A Morphometric Analysis of the Foramen Ovale and the Zygomatic Points Determined by a Computed Tomography in Patients with Idiopathic Trigeminal Neuralgia

  • Objective : The purpose of this study is to measure the dimensions of foramen ovale and to localize the zygomatic point using computed tomography[CT] in Korean adults with idiopathic trigeminal neuralgia. Methods : Facial axial CT scans using the orbitomeatal plane were performed in 67patients [39males and 28females; mean age 58.8years] with idiopathic classic trigeminal neuralgia. We measured the size of the foramen ovale and localized the zygomatic point which was a skin marker over the ipsilateral zygoma that approximates the lateral projection of a straight line joining the centers of the two foramen ovale. Results : The axial dimensions of the foramen ovale on the orbitomeatal plane were of average length : $8.18{\pm}0.82mm$ [range $6.9{\sim}11.5mm$]. width : $4.06{\pm}0.86mm$ [$2.5{\sim}5.7mm$]. The average distance between the external acoustic meatus and the zygomatic point was $21.64{\pm}1.99mm$ [$16.3{\sim}25.0mm$] and the average distance of anterior margin of condylar process of mandible to zygomatic point was $4.29{\pm}1.19mm$ [$1.0{\sim}7.0mm$]. Conclusion : The anatomical understandings including the size of the foramen ovale and localization of the zygomatic point could be helpful in determining a plan of percutaneous approaches to foramen ovale.

  • This paper proposes the web-based virtual collaboration among experts. The proposed system supports the presentation tool using Synchronous Multimedia Integration Language(SMIL) which is the easy and efficient way of adding multimedia to presentations. The presentation gives essential information to the participants before actual discuss. The participants use texts in discussing over the presented medical image. The spatial elements such as point or line, and some type of marker with their relative participants' comments can be set or removed dynamically to represent areas of interest in digital images. XML files are used for recording experts' opinions as well as the spatial elements that are associated with digital images during the discussion and stored for future reference. The participants can also set and reset a polygon in the image to select the interested area and refer to the stored relating information.

Development and Evaluation of QTL-NILs for Grain Weight from an Interspecific Cross in Rice

  • In a previous study, we mapped 12 QTLs for 1,000 grain weight (TGW) in the 172 $BC_2F_2$ lines derived from a cross between Oryza sativa ssp. Japonica cv. Hwaseongbyeo and O. rufipogon. These QTLs explained 5.4 - 11.4% of the phenotypic variance for TGW. Marker-aided selection combined with backcrosses was employed to develop QTL-NILs for each QTL. $BC_2F_2$ lines with each target QTL were backcrossed to Hwaseongbyeo twice and then allowed to self to produce $BC_4F_5$ populations. SSR markers linked to TGW were employed to select QTL-NILs with the respective target QTL. Six QTL-NILs with the recurrent parent, Hwaseongbyeo were evaluated for nine traits for three years from 2007 and 2009. Differences were observed between each of the 6 QTL-NILs and Hwaseongbyeo in TGW. In addition to TGW, these QTL-NILs displayed differences in other agronomic traits possibly indicating a tight linkage of genes controlling these traits. The direction of the QTL for TGW in 6 QTL-NILs was consistent as in the $BC_2F_2$ lines from the same cross. Difference in TGW between each of the QTL-NILs and Hwaseongbyeo was associated with the difference in one or two grain shape traits; grain length, grain width, and grain thickness. SSR markers linked to the QTL for TGW will facilitate selection of the grain shape character in a breeding program to diversify grain shape and provide the foundation for map-based gene isolation. Also, the QTL-NILs developed in this report and the progenies from crosses between the QTL-NILs will be useful in clarifying epistatic interactions among QTLs for TGW.

Involvement of a Novel Organic Cation Transporter in Paeonol Transport Across the Blood-Brain Barrier

  • Paeonol has neuroprotective function, which could be useful for improving central nervous system disorder. The purpose of this study was to characterize the functional mechanism involved in brain transport of paeonol through blood-brain barrier (BBB). Brain transport of paeonol was characterized by internal carotid artery perfusion (ICAP), carotid artery single injection technique (brain uptake index, BUI) and intravenous (IV) injection technique in vivo. The transport mechanism of paeonol was examined using conditionally immortalized rat brain capillary endothelial cell line (TR-BBB) as an in vitro model of BBB. Brain volume of distribution (VD) of [$^3H$]paeonol in rat brain was about 6-fold higher than that of [$^{14}C$]sucrose, the vascular space marker of BBB. The uptake of [$^3H$]paeonol was concentration-dependent. Brain volume of distribution of paeonol and BUI as in vivo and inhibition of analog as in vitro studies presented significant reduction effect in the presence of unlabeled lipophilic compounds such as paeonol, imperatorin, diphenhydramine, pyrilamine, tramadol and ALC during the uptake of [$^3H$]paeonol. In addition, the uptake significantly decreased and increased at the acidic and alkaline pH in both extracellular and intracellular study, respectively. In the presence of metabolic inhibitor, the uptake reduced significantly but not affected by sodium free or membrane potential disruption. Similarly, paeonol uptake was not affected on OCTN2 or rPMAT siRNA transfection BBB cells. Interestingly. Paeonol is actively transported from the blood to brain across the BBB by a carrier mediated transporter system.

Phylogenetic study of the Genus Suaeda(Chenopodiaceae) based on chloroplast and nuclear DNA sequences from Korea (엽록체 DNA 및 핵 DNA 염기서열에 근거한 한국산 나문재속(명아주과)의 분류학적 연구)

  • The purpose of this study was to identify the phylogenetic relationships of the plants in the Korean genus Suaeda and to find out the molecular markers that could confirm the interspecies relationships in the family tree through molecular phylogenetic studies. We used the nuclear ribosomal DNA ITS and the chloroplast DNA matK, psbA-trnH, and trnL-trnF as the molecular markers. We could not distinguish between S. japonica and S. maritima and between S. maritima and S. australis in the ITS region and could not distinguish between S. japonica and S. australis with the base sequence in the psbA-trnH and trnL-trnF region. However, we analyzed the combinations of four molecular marker regions and confirmed that each of five plant species of the genus Suaeda formed the independent line. Therefore, it is considered that combinations of molecular markers would be useful for the analysis of phylogenetic relationships in the genus Suaeda. Further investigations of the ecological and morphological characteristics would be needed to understand the phylogenetic relationship and lineage diversification in the genus Suaeda.

Comparison of energy and nutrient utilization in the aged laying hens with different eggshell strengths or different intensities of brown eggshell color

  • The current experiment was conducted to compare nutrient utilization in the aged laying hens with different eggshell strengths or different intensities of brown eggshell color. Two metabolic trials with 96-week-old Hy-Line Brown laying hens were conducted. Twelve hens with a strong eggshell strength (SES) and 12 hens with a weak eggshell strength (WES) were selected and used in the first experiment, whereas 12 hens with dark brown eggshells (DBE) and 12 hens with light brown eggshells (LBE) were selected and used in the second experiment. All hens were fed a common commercial layer diet. Diets included chromic oxide as an indigestible marker to determine the apparent total tract retention (ATTR) of the gross energy (GE), dry matter, acid-hydrolyzed ether extract, nitrogen, neutral detergent fiber, acid detergent fiber, ash, and minerals. The results indicate that the ATTR of Ca in the aged laying hens with SES (59.7%) was greater (p < 0.05) than in the aged laying hens with WES (48.6%). However, the ATTR of the GE and other nutrients was not different between the hens with SES or WES. There were no differences in the ATTR of GE and nutrients between the hens with DBE or LBE. In conclusion, aged laying hens with SES have a greater ability to utilize dietary Ca than those with WES, indicating that the extent of dietary Ca utilization determines the eggshell strength of aged laying hens. However, nutrient utilization in diets may have little impact on eggshell brown coloration in aged laying hens.

Qualitative and quantitative determination of oleanolic acid in a scalp tonic products by HPLC using response surface methodology for extraction optimization

  • The simple and effective analytical method for the quality control of a novel scalp tonic formulation has been developed and optimized in terms of HPLC conditions and sample preparation method, meanwhile, the optimization of preparation condition was using response surface methodology (RSM) based on central composite design (CCD). Oleanolic acid was selected as marker compound because of its bioactivities for alopecia therapy. The developed analytical method and extraction condition were successfully qualified. Coefficient of determination ($r^2$) for the calibration was 0.9997 with a line passing through the origin point in the range of 0.1-100 mg/mL. The limit of detection (LOD) and the limit of quantitation (LOQ) were 17.5 ng/mL and 55.0 ng/mL, respectively. The intra-day and inter-day precision of the method were 0.5-1.4 % and 0.7-1.8 % in relative standard deviation, respectively, while those accuracy were 99.5-100.9 % and 100.0-102.2 %, respectively. The repeatability of oleanolic acid in samples ranged of 0.3-1.9 % based on peak area and 0.3-0.7 % for retention time. Recoveries from samples were 95.0-99.4 % with lower than 1.8 % in relative standard deviation. Overall, the developed analytical method will be used for quality control of this commercial scalp tonic products successfully.

Development of Rainfed-adapted, Fertilizer-efficient Temperate Rice Varieties by Pup1 Introgression

  • Water and phosphorus (P) fertilizer are two of the most critical inputs in rice cultivation. Under climate change scenarios and urbanization, irrigation and fertilizer are becoming limiting factors often leading to significant decrease in yield whenever supplied scarcely. It has been shown that the Pup1 QTL confers tolerance to P starvation and improved early-stage root vigor in indica rice grown in the tropics. However, whether the QTL works in japonica rice genetic background grown in temperate regions remains to be elucidated. Here, we have introgressed the Pup1 QTL into three temperate rice varieties MS11, TR22183, and Dasan using marker-assisted backcrossing and next generation sequencing. The selected lines all harbored the full Pup1 QTL with recurrent parent genome recovery rates ranging from 66.5% to 99.8%. Evaluation of the introgression rice lines grown in South Korea under low inputs of P and water revealed early vegetative growth advantage relative to that of the recurrent parents. Under rainfed condition, Pup1 introgression lines had yield advantage ranging from 7.2 to 19.9% and 24 to 26% in P non-supplied soil and P-supplied soil, respectively compared to that of the recurrent parents suggesting that Pup1 confers enhanced yield under low P and water inputs in temperate rice genetic background grown in temperate climate. In terms of early vegetative growth, temperate Pup1 introgression lines showed a similar trend on the extent to which Pup1 promotes yield advantage in temperate rice in comparison with indica control Pup1 introgression line IR64-Pup1.

Identification of a Novel Bakanae Disease Resistance QTL in Zenith Cultivar Rice (Oryza sativa L.)

  • Bakanae disease, caused by several Fusarium species, imposes serious limitations to the productivity of rice across the globe. The incidence of this disease has been shown to increase, particularly in major rice-growing countries. Thus, the use of high resistant rice cultivars offers a comparative advantage, such as being cost effective, and could be preferred to the use of fungicides. In this research, we used a tropical japonica rice variety, Zenith, a bakanae disease resistant line selected as donor parent. A RIL population (F8:9) composed of 180 lines generated from a cross between Ilpum and Zenith was used. In primary mapping, a QTL was detected on the short arm of chromosome 1, covering about 3.5 Mb region flanked by RM1331 and RM3530 markers. The resistance QTL, qBK1Z, explained about 30.93% of the total phenotype variation (PVE, logarith of the odds (LOD) of 13.43). Location of qBK1Z was further narrowed down to 730 kb through fine mapping using additional RM markers, including those previously reported and developed by Sid markers. Furthermore, there is a growing need to improving resistance to bakanae disease and promoting breeding efficiency using MAS from qBK1Z region. The new QTL, qBK1Z, developed by the current study is expected to be used as foundation to promoting breeding efficiency with an enhanced resistance against bakanae disease. Moreover, this study provides useful information for developing resistant rice lines carrying single or multiple major QTLs using gene pyramiding approach and marker-assisted breeding.

Human Lung Cancer Cell Xenografts Implanted under the Capsule of Kidney, Spleen and Liver (폐암 세포주를 사용한 신, 비장 및 간 피막하 분식법의 비교)

  • Bakground : Complete resection by the surgery has been selected as the treatment of choice in lung cancer patients, but in cases of recurrence after excision or inoperable cases, the importance of anticancer chemotherapy has been emphasized. If one can select a set of the sensitive chemotherapeutic agents before anticancer chemotherapy, it will give more favourable results. Subrenal capsular assay has been recognized as a useful in-vivo chemosensitivity test of thoracic and abdominal tumors and it can be done in a short time for a rapid interpretation of tumor responsiveness to anticancer chemotherapeutic drugs. It has been reported that various kinds of cancer cells can be implantable to the kidney, but so far there is no comparative study of xenogeneic cell implantation on liver, spleen and kidney. The author implanted the human lung cancer cells under the capsule of S.D rat's liver, spleen and kidney respectively and compared the pattern of growth and histology. Material and Method: After incubation of human lung cancer cell line (SW-900 G IV) in RPMI 1640 (Leibovitz L-15 medium) culture media, 3${\times}$3${\times}$3 mm size fibrin clots which contain 108 cancer cells were made. Thereafter the fibrin clots were implanted at subcapsule area of liver, spleen and kidney of S.D. female rat. For immune suppression, cyclosporin-A (80 mg/Kg) was injected subcutaneously daily from post-implantation first day to sixth day. The body weight was measured at pre and post implantation periods. The growth pattern and the size of tumor mass were observed and the pathologic examination and serum tumor marker tests were performed. Result: Body weight increased in both of control and experimental groups. Serum Cyfra 21-1 was not detected. Serum levels of CEA and NSE revealed no significant change. The SCC-Ag increased significantly in implanted group. The growth rate of human lung cancer cells which was implanted on spleen was higher than on liver or kidney. The surface area, thickness, and volume of tumor mass were predominant at spleen. The success rates of implantation were 80% on kidney, 76.7% on spleen and 43.3% on liver. Pathologic examination of implanted tumors showed characteristic findings according to different organs. Tumors that were implanted on kidney grew in a round shape, small and regular pattern. In the spleen, tumors grew well and microscopic neovascularization and tumor thrombi were also found, but the growth pattern was irregular representing frequent daughter mass. Human lung cancer cells that were implanted in the liver, invaded to the liver parenchyme, and had low success rate of implantation. Microscopically, coagulation necrosis and myxoid fibrous lesion were observed. Conclusion: The success rate of implantation was highest in the kidney. And the mass revealed regular growth that could be measured easily. The SCC-Ag was presented earlier than CEA or Cyfra21-1. The Cyfra21-1 was not detected at early time after implantation. The best model for tumor implantation experiment for chemosensitivity test was subrenal capsular analysis than liver and spleen and the useful serum tumor marker in early period of implantation was the SCC-Ag.
