Isolation of the Pathogenic Bacteria from Chicken and Antimicrobial Drug Sensitivity of the Strain Isolated (가금유래 주요병원성세균의 분리와 분리균주에 대한 약제감수성조사)

  • A total of 1503 specimens were submitted to the Poultry Disease Diagnostic Service Laboratory during the year 1966 and 1978. The most frequently diagnosed diseases in order of prevalence were avian mycoplasmosis, staphylococcosis, colibacillosis, salmonellosis and pullorum disease, the percentages of the conditions being 24.6%, 20.0%, 18.0%, 12.6% and 6.4%, respectively, The drug resistance of pathogenic mirnoorganisms isolated during the year 1978 from chicken with colicabacillosis, staphylococcosis or salmonellosis were investigated by the use of disc diffusion technique, the results being as follow. 1) Drug resistance of 63 strains of Escherichia coli More than 95% of the strains tested were sensitive to colistin and gentamicin. The percentages of strains sensitive to kanamycin, chloramphenicol, ampicillin and nitrofurantoin were 66.7%, 60.3%, 60.3% and 47.6%, respectively. Majority of the strains were highly resistant to streptomycin and tetracyline. All the strains were resisistant to bacitracin lincomycin, oleandomycin, penicillin and erythromycin. All the strains tested were resistant to more than two among 10 drugs in common use such as penicillin, erythromycin, streptomycin, tetracycline, neomycin, chloramphenicol, kanamycin, ampicillin and gentamicin, and 27 different resistance patterns were noted. The most frequent multiple resistance pattern was PC, EM, SM and TC (11.1%). 2) Drug resistance of 48 strains of Salmonella More than 95% of the strains tested were sensitive to colistin, gentamicin ana ampicillin. The percentages of st rains sensitive to kanamycin, tetracycline, neomycin and nitrofurantoin were 81,3%, 79%, 72.9%, and 68.0% respectively. None of them was sensitive to streptomycin, oleandomycin, erythromycin, lincomycin and bacitracin. All the strains were resistant to more than one among 7 drugs in common use such as streptomycin, erythromycin, neomycin, tetracycline, kanamycin, ampicillin and gentamicin. The most frequent resistance pattern was SM and EM(66.7%). 3) Drug resistance of 54 strains of Staphylococci All the strains tested were sensitive to gentmaicin, kanamycin and cephalothin. Majority of them were highly sensitive to bacitracin, methicillin, nitrofurantoin and chloramphenicol. The Percentages of strains sensitive to streptomycin, ampicillin, lincomycin and tetracycline were 66.7%, 55.6%, 44.4% and 27.8%, respectively. Among them, 51 strains were resistant to more than one among 11 drugs in common use such as tetracycline, lincomycin, ampicillin, penicillin, streptomycin, erythromycin, neomycin, oleandomycin, chloramphenicol, methicillin and bacitracin, and thirty one different resistance patterns were noted.

Reconsideration of Classical Antibiotic Lincomycin: Anti-inflammatory Effect in LPS-stimulated RAW 264.7 Cells

  • Since, side effects of antibiotics are frequently emphasized these days, their use is gradually diminishing, and alternative drugs are being developed. We have sought to reintroduce them as raw materials for human health as conventional 'weapons' that have been retired after their historical duties. In this study, we investigated the anti-inflammatory effects of lincomycin (LIN), on lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. Our findings show that LIN potently inhibited production of LPS-induced proinflammatory mediators, such as nitric oxide (NO) and prostaglandin E2 (PGE2), without cytotoxicity. Consistent with these findings, LIN strongly decreased protein expression levels of inducible NO synthase (iNOS) and cyclooxygenase (COX-2). Furthermore, LIN reduced pro-inflammatory cytokines such as tumor necrosis factor (TNF)-α, interleukin (IL)-6, and IL-1β. To further elucidate the mechanisms of these inhibitory effects of LIN, we studied LPS-induced IκB-α degradation, and mitogen-activated protein kinase (MAPK) phosphorylation. LIN suppressed downregulation of inhibitory κB (IκB-α) degradation, and the phosphorylation of the c-Jun N-terminal kinase (JNK) pathway. Based on these results, we suggest that LIN may be considered a potential candidate as an anti-inflammatory cosmetic or a medicine for human health.

  • This study was performed to delineate pharmacokinetic residue characteristics of lincomycin (LM) in olive flounder at low water temperature (13℃), and to examine whether LM is effective against main Gram-negative pathogens. It was observed that the times for residue declined below the maximal residue limit (0.1 mg/kg) in the muscle was, respectively, 32, 33 and 55 days following 10, 20 and 40 mg/kg intramuscular LM injections. The in vitro MIC value of LM against Edwardsiella piscida was higher than 256 ㎍/mL and against Vibrio harveyi 32 ㎍/mL when examined with several clinical isolates. These results suggest that the withdrawal time of LM should be more than 30 days in consideration of the 10 mg/kg employed as the regular clinical dose. It was also found that LM administered to treat streptococosis in olive flounder is not likely to be effective against some pathogenic Gram-negative bacteria.

  • Previously, we reported on the antibiotic resistance patterns of 50 strains of Staphylococcus aureus which were isolated from a hospital in Busan, Korea from July 2005 to December 2006. We have isolated small plasmids and classified plasmid types by agarose gel electrophoresis. We have selected 5 plasmids and determined complete nucleotide sequences of those plasmids. The aim of this paper is to report on the characteristics of cadmium, erythromycin, lincomycin resistance plasmids and a cryptic plasmid based on the sequence analysis obtained by using the BLAST program.

Antibiotic susceptibility of Porphyromonas gingivalis and Prevotella intennedia from the Patients with Adult Periodontitis (성인성 치주염환자에서 분리한 Porphyromonas gingivalis와 Prevotella intennedia의 항생제 내성에 관한 연구)

  • The purpose of this study was to an in estimate the antibiotic susceptibility of P. gingivalis and P. interrnedia isolate from the subgingival plaque to adult periodontitis. Six P. gingivalis and five P. intermedia bacterial strains were tested for their susceptibility to 10 antimicrobial agents under disc diffusion method and broth dilution method. Ten patients with deep pocket(6mm) were selected for this study. They had not taken antibiotics for 6 months and no history of dental treatment for 6 months before this study. The result were as follow : 1. For antibiotic disc diffusion method, six P. gingivalis and five P. interrnedia were tested with 10 antimicrobial agents which comprised penicillin, gentamycin, clindamycin, lincomycin, ampicillin, erythromycin, tetracycline, amikacin, chloramphenicol, vancomycin. The sensitive antibiotics were tetracycline, penicillin, ampicillin, vancomycin, chloramphenicol and resisitent antibiotics were lincomycin. The other antimicrobial agents were less active. 2. From the study of determination on the minimal inhibitory concentration(MIC) by broth dilution method, the MIC of tetracycline to P. gingivalis and P. intermedia were $0.5-1.0{\mu}g/ml$, $0.5{\mu}g/ml$, that of ampicillin were $1-8{\mu}g/ml$, that of clindamycin were $1-32{\mu]g/ml$, $8-16{\mu}g/ml$, that of lincomycin were $16-32{\mu}g/ml$, $2-32{\mu}g/ml$. These data suggest that tetracycline and ampicillin may be valuable drug in the elemination of P. gingivalis and P. interrnedia from the patients with adult periodontitis.

Differential Recovery of Photosystem II Complex from Low-Temperature Photoinhibition in Plants with Different Chilling Sensitivity

  • To examine the chilling tolerance lipids, we compared the chilling susceptibility of photosystem II of wild type tobacco plants with that of transgenic tobacco plants, in which the sensitivity to chilling had been enhanced by genetic modification of fatty acid unsaturation of chloroplast membrane lipids. The transgenic tobacco plants were found to contain reduced levels of unsaturated membrane fatty acids by being tansformed with cDNA for glycerol-3-phosphate acyltransferase from squash. For the purpose of studying on the functional integrity of photosystem II during low-temperature photoinhibition, the photochemical efficiency was measured as the ration of the maximun fluorescence of chlorophyll (Fv/Fm) of photosystem II. In parallel with an investigation on the transgenic plants, susceptibility of chilling-resistant species, such as spinah and pea, and of chilling-sensitive ones, such as squash and sweet potato, to low-temperature photoinhibition was also compared in terms of room temperature-induced chlorophyll fluorescence from photosystem II. When leaf disks from the two genotypes of tobacco plants were exposed to light at 5$^{\circ}C$, the transgenic plants showed more rapid decline in photochemical activity of photosysytme II than wild-type plants. When they were pretreated with lincomycin, an inhibitor of chloroplast-encoded protein synthesis, the extent of photoinhibition was even more accelerated. More impottantly, they showed a comparable extent of photoinhibition in the presence of lincomycin, making a clear contrast to the discrepancy observed in the discrepancy observed in the absence of lincomycin. Restoration of Fv/Fm during recovery from low-temperature photoinhibition occurred more slowly in the transgenic tobacco plants than the wild-type. These findings are discussed in relation to fatty acid unsaturation of membrane phosphatidylglycerol. It appears that the ability of plants to rapidly regenerate the active photosystem II complex from might explain, in part, why chilling-resistant plants can toleratlow-temperature photoinhibition.

Effects of Chilling Injury in the Light on Chlorophyll Fluorescence and D1 Protein Turnover in Cucumber and Pea Leaves

  • Light-chilling effects were investigated in chilling-sensitive cucumber (Cucumis sativus L. cv. Ilmichungjang) and chilling-resistant pea (Pisum sativum L. cv. Giant) leaf discs in relation to possible damage in D1 protein. In both plants, dark-chilling did not cause any noticeable changes in (Fv)m/Fm and lincomycin did not affect the decrease in (Fv)m/Fm caused by light-chilling. This result suggests that the de novo synthesis of D1 protein did not occur actively during light-chilling. In pea light-chilled for 6 h. the decreased (Fv)m/Fm was partly recovered in the dark, and almost complete recovery was observed in the light. In cucumber light-chilled for 3 h. the reduced (Fv)m/Fm decreased further for the initial 2 h recovery process in the light regardless of the treatment of lincomycin and recovered very slowly. In both plant species, the treatment of lincomycin inhibited the recovery process in the light, but did not significantly inhibit the process in the dark. In cucumber leaves pulse-labeled with $[^{35}S]Met$, the labeled band intensities of isolated pigment-protein complexes were almost the same during the 6 h light-chilling, but significant decreases in band intensities were observed during the 3 h recovery period. This result suggests that the irreversibly damaged D1 protein was degraded during the recovery period. However, no noticeable changes were observed in the pea leaves during the 12 h chilling and 3 h recovery period. The polyacrylamide gel electrophoresis of the pigment-protein complexes showed that the principal lesion sites of light-chilling were different from those of room temperature photoinhibition.

The Production of Lincomycin by Repeated Batch Cultures of Immobilized Streptomyces lincolnensis (고정화된 Streptomyces lincolnensis의 반복 회분식 배양에 의한 린코마이신 생산)

  • The production stability of high-yielding mutants of Streptomyces lincolnensis immobilized on celite beads was examined in repeated batch cultures. We also explored the feasibility of immobilization of vegetative mycelial cells on pre-wetted celite beads, which is practical method for cell immobilization. Repeated transfer of immobilized cells into fresh medium every 10 days increased productivity of immobilized cells and maximum concentration of lincomycin, 1007 $({\pm}256)$ mg/L, was obtained at the end of the ninth cycle. A 1.4-fold higher productivity was obtained in immobilized-cell culture than that obtained by suspended-cell culture. When pre-wetted beads were inoculated with vegetative mycelia and cultured a slightly higher amount of immobilized cells and lincomycin was obtained more than those obtained by culture of spores immobilized on dry beads. This result indicates that immobilization of mycelial cells on pre-wetted beads was readily available. This technique is simple and no additional facilities are required for cell immobilization.

Effects of Dietary Supplemental Lincomycin and Food Waste on Performance and Blood Components of Broiler Chicks (린코마이신과 남은 음식물의 첨가.급여가 육계의 생산성 및 혈액의 성상에 미치는 영향)

  • Two experiments were conducted to investigate the effect of dietary supplemental lincomycin at different food waste supplements on performance of broiler chicks. Diets were based corn, soybean meal and contained 3,078, 3,089 ㎉/kg ME, 21% CP for the first three weeks and 3,116 ㎉/kg and 19% CP for the rest two weeks. Three levels of lincomycin(LM; 0, 2.2, 4.4mg/kg) and food waste (FW;0, 5, 10%) were differently used for both experiments. Weight gain, feed intake, feed conversion ratio were weekly measured for five weeks. Blood components, liver and abdominal fat were sampled at the end of experiments. In experiment, 1 weight gain of birds fed 4.4mg/kg LM was significantly higher than that of no LM treatments (p<0.05). It was tended to decrease in 10% food waste treatment compared to that of control but was not significantly different. FCR of chocks fed LM supplement tended to improve as dietary LM level increased. It was not superior in 10% food waste treatment to that of control, but tended to improve compared to that of other treatments. Weight gain and FCR of birds fed 2.2mg/kg LM without food waste tended to improve compared to those of other treatments, but were not significantly different. Liver to body weight ratio birds fed LM supplements regardless of FW decreased significantly compared to that of control (p<0.05), but was not consistency in FW treatments. There was no significance in blood components except blood protein. In experiment 2, weight gain of chicks fed no FW showed quadratic, but increased as dietary supplemental LM increased. Birds fed 5% FW was not superior weight gain to that of other FW treatments. It also decreased in no LM treatment with 10% FW compared to that of control. However, the birds fed 4.4 mg/kg LM with 10% FW exhibited significantly higher weight gain the trend of weight gain. It tended to increase up to 10% FW combined with 4.4 mg/kg LM, but was not significantly different. Liver to body weight ratio of chicks fed LM was significantly decreased more than the control(p<0.05).The results of these experiments indicated that chicks fed 10% FW with 4.4 mg/kg LM supplements maximized growth rate of all treatments, but was not improve FCR in both experiments.

  • These studies were conducted to investigate the effect of dietary supplemental lincomycin at different food waste supplements on performance and blood components of broiler chicks. Diets were based corn, soybean meal and contained 3,100㎉/kg ME, 21% CP for the rest two weeks. Three levels of dietary lincomycin(LM ; 0, 2.2, 4.4 mg/kg) and food waste(FW : 0, 7.5, 15%) were added for both experiments. Weight gain, feed intake and feed conversion ratio(FCR) were measured for five weeks. Blood components, liver weight and abdominal fat ratio to body weight were examined at the end of experiment. Metabolizable energy of food waste used in formulating diet prior to feeding trials were measured 2,504(Expt 1) and 2,734㎉/kg(Expt 2), respectively. In Expt 1, weight gain of birds fed 4.4mg/kg LM was significantly higher than that of no LM treatment(P〈0.05). It also tended to increase in 7.5% food waste treatment compared to that of control but was not significantly different. FCR of chicks fed 2.2mg/kg LM supplement was significantly improved compared to that of other treatments. However, chicks fed food waste supplements had higher FCR than control as a dietary FW level increased(P〈0.05). There were no significance in blood components of all treatments. Liver to body weight ratio of birds fed 7.5% FW supplements decreased significantly(P〈0.05), but was no consistency in LM treatments. In Expt 2, weight gain of chicks fed 15% FW was not significantly different from the control and 7.5% FW. It tended to increase as dietary supplemental LM increased. It also increased in no LM treatment with 15% FW compared to that of control. However, the birds fed 2.2mg/kg LM with 15% FW exhibited higher weight gain than dther treatments. As FW supplement increased, the feed intake increased significantly(P〈0.05). There were no significance in blood components except serum calcium. Weights of liver were not affected by dietary FW or LM. The results of these experiments indicated that chicks fed 4.4mg/kg LM supplements maximized growth rate of all treatments and improved FCR in 2.2mg/kg LM.
