Structural Design for Performance Improvement of Line Center (라인센터의 성능향상을 위한 구조설계)

  • Recently, the field of the engineering has been studied about optimum design continuously. Verified data by comparison with simulation and dynamic characteristic analysis enables the design of a machine tool to be modified easily and effectively concerning to the mode shape of the vibration. Especially, BC-500 Line Center has got some problems causing vibration which mainly come from Column and ATC part. So it is necessary to solve those problems by the two kinds of method such as changing structural design and reinforcing with ribs. In this paper, column and ATC part of BC-500 Line center are modified by an optimum design by the analysing method of FEM to avoid vibration. As a result, a more stable machine tool was designed by this simulation as optimum condition.

  • The curved line in construction of railroad is inevitable. Also it needs a transition curve when the straight line connects to curved line. In this research, by establishing the reciprocal relations of inner, center and outer track, we will present the calculation method of construction-center line in normal double-track railroad which uses cubic parabolic type as a transition curve.

  • A fullness conjecture of Kuznetsov says that if a smooth projective variety X admits a full exceptional collection of line bundles of length l, then any exceptional collection of line bundles of length l is full. In this paper, we show that this conjecture holds for X as the blow-up of ${\mathbb{P}}^3$ at a point, a line, or a twisted cubic curve, i.e., any exceptional collection of line bundles of length 6 on X is full. Moreover, we obtain an explicit classification of full exceptional collections of line bundles on such X.

  • Cell line development is the most critical and also the most time-consuming step in the production of recombinant therapeutic proteins. In this regard, a variety of vector and cell engineering strategies have been developed for generating high-producing mammalian cells; however, the cell line engineering approach seems to show various results on different recombinant protein producer cells. In order to improve the secretory capacity of a recombinant tissue plasminogen activator (t-PA)-producing Chinese hamster ovary (CHO) cell line, we developed cell line engineering approaches based on the ceramide transfer protein (CERT) and X-box binding protein 1 (XBP1) genes. For this purpose, CERT S132A, a mutant form of CERT that is resistant to phosphorylation, and XBP1s were overexpressed in a recombinant t-PA-producing CHO cell line. Overexpression of CERT S132A increased the specific productivity of t-PA-producing CHO cells up to 35%. In contrast, the heterologous expression of XBP1s did not affect the t-PA expression rate. Our results suggest that CERT-S132A-based secretion engineering could be an effective strategy for enhancing recombinant t-PA production in CHO cells.

Gene Silencing Induced by Cytosine Methylation in Transgenic Tomato (형질전환 토마토에서 Cytosine Methylation에 의한 유전자발현 억제)

  • Transgene expression was analyzed in tomato plants. Four lines of neomycin phosphotransferase II gene (NPTII) and the trehalose biosynthetic fusion gene (TPSP) transformed $T_0$ plants showed kanamycin resistance on selection medium. However, the analysis of phenotype (kanamycin resistance) and mRNA expression in $T_1$ plants indicated that the expression of the NPTII and TPSP transgenes was down-regulated to an undetectable level in two independent lines 1 and 11. Southern analysis demonstrated that the lines 1 and 11 had multicopies of the transgenes, whereas the typical transgenic lines 2 and 10 had 1 or 2 copies. DNA methylation analysis using methylation sensitive enzyme detected accumulated CpG DNA methylation on TPSP coding region and CaMV35S promoter region in the line 11, but not the typical transgenic line 2. These results suggest that multicopy transgene in plants is attributed to down-regulation of the transgene expression via transcriptional gene silencing.

A Study on The Design of Planer Comb-Line Bandpass Filter Using Equivalent Circuits of Asymmetrical Coupled Line (비대칭 결합선로 등가회로를 사용한 Comb-line 구조의 대역통과 여파기 설계)

  • In this paper, we introduce a procedure to obtain a equivalent circuit of comb-line band pass filter. By employing equivalent circuits of each asymmetrical coupled line. we composed the full equivalent circuit of comb-line bandpass filter and derived simple design equations for extracting each line's impedance. To show the validity of design equations, we simulated and fabricated a planar type comb-line bandpass filter, which has center frequency 1.8㎓, band-width 50㎒ and four resonators. The resulting filter is very compact, have broad stop band with the second pass band centered at four times the center frequency of the first pass band. The experimental results show exact performances of design specification.

  • Product-Line 기반의 소프트웨어 개발은 특정 도메인의 소프트웨어 제품군 내의 공통성과 가변성 분석을 통한 공통 아키텍처의 추출과 재사용으로 개발의 생산성과 효율성을 향상시킨다. 일반적인 Product-Line 기반 방법론에 따르면, 재사용되는 핵심자산은 아키텍처, 컴포넌트, 등의 소프트웨어 내부 비즈니스 로직에 한정되어 있으며, 실제로 소프트웨어 개발에 있어 많은 비용이 지출되는 UI(User Interface), 데이터베이스 등의 설계 및 개발에 대한 내용은 언급되어 있지 않은 실정이다. 본 논문에서는 소프트웨어 개발에 필요한 UI 모듈을 핵심자산의 형태로 Product-Line 에서 사용할 수 있도록 하는 방법을 제안한다. UI 모듈을 설계하여 명세하는 방법과 설계된 UI를 디자인 템플릿과 연결하여 내부 로직과 연계하는 방법에 대해 설명한다. 이 방법을 이용하면 Product-Line을 위한 핵심자산 구성시 UI 모듈을 포함할 수 있어 생산성과 효율성을 향상시킬 수 있을 것으로 생각된다.

Study on the Compositions of Photosensitive Ag Paste for Patterning Embedded Fine-Line Inductor in LTCC (LTCC 내장형 미세 라인 인덕터 구현을 위한 감광성 Ag Paste 조성에 관한 연구)

  • Line width under $100\;{\mu}m$ with good resolution is difficult to achieve using conventional thick-film process utilizing screen printing method. However combined with lithography technology finer line and space for miniaturization and highly integrated package is achievable. In this study, photosensitive Ag paste of optimum formulation used for thick film lithography technology was fabricated by various Ag powder, glass powder and additives. As the result, line width of $30\;{\mu}m$ with good definition and reduced mismatch during co-firing with LTCC substrate was acquired. Formulated Ag paste was used to pattern embedded fine line inductor with over 90% yield.

  • Establishment of immortalized human dermal papilla cells (DPCs) retaining the characteristics of DPCs would be a great help for hair researchers. We recently established a simian virus 40T (SV40T)-transformed human DP cell line (SV40TDPC). However, the cell line senesced around passage 25 and ceased proliferation. In this study, we introduced the human telomerase reverse transcriptase (hTERT) gene into SV40T-DPC and established an immortalized human DP cell line. The cell line, SV40T-hTERT-DPC, did not induce tumors when inoculated into nude mice. SV40T-hTERT-DPC maintained morphology of early passage DPCs, expressed markers of DPCs, and retained responses to Wnt/${\beta}$-catenin and bone morphogenic protein (BMP) signaling pathways known to be required for hair-inducing activity of DPCs. The data strongly suggest that SV40T-hTERT-DPC retains many characteristics of human DPCs in vivo without malignant transformation.

Study on longitudinal variation of subcooling with high elevated liquid line in a modular heat pump system (모듈형 동시냉난방 열펌프의 장배관/고낙차에 따른 액선 과냉도 변화에 대한 연구)

  • This study is simulation of high elevated liquid line of a modular heat pump system to observe longitudinal subcooling variation. In a high elevated tube, subcooled refrigerant(R410A) through a condenser changes its states by heat transfer with surrounding air and by pressure drop from elevation. In this study, the liquid line was simulated through correlations of heat transfer and pressure drop for the variation from single-phase into two-phase flow. Pressure drop, heat transfer rate and vapor quality were calculated as key parameters. Two-phase turning heights and variations of the key parameters were confirmed from the simulation. As a result, high elevation of liquid line has great influence on upward flow, which requires additional equipment to compensate the variation.
