Validation of a Selective Method for Simultaneous Determination of Doxifluridine and 5-Fluorouracil in Dog Plasma by LC-MS/MS (LC/MS/MS를 이용한 비글견의 혈장 중 Doxifluridine 및 5-Fluorouracil의 동시 분석법 Validation)

  • A simple, sensitive and selective liquid chromatographic/tandem mass spectrometric method (LC-MS/MS) was developed and validated for doxifluridine and 5-fluorouracil (5-FU) quantification in dog heparinized plasma. Sample preparation was based on liquid-liquid extraction using a mixture of isopropanol/ethyl acetate (1/9 v/v) to extract doxifluridine, 5-FU and 5-chlorouracil (5-CU, an internal standard) from plasma. Chromatography was performed on a C-18 analytical column and the retention times were 2.7, 1.5 and 1.7 min for doxifluridine, 5-FU and 5-CU, respectively with shorter analysis time within 5 min than previously reported methods. The ionization was optimized using ESI negative mode and selectivity was achieved by tandem mass spectrometric analysis by multiple reaction monitoring (MRM) using the transformations of m/z 244.8>107.6, 129.0>42.0 and 144.9>42.1 for doxifluridine, 5-FU and 5-CU, respectively. The achieved low limit of quantification was 20.0 ng/mL and the assay exhibited linear range of 20-2000 ng/mL ($R^2>0.99957$ for doxifluridine and $R^2>0.99857$ for 5-FU), using $100{\mu}L$ of plasma. Accuracy and precision of quality control samples for both doxifluridine and 5-FU met KFDA and FDA Guidance criteria of 15% for accuracy with coefficients of variation less than 15%. This method demonstrated adequate sensitivity, specificity, accuracy, precision and stability to support the simultaneous analysis of doxifluridine and 5-FU in dog plasma samples in pharmacokinetic and bioequivalence studies.

Optimized Sigma-Delta Modulation Methodology for an Effective FM Waveform Generation in the Ultrasound System (효율적인 주파수 변조된 초음파 파형 발생을 위한 최적화된 시그마 델타 변조 기법)

  • A coded excitation has been studied to improve the performance for ultrasound imaging in term of SNR, imaging frame rate, contrast to tissue ratio, and so forth. However, it requires a complicated arbitrary waveform transmitter for each active channel that is typically composed of a multi-bit Digital-to-Analog Converter (DAC) and a linear power amplifier (LPA). Not only does the LPA increase the cost and size of a transmitter block, but it consumes much power, increasing the system complexity further and causing a heating-up problem. This paper proposes an optimized 1.5bit fourth order sigma-delta modulation technique applicable to design an efficient arbitrary waveform generator with greatly reduced power dissipation and hardware. The proposed SDM can provide a required SQNR with a low over-sampling ratio of 4. To this end, the loop coefficients are optimized to minimize the quantization noise power in signal band while maintaining system stability. In addition, the decision level for the 1.5 bit quantizer is optimized for a given input waveform, which results in the SQNR improvement of more than 5dB. Computer simulation results show that the SQNR of a FM(frequency modulated) signal generated by using the proposed method is about 26dB, and the peak side-lobe level (PSL) of its compressed waveform on receive is -48dB.

Genetic Transformation of the Yeast Dekkera/Brettanomyces bruxellensis with Non-Homologous DNA

  • Yeast Dekkera/Brettanomyces bruxellensis is probably the most common contaminant in wineries and ethanol production processes. The considerable economic losses caused by this yeast, but also its ability to produce and tolerate high ethanol concentrations, make it an attractive subject for research with potential for industrial applications. Unfortunately, efforts to understand the biology of D. bruxellensis and facilitate its broader use in industry are hampered by the lack of adequate procedures for delivery of exogenous DNA into this organism. Here we describe the development of transformation protocols (spheroplast transformation, LiAc/PEG method, and electroporation) and report the first genetic transformation of yeast D. bruxellensis. A linear heterologous DNA fragment carrying the kanMX4 sequence was used for transformation, which allowed transformants to be selected on plates containing geneticin. We found the spheroplast transformation method using 1M sorbitol as osmotic stabilizer to be inappropriate because sorbitol strikingly decreases the plating efficiency of both D. bruxellensis spheroplast and intact cells. However, we managed to modify the LiAc/PEG transformation method and electroporation to accommodate D. bruxellensis transformation, achieving efficiencies of 0.6-16 and 10-20 transformants/${\mu}g$ DNA, respectively. The stability of the transformants ranged from 93.6% to 100%. All putative transformants were analyzed by Southern blot using the kanMX4 sequence as a hybridization probe, which confirmed that the transforming DNA fragment had integrated into the genome. The results of the molecular analysis were consistent with the expected illegitimate integration of a heterologous transforming fragment.

Validation of and HPLC Method for Nadolol in Human Plasma and Evaluation of Its Pharmacokinetics after a Single-dose in Korean Volunteers (인체 혈장 중 나돌올의 HPLC 분석법 검증 및 단회투여 후 약물동태 연구)

  • A high-performance liquid chromatographic method was validated for quantitation of nadolol in human plasma. Nadolol and internal standard, pindolol, were extracted with tert-butyl methyl ether after addition of 10 M sodium hydroxide solution. The analytes were separated on a reverse phased C18 column using a mobile phase consisting of 0.05 M ammonium phosphate monobasic buffer, acetonitrile and methanol (81: 17:2 v/v/v) and detected using a fluorescence detector (excitation wavelength 230 nm, emission wavelength 294 nm). The method was specific and sensitive enough to detect as low as 3 ng/mL of nadolol in human plasma. Linear calibration range was 3-150 ng/mL with correlation coefficient greater than 0.999. The overall accuracy was in the range of 96.8 to 103% and precision C.V.(%) 7.30 to 12.2%. The recovery was approximately 100% and stability was confirmed during storage and sample preparation. The present HPLC method was successfully applied to study bioavailability after oral administration of 80 mg of nadolol in healthy Korean subjects. The mean $AUC_{t}$ was $1968{\pm}397\;ng{\cdot}hr/mL$ and $C_{max}$ of $186{\pm}79.3\;ng/mL$ was reached at $3.5{\pm}0.76\;hr$. The mean $t_{1/2}$ of nadolol was $17.3{\pm}2.59\;hr$.

The Simulator Design for the Analysis of Aircraft Longitudinal Dynamic Characteristics (항공기 세로 동특성 해석을 위한 시뮬레이터 설계)

  • State-space method for the analysis of the dynamic characteristics of a body motion is set up as mathematical tool for the solution of differential equation by computer. Representation of a system is described as a simple form of matrix calculation and unique form of model is available for the linear or nonlinear, time variant or time invariant, mono variable or multi variable system etc. For the analysis of state-space method a complicated vector calculation is required, but this analysis can be simplified with the specific functions of a software package. Recently as the Graphical User Interface softwares are well-developed, then it is very simplified to execute the simulation of the dynamic characteristics for the state-space model with the interactive graphics treatment. The purpose of this study is to developed the simulator for the educational analysis of the dynamic characteristics of body motion, and for the analysis of the longitudinal dynamic characteristics of an aircraft that is primarily to design the simulator for the analysis of the transient response of an aircraft longitudinal stability.

Video Augmentation of Virtual Object by Uncalibrated 3D Reconstruction from Video Frames (비디오 영상에서의 비보정 3차원 좌표 복원을 통한 가상 객체의 비디오 합성)

  • This paper proposes a method to insert virtual objects into a real video stream based on feature tracking and camera pose estimation from a set of single-camera video frames. To insert or modify 3D shapes to target video frames, the transformation from the 3D objects to the projection of the objects onto the video frames should be revealed. It is shown that, without a camera calibration process, the 3D reconstruction is possible using multiple images from a single camera under the fixed internal camera parameters. The proposed approach is based on the simplification of the camera matrix of intrinsic parameters and the use of projective geometry. The method is particularly useful for augmented reality applications to insert or modify models to a real video stream. The proposed method is based on a linear parameter estimation approach for the auto-calibration step and it enhances the stability and reduces the execution time. Several experimental results are presented on real-world video streams, demonstrating the usefulness of our method for the augmented reality applications.

  • Crane systems are very important in industrial fields to carry heavy objects such that many investigations about control of the systems are actively conducted for enhancing its control performance. This paper presents an adaptive control approach using the model matching for a complex 3-DOF nonlinear crane system. First, the system model is linearized through feedback linearization method and then PD control is applied in the approximated model. This linear model is considered as nominal to derive corrective control law for a perturbed crane model using Lyapunov theory. This corrective control is primitively aimed to compensate real-time control deviation due to partially known perturbation. We additionally study stability analysis of the crane control system using Lyapunov perturbation theory. Evaluation of our control approach is numerically carried out through computer simulation and its superiority is demonstrated comparing with the classical control.

  • An active steering bogie has been theoretically proved to improve both stability and steering performance remarkably. However, It has not been commercialized yet even though many researchers have been trying to develop it because some technical difficulties still exist such as information acquisition fer active control, increasing mechanical components, high energy consumption, fail-safe problem and so on. To solve those problems, an advanced active steering mechanism is proposed in this paper. With this mechanism, required control force is small enough to use direct drives. Therefore, the number of additional mechanical components can be minimized since mechanical transducers like gears are not necessary. Fail-safe function can be also inserted easily. In this paper, concept design of the proposed active steering bogie is introduced and the possibility is verified through computer simulation using linear dynamic model.

  • With increasing demand for LNG as energy resources and need for $CO_2$ sequestration as greenhouse gas, more storage facilities are required in Korea. Due to the recent acute safety concerns and land shortage, storage facilities tend to be located underground. In design and construction of underground storage for low and high temperature materials, besides their mechanical characteristics, the thermal characteristics of rock under temperature variation should be understood. In this study, laboratory experiments for the measurement of the thermal expansion coefficient of rock were performed using strain gauge in consideration of the particle size of mineral and experiment temperature range. Experiment results show that thermal expansion coefficient decreased as the temperature decreases. In addition, linear thermal expansion coefficient was developed for typical Korean rocks such as granite. The results of this study can be utilized for the evaluation of thermal propagation in rock mass and the thermo-mechanical stability of underground facilities.

Dispersion polymerization of styrene and Methylmethacrylate using macromonomers as a reactive stabilizer

  • The novel linear- (V-LUM) and cross-type macromonomers (C-VUM) of vinyl-terminated bifunctional polyurethane were synthesized and applied to the dispersion polymerization of styrene and MMA in ethanol. The existence of the vinyl terminal groups and the grafted macromonomer with styrene and PMMA was verified using 1H NMR and 13C NMR. Monodisperse polystyrene (PS) microspheres were successfully obtained above 15 wt % of macromonomer relative to styrene. The macromonomer can efficiently stabilize higher surface area of the particles compared to a conventional stabilizer, PVP. The grafting ratio of the PS calculated from 1H NMR linearly increased up to 0.048 with 20 wt % of the macromonomer and the high molecular weights (501,300 g/mol) of PS with increased glass transition and enhanced thermal stability were obtained. Furthermore, the stable and monodisperse PMMA microspheres having a weight-average diameter of $5.09{\mu}m$ and a good uniformity of 1.01 were obtained with 20 wt% L-VUM. The molecular weight increased, but the size of the PMMA particles decreased with the macromonomer concentration due to the increased stabilizing effect. The molecular weight of the PMMA was approximately two fold higher than that by a conventional PVP. The L-VUM acts as a reactive stabilizer, which gives polyurethane-grafted PS or PMMA with a high molecular weight. In addition, the XPS result showed that the C-PS (PS using the C-VUM) was anchored with a larger amount of PEG than that of the L-PS (PS using the L-VUM) on the particle surface. Thus, the reaction and stabilizing mechanism of the macromonomers for the formation of PS particles is proposed.
