Production, Immobilization, and Characterization of Croceibacter atlanticus Lipase Isolated from the Antarctic Ross Sea (남극 로스해에서 분리한 Croceibacter atlanticus균 유래 리파아제의 생산, 고정화, 효소특성 연구)

  • The Antarctic Ocean contains numerous microorganisms that produce novel biocatalysts that can have applications in various industries. We screened various psychrophilic bacterial strains isolated from the Ross Sea and found that a Croceibacter atlanticus strain (Stock No. 40-F12) showed high lipolytic activity on a tributyrin plate. We isolated the corresponding lipase gene (lipCA) by shotgun cloning and expressed the LipCA enzyme in Escherichia coli cells. Homology modeling of LipCA was carried out using the Spain Arreo lake metagenome alpha/beta hydrolase as a template. According to the model, LipCA has an ${\alpha}/{\beta}$ hydrolase fold, Gly-X-Ser-X-Glymotif, and lid sequence, indicating that LipCA is a typical lipase enzyme. Active LipCA enzyme was purified fromthe cell-free extract by ammonium sulfate precipitation and gel filtration chromatography. We determined its enzymatic properties including optimum temperature and pH, stability, substrate specificity, and organic solvent stability. LipCA was immobilized by the cross-linked enzyme aggregate (CLEA) method and its enzymatic properties were compared to those of free LipCA. After cross-linking, temperature, pH, and organic solvent stability increased considerably, whereas substrate specificities did not changed. The LipCA CLEA was recovered by centrifugation and showed approximately 40% activity after 4th recovery. This is the first report of the expression, characterization, and immobilization of a C. atlanticus lipase, and this lipase could have potential industrial application.

  • For many years a wide variety of surgical techniques for closure of cleft lip has been used. Still many surgeons prefer the Millard's rotation-advancement lip repair because the surgical scar is masked in the philtral crest and the nostril floor, and it improves the relationship of the alar base of the cleft side, producing harmonious symmetry of the nostril and the nostril sill. In addition, it uses and preserves the lip anatomy, returning lip tissue into its normal position, minimizing the amount of tissue that is discarded, and reconstructing the orbicular oris muscle. One of the major disadvantages of this procedure is the lack of accurate measurements. The object of this study is to help in the cleft lip surgery with investing its features and design.

  • Bimodal speech recognition based on lip reading has been studied as a representative method of speech recognition under noisy environments. There are three integration methods of speech and lip modalities as like direct identification, separate identification and dominant recording. In this paper we evaluate the robustness of lip reading methods under the assumption that lip parameters are estimated with errors. We show that the dominant recording approach is more robust than other methods with lip reading experiments. Also, a measure of lip parameter degradation is proposed. This measure can be used in the determination of weighting values of video information.

Finite Element Analysis of Contact Behaviors of Rubber Lip Seals (고무립시일의 접촉거동에 관한 유한요소해석)

  • This paper deals with a FEM computation as well as measurement of the contact force and distribution of the temperature in rubber lip seals when the sealing interference between the rotating shaft and the lip edge is present. The study of the contact forces and distributions of the temperature and the sealing contact stress has always been one of the basic steps in the process of designing a lip seal. The calculated FEM results indicate that as the sealing interference increases, the contact force moderately increases compared with decreased sealing interference at the seal lip edge and radically increases the contacting width. And the FEM computation of oontact forces including nonlinear problems has been compared with experimental measurements with good agreement. The frictional heat does not dissipate promptly in the rubber seal lip and tends to accumulate at the contacting lip edge especially.

  • The treatment of cleft lip and palate patients requires multidisciplinary coorperation, and the involved clinicians rely on the completeness and accuracy of the patient's medical records in developing comprehensive treatment plans. There are so many classifications in cleft lip and palate but each classification has advantages and disadvantages. Furthermore there are few classification or assessment in secondary cleft lip and palate deformities. A modification of Kenahan's Y classification in primary cleft lip and palate and new classification in secondary cleft lip and palate deformities are proposed as a simple and reproducible method. These reproducible classification may be used to facilitate not only storing and analyzing of medical informations in computer but also the planning of secondary repairs

  • Unilateral cleft lip is a common congenital anomaly that affects the appearance and function of the upper lip and nose. Surgical repair of cleft lip aims to restore the normal anatomy and functionality of the affected structures. In recent years, several advances have been made in the field of cleft lip repair, including new surgical techniques and approaches. This comprehensive review discusses the surgical management of patients with unilateral cleft lip and palate and provides step-by-step instructions for the surgical procedures.

A survey of trend in cleft lip and palate management in oral & maxillofacial surgery in Korea. (한국 구강악안면외과 수련병원의 구순구개열 환자의 치료경향에 관한 조사)

  • The purpose of this survey was to obtain an overall view of the status of cleft lip and palate management in Oral and Maxillofacial Surgery(OMFS) in Korea. Korea Cleft lip and Palate Association conducted mailed survey which was composed of 29 questions conceming pre-operative and post-operative management in CLP surgery and completed by 58 training hospital. Of 58 questionnaires sent, 23 were returned(response rate of 40%). Authors compared this results with those of other similar surneys, which reveal much difference between OMFS and Plastic Surgery(PS) in CLP treatment. In OMFS, many surgeons were in favor of presugical orthopaedics than lip adhesion to manage protruded premaxilla. It reflects interdisciplinary team approach between OMFS and orthodontic department reduces the need of lip adhesion through presurgical orthopaedics. Timing of palatal surgery was later than that of PS, which reflects concern for an impediment of maxillaty growth. To our knowledge, this survey may be the first on organization and management for cleft patient in OMFS in Korea.

  • Bimodal speech recognition systems have been proposed for enhancing recognition rate of ASR under noisy environments. Visual feature extraction is very important to develop these systems. To extract visual features, it is necessary to detect exact lip position. This paper proposed the method that detects a lip position using color similarity model and SVM. Face/Lip color distribution is teamed and the initial lip position is found by using that. The exact lip position is detected by scanning neighbor area with SVM. By experiments, it is shown that this method detects lip position exactly and fast.

  • More than 50% of sepsis cases are associated with pneumonia. Sepsis is caused by infiltration of bacteria into the blood via inflammation, which is triggered by the release of cell wall components following lysis. However, the regulatory mechanism of lysis during infection is not well defined. Mice were infected with Streptococcus pneumoniae D39 wild-type (WT) and lipase mutant (${\Delta}lipA$) intranasally (pneumonia model) or intraperitoneally (sepsis model), and survival rate and pneumococcal colonization were determined. LipA and autolysin (LytA) levels were determined by qPCR and western blotting. S. pneumoniae Spd_1447 in the D39 (type 2) strain was identified as a lipase (LipA). In the sepsis model, but not in the pneumonia model, mice infected with the ${\Delta}lipA$ displayed higher mortality rates than did the D39 WT-infected mice. Treatment of pneumococci with serum induced LipA expression at both the mRNA and protein levels. In the presence of serum, the ${\Delta}lipA$ displayed faster lysis rates and higher LytA expression than the WT, both in vitro and in vivo. These results indicate that a pneumococcal lipase (LipA) represses autolysis via inhibition of LytA in a sepsis model.

  • A comparative study was undertaken to investigate the collapse of maxillary dental arch and palate in unilateral cleft lip and palate individuals. The material for this study consisted of 39 subjects with repaired unilateral cleft lip and palate (30 males, 9 females). The measurements of unilateral cleft lip and palate individuals were compared with the measurements of normal individuals (30 males, 30 females). All the subjects were in the mixed dentition stage and the mean age was almost the same. The following conclusions were obtained. 1. A large number of the maxillary dental arch of the unilateral cleft lip and palate individuals showed ${\Omega}$-shape, and the arch length was shorter than that of normal individuals. The intermolar width did not show significant difference between cleft group and group, but the intercanine width was mcuh smaller than that of normal individuals. 2. The palate of the unilateral cleft lip and palate subjects showed shorter and shallower form than that of normal subjects. 3. The palatal area of the unilateral cleft lip and palate subjects was smaller than that of normal subjects, and the cleft side area was much smaller than the opposing side area. 4. There was no significant sexual difference in measurements of maxillary dental arch and palate of the unilateral cleft lip and palate subjects.
