  • The assimilability of glucose and xylose of Rhodopseudomonas K-7, whose hydrogen evolution has been characterized previously, was investigated under the anaerobic photosynthetic and the aerobic dark conditions. This organism is able to grow well in the medium containing glutamate and malate as organic substances under the anaerobic light condition. However, the substitution of glucose for malate retarded the growth rate, while the addition of glucose to the seed culture remarkably promoted the utilization of glucose added in the main culture. Optimal glucose concentration in the seed culture to induce glucose assimilability of the organism was around the concentration of 60 mM of glucose. Then, the seed culture grown in the medium containing 60 mM of glucose were inoculated in the medium containing 10, 20, 30, 60 and 100 mM of glucose respectively. The results were revealed that the consumable content of glucose was limited even though the high concentrations of glucose was contained in the medium. The consumption of considerable amount of glucose was observed when cultured under the aerobic dark conditions than the anaerobic illuminated conditions.

  • Since Sirimavo Bandarinaike became the first female prime minister in 1960, women all over the world has been trying to break the highest glass ceiling. In a place where men had been dominating throughout history, women political leaders had to find a way to look the part by wearing power suits. Feminity was out of the question until the digital era of 21st century started. The world has evolved into a place where feminity could be strength instead of weakness. Now women political leaders are showing body curves, cleavage, using vivid and pastel colors, soft fabrics, elaborate details, accessories, long and curly hair. The padded shoulders of dark coloured power suits were replaced by soft colorful flowing feminine suits. Yulia Tymoshenko of Ukraine is the best example of 21st century woman political leader using feminity in her fashion style while enacting her role. When she first started politics in 1990's, she wore dark colored power suits like all other women political leaders did with short dark hair. As time passed her suits became more feminine with light colors and elaborate details. She has changed her hair into a neat braid, based on a traditional Ukrainian hair style. She used orange color for the Orange Revolution and white for pure image. Yulia Tymoshenko's fashion style as a way of role enactment showed professionalism, feminity, integrity, and ethnicity.

  • Zoeae of Alpheus euplirosyne rlchardsoni Valdwyne, 1971 reared in the laboratory are described and Illustrated The culture was carded out under the laboratory condition with the photoperiod l4hr light and lOhr darkness and salinity of 33.3$\textperthousand$ at 25$^{\circ}$C. This species exhibits extended development on the egg size just before hatching and on the length of the first zoea. The morphological differences between the first zoea of this species and those of five other species in the same genus are discussed.

Effects of Low Dose Gamma Radiation on the Formation of Shikonin Derivatives on Callus Cultures of Lithospermum erythrorhizon (지치(Lithospermum erythrorhizon S.)의 캘러스배양에서 Shikonin 유도체 생산에 미치는 저선량γ선의 효과)

  • The effects of low dosage ${\gamma}$-radiation on the cell growth and the formation of shikonin derivatives were investigated in callus cultures of Lithospermum erythrorhizon under different medium and light conditions. Gamma radiation significantly affected the cell growed and formation of shikonin derivatives, depending on the culture conditions. In the cell cultures grown on M-9 medium, 2Gy and 16Gy of ${\gamma}$-radiation increased the calli growth and the formation of shikonin derivatives, respectively under 16hr day light condition. When calli were cultured for 60 days in the dark after irradiation of ${\gamma}$-radiation, cell growth was increased at low dosage of 1Gy and 2Gy in LS medium containing BA 2mg/L and IAA 0.2mg/L. Interestingly, calli grown in M-9 medium by 2Gy irradiation for 60 days significantly stimulated the formation of shikonin derivatives(13.21mg/g cell fresh wt), which was approximately 6 times higher than untreated cells.

  • Effect of light and pH on mycelial growth and sporangium formation of Phytophthora capsici Leon, causal fungus of fruit rot of red pepper was carried out on culture media. The maximum mycelial growth occurred at $25^{\circ}C$ and pH 6.0 on potato sucrose agar. The fungus did not produce sporangia readily in dark condition. However, abundant sporulation occurred by illuminating continuous cool white fluorescent light with 2000 lux for 48 hrs. at $20{\pm}2^{\circ}C$. Oat meal agar was one of the best media for sporangia formation.

  • We investigated the developmental characteristics and life cycle of the lawn cutworm, Spodoptera depravata (Lepidoptera: Noctuidae), which is one of the most important pests causing economic damage in grass production. For larval culture, we provided the zoysiagrass at $25^{\circ}C$ and $60{\pm}5%$ humidity. The durations of the developmental stages were as follows: $4.11{\pm}0.19$ days for eggs, $25.17{\pm}3.02$ for larvae, $8.80{\pm}0.28$ for pupae, and $7.57{\pm}0.95$ for adults. We grew the larvae to the 7th instar stage, unlike previous studies, in which it was assumed that the 6th instar was the final age. There was a significant positive correlation between the body length and head capsule width of each instar larvae. In terms of morphology, the eggs changed from light green immediately following oviposition to black as they developed, and the grass-fed larvae changed from light yellow immediately after hatching to green as development continued. We observed a pattern of black spots at regular intervals on the dorsal sides of the abdomens of the final instar larvae. Furthermore, we detected two notable designs on the dorsal side of the front of the head. The pupal colors changed from light brown and green immediately after pupation, to dark brown as the pupal cuticle hardened. The wingspans of the adults were similar in both sexes. However, the forewings of the males had obvious outer lines and eyespots with dark gray-brown backgrounds, whereas the corresponding features on the female forewings were less obvious. The oviposition preperiod was 2.11 days, the oviposition period was 4.2 days, the average fecundity per female was approximately 341 eggs, and the hatching rate was approximately 76.1%.

  • A new light pink spray-mum, 'Noble ND' was bred by crossing 'Mona Lisa', a white anemone type flower, and 'Dark Weldon', a pink anemone type flower in October, 2004. The growth and flowering characteristics were investigated in forcing, natural, and retarding cultures from 2006 to 2007. The 'Noble ND' was a medium-flowering fall-mum with light pink anemone type flower and had 9.2 flowers per stem, with 4.8 cm flower diameter in natural culture. The number of days to flowering and flowers per plant was 57 and 7.4, respectively, in forcing culture of 'Noble ND', whereas the number of days to flowering was 48 in retarding culture.

  • This study was conducted to determine the appropriate culture conditions for developing photoautotrophic cells from tobacco using plant growth regulators, sucrose and light condition in LS medium. The greatest callus induction was observed in the LS medium supplemented with $10^{-5}$ M NAA and $10^{-4}$ M BA, showing 3.08 g of callus fresh weight determined at 30 days after incubation. The highest chlorophyll content of callus was obtained in the LS medium supplemented with $10^{-5}$M NAA and $10^{-6}$ M BA, showing 28.42${\mu}g/g$. In both light and dark conditions, callus induction increased as the concentration of sucrose increased from 0.5%to 3.0%. In particular, in the light condition, the greatest callus induction was made in the LS medium supplemented with 2% sucrose. On the other hand, the heighest chlorophyll content was observed at 0.5% sucrose and chlorophyll wasn't induced in dark condition. The chlorophyll content of callus cultured in LS medium containing 0.75% sucrose was similar to 1% sucrose, as far as the chlorophyll content of callus was concerned.

Practical Propagation Methods for Production of Prothalli and Sporophytes in Deparia pycnosora (Christ) M. Kato

  • Deparia pycnosora (Christ) M. Kato is a fern used as ornamental plant. In addition, it is called "Teol-go-sa-ri" in Korean name. The aim of this study was to develop a practical propagation method of D. pycnosora using tissue culture technique. Prothallus obtained from spore germination was the used as experiment materials. The prothalli (300 mg) used in all experiments were sub-cultured for 8-week intervals. The most suitable media for prothallus propagation were identified by culturing 300 mg of prothalli in $1/4{\times}$, $1/2{\times}$, $1{\times}$, $2{\times}$ MS medium and in Knop medium for 8 weeks. Also, the prothalli were cultured by chopping with a scalpel. In addition, sucrose, activated charcoal, and total nitrogen source were added in different concentrations based on the culture medium selected. Cultures were maintained at a temperature of $25{\pm}1^{\circ}C$, light intensity of $30{\times}1.0{\mu}mol-m-2{\cdot}s-1$, and a photoperiod of 16/8 h (light/dark) in in vitro. The results showed that optimum was achieved prothallus fresh weight and development in $1{\times}$ MS medium. When other components were added to the basic $1{\times}$ MS medium, prothallus propagation was maximized in $1{\times}$ MS medium supplemented with 2% sucrose, 0.2% activated charcoal, and 60 mM total nitrogen. To select a suitable soil mixture for sporophyte formation, 1.0 g of prothallus was blended with distilled water, spread on five combinations of different soil substrates (decomposed granite, horticultural substrates, peat moss, and perlite), and cultivated for 12 weeks. The sporophyte cultures were maintained at a temperature of $25{\pm}1^{\circ}C$, light intensity of $43{\pm}2.0{\mu}mol-m-2{\cdot}s-1$, humidity of $84{\pm}1.4%$, and a photoperiod of 16/8 h (light/dark). As a results, horticultural substrate alone, 2:1 (v:v) mixtures of horticultural substrate and perlite, and 2:1 mixtures of horticultural substrate and decomposed granite induced 208.0, 201.3 and 248.8 sporophytes per pot, respectively. Therefore, this result could provide a practical mass propagation method of D. pycnosora

Temperature and Light Responses on the Growth and Maturation of Gametophytes of Undaria pinnatifida (Harvey) Suringar in Korea (한국산 미역 배우체의 생장과 성숙에 대한 온도 및 광반응)

  • Temperature and light responses on the growth and maturation of gametophytes of Undaria pinnatifida were studied in laboratory culture. The effect of the environmental factors on formation of young sporophyte of U. pinnatifida was also examined in the same culture system. Maximum growth and rapid maturation of the gametophytes were observed at $12:12LD,\;17^{\circ}C$ and 60{\mu}mol\;m^{-2}s^{-1}. However, they survived in a wide range of the examined temperature, light intensity and photoperiod. Particularly they survived under continuous dark condition (0:24LD) until 210 days without any growth and maturation, but died within $10\~40$ days at $30^{\circ}C$. This suggests that optimum rendition for conservation of Undaria gametophytes is under continuous dark photoperiod at $17\~25^{\circ}C$.
