Melting curve analysis of fluorescently labeled DNA fragments is used extensively for genotyping single nucleotide polymorphism(SNP). Here, we evaluated a SNP genotyping method by melting curve analysis with the two probe chemistries in a 384-well plate format on a Roche LightCycler 480. The HybProbe chemistry is based on the fluorescence resonance energy transfer(FRET) and the SimpleProbe chemistry uses a terminal self-quenching fluorophore. We evaluated FRET HybProbes and SimpleProbes for two SNP sites closely linked to two quantitative trait loci(QTL) for southern root-knot nematode resistance. These probes were used to genotype the two parents and 94 $F_2$ plants from the cross of PI 96354$\times$Bossier. The SNP genotypes of all samples determined by the LightCycler software agreed with previously determined SSR genotypes and the SNP genotypes determined on a Luminex 100 flow cytometry instrument. Multiplexed HybProbes for the two SNPs showed a 98.4% success rate and 100% concordance between repeats two of the same 96 DNA samples. Also, we developed a HybProbe assay for the Rcs3 gene conditioning broad resistance to the frogeye leaf spot(FLS) disease. The LightCycler 480 provides rapid PCR on 384-well plate and allows simultaneous amplification and analysis in approximately 2 hours without any additional steps after amplification. This allowed for a reduction of the potential contamination of PCR products, simplicity, and enablement of a streamlined workflow. The melting curve analysis on the LightCycler 480 provided high-throughput and rapid SNP genotyping and appears highly effective for marker-assisted selection in soybean.

Objectives: Sprague-Dawley rats were fed with high fat diet, and atherogenic changes were seen in the aorta. However, when Sprague-Dawley rats were fed with a high fat diet and administered Youngyopaedoc-san together, these atherogenic changes were rarely seen. This study was aimed to find the inflammatory marker level changes in Sprague-Dawley rats with Youngyopaedoc-san-related anti-atherogenic effect. Methods: The extract from Youngyopaedoc-san was made by the pharmacy department of Kyung-hee Oriental Medical Hospital. The animals were divided into five groups: normal diet, high fat diet, high fat diet with Youngyopaedoc-san, high fat diet with Vytorin, and high fat diet with Youngyopaedoc-san and Vytorin. A light microscopic image of a cross section taken from the aorta of the Sprague-Dawley rat was analyzed. We compared inflammatory marker levels among the five groups. Results: The complex of Youngyopaedoc-san and Vytorin has more anti-atherogenic effects in the aorta of Sprague-Dawley rats fed with high fat diet than Vytorin alone. Youngyopaedoc-san has inhibitory effect on the increase of IFN-${\gamma}$ and IL-2 levels. The difference on eosinophil levels of each group was statistically significant, but the eosinophil level of each group was within normal limits, so the difference on eosinophil levels was not clinically significant. Conclusions: Youngyopaedoc-san-related anti-atherogenic effect could be a result of inhibitory mechanism on IFN-${\gamma}$ and IL-2.
Stress is accompanied by changes in the responses of the autonomic nervous system, and the heart rate variability (HRV) index is a quantitative marker that reflects autonomic responses induced by stressors. In this study, we observed changes in the autonomic responses induced by combinations of 30% oxygen administration and color light for stress relaxation. In all, 42 participants produced stress symptoms over the preceding two weeks, as rated on the stress response scale. After stress assessment, they were exposed to three therapeutic conditions, and electrocardiogram (ECG) signals were recorded before, during, and after therapy. The three therapy conditions consisted of only 30% oxygen administration with white light, a combination of 30% oxygen and orange light, and a combination of 30% oxygen and blue light. The HRV indices extracted from ECG signals were heart rate (HR), the standard deviation of the RR interval (SDNN), the mean square root of consecutive RR interval difference values (RMSSD), the low frequency component of HRV (LF), the high frequency component (HF), and the LF/HF ratio. These indicators were used to compare mean values before and after therapy. The results showed that HR and the LF/HF ratio were significantly lower after therapy than before it. In particular, the condition with 30% oxygen and blue light yielded significantly greater RMSSD and HF increases, as well as decreases in LF/HF ratio than in other two conditions. Our results suggest that therapy with 30% oxygen and blue light is the most effective for the relaxation of stress, which implies autonomic balance by parasympathetic activation.

Constrained IoT devices cannot achieve full coverage of software attestation even though the integrity of software is critical. The limited modification attacks on control flow of software aim at the shadow area uncovered in software attestation processes. In this paper, we propose a light-weight protection system that detects modification by injecting markers to program code.
Most genes are processed by alternative splicing for gene expression, resulting in the complexity of the transcriptome in eukaryotes. It allows a limited number of genes to encode various proteins with intricate functions. Alternative splicing is regulated by genetic mutations in cis-regulatory factors and epigenetic events. Furthermore, splicing events occur differently according to cell type, developmental stage, and various diseases, including cancer. Genome instability and flexible proteomes by alternative splicing could affect cancer cells to grow and survive, leading to metastasis. Cancer cells that are transformed by aberrant and uncontrolled mechanisms could produce alternative splicing to maintain and spread them continuously. Splicing variants in various cancers represent crucial roles for tumorigenesis. Taken together, the identification of alternative spliced variants as biomarkers to distinguish between normal and cancer cells could cast light on tumorigenesis.

The SMAC (Simplified Marker And Cell) algorithm is extended for an application to thermal non-equilibrium two-phase flows in light water nuclear reactors (LWRs). A two-fluid three-field model is adopted and a multi-dimensional unstructured grid is used for complicated geometries. The phase change and the time derivative terms appearing in the continuity equations are implemented implicitly in a pressure correction equation. The energy equations are decoupled from the momentum equations for faster convergence. The verification of the present numerical method was carried out against a set of test problems which includes the single and the two-phase flows. The results are also compared to those of the semi-implicit ICE method, where the energy equations are coupled with the momentum equation for pressure correction.
It has been widely accepted that Korean -i/ka, known as the nominative marker, can give rise to exhaustivity effect. However, it has been rarely discussed and still remains unclear how and why the particle can convey exhaustivity. The purpose of this paper is to shed light on this issue, based on Kim's (2014) analysis of the function of -i/ka. The main claim is that the exhaustivity expressed by -i/ka is a pragmatic effect derived from the interaction of the function of -i/ka, namely, unique specification, and the existence of alternatives to the denotation of an -i/ka-marked phrase in discourse context. Thus, the nature of the exhaustive implicature induced by -i/ka is not conventional but conversational.
Alzheimer's disease (AD) is the most common age-dependent neurodegenerative disorder and shows progressive memory loss and cognitive decline. Intraneuronal filaments composed of aggregated hyperphosphorylated tau protein, called neurofibrillary tangles, along with extracellular accumulations of amyloid $\beta$ protein (A$\beta$), called senile plaques, are known to be the neuropathological hallmarks of AD. In light of recent studies, epigenetic modification has emerged as one of the pathogenic mechanisms of AD. Epigenetic changes encompass an array of molecular modifications to both DNA and chromatin, including transcription factors and cofactors. In this review, we summarize how DNA methylation and changes to DNA chromatin packaging by post-translational histone modification are involved in AD. In addition, we describe the role of SIRTs, histone deacetylases, and the effect of SIRT-modulating drugs on AD. Lastly, we discuss how amyloid precursor protein (APP) intracellular domain (AICD) regulates neuronal transcription. Our understanding of the epigenomes and transcriptomes of AD may warrant future identification of novel biological markers and beneficial therapeutic targets for AD.
