광환경과 기상조건에 따른 야간조도 영향 분석 (An Analysis for the Night illuminance Affected on Light Environments and Weather Conditions)

  • This study deals with the light environments and weather conditions affecting to the night illuminance over the Korean peninsula. The experiment was executed to analyze the effects on the illuminance at separate sites(Gyeryong and Pilseung) considering the different light environments. The analysis was applied to illuminance measurement from the lightmeter, which was developed for the IYA(International Year of Astronomy) 2009, in order to observe the illuminance of areal networks. The weather observations, such as the cloud cover and visibility, were used to understand the quantitative influence of the illuminance to the selected sites. The results show that the illuminance measurements are significantly different from data of the operational illuminance prediction model which simply applies extinction effect for the illuminance. It shows that these differences are caused by the light environments and weather conditions for each site. Therefore, it can be confirmed that the night illuminance is the output of interaction with the characteristics of light for luminous sources.

Effects of the Net Contrast of a Model Codend on the Escapement of Juvenile Red Seabream Pagrus major

  • Visual stimuli of nets, which affect fishing selectivity, vary by twine diameter, color, and material under different light conditions and visual geometries. In this study, two cylindrical model codends of two mesh sizes, 28 and 43 mm, were made of high-contrast, dark brown polyethylene (PE) netting twine and low contrast, light-blue polyamid (PA) monofilament twine. Each model codend was filled with juvenile seabream and set in the water channel of a light-blue circular tank under a flow speed 0.8 m/s for 30 min. Light conditions were set to relatively bright, dim, and dark. The resulting retention rates of juvenile seabream were 15-35% lower for the low-contrast codend with PA monofilament than for the high contrast PE twine netting under bright and dim light conditions, while no difference was observed under dark conditions. The effects of mesh size and netting contrast on the retention rate were dependent on the light level, while the retention rate due to netting contrast was independent of mesh size. Therefore, low-contrast nets in the codend could help to reduce juvenile bycatch by disturbing the orderly optomotor response.

Photocatalytic Degradation Characteristics of Organic Compound by Boron-doped TiO2 Catalysts

  • Boron-doped $TiO_2$ photocatalysts were synthesized by a modified sol-gel method and their photocatalytic activities were performed and compared with those of pure synthetic and commercial $TiO_2$ catalysts under UV or visible light conditions. Pure $TiO_2$ itself exhibited very negligible photocatalytic performance under visible light conditions in the aspects of toluene decomposition reactions, although significant decomposition potential was observed as expected with UV light conditions. However, boron doping over $TiO_2$ significantly improved photocatalytic activity particularly under visible conditions, where over 95% degradation of toluene was achieved with 1wt% $B-TiO_2$ within 2 hrs. All the decomposition reactions seemed to follow pseudo first-order kinetics. The effects of boron-doping and its characteristics are further discussed through the kinetic studies and comparison of results.

  • The variation of glucosinolates (GSLs) in Chinese cabbage ('Sinhongssam') (Brassica rapa L. spp. pekinensis) cultivated under lights to control plant growth conditions was evaluated at different development stages. Under experimental conditions in plant factory system, plant growth conditions including light, temperature, and nutrients were designed to enhance GSLs. The variation of glucosinolates (GSLs) in Chinese cabbage ('Sinhongssam') (Brassica rapa L. spp. pekinensis) cultivated under lights to control plant growth conditions was evaluated at different development stages. Under experimental conditions in plant factory system, plant growth conditions including light, temperature, and nutrients were designed to enhance GSLs. The contents of GSLs were quantified in Chinese cabbage according to different light sources (Red+White, RW; Red+Blue+White, RBW, Fluorescence lamp, FL) at development stages (28, 42, and 56 days after sowing, DAS) using HPLC. Nine GSLs including five aliphatic (progoitrin, sinigrin, glucoalyssin, gluconapin, and glucobrassicanapin) three indolyl (glucobrassicin, 4-methoxyglucobrassicin, and neoglucobrassicin), and one aromatic (gluconasturtiin) GSLs were identified based on peak retention time in previous results of our laboratory. GSL contents were higher in RBW (36.55) and lower in FL ($15.24{\mu}mol/g/\;DW$). Results revealed that GSL contents were higher under controlled photoperiods (20/4 h) ($58.35{\mu}mol/g\;DW$) and controlled light intensity ($160{\mu}mol/m^2/s$) ($34.02{\mu}mol/g\;DW$), respectively. Lower amount of progoitrin and comparatively higher amount of glucobrassicin and gluconasturtiin was noted in Chinese cabbage cultivated under FL light (2.38, 9.82, and 2.10) at 42 DAS, photoperiod 20/4 h (3.16, 2.52, and 1.30) at 28 DAS, and light intensity at $130{\mu}mol/m^2/s$ (2.28, 2.24, and $1.51{\mu}mol/g\;DW$) at 42 DAS. Therefore FL light, photoperiod (20/4 h), and light intensity ($130{\mu}mol/m^2/s$) were considered as most suitable for the enhancement of GSLs in Chinese cabbage.

Nitrate uptake of the red tide dinoflagellate Prorocentrum micans measured using a nutrient repletion method: effect of light intensity

  • The ability of a red tide species to take up nutrients is a critical factor affecting its red tide dynamics and species competition. Nutrient uptake by red tide species has been conventionally measured by incubating nutrient-depleted cells for a short period at 1 or 2 light intensities. This method may be applicable to certain conditions under which cells remain in oligotrophic water for a long time and high nutrients are suddenly introduced. Thus, a new method should be developed that can be applicable to the conditions under which cells are maintained in eutrophicated waters in healthy conditions and experience light and dark cycles and different light intensities during vertical migration. In this study, a new repletion method reflecting these conditions was developed. The nitrate uptake rates of the red tide dinoflagellate Prorocentrum micans originally maintained in nitrate repletion and depletion conditions as a function of nitrate concentration were measured. With increasing light intensity from 10 to $100{\mu}E\;m^{-2}s^{-1}$, the maximum nitrate uptake rate ($V_{max}$) of P. micans increased from 3.6 to $10.8 pM\;cell^{-1}d^{-1}$ and the half saturation constant ($K_{s-NO3}$) increased from 4.1 to $6.9{\mu}M$. At $20{\mu}E\;m^{-2}s^{-1}$, the $V_{max}$ and $K_{s-NO3}$ of P. micans originally maintained in a nitrate repletion condition were similar to those maintained in a nitrate depletion condition. Thus, differences in cells under nutrient repletion and depletion conditions may not affect $K_{s-NO3}$ and $V_{max}$. Moreover, different light intensities may cause differences in the nitrate uptake of migratory phototrophic dinoflagellates.

Growth rates and nitrate uptake of co-occurring red-tide dinoflagellates Alexandrium affine and A. fraterculus as a function of nitrate concentration under light-dark and continuous light conditions

  • The dinoflagellate genus Alexandrium is known to often form harmful algal blooms causing human illness and large-scale mortality of marine organisms. Therefore, the population dynamics of Alexandrium species are of primary concern to scientists and aquaculture farmers. The growth rate of the Alexandrium species is the most important parameter in prediction models and nutrient conditions are critical parameters affecting the growth of phototrophic species. In Korean coastal waters, Alexandrium affine and Alexandrium fraterculus, of similar sizes, often form red-tide patches together. Thus, to understand bloom dynamics of A. affine and A. fraterculus, growth rates and nitrate uptake of each species as a function of nitrate ($NO_3$) concentration at $100{\mu}mol\;photons\;m^{-2}s^{-1}$ under 14-h light : 10-h dark and continuous light conditions were determined using a nutrient repletion method. With increasing $NO_3$ concentration, growth rates and $NO_3$ uptake of A. affine or A. fraterculus increased, but became saturated. Under light : dark conditions, the maximum growth rates of A. affine and A. fraterculus were 0.45 and $0.42d^{-1}$, respectively. However, under continuous light conditions, the maximum growth rate of A. affine slightly increased to $0.46d^{-1}$, but that of A. fraterculus largely decreased. Furthermore, the maximum nitrate uptake of A. affine and A. fraterculus under light : dark conditions were 12.9 and $30.1pM\;cell^{-1}d^{-1}$, respectively. The maximum nitrate uptake of A. affine under continuous light conditions was $16.4pM\;cell^{-1}d^{-1}$. Thus, A. affine and A. fraterculus have similar maximum growth rates at the given $NO_3$ concentration ranges, but they have different maximum nitrate uptake rates. A. affine may have a higher conversion rate of $NO_3$ to body nitrogen than A. fraterculus. Moreover, a longer exposure time to the light may confer an advantage to A. affine over A. fraterculus.

Role of OrfQ in Formation of Light-Harvesting Complex of Rhodobacter sphaeroides under Light-Limiting Photoheterotrophic Conditions

  • A puc-deleted cell of Rhodobacter sphaeroides grows with a doubling time longer than 160 h under light-limiting photoheterotrophic (3 Watts [W]/㎡) conditions due to an absence of the peripheral light-harvesting B800-850 complex. A spontaneous fast-growing mutant, R. sphaeroides SK101, was isolated from the puc-deleted cells cultured photoheterotrophically at 3 W/㎡. This mutant grew with an approximately 40-h doubling time. The growth of the mutant, however, was indistinguishable from its parental strain during photoheterotrophic growth at 10 W/㎡ as well as during aerobic growth. The membrane of SK101 grown aerobically did not reveal the presence of any spectral complex, while the amounts of the B875 complex and photosynthetic pigments of SK101 grown anaerobiclly in the dark with dimethylsulfoxide (DMSO) were the same as those of the parental cell. These results indicate that the oxygen control of the photosynthetic complex formation remained unaltered in the mutant. The B875 complex of SK101 under light-limiting conditions was elevated by 20% to 30% compared with that of the parental cell, which reflected the parallel increase of the bacteriochlorophyll and carotenoid contents of the mutant. When the puc was restored in SK101, the B875 complex level remained unchanged, but that of the B800-850 complex increased. The mutated phenotype of SK101 was complemented with orfQ encoding a putative bacteriochlorophyll-mobilizing protein. Accordingly, it is proposed that the mutated OrfQ of SK101 should have an altered affinity towards the assembly factor specific to the most peripheral light-harvesting complex, which could be either the B875 or the B800-850 complex.

Effects of light and nutrient on flower formation and vegetative growth of Viola collina

  • Background: Mixed breeding herb Viola collina Besser, which produces both chasmogamous and cleistogamous flower, has limited habitats under closed canopy and short and early flowering timing, making it relatively more vulnerable to climate change. To better understand the effect of light and nutrient on the flower formation and vegetative growth of V. collina, a mesocosm experiment was conducted. Two-by-two factorial treatments of two light conditions (100% and 60% of natural light) and two fertilizer treatment conditions (fertilized and not fertilized) were applied in the mesocosm experiment. Results: The number of flowers, including chamogamous and cleistogamous flowers, was highest (5.65/pot) under 60% light and fertilized condition and lowest (1.41/pot) under 100% light and not-fertilized condition. However, above ground vegetative growth was highest (2.89 g/pot) under 100% light and fertilized condition and lowest (2.38 g/pot) under 60% light and not-fertilized condition. Above ground biomass to belowground biomass ratio was highest (1.50) under 60% light and fertilized condition and lowest (1.26) under 100% light and fertilized condition. Conclusions: This study showed that high light and nutrient are responsible for the vegetative growth, though the effect of fertilizer was reduced due to allocation and retainment of nutrients. In addition, the low light is necessary to make flowers, especially chasmogamous flowers.

광 조건에 따른 식물공장 재배 쌈 배추의 기능성 성분 변화 (Variation of Functional Compounds in Leafy Chinese Cabbage Grown Under Different Light Conditions in a Plant Factory)

  • 최근 육종되어 시장에 소개된 신홍쌈 배추를 다양한 광 조건의 식물공장에서 재배하여 그 영양적 특성, 특히 기능성 물질인 phenolic compounds류의 함량을 중심으로 비교분석을 수행하였다. LED 광원을 이용하였으며 LED는 적+청, 적+백, 적+청+백 세가지 조합으로 신홍쌈 재배에 사용하였고 형광등을 광원으로 이용하여 재배한 경우와 비교하였다. LED 적+청 조건에서는 배추가 정상적으로 성장하지 못하여 성분 분석연구에 사용하지 않았으며 형광등을 포함한 다른 광 조건에서는 서로 유사한 성장속도를 보였으며 정상적인 형태 및 크기로 성장하였다. Anthocyanidin 중에서는 cyanidin만이 배추 내에 축적되었으며 그 함량은 LED 조건이 형광등에 비하여 2배 이상 높은 것으로 나타났다. Phenolic acid와 flavonol의 총 함량은 광 조건에 따른 차이가 크게 나타나지 않았다. 특이하게도 caffeic acid는 형광등 조건에서는 거의 축적되지 않았으나 LED 광에서는 형광등 재배 배추에 비하여 70-160배 이상의 높은 함량을 나타내었다. 유리 아미노산과 유리당의 전체 함량은 대체로 LED 광 조건에서 높게 측정되었으나 그 차이는 크지 않았다. 결론적으로, LED 광 중 R+W 조합이 신홍쌈의 기능성 성분 축적 및 식품적 가치상승에 가장 유리한 것으로 나타났다.

Behavioral characteristics of a chondrostean sturgeon species Acipenser baerii prelarvae in response to different environmental light intensities in a diel photoperiodic cycle

  • Behavioral response to a diel photoperiodicity (500 lx for 16 h, 5 lx for 4 h and < 0.5 lx for 4 h) and phototactic characteristics in dark conditions were examined with Siberian sturgeon Acipenser baerii (Chondrostei, Actinopterygii) prelarvae. Siberian sturgeon prelarvae represented both qualitative and quantitative changes in their behavioral patterns according to different light intensities in a diel photoperiodicity. Under daylight conditions (500 lx), prelarvae displayed saltatory changes of behavioral features with ages (Day 0-Day 9) in a general order of swimming-up/drifting, swimming in the upper water column, benthic swimming with rheotaxis, schooling and post-schooling behavior. Compared to daylight conditions, prelarvae tended to show more benthic performances and quantitative reductions of schooling and post-schooling behaviors under dimlight conditions (5 lx). Under dark conditions (< 0.5 lx), prelarvae exhibited a fairly uniform behavioral pattern characterized by the benthic swimming across the bottom of the tank. From phototaxis tests under dark conditions, navigational responses of prelarvae to a spotlight illumination were quantitatively changed as their ages increased. The phototactic responses reached the peak on Day 2, continued until Day 4, and then gradually decreased until Day 8. A partial recovery of positive phototaxis was observed on Day 9. Data from this study suggest that the diel light cycle as well as the light intensity of each interval in the cycle should be considered as important components of a practical guide for evaluating fitness and developmental states of artificially propagated Siberian sturgeon prelarvae.
