Studies on the Immunomodulating Effects of Polysaccharide Extracted from Ganoderma lucidum on Macrophage (영지버섯 다당체의 마우스 대식세포 면역증강 효과)

  • The immunomodulating effects of polysaccharide extracted from Ganoderma lucidum(PSG) on macrophage were evaluated using murine macrophage cell line ATCC TIB 71 cells or peritoneal exudate cells of BALB/c mice. The cell were incubated with various content of PSG for 24 hours at 0.5% $CO_2$ incubator under varying experimental conditions. PSG stimulated the non-specific activites of macrophage such as mitotic activity and expression of surface interleukin-2 receptors by dose-dependent pattern with statistic significance(p<0.001): however, PSG had little immunoregulatory effects on cytokines derived from peritoneal macrophages of BALB/c mice. There were no significant changes in the se-cretion of interleukin-6, interleukin-6, or tumor necrosis factors(Tn) of PSG treated cells compared to the control group. But PSG increased secretion of cytokines(IL-1 and TNF) when the cells were primed and trigged with BCG and IFN-${\gamma}$.

  • Orthodontic force is a mechanical stress controlling both of tooth movement and skeletal growth. The mechanical stress stimulate bone cells that may exert some influence on bone remodeling. The purpose of this study was to evaluate the difference in cellular activity depending on mechanical stresses such as compressive and tensile force by determining the alkaline phosphatase(ALP) activity. A clonal osteogenic cell line MC3T3-E1 was seeded into a 24-well plate($2{\times}10^4/well$). At the confluent phase, a continuous compressive hydrostatic pressure($25g/cm^2$, $300g/cm^2$) and continuous tensile hydrostatic pressure($-25g/cm^2$, $-300g/cm^2$) were applied for 4, 6, 10, 14, 18, 20 days respectively by a diaphgragm pump. At the end of the stimulation period, cell layers were prepared for ALP activity assay. The ALP activity of the compressive group increased more than that of the tensile group at same force magnitude, whereas the cells responded to a similar pattern regardless of the type of mechanical stress The ALP activity of the compressive and tensile group turned into the level of the control group as the length of time increased. These results indicated that a mechanical stress may be more effective on cellular activity during active cellular proliferation and differentiation periods. The time to achieve maximum ALP activity was delayed as the mechanical stress increased in both the compressive and the tensile group. Accordingly, the magnitude of the stress rather than the type of mechanical stress may have more influence on cellular activity.

Wave and surface current measurement with HF radar in the central east coast of Korea (동해중부에서 HF Radar를 이용한 파랑 및 해수유동 관측)

  • We installed HF Radar of Array type in Site A and Site B, observing the real-time wave and current in the central East coast of Korea. WERA(WavE RAdar) in this research uses HF Radar of Array Type with frequency range of 24.525 MHz, developed by Helzel, Germany. Each site is a 8-Channel system consisting of four transmitters and eight receivers, generating wave and current data, being observed every thirty minutes at the present time. HF Radar has grid resolution of an interval of 1.5 km using bandwidth of 150 kHz; The wave data covers an observation range of about 25 km, and the current data covers the maximum observation range of about 50 km. The Wave data observed by HF Radar was compared and verified with the AWAC data observed in the research sites. MIT also compared the Current data observed by HF Radar with Monthly the East sea average surface current and current flow pattern provided by KOHA(Korea Hydrographic and oceanographic Administration). The regression line and deviation of the comparison data of Wave was calculated by Principal Component Analysis, which showed correlation coefficient 0.86 and RMSD 0.186. Besides, data analysis of long-term changes of the current in the East coast showed that, during August and September, the North Korean Cold Current flow into the southward direction and the East Korean Warm Current flow into the northward direction in the coast.

Wavelength-resolved Thermoluminescence of Chemical-vapor-deposited Diamond Thin Film (화학증착된 다이어몬드 박막의 파장 분해된 열자극발광)

  • Diamond thin films were synthesized by a chemical vapor deposition (CVD). Raman spectrum showed the diamond line at 1332 $cm^{-1}$ / and x-ray diffraction pattern exhibited a strong (111) peak of diamond. The scanning electron microscopy analysis showed that the CVD diamond thin film was grown to be unepitaxial crystallites with pyramidal hillocks. A wavelength-resolved thermoluminescence (TL) of the CVD diamond thin film irradiated with X-rays showed one peak at 430 nm around 560 K. The glow curve of the CVD diamond thin film produced one dominant 560-K peak that was caused by first-order kinetics. Its activation energy and the escape frequency were calculated to be 0.92 ~ 1.05 eV and 1.34 $\times$ 10$^{7}$ sec$^{-1}$ , respectively. The emission spectrum at 560 K was split into 1.63-eV, 2.60-eV, and 3.07-eV emission bands which is known to be attribute to silicon-vacancy center, A center, and H3 center, respectively.

  • Ring in the scroll painting is one of the mounting elements which are fixed string and tassel to hang, these differ widely in shape and produce a variety of metals. Most traditional shapes of ring have been lost, because of remount, there are used Japanese style rings in present.Therefore, this study examines to shape, production technique and analyze component traditional style rings of 19 traditional scroll paintings in National Museum of Korea for restoration of original style. Ring has been recorded official names; Wonhwan(Ring), Gukhwadong(Chrysanthemum shaped ornaments), Baemok(Ring-shaped nail) in Uigwe. Result of an optical microscope (Leica, M205A), Wonhwan has two type of production technique; one is cutting and bending a metal rod and other is cast. Baemok is made to forging process after metal rod or plate by casting alloy. Baemok decoration is metal plate cutting shape, and then decorates it with pattern by using kicking line engraving, chasing and so on. Component analysis result from portable X-ray fluorescence found various metals, such as, brass, iron silver-cooper. Brass based on copper and zinc used rings of 17 scroll painting. Baemok of Yun Sidal portrait is used iron and plated with a tin-lead alloy. Yi Seogu portrait is silver-cooper alloy in whole ring.

Study of the Behavior of Concrete Slab Track on Earthwork According to the Variation of Train Axle Load and Speed (열차하중, 속도변화에 따른 토공상 콘크리트 슬래브궤도의 거동특성연구)

  • In recent high speed rail way, the operating speed of train is enhanced and the introduction of EMU train vehicles is increased. In addition, as expected the demand of the concrete slab track and the trend of design cross-section reduction, the clear behavior of evaluation of internal slab layer is demanded about the variation of design load and speed. The purpose of this study is to evaluate and identify the mechanical behavior pattern of concrete slab track and track-road bed with the variation of axle load and train speed. To this end, the behavior of TCL and HSB was evaluated in according to the variation of axle load and speed. And the analysis results and the data measured TCL strain sensor, which was embedded in TCL slab under installation on Honam high speed railway, was analyzed. The analysis result shows that the strain are increasing in according to the speed-up of train, and line regression was obtained from measured data. Analysis data of the state of bonding condition of slab layer and measured data was analyzed. It is conducted that the TCL layer stress of HEMU 430X, which of axle load, is lighter was similar to the stress of KTX-Honam, the standard deviation of measured stress is dramatically increased.

Design of swelling gastroretentive drug delivery system for sustained release of metformin (메트포르민의 서방출을 위한 팽윤성 위체류 약물전달시스템 설계)

  • Metformin is a recommended first-line therapy drug for type 2 diabetes patients. However, compared to other oral antidiabetic drugs (OAD), metformin has a large unit dosage, with bioavailability of 40-60%. This limiting bioavailability is because metformin is absorbed only in the upper gastrointestinal tract as a BCS class 3 drug. Hence, we propose that applying the Gastroretentive Drug Delivery System (GRDDS) and extending drug release time in the stomach will result in improved bioavailability. We selected the swelling type delivery system, as it is considered the most stable gastroretention technology compared to other GRDDSs. We modified the swelling excipient by using a natural swelling excipient to form a swelling tablet made of carrageenan and hydroxypropyl methylcellulose (HPMC). Our results indicate that the swelling complex tablet made of carrageenan and HPMC has a good swelling ability and shows required sustained release in a dissolution pattern. In addition, the carrageenan complex has a better swelling ability than the marketed metformin tablet, as determined by the ratio, (swelling ability)/(excipient weight). Taken together, our results indicate that the carrageenan complex can be developed as a good swelling excipient. Further optimizations are required for the commercialization of the carrageenan complex.

Lipid Contents and Fatty Acid Composition of Three Lipid Classes in Korean Pinenut (잣지질 성분의 분획정량 및 각 획분의 지방산 조성)

  • Total lipids (TL) from Korean pinenut (Pinuskoraiensis S & Z) were extracted, purified and fractionated into three lipid classes (neutral lipid: NL, glycolipid; GL, phospholipid; PL). Lipid contents(constituent components) and fatty acid composition of three lipid classes were determined by thin layer chromatography and gas chromatography. TL ranged from 69.0% to 69.8% in fresh pinenut and consisted of 95.9% to 96.7% NL, 3.2% to2.5% GL and 0.9% to 0.8% PL. In the NL, triglycerides were predominant (80.8%) with the smaller amounts of sterol, diglycerides, free fatty acids, sterol esters and hydrocarbons. Monogalactosyl diglycerides and esterified steryl glycosides (23.5%) were the major components of GL, but cerebrosides, steryl glycosides and digalactosyl diglycerides were also found as minor components. Of the PL, phosphatidyl choline (40.2%) and phosphatidyl ethanolamine (19.4%) were the major components, comprising over 60% of this class. Phosphatidyl inositol, lysophosphatidyl choline were also present in the PL. The major fatty acids in the NL were linoleic acid (48.6%), oleic acid (28.8%) and arachidic acid(14.4%), The fatty acid composition in the GL was similar to the pattern in the NL, but PL contained a higher percentage of palmitic acid (17.7%) and stearic acid (6.0%) than other lipid classes.

  • This study can be expressed by gathered and formed into the exhibition space's structure practical progress/ application. For the one model of fascinating exhibition's space thru inter-space's pertinent adjustment, production between spactator and exhibition constituent on an exhibition story-line, I adjusted the focus to follow mentioned methods systematically for the example; An exhibition space is completed to be very impressive and attractive space by proper adjustment and production of M-M/C interface in exhibition storyline. Quantity space is transfered into quality space through the transmission of an exhibition which can be define as the point of an exhibition structure. And also could de transferd into a bodily sensation space that inherent full of interactive constituent. Changeable exhibition constituents (exhibition item and text) that sporadically expatiated (not trimed, the original form) in the process of an exhibition structure draw much higer quality of the optimal solution in optimize process which is given when aggregate again to contextual flow of synthetic exhibition scenario. Reconstruction of individual exhibition constituents to the new story, that is, transference of exhibition text to exhibition context is inspirit to an exhibition by maximize the exhibition effect in connection can be systematized through carrying out an outcentripetalpart. However, since pattern of an exhibition structure that consequtively meet variety spacetime of an exhibittion environmental can't be exist, this study presents centralizing the exhibition plan of Korea communication pavilion of the annual Expo Comm Wireress Korea, sustained process from design proposal, research and analysis to synthesis, development, transmission and management to an example of an applying crystallization.

Effects of Verapamil and Tetracaine on Acetylcholine-and Oxytocin-induced Uterine Contraction Pattern (Acetylcholine및 Oxytocin에 의하여 야기되는 렛드 자궁수축에 미치는 Verapamil 및 Tetracaine의 영향)

  • The effects of verapamil and tetracaine on acetylcholine-and oxytocin-induced contraction of uterus from estrogen-treated rat were examined. Isometric tensions were recorded on the Physiograph and stored in TriGem 20XT computer as digitized data for off-line analysis of the components, which described the contraction patterns: trought tension (T), peak tension (P), contraction frequency (F), and duration (D). In the acetylcholine-induced contraction, verapamil $(0.25\;{\mu}M)$ significantly decreased P and D. In contrast, tetracaine $(42{\mu}M)$ decreased F, but increased D. In the low oxytocin-induced contraction, verapamil $(0.25\;{\mu}M)$ decreased P and D, and tetracaine $(42{\mu}M)$ decreased F but increased D. In the high oxytocin-induced contraction, verapamil decreased P and D, but tetracaine decreased P without affecting on other components. These results suggest that the analysis of effects of a certain inhibitor on the components of contraction allow to postulate its specific inhibitory mechanism of the smooth muscle contraction.
