Tea Breeding in Korea (우리나라 차나무 품종육성)

  • When tea trees were introduced to Korea peninsular from China? Historically, Mr. Taeryum, an envoy of Shilla dynasty brought tea seeds from China during Tang dynasty and the seeds were planted at Jiri Mt. by the order of King Heungduk at AD828. During Koryo Dynasty(918 1392), Buddhism spread rapidly all over the country and the tea culture reached its highest stage of prosperity. At the Chosun Dynasty, however, the ceremonial drinking of tea vanished almost completely due to the flourishing Confucian tradition, a kind of substitution of Buddhism. But a few people have supported the traditional tea culture by themselves. Since the independence of Korea soon after the World War II at 1945, Korean War have been exploded at 1950. After economic evolution have been succeeded at 1980, the cultivation area of tea trees has been increased about 2,000ha and the cultural tradition of tea drinking has become popular again at a tea consume quantities amounted to 100g per capita at 2004. The northern limited area of tea plant is lined on the southern part of Korea peninsular. It is very small region compared to China about one million ha and to Japan over 60 thousand ha. It is problem not only the area of tea fields but also the methodology of tea cultivation, for examples without clonal cultivars and mechanical systems. WTO treatments was discussing with Korea, China and Japan government at 2005. Green tea custom is very high at 514% in Korea. If three countries will be agreed the imported tax will be cut off, the Korean tea farmers will be confused because of unstable situation of tea markets. All most of tea farmers should be made the tea fields by seeding not clonal propagation. Because of clonal cultivars have not developed in Korea, there have not been the research institutes for tea plants and manufactures before 1992. Now there are three research institute of tea in Korea; Tea Experiment Station at Bosung of Jeonnam Agricultural Research & Extension Services, Mokpo Experimental Station of National Institute of Crop Science, and Green Tea Cluster Institute of Hadong. Mokpo and Hadong Research Station were established at 2004 and at 2005 but Bosung Station was established at 1992. Seven clonal tea cultivers were selected at Bosung Station; Bohyang, Myngsun, Chanlok, Sunhyang, Mihyang, Jinhyang and Ohsun until 2004. Mokpo Experimental Institute was started the tea provenance testing about 4 provenances: Kangwon-do, Jeonlabuk-do, Jeonlanam-do, and Kyungsangnam-do. Korean new tea cultivers should be selected because Koran wild tea population have been high genetic variation. If tea breeding research will be successful to select new clonal cultivers, the tea farmers of Korea will be stable after WTO treatment with each country.

A study of on site Pilot plant test of drying sewage sludge using Chain crusher flash dryer (타격기류 건조장치에 의한 하수슬러지의 건조 실증실험에 관한 연구)

  • Effective drying method of sewage sludge is researched in this study. To dry the sludge, chain crusher flash dryer was adopted to remove moisture content in the cell which is mostly responsible for the sludge moisture content. And Pilot plant experiment was conducted in real life sewage treatment plant to study effect and characteristics of operating conditions. Operating variables include sludge feeding rate, rotational speed of chain, process temperature and feed moisture content. As rotational speed of chain increased, product yield of sludge increased, and the performance of the testing system increased. And, as process temperature increased, the sludge drying efficiency increased. It is found that optimum feed moisture content is at 60% which shows the maximum sludge product yield and about 10 moisture content(%) of sludge product. Sludge feed rate showed optimal value, and when the sludge feed rate is exceeded, sludge product yield did not increased but the amount of residue increased. Pilot plant experiment results are as follow. The optimal condition for the rotational speed of chain 1600rpm(max. speed), final sludge discharge temperature $80^{\circ}C$, feed moisture content 60%, and feed rate 60kg/h. When the plant was operated at the optimal conditions, the final product showed fairly good results such as sludge product yield 85.5%, moisture content 11.0% and sludge drying efficiency 81.7%.

Novel Nucleotide Variations, Haplotypes Structure and Associations with Growth Related Traits of Goat AT Motif-Binding Factor (ATBF1) Gene

  • The AT motif-binding factor (ATBF1) not only interacts with protein inhibitor of activated signal transducer and activator of transcription 3 (STAT3) (PIAS3) to suppress STAT3 signaling regulating embryo early development and cell differentiation, but is required for early activation of the pituitary specific transcription factor 1 (Pit1) gene (also known as POU1F1) critically affecting mammalian growth and development. The goal of this study was to detect novel nucleotide variations and haplotypes structure of the ATBF1 gene, as well as to test their associations with growth-related traits in goats. Herein, a total of seven novel single nucleotide polymorphisms (SNPs) (SNP 1-7) within this gene were found in two well-known Chinese native goat breeds. Haplotypes structure analysis demonstrated that there were four haplotypes in Hainan black goat while seventeen haplotypes in Xinong Saanen dairy goat, and both breeds only shared one haplotype (hap1). Association testing revealed that the SNP2, SNP5, SNP6, and SNP7 loci were also found to significantly associate with growth-related traits in goats, respectively. Moreover, one diplotype in Xinong Saanen dairy goats significantly linked to growth related traits. These preliminary findings not only would extend the spectrum of genetic variations of the goat ATBF1 gene, but also would contribute to implementing marker-assisted selection in genetics and breeding in goats.

Analysis of BWIM Signal Variation Due to Different Vehicle Travelling Conditions Using Field Measurement and Numerical Analysis (수치해석 및 현장계측을 통한 차량주행조건에 따른 BWIM 신호 변화 분석)

  • Bridge Weigh-in-Motion(BWIM) system calculates a travelling vehicle's weight without interruption of traffic flow by analyzing the signals that are acquired from various sensors installed in the bridge. BWIM system or data accumulated from the BWIM system can be utilized to development of updated live load model for highway bridge design, fatigue load model for estimation of remaining life of bridges, etc. Field test with moving trucks including various load cases should be performed to guarantee successful development of precise BWIM system. In this paper, a numerical simulation technique is adopted as an alternative or supplement to the vehicle traveling test that is indispensible but expensive in time and budget. The constructed numerical model is validated by comparison experimentally measured signal with numerically generated signal. Also vehicles with various dynamic characteristics and travelling conditions are considered in numerical simulation to investigate the variation of bridge responses. Considered parameters in the numerical study are vehicle velocity, natural frequency of the vehicle, height of entry bump, and lateral position of the vehicle. By analyzing the results, it is revealed that the lateral position and natural frequency of the vehicle should be considered to increase precision of developing BWIM system. Since generation of vehicle travelling signal by the numerical simulation technique costs much less than field test, a large number of test parameters can effectively be considered to validate the developed BWIM algorithm. Also, when artificial neural network technique is applied, voluminous data set required for training and testing of the neural network can be prepared by numerical generation. Consequently, proposed numerical simulation technique may contribute to improve precision and performance of BWIM systems.

Development of the Functional Films Coated with Nano-TiO2 Particles for Food Packaging and Removal of Off-flavor from Soybean Sprouts (나노 TiO2를 적용한 식품 포장 필름 개발 및 콩나물의 이취 제거)

  • For testing the ultraviolet (UV)-blocking property of functional films coated with Nano-$TiO_2$ particles, UV-Vis spectra of oriented polypropylene (OPP) films uncoated and coated with $TiO_2$ of 3% and 5% in Polyurethane (PU) and polyvinyl butyral (PVB)-Cellulose binders were measured. The result of UV-Vis analyses showed that the film coated with 5% $TiO_2$ in PVB binders had a significant effect on UV protection of 90% compared with the film uncoated. Also The result of The photodegradation of methylene blue (MB), OPP films coated with 5% in both PU and PVB binders had a high photocatalytic activity for MB degradation. To evaluate the effect of the developed functional film coated with Nano-$TiO_2$ particles, fresh soybean sprouts were used. Nano-$TiO_2$ coated film was observed to decompose the off-flavor produced by soybean sprouts within packages during distribution, but uncoated film did not. Therefore, Nano-$TiO_2$ coated film package could give the greatest effect in extending the shelf life of soybean sprouts.

The Effect of Health Education on the Performance of Health Promoting Behavior in College Students (건강교육이 대학생의 건강증진 행위에 미치는 영향)

  • This study has been done for the purpose of testing the effect of health education on the performance of health promoting behavior in college students, and identifying the factors affecting health promoting behaviors. A Nonequivalent control group posttest research design was used. Two hundred thirty college students at K College in T city were studied. Of them, 114 who attended a systematic health education session for three hours a week during one semester were the experimental group. And 116 college students who were chosen of matched sampling of grade, class and sex were the control group. This study was conducted from March 1 to July 2, 1995. The instruments used for this study included a survey of general characteristics, perceived health status, self-esteem, health promoting behavior and health locus of control. Analysis of data was done by use of mean, 1-test, Pearson correlation coefficient and multiple regression. The results of this study are summarized as follows : 1) The average item score for the health promoting behavior was low at 2.52. In the sub-categories, the highest degree of performance was ‘harmonious relationships’, following ‘sanitary life’, ‘self-esteem’, ‘rest and sleep’, and ‘emotional support’ and the lowest degree was ‘professional health management’. 2) Hypothesis 1 that the college students who get health education will have a higher degree of health promoting behavior than the college students who do not get health education was accepted. There was a statistically significant difference between the average of the experimentalgroup, 2.60, and the average of the control group, 2.45.(t=11.30, p=0.0009). 3) Hypothesis 2 that the college students who get health education will have a higher score of perceived health status than college students who do not get the health education was rejected. (t=1.13, p=0.289) 4) Performance of health promoting behavior was positively correlated with self-esteem and grade and negatively correlated with perceived health status. 5) The most important factor affecting performance of health promoting behavior was self-esteem. The following suggestions are made based on the above results : 1) Replication of the research is needed to confirm effects of health education. 2) More effective health education programs need to be developed through by modification of teaching methods and content analysis of health education. 3) Other factors affecting health promoting behavior should be identified. 4) Nursing colleges or departments of nursing should make an effort to develop and carry out various health education programs for the health promotion of all college students.

Psychometrics of Task Self-Efficacy Scale for Korean Elderly (노인의 일상활동수행에 대한 자기효능척도의 신뢰도 및 타당도 평가)

  • The purpose of the study was to test the reliability and validity of the Korean version of Task Self-Efficacy Scale for activities of dally living (ADL). The Task Self-Efficacy Scale was developed by Roberts(1996) for low-intensity exercise study with older people to predict their performance of ADL. The scale was translated and back translated by bilingual persons, and then was modified to resolve variations in the translations. The Korean version of Self-Efficacy Scale for ADL was then administered to 193 elderly people including 95 hospitalized patients and 98 outpatients or healthy people. Face to face interview was used to fill out the structured questionnaire, and each interview took approximately 30 minutes. The subjects for the study were 80 women and 112 men with an age range of 65 to 95 years(M=71 years) of whom 82.6% classified themselves as moderate or quite active Most subjects(80.2%) had an education level of elementary school or less. The Self-Efficacy Scale for ADL is measured on a 0 to 10 VAS, assessing three areas of ADL : self care activities, household tasks, and motor tasks. The higher the score is, the higher person's confidence in performing ADL. Psychometric testing revealed that the scale was found to be internally consistent, showing a Cronbach's alpha of .97 The scale was significantly correlated with subjects' level of activity and subjective assessment of their health status. Moderate correlation with health-related hardiness scale also supported the validity. Factor analysis was performed to confirm whether the scale represents the three sub-areas as suggested in the literature. The results of the factor analysis led to a three factor solution according to Kaiser's criterion, but the items were not strongly and cleanly loaded for the third factor. This can be explained in that, among the three sub-ADL areas of the self-efficacy scale, the areas of self care activities and household tasks seem to have similar levels of difficulty in performance with not enough differences for the self-efficacy scale to distinguish between the two areas. Therefore, one factor solution was suggested since ADL can be seen as a unit of activities at similar level of difficulty in performance. One factor solution explained 68.1% of variance of the 19-item scale and all items were correlated over .6 with the factor, showing that the selected factor solution fits the model. The results indicated that the Korean version of Task Self-Efficacy Scale for ADL was reliable and valid in producing useful information to evaluate the effects of various interventions toward promoting health and quality of life for elderly people.

Expression of HLA and Mixed Lymphocyte Reaction of Mesenchymal Stem Cells Derived from Human Umbilical Cord Blood (제대혈 유래 중간엽줄기세포에서 HLA의 발현과 Mixed Lymphocyte Reaction)

  • In recent years, the mesenchymal stem cells (MSC) derived from various tissues have been widely tested for developing cell therapies, tissue repair and transplantation. Although there has been much interest in the immunomodulatory properties of MSC and their immunologic reactions following autologous, allogeneic and xenogenic transplantation of MSC in vivo, up to date, the expression of immunogenic markers, such as class I and II human leukocyte antigens (HLA), after differentiation of human umbilical cord blood (hUCB)-derived MSC has been poorly investigated and require extensive in vitro and in vivo testing. In this experiment, the expression of the HLA-ABC and HLA-DR on hUCB-derived MSC have been tested by immunocytochemical staining. The undifferentiated MSC were moderately stained for HLA-ABC but very weakly for HLA-DR. In order to investigate the inhibitory effect of allogeneic lymphocytes on proliferation of MSC, the MSC were cultured in the presence or absence of peripheral allogeneic lymphocytes stimulated with concanavalin A. The allogeneic lymphocytes did not significantly inhibit MSC proliferation. We conclude that hUCB-MSC expressed moderately class I HLA antigen while almost negatively class II HLA antigen. The MSC have an immunomodulatory effect which can suppress the allogeneic response of lymphocytes. These in vitro data suggest that allogeneic MSC derived from cord blood can be useful candidate for allogeneic cell therapy and transplantation without a major risk of rejection.

SPAWNING AND LARVAL DEVELOPMENTS OF TWO SPECIES OF POLYCLAD WORMS, STYLOCHUS IJIMAI & KABURAKI AND PSEUDOSTYLOCHUS OBSCURUS (STIMPSON) (납작벌레류(Stylochus ijimai, Pseudostylochus obscurus)의 산란 및 유생발생에 관하여)

  • 1. Stylochus ijimai spawns from May to October with peak spawning from July to September, P. obscurus spawns from June to October with peak spawning from July to September. 2. S. ijimai spawns approximately 96,000-132,000 eggs by one spawning. 1. Just after spawnings, the eggs of S. ijimai were $102\mu$ in diameter, and those of P. obscurus were $108\mu$. 4. S. ijimai and P. obscurus have simple eggs. S. ijimai develop indirectly having Gotte larvae, while P. obscurus develop directly. 5. These two species show very similar patterns of early developments from maturation division of eggs to the gastrula stage. 6. The zygots of S. ijimai reaches mesentoblast stage 48 hours after fertilization, and Gotte larvae hatch out 7 days after fertilization. The zygots of P. obscurus reaches mesentoblast stage 72 hours after fertilization, and juveniles hatch out 14 days after fertilization. 7. S. ijimai, have 14 days of planktonic larvae stages bearing strong phototacic behavior : P. obscurus have 7 days of planktonic life without phototacic behavior. 8. Newly hatched larvae of S. ijimai and P. obscurus are $138\mu\;and\;170\mu$ in length respectively. The early creeping forms of S. ijimai and P. obscurus are $152\mu\;and\;185\mu$ in length respectively. 9. In the early creeping stage S. ijimai are characterized by testing processes and flattening of the body. In the same stage P. obscurus lost 2 eye-spots in the cerebral area. 10. The early creeping larvae of these two species were found only in mud-flat substrates.

Performance-tests of honeybee queens(Apis mellifera) in Korea (여왕봉(女王蜂)의 능력검정(能力檢定)에 관(關)한 연구(硏究))

  • The study was conducted to obtain some basic information to establish the system of performance-tests and selection of honeybee queens(Apis mellifera) under Korean circumstances, Colony performances were tested with thirty colonies of Apis mellifera at two apiaries in Taegu, Korea from September, 1988 to August 1989. The results of performance-testing on the colonies are summarized as follows : The colony weight measured before wintering was averaged $23.6{\pm}1.90kg$ and the colony weight was decreased by $2.9{\pm}0.82kg$ in average during winter season. Thirteen colonies were entered in two story hive from thirty single box colonies from April 17 to May 5, 1989 with increase of bee population and, consequently, the ability of enter-supers of the colonies apperared to be low. The ability of collecting pollen was measured to be $14.8{\pm}2.15gr$ per colony during 24 hours in April, and the number of swarm cells was counted $12.5{\pm}3.43$ cells per colony in aveage. Tendency to use propolis appeared to be moderate, and the number of returning foragers for a minute per colony was counted $108.7{\pm}18.31$ bees in average. Brood area was measured $2,464{\pm}628,67cm^2$ per colony in the post nectar flow season of acasia, and 30.8 percent of the colonies appeared to be infected with chalkbrood disease, The amount of honey production was $14.9{\pm}8.49kg$ per colony, which was harvested two times during the main nectar flow season of acasia.
