A Convergent Synthesis of Bis-2-oxo Amide Triacylglycerol Analogues as Potent Lipase Inhibitors Using Acyl Cyanophosphorane Methodology

  • A number of bis-2-oxo amide triacylglycerol analogues, a recently reported potent human gastric lipase inhibitor and its new analogues, have been prepared starting from 1,3-dibromo-2-propanol utilizing acyl cyanophosphorane methodology as a key step in a convergent manner. The key coupling reaction has been accomplished at -$78^{\circ}C$ between 1,3-diamino-2-propanol derivative and the labile diketo nitriles, derived from acyl cyanotriphenylphosphoranes upon oxidizing with $O_3$, under mild condition in moderate yields.

Inhibitory Effect of Thujae orientalis Semen Extract on Pancreatic Lipase Activity (백자인 추출물에 의한 pancreatic lipase의 저해 효과)

  • The possible presence of inhibitors of pancreatic lipase (tricaylglycerol acylhydrolase EC 3.1.1.3) was screened from Korean traditional edible or medicinal herbs. Among tested herbs, Arecae pericarpium, Mucunae Caulis, Rhus javanica, Thujae orientalis were shown to have strong inhibitory effect against pancreatic lipase. Thujae orientalis was finally selected as a candidate for pancreatic lipase inhibitor. The extract of Thujae orientalis was showed selective inhibition on porcine pancreatic lipase activity. Active inhibitors, TF-1, TF-2, TF-3, were purified from an extract of Thujae orientalis, using chloroform extraction, followed by successive chromatography in silica gel and LH-20 and high performance liquid chromatography (HPLC). The $IC_{50}$ values of TF-1, TF-2, TF-3 and orlistat were 44.7, 98.7, 46.1 and $27.6{\mu}g/ml$, respectively. And also the TF-2 and orlistat were shown to be inhibitory effect on the differentiation of preadipocyte NIH-3T3 L1 cells at a concentration of $10{\mu}g/ml$.

  • Activity-guided fractionation of an ethyl acetate (EtOAc)-soluble portion of an ethanolic extract from the leaves of cultivated mountain ginseng, using pancreatic lipase inhibition assay, led to the isolation and identification of three flavonoids of a previously described structure, kaempferol-3-O-sophoroside (I), kaempferol-3-O-${\beta}$-Dglucopyranoside (astragalin, II) and kaempferol (III). All compounds (I.III) showed pancreatic lipase inhibitory activities, with $IC_{50}$ values ranging from $20.3{\pm}2.2$ to $9.1{\pm}1.5$ ${\mu}M$, kaempferol (III) showed the most potent inhibitory activity with an $IC_{50}$ of $9.1{\pm}1.5$ ${\mu}M$. The level of activity may depend on the number of C-3 glucosyl group(s) linked to the kaempferol backbone, and the isolated compounds may have promise as pancreatic lipase inhibitors.

  • To investigate the effects of trypsin inhibitors, aprotinin and urinary trypsin inhibitor (UTI), on the cute pancreatitis, this study was carried out in dogs of acute pancreatitis induced by oleic acid (0.28 mg/kg). Administration with aprotinin and UTI seemed to have a therapeutic effect on the clinical sign, ultrasonographic finding, histopathologic finding. But in amylase and lipase activity, there were no significant differences among three groups.

Pancreatic lipase Inhibitory Compound from Apis mellifera venome

  • While searching for pancreatic lipase inhibitors, the active compound was found in a methanol extract of Apis mellifera venome. The active compound was isolated by Diaion HP-20 column chromatography, thin layer chromatography and HPLC. The active compound is stable to the extreme pH and heat. There is no loss of activity both in acidic and alkaline solution in the pH range of 2 to 11 by heating for 15 minutes at $90^{\circ}C$. The rf value of the compound was 0.51 at TLC with butanol : methanol: water (4:1:2) solvent system. The molecular weight of the compound was determined to be 293 by EI-MS.

  • Activity-guided isolation from an ethylacetate-soluble fraction of a 70% (v/v) ethanolic extract from the roots of Taraxacum ohwianum, using a pancreatic lipase inhibition assay, resulted in isolation and identification of five phenolic metabolites of previously known structure; these were 3,5-di-O-caffeoylquinic acid, chicoric acid, caffeic acid, protocatechuic aldehyde, and luteolin. All structures were confirmed by NMR and MS scpectroscopic data. Of these compounds 3,5-di-O-caffeoylquinic acid exhibited the most potent inhibitory activity, with $IC_{50}$ of $65.1{\pm}0.7\;{\mu}M$ against pancreatic lipase.

  • A series of ${\gamma}$-aminobutyric acid-derived, potent human cytosolic phospholipase A$_2$ inhibitors have been prepared from acyl cyanophosphoranes and amine derivatives in a convergent manner. The ${\alpha}$-keto amide functionalities in the inhibitors have been introduced as electrophilic fragments via direct coupling reactions between the labile ${\alpha},{\beta}$-diketo nitriles and ${\gamma}$-aminobutyric acid t-butyl ester derivatives at -78 $^{\circ}C$ in moderate to good yields.

Screening of Medicinal Plants for Development of Functional Food Ingredients with Anti-Obesity (항비만 기능성 식품소재 개발을 위한 생약 추출물의 탐색)

  • In order to search anti-obesity agents, the methanol extracts of 155 herbal medicines were screened using with pancreatic lipase, which is involved in conversion of triglycerol to fatty acid. Among the tested medicinal plants, methanol extracts of Amsonia elliptica, Arecae pericarpium, Biota orientalis, Cinnamomum cassia, Curcuma aromatia, Elsholtzia ciliate, Glycyrrhiza uralensis, Mucunae Caulis, Rhus javanica, and Rubus coreanus showed potent inhibition at final concentration of $200\;{\mu}g/ml$ on pancreatic lipase activity. All of them were extracted into chloroform fraction. The relative inhibitory activities against pancreatic lipase by orlistat, the chloroform fraction of Arecae pericarpium and Cinnamomum cassia were 89, 80 and 80%, respectively. These results demonstrated that the screened medicinal plants could be develop as the effective lipase inhibitors in preventing and ameliorating obesity of human beings.

Anti-lipase and Lipolytic Activities of EtOH Extract from Juniperus rigida (노간주나무 에탄올추출물의 지방 흡수 억제 및 지방분해 효능)

  • Obesity is an important risk factor that significantly increases mortality and disease rates in the cardiovascular disease, diabetes, and various diseases. So far, the most powerful way to inhibit fat absorption is pancreatic lipase inhibitors. In this study, we investigated the anti-obesity effect of the extract of Juniperus rigida. Juniperus rigida extract (JRE) had a inhibitory effect on pancreatic lipase activity ($IC_{50}$=8.63 ${\mu}g$/ml). In in vivo oil-emulsion loading test, this extract also inhibited the intestinal fat absorption. In addition, we measured inhibitory effects of JRE on activity of phosphodiesterase (PDE) and hormone sensitive lipase (HSL) among the important enzymes associated with lipolysis. JRE strongly inhibited PDE activity ($IC_{50}$=4.56 ${\mu}g$/ml), whereas inhibitory effect on HSL activity was very weak compared with orlistat. As a result, JRE inhibited the absorption of fat by inhibiting the activity of pancreatic lipase and induced lipolysis through inhibition of PDE activity. Therefore, we suggest that Juniperus rigida may be a potential therapeutic agent improving obesity.
