  • T-2 toxin is one of mycotoxins produced by fungi such as Fusarium spp. and possesses a potent cytotoxicity to eukaryotic cell. The contamination of mycotoxins in cereals and feedstuffs is one of the great concerns in health authorities. Therefore, the development of the specific, sensitive and simplified analysis method for T -2 toxin is required. During more than ten years, several chemical and biological analysis methods were proposed and applied for the detection and quantification of T-2 toxin. TLC, GLC-FID and GC-MS are widely employed, but these methods required numerous clean-up procedures before analysis, and the detection limit for T-2 toxin is more than 10 ppb. Biological analysis methods with dermal tissues and cultured cells are not specific to T-2 toxin, since T-2 toxin and other related derivatives possess a similar toxicological activity although their relative activity is different each otber. Based on tbe specific reaction between antibody and antigen, the authors tried to introduce the immunochemical methods for determination of T-2 toxin. The enzyme-linked immunosorbent assay method using monoclonal antibody for T-2 toxin was applied to analyse T-2 toxin. The detection limit of T-2 toxin by ELISA method was 0.1 ppb. The correlation between ELISA and GC-MS method on these samples was very high. ELISA method developed for the detection and quantification of T -2 toxin in this paper possesses simplicity, high sensitivity and specific for T-2 toxin. Furthermore, the ELISA method with T-2 toxin monoclonal antibody was an excellent tool for the screening of Fusarium spp. which was suspected to produce T-2 toxin.

  • The scale and operating conditions of logistical systems very sensitively varies according to the variation of traffic intensity that is affected by the arrival characteristics of trucks and the attributes of loading/unloading services in logistics facilities. More exactly, logistics costs are incurred according to variations of traffic intensity. which are intimately linked with in a given time period. Also. although traffic intensity changes minutely, the range of cost variation is wide. Nevertheless, with regard to operating logistics systems, the existing studies make no attempt to analyze these factors. Therefore, it was the purpose of this study to determine the optimal traffic intensity to minimize excessive logistics costs resulting from the generation of unnecessary costs such as waiting costs and overcosts in operating a facility. For the purposes of this analysis. a determination model of optimal traffic intensity was constructed according to queuing theory. The inflow/outflow conditions of trucks and the terminal operational conditions were collected from an off-dock container terminal in Busan. On the basis of this data. the optimal traffic intensity that could off-set excessive waiting and operating costs was determined quantitatively. Also. using the optimal traffic intensity to be determined. we consider the improvements of operating system in the logistics facilities.

  • The aim of this study was to determine the brittle fracture behavior of reactor pressure vessel steel by considering the temperature dependence of a damage model. A multi-island genetic algorithm was linked to a Weibull stress model, which is the model typically used for brittle fracture evaluation, to improve the calibration procedure. The improved calibration procedure and fracture toughness test data for SA508 carbon steel at the temperatures $-60^{\circ}C$, $-80^{\circ}C$, and $-100^{\circ}C$ were used to decide the damage parameters required for the brittle fracture evaluation. The model was found to show temperature dependence, similar to the case of NUREG/CR-6930. Finally, on the basis of the quantification of the difference between 2- and 3-parameter Weibull stress models, an engineering equation that can help obtain more realistic fracture behavior by using the simpler 2-parameter Weibull stress model was proposed.

  • To evaluate the involvement of translation initiation factors eIF4E and eIFiso4E in Chilli veinal mottle virus (ChiVMV) infection in pepper, we conducted a genetic analysis using a segregating population derived from a cross between Capsicum annuum 'Dempsey' containing an elF4E mutation ($pvr1^2$) and C. annuum 'Perennial' containing an elFiso4E mutation (pvr6). C. annuum 'Dempsey' was susceptible and C. annuum 'Perennial' was resistant to ChiVMV. All $F_1$ plants showed resistance, and $F_2$ individuals segregated in a resistant-susceptible ratio of 166:21, indicating that many resistance loci were involved. Seventy-five $F_2$ and 329 $F_3$ plants of 17 families were genotyped with $pvr1^2$ and pvr6 allele-specific markers, and the genotype data were compared with observed resistance to viral infection. All plants containing homozygous genotypes of both $pvr1^2$ and pvr6 were resistant to ChiVMV, demonstrating that simultaneous mutations in elF4E and eIFiso4E confer resistance to ChiVMV in pepper. Genotype analysis of $F_2$ plants revealed that all plants containing homozygous genotypes of both $pvr1^2$ and pvr6 showed resistance to ChiVMV. In protein-protein interaction experiments, ChiVMV viral genome-linked protein (VPg) interacted with both eIF4E and eIFiso4E. Silencing of elF4E and eIFiso4E in the VIGS experiment showed reduction in ChiVMV accumulation. These results demonstrated that ChiVMV can use both eIF4E and eIFiso4E for replication, making simultaneous mutations in eIF4E and eIFiso4E necessary to prevent ChiVMV infection in pepper.

  • The purpose of this study was to determine the association between blood lipids, fibrinogen, fasting blood sugar, lifestyle-related factors and lipoprotein(a). This study was performed with 140 healthy adults (male:80, female:60) among the teachers in primary, middle, and high school in Seoul in November 1996. Lipoprotein(a) concentration was measured with an enzyme-linked-immunosorbents assay(ELISA), and the results showed that the distribution was highly skewed and the average concentrations of male and female were $16.94{\pm}12.70\;mg/dL\;and\;17.92{\pm}7.58\;mg/dL$, respectively. The percentage of subject with lipoprotein(a) concentrations higher than 30 mg/dL was 26.4%. Univariate analysis showed significant association between lipoprotein(a) and age, alcohol, meat, and NaCl consumption (p<0.05). In multivariate analysis, lipoprotein(a) correlated positively with age (p<0.05) and meat consumption (p<0.01). These data suggest that lifestyle-related factors may affect the lipoprotein(a) concentration regarded as a risk factor of atherosclerosis disease.

  • The main objective of water distribution system is to supply enough water to users with proper pressure. Hydraulic analysis of water distribution system can be divided into Demand Driven Analysis (DDA) and Pressure Driven Analysis (PDA). Demand-driven analysis can give unrealistic results such as negative pressures in nodes due to the assumption that nodal demands are always satisfied. Pressure-driven analysis which is often used as an alternative requires a Head-Outflow Relationship (HOR) to estimate the amount of possible water supply at a certain level of pressure. However, the lack of data causes difficulty to develop the relationship. In this study, effective supply, which is the possible amount of supply while meeting the pressure requirement in nodes, is proposed to estimate the serviceability and user's convenience of the network. The effective supply is used to calculate Subsystem Importance Index (SII) which indicates the effect of isolating a subsystem on the entire network. Harmony Search, a stochastic search algorithm, is linked with EPANET to maximize the effective supply. The proposed approach is applied in example networks to evaluate the capability of the network when a subsystem is isolated, which can also be utilized to prioritize the rehabilitation order or evaluate reliability of the network.

  • Duroc is widely used to improve the meat quality and productivity. To elucidate the phylogenetic relation and the sequence specificity for the maternal property, the complete sequence of mitochondrial genome was determined and the population diversity of Duroc was investigated in this study. The length of mtDNA tested is 16,584-bp. There are several insertion/deletion mutations in the control region and coding regions for tRNA and rRNA, respectively, but not in peptide-coding regions. Four peptide-coding genes(COⅡ, COⅢ, ND3 and ND4) showed incomplete termination codon sequences such as T--, and two(ND2 and ND4L) did alternative initiation codons(AIC), respectively. Especially, the initiation codon sequences of ND2 gene were polymorphic in this population. Polymorphisms were detected in 11-bp duplication motif within control region as well as ND2 and CYTB. Variation patterns observed from the tests on three mtDNA regions were linked completely and then two haplotypes obtained from combining the data dividing this population. Duroc mtDNA is observed at the European pig cluster in the phylogenetic tree, however, the results from the population analyses supported previous opinions. This study suggests that the breed Duroc was mainly originated from the European pig lineage, and Asian lineage was also used to form the pig breed Duroc as maternal progenitors.

  • Dopamine (DA) is an important neurotransmitter molecule of catecholamines. Its deficiency could lead to brain disorder such as Parkinson's disease and schizophrenia. Therefore, it is necessary to establish a suitable analytical technique with sensitivity and simplicity. A competitive enzyme-linked immunosorbent assay for DA has been optimized and characterized. Assay sensitivity is controlled by two factors in competitive immunoassay. One is a nature and concentration of competitor, and the other is those of binder, antibody. Thus, optimization was performed: BSA-DA conjugate and antibody-avidin conjugate were prepared by dual heterobifunctional coupling method using SATA and SMCC. Assay condition was optimized with $6.66\;{\mu}gmL^{-1}$ of BSA-DA and $4.17{\times}10^{-10}\;M$ of antibody-avidin conjugate. A dose-response curve was constructed, and a limit of detection and a dynamic range for DA were accomplished to $2.3{\times}10^{-2}\;{\mu}g\;mL^{-1}$ and four orders of magnitude ($1.0{\times}10^{-7}\;M$ to $1.0{\times}10^{-3}\;M$), respectively. Calibration curve was constructed on dynamic range and least-squares regression of this data gave the following relationship: absorbance = -0.1098 log[DA]+0.0353 ($R^2$ = 0.9956).

  • This paper suggests that LOD establishes its own link policy and publishes it to LOD cloud to provide identity among entities in different LODs. For specifying the link policy, we proposed vocabulary set founded on RDF model as well. We implemented Policy based In-depth Searching and Cleansing(PISC for short) system that proceeds in-depth searching across LODs by referencing the link policies. PISC has been published on Github. LODs have participated voluntarily to LOD cloud so that degree of the entity identity needs to be evaluated. PISC, therefore, evaluates the identities and cleanses the searched entities to confine them to that exceed user's criterion of entity identity level. As for searching results, PISC provides entity's detailed contents which have been collected from diverse LODs and ontology customized to the content. Simulation of PISC has been performed on DBpedia's 5 LODs. We found that similarity of 0.9 of source and target RDF triples' objects provided appropriate expansion ratio and inclusion ratio of searching result. For sufficient identity of searched entities, 3 or more target LODs are required to be specified in link policy.
