxidative stability of the fried products during storage in the dark, and the PL in the egg yolk might have contributed to the improvements in lipid oxidative stability.

Effects of Cervus elaphus extract solution for Aqua-acupuncture on Antioxidation in Rat's Brain induced by ischemia and reflow (허혈(虛血)/재관류(再貫流) 뇌(腦) 조직(組織)에서 녹용(鹿茸) 약침액(藥鍼液)의 항산화작용(抗酸化作用)에 대한 연구(硏究))

  • Cervus elaphus, being known to reinforce Kidney, have tested to study the effects concerning damages of brain tissue induced by lipid peroxidation. In vitro, the level of lipid peroxide in brain tissue was decreased proportinally according to dose by Cervus elaphus extract solution for aqua-acupuncture (CESAA). It was much more decreased, when lipid peroxidation was induced with Fe(II). And, it was seen proportinally decrease according to the dose of CESAA on xanthine oxidase activities and type conversion ratio. However, I can not find special changes about aldehyde oxidase activities. And, I had observed the effects of CESAA on damages of rat's brain following ischemia and reflow. Before ischemia was caused, CESAA was applied 0.2 ml per 250 g through femoral vein in ischemia and reflow group and normal sailine was applied in normal group. Ischemia was caused by cervical artery's clamp for 30 min and reflowed by clamp remove after 15 min. It was increased on the content of lipid peroxidation, activies and type conversion ratio of xanthine oxidase following ischemia and reflow. However, they were decreased when CESAA was pre-appllied. However, it could not seen special changes on aldehyde oxidase activities, either. In conclusion. CESAA recovers the damage of brain due to ischemia and reflow by decreasing the lipid peroxidation through decreasing of xanthine oxidase activies and type conversion ratio.

Changes of Fatty Acid Composition and Lipid Oxidation in Anchovy During Fermentation with Salt (멸치젓 숙성중 지질의 산화와 지방산 조성의 변화)

  • Anchor Engraulis japonica was fermented with $10\%$ and $20\%$ salt at $10^{\circ}C$ ($10\%$ 5A) and $20^{\circ}C$ ($20\%$ SA), respectively, and the changes in lipid content, fatty acid composition and lipid oxidation was observed for 105 days. Peroxide value (POV) in $10\%$ SA reached to the maximum (46.4 meq/kg) on 30 days of fermentation, while in $20\%$ SA, it reached the maximum (54.7 meq/kg) on 45 days of fermentation, and then decreased in both samples during fermentation. Thiobarbituric acid values revealed a similar tendency with the change of POV during the fermentation. These results indicated that lipid of the anchovy was oxidized faster in lower salt than high salt in the early stage. Total lipid (TL) content during the fermentation for 105 days decreased approximately $16\%$ in $10\%$ SA and approximately $33\%$ in $20\%$ SA. Phospholipid (PL) content also decreased in both samples and the content of neutral lipid (NL) was unchanged in $10\%$ SA, while it decreased in the same amount as PL in $20\%$ SA. The prominent fatty acids in TL of the anchovy sample were 14 : 0, 16 : 0, 16 : In-7, 18 : in-9, 20 5n-3 and 22 : 6n-3. After fermentation for 105 days, approximately $87\%$ and $67\%$ of the prominent fatty acids remained in $10\%$ SA and $20\%$ SA, respectively, but the kind of the fatty acids was unchanged.
