Comparative study of linear and cyclic forms of apoptosis-targeting peptide

  • Apoptosis, a genetically determined process of programmed cell death, is considered a vital component of various processes including normal cell turnover, animal development, and tissue homeostasis. It has a crucial role in many medical disorders and hence the development of non-invasive imaging tool is highly demanded. Recently, we have developed a peptide-based radioactive probe (ApoPep-1) for apoptosis detection. In that work the potential of probe for apoptosis detection was verified, however in vivo stability of radiolabeled peptide was not enough to monitor apoptosis for extended period. In current study, we prepared cyclic ApoPep-1 peptides to improve the stability of origianl linear ApoPep-1 and carried out direct comparison studies in vitro and in vivo. A targeting efficacy of newly synthesized cyclic ApoPep-1 peptide for apoptosis was confirmed in acute myocardial infarct model.

피부 조직의 라만 스펙트럼에서 NMF 알고리즘을 통한 기저 세포암 진단 방법 (A Diagnosis Method of Basal Cell Carcinoma by Raman Spectra of Skin Tissue using NMF Algorithm)

  • 기저 세포암은 가장 일반적인 피부암이고 그 발병이 급속도로 증가하고 있다. 본 연구에서는 피부 조직에서 측정한 라만 스펙트럼에서 기저 세포암 진단을 위해 NMF(non-negative matrix factorization) 알고리즘을 사용하는 방법을 제안하였다. 측정된 라만 스펙트럼은 영역 선택과 정규화 등의 몇 가지 전처리 과정을 거쳐 분류 실험에 사용한다. 전처리 과정을 수행한 라만 스펙트럼은 NMF 알고리즘을 이용하여 분해된 행렬의 열벡터를 기저로 사용한다. 이 기저들을 선형 결합하여 각 클래스의 평균 스펙트럼에 근사하기 위한 가중치는 행렬 연산으로 결정한다. 분류 실험은 스펙트럼과 NMF에 의한 기저와 가중치의 선형 결합 스펙트럼의 차에 대한 제곱평균제곱근을 최소로 하는 클래스를 선택하는 것으로 수행한다. 기저 세포암의 진단을 위한 분류 실험에서 제안한 방법을 사용하는 경우가 약 99.1%의 평균 분류율로 이전의 BCC 진단에 사용한 방법보다 약 2-3% 정도의 향상된 성능을 보였다.

White-Cell 구조를 응용한 비분산 적외선 이산화탄소 센서의 온도특성 (Temperature Dependency of Non-dispersive Infrared Carbon Dioxide Gas Sensor by Using White-Cell Structure)

  • NDIR $CO_2$ gas sensor was prototyped with ASIC implemented thermopile sensor which included temperature sensor and White-Cell structure in this paper. The temperature dependency of dual infrared sensors ($CO_2$ and reference IR sensors) has been characterized and their output voltage ratios according to the temperature and gas concentration were presented in this paper for achieving temperature compensation algorithm. The initial output voltages of NDIR $CO_2$ gas and reference IR sensors showed $3^{rd}$ order polynomial and linear output voltages according to the variation of ambient temperatures from 253 K to 333 K, respectively. The output voltages of temperature sensor presented a linear dependency according to the ambient temperature and could be described with V(T) = -3.0069+0.0145T(V). The characteristics of output voltage ratios could be modeled with five parameters which are dependent upon the ambient temperatures and gas concentration. The estimated $CO_2$ concentrations showed relatively high error below 300 ppm (maximum 572 % at 7 ppm $CO_2$ concentration), however, as the concentration increased from 500 ppm to 2,000 ppm, the overall estimated errors of $CO_2$ concentrations were less than ${\pm}10%$ in this research.

방사선에 전신 조사된 마우스 음와 세포의 아포토시스 유도를 이용한 생물학적 선량 측정 모델 개발 연구 (Mouse model system based on apoptosis induction to crypt cells after exposure to ionizing radiation)

  • 방사선 피폭선량의 예측을 위한 방사선 민감 지표 모델 개발의 일환으로 apoptotic fragment assay법이 방사선에 피폭된 후 체내 피폭선량을 예측할 수 있는 지표로의 이용 가능성을 평가하기 위하여 코발트-60 감마선과 의료용 싸이크로트론 50MeV($p{\rightarrow}Be^+$) fast neutron 을 0.25Gy에서 1Gy의 선량을 마우스에 각각 전신 조사한 후 소장 음와세포내 apoptotic crypt cell의 수적 변화를 관찰하였다. 저선량 조사군에서 apoptotic crypt cell의 출현 빈도가 1Gy까지 급격하게 증가한 것으로 보아 방사선이 stem cell 지역에 있는 crypt cell의 형태학적 변화를 유발하는 것으로 나타났다. 이상의 결과는 아포토시스가 손상된 세포를 제거하므로 손상된 방사선 민감 표적 장기의 항상성 유지에 중요한 역할을 하는 것으로 판단되었다. Apoptotic fragments의 발생빈도에 대한 선량-반응 곡선에 있어서 음와세포는 중성자조사군이 $y=0.18+(9.728{\pm}0.887)D+(-4.727{\pm}1.033)D^2$ ($r^2=0.984$)으로, 반면에 감마선조사군은 $y=0.18+(5.125{\pm}0.601)D+(-2.652{\pm}0.7000)D^2$ ($r^2=0.970$)의 식을 얻었다. 이와 같이 중성자조사군과 감마선조사군은 공히 linear quadratic model 로 관찰되었다. apoptotic fragments 의 발생빈도와 조사 선량간에 유의한 효과가 있는 것으로 확인되었다. 이상의 결과에서 조사선량의 증가에 비례하여 방사선 민감 세포의apoptotic fragments 가 수적으로 증가하였으며, 고준위 방사선과 저준위 방사선은 선량 반응 관계식과 시간 경과에 따른 영향이 매우 유사하였으며, 마우스 음와세포의 apoptosis 유도에 대한 중성자선의 방사선 생물학적 효과비(RBE)는 2.072이였다. 그리고 모든 방사선조사군에서 방사선피폭 후 4시간과 6시간에 apoptosis 유도가 가장 많았으며, 음와세포의 형태학적 소견은 정상 대조군에서 관찰되지 않는 전형적인 apoptotic fragments 가 나타났다. 따라서 음와 세포에서의 아포토시스 유도는 방사선 피폭으로 발생된 세포 손상의 생물학적 영향 평가검색, 방사선 방호제의 민감도 검사, 방사성 동위원소의 체내 오염에 대한 체내 피폭선량 예측의 지표 및 방사선 민감 표적장기의 손상정도 파악에 이용 가능할 것임. Apoptotic fragment assay 법은 0.25Gy에서 1Gy 까지의 선량에서 간편하고 빠르며 재현성이 있는 지표로서 방사선 민감 표적 장기의 선량 반응 평가와 방사선 피폭후 조기 피폭선량 예측을 위한 방사선 생물학적 선량측정법의 좋은 지표로 사용할 수 있을 것으로 사료됨.

세포 노화에 있어서 복제 세네센스 현상과 산화적 스트레스의 영향 (Replicative Senescence in Cellular Aging and Oxidative Stress)

  • Explanted mammalian cells perform a limited number of cell division in vitro and than are arrested in a state known as replicative senescence. Such cells are irreversibly blocked, mostly in the G1 phase of cell cycle, and are no longer sensitive to growth factor stimulation. Thus replicative senescence is defined as a permanent and irreversible loss of replicative potential of cells. For this characteristic, replicative senescence seems to evolve to protect mammalian organism from cancer. However, senescence also contributes to aging. It seems to decrease with age of the cell donor and, as a form of cell senescence, is thought to underlie the aging process. Extensive evidence supports the idea that progressive telomere loss contributes to the phenomenon of cell senescence. Telomeres are repetitive structures of the sequence (TTAGGG)n at the ends of linear chromosomes. It has been shown that the average length of telomere repeats in human somatic cells decreases by 30∼200 bp with each cell division. It is generally believed that when telomeres reach a critical length, a signal is activated to initiate the senescent program. This has given rise to the hypothesis that telomeres act as mitotic clocks to regulate lifespan. One proposes that cumulative oxidative stress, mainly reactive oxygen species generated from mitochondria, may mainly cause telomere shortening, accelerating aging. Here, the biological importance and mechanism of replicative senescence were briefly reviewed. Also it was summarized that how oxidative stress affects replicative senescence and telomere shortening.

청소년의 휴대폰 의존에 미치는 영향: 자존감의 조절효과 검정 (The Effects of Cell-Phone Dependency of Adolescents: Moderated Analysis of Self-Esteem)

  • Objectives : This study aimed to identify factors of adolescents' cell-phone dependency using Agnew's General Strain Theory (GST) and the perspective of adolescents' micro-systems. A second aim was to verify the moderating effect of self-esteem. Methods : In total, 2,046 students who participated in a study on KCTPS mid. 1 panel 3 Wave by the National Youth Policy Institute were examined in this research. Data were analyzed through descriptive statistics, factor analysis, reliability tests, and hierarchical linear regression using SPSS Ver. 22.0. Results : The strain factors had a positive effect on adolescents' depression, and depression had a positive effect on cell-phone dependency. In addition, self-esteem had a negative moderating effect between depression and cell-phone dependency. Conclusions : Factors of cell-phone dependency were identified. The findings of the study indicate that an intervention is required to lower strain and depression and to enhance self-esteem among adolescents.

Optimized and Portable FPGA-Based Systolic Cell Architecture for Smith-Waterman-Based DNA Sequence Alignment

  • The alignment of DNA sequences is one of the important processes in the field of bioinformatics. The Smith-Waterman algorithm (SWA) performs optimally for aligning sequences but is computationally expensive. Field programmable gate array (FPGA) performs the best on parameters such as cost, speed-up, and ease of re-configurability to implement SWA. The performance of FPGA-based SWA is dependent on efficient cell-basic implementation-unit design. In this paper, we present an optimized systolic cell design while avoiding oversimplification, very large-scale integration (VLSI)-level design, and direct mapping of iterative equations such as previous cell designs. The proposed design makes efficient use of hardware resources and provides portability as the proposed design is not based on gate-level details. Our cell design implementing a linear gap penalty resulted in a performance improvement of 32× over a GPP platform and surpassed the hardware utilization of another implementation by a factor of 4.23.

2 kW급 개방 캐소드형 연료전지 출력 향상을 위한 온습도 제어 (Performance Increase for a 2 kW Open Cathode Type Fuel Cell Using Temperature/Humidity Control)

  • Temperature and humidity regulations of an open-cathode PEM fuel cell with balance of plant (BOP) are developed in this study. The axial fan, a bubble humidifier, set of solenoid valves and a controller are used to perform temperature and humidity control simultaneously. A fuzzy controller is designed, and it shows its superiority in real-time controlling for strong non-linear dynamical fuel cell system. The axial fan speed is used for temperature control and solenoid valve on/off signal of the bubble humidifier is used for humidity control. The axial fan speed is controlled to keep the fuel cell temperature within the desired point. Meanwhile, the bubble humidifier is utilized to moisture hydrogen to manage the water content of membrane. The results show that the proposed fuzzy controller effectively increases the output power of 10% for a PEM fuel cell.

AgO-Zn 전지의 충전상태에 따른 임피던스 특성 (Impedance characteristics of AgO-Zn cell at various State of Charge)

  • In the present study, impedance characteristics for AgO-Zn cell at various State-of-Charge (SoC) has been studied. The impedance measurements of AgO-Zn cell at various SoC were made over the frequency range from 100kHz to 10mHz with an amplitude 5mV. The impedance parameters have been evaluated by the analysis of the data using an equivalent circuit and a Non-linear least squares (NLLS) fitting method. The total resistances reflects the SoC of the cell. This indicates that the total resistance is important parameter for predicting SoC of AgO-Zn cell.

Chemical Inhibition of Cell Recovery after Irradiation with Sparsely and Densely Ionizing Radiation

  • The dependence of cell survival on exposure dose and the duration of the liquid-holding recovery (LHR) was obtained for diploid yeast cells irradiated with ionizing radiation of different linear energy transfer (LET) and recovering from radiation damage without and with various concentrations of cisplatin - the most widely used anticancer drug. The ability of yeast cells to recover from radiation damage was less effective after cell exposure to high-LET radiation, when cells were irradiated without drug. The increase in cisplatin concentration resulted in the disappearance of this difference whereas the fraction of irreversible damage was permanently enlarged independently of radiation quality. The probability of cell recovery was shown to be constant for various conditions of irradiation and recovery. A new mechanism of cisplatin action was suggested according with which the inhibition of cell recovery after exposure to ionizing radiations was completely explained by the production of irreversible damage.
