A Study on Improving the Operation of Reading Culture Programs for Elementary School Students in Public Libraries in Suwon City (수원시 공공도서관 초등학생 독서문화프로그램의 운영 개선을 위한 연구)

  • With the cultural and educational functions of public libraries becoming revitalized according to social changes, there has been a development of reading cultural programs. This study researched the operating conditions of reading culture programs for elementary school students in public libraries in Suwon City for the past 5 years and presented the programs' drawbacks and improvement plans drawn from surveys on user satisfaction. An analysis of the survey results yielded the following suggestions for improvement: forming a consultative group with other managers of reading culture programs in Suwon's public libraries, promoting using various media, forming a human resource pool to hire professional instructors, operating of the educational programs for potential volunteers, expanding of the facilities for reading culture programs, and linking with the other folk culture and art organizations in order to develop classes folk culture and art organizations. The results of this paper will be used as a reference when operating conditions of reading culture programs for elementary school students in public libraries in Suwon City, and they will offer the opportunity to implement problems according to needs of participants.

A Study on the Impact of Volunteering Experience in a Small Library Within an Apartment Complex on the Sense of Community: A Case of Apartment Small Libraries in D Metropolitan City (아파트 단지 내 작은도서관 자원봉사 경험이 지역공동체 의식형성에 미치는 영향: D 광역시 아파트 작은도서관 사례를 중심으로)

  • The purpose of this study is to analyze the effect of volunteering experience at a small library in an apartment complex on the formation of a sense of community through a qualitative method. To achieve the study purpose, thirty-three volunteers from five small libraries in an apartment complex in DS-gu at D Metropolitan City were interviewed. According to the study results, they started volunteering at the libraries as a main user of the libraries. Through the volunteering experience, they had positive feelings about the libraries and were aware of the problems with the libraries. Furthermore, the volunteering experience facilitated communication among the residents of the apartment and they recognized the libraries as a shared community space. Finally, through the above processes, the volunteering experience contributes to formimg a sense of community.

  • 이 연구는 대구광역시의 공공도서관 자원봉사자 112명의 행동요인들을 조사하가 위해, 사회복지분야에서 연구된 자원봉사의 여러 가지 행동 모델들이 공공도서관에서도 적용될 수 있다는 것을 보여준다. 선행연구의 환경적 요인, 역할모델 요인, 보상요인, 인지 요인 등이 자원봉사의 참여지속과 강도에 미치는 영향을 분석한 결과, 선정된 독립변수들이 종속변수에 통계적으로 유의한 영향을 미치는 것으로 나타났고, 이 연구에서 새롭게 투입된 공공도서관에 대한 충성도는 공공도서관 자원봉사자의 행동요인을 설명하는 중요한 요인이 될 수 있는 것으로 조사되었다.

A Case Study of Health Information Service for Patient Users in Overseas Hospital Libraries (국외 병원도서관에서의 환자이용자를 위한 건강정보서비스 사례연구)

  • The purpose of this study is to suggest implications from the case of health information service for patients in overseas hospital libraries. A total of 89 overseas hospital libraries were selected, including general hospitals, specialized hospitals, women's hospitals, children's hospitals, and veterans' hospitals. The health information service that is provided in general and common in 89 hospital libraries was surveyed and the differentiated health information service was investigated next. As a result, first, it can be seen that the establishment of hospital libraries and the provision of health information services are common outside of Korea. Second, various human resources such as librarians, health information specialists, medical specialists, social workers, clinical librarians, health education specialists, and volunteers are utilized. Third, it provides not only print materials but also various information sources such as electronic materials, websites, pamphlets, brochures, and provides health information in various languages. Fourth, in providing health (information literacy) education and programs, services are provided through linkage with hospitals, local public libraries, and local communities. The implications for domestic hospital libraries are as follows: First, the change of awareness of the establishment of hospital libraries and the provision of health information services; second, the support of the curriculum and associations and the need for continuing education; third, it is necessary to link with related organizations for mandatory and diversification of health information services in hospital libraries.

Current evidence on acupuncture from sham needle studies (거짓침을 이용한 침연구의 현황)

  • Objectives : There has been a considerable debate about how to best control for placebo effects in clinical trials of acupuncture. Recently several sham needles were developed and validated. This study aimed at summarising the validation studies of these needles and evaluating the outcomes of the randomised controlled trials (RCTs) using them. Methods : Computerised literature searches were performed using 'acupuncture' AND 'placebo OR sham' with a limitation of the results to RCTs in Medline via PubMed and the Cochrane Library. Only formally validated sham needle controlled studies were included. Data were extracted regarding study design, condition, sample size, credibility testing, intervention and outcomes. Methodological quality was assessed using a modified Jadad scale. Results : Three validated sham needles by Streitberger, Park, and Fink, were identified. Acupuncture's effectiveness for various conditions was tested using these needles in 12 RCTs. Real acupuncture was superior to sham acupuncture for rotator cuff tendonitis and hypertension. No significant differences between real acupuncture and sham acupuncture emerged for chemotherapy-related nausea and vomiting, postoperative nausea and vomiting, menstrually related migraine, acute stroke rehabilitation, chronic/episodic tension-type headache, neutrophil respiratory burst in healthy volunteers, alcohol withdrawal symptoms and chronic poststroke leg spasticity. Conclusions : The new sham devices have been adequately validated and may be useful tools for investigating specific research question. In spite of the sham needle's limits, the results of RCTs using such devices tend to suggest that the clinical effects of acupuncture are largely due to a placebo response.

Development of Human Antibody Inhibiting RNase H Activity of Polymerase of Hepatitis B Virus Using Phage Display Technique (Phage Display 기법을 이용한 B형 간염 바이러스 Polymerase의 RNase H 활성을 억제하는 인간 단세포군 항체의 개발)

  • Background: To develop a novel treatment strategy for hepatitis B virus infection, a major cause of liver chirosis and cancer, we aimed to make human monoclonal antibodies inhibiting RNase H activity of P protein playing in important role in HBV replication. In this regard, phage display technology was employed and demonstrated as an efficient cloning method for human monoclonal antibody. So this study analysed the usability of human monoclonal antibody as protein based gene therapy. Methods: RNase H of HBV was expressed as fusion protein with maltose binding protein and purified with amylose resin column. Single chain Fv (scFv) phage antibody library was constructed by PCR cloning using total RNAs of PBMC from 50 healthy volunteers. Binders to RNase H were selected with BIAcore 2000 from the constructed library, and purified as soluble antibody fragment. The affinity and sequences of selected antibody fragments were analyzed with BIAcore and ABI automatic sequencer, respectively. And finally RNase H activity inhibiting assay was carried out. Results: Recombinant RNase H expressed in E. coli exhibited an proper enzyme activity. Naive library of $4.46{\times}10^9cfu$ was screened by BIAcore 2000. Two clones, RN41 and RN56, showed affinity of $4.5{\times}10^{-7}M$ and $1.9{\times}10^{-7}M$, respectively. But RNase H inhibiting activity of RN41 was higher than that of RN56. Conclusion: We cloned human monoclonal antibodies inhibiting RNase H activity of P protein of HBV. These antibodies can be expected to be a good candidate for protein-based antiviral therapy by preventing a replication of HBV if they can be expressed intracellularly in HBV-infected hepatocytes.

Development of HLA-A, -B and -DR Typing Method Using Next-Generation Sequencing (차세대염기서열분석법을 이용한 HLA-A, -B 그리고 -DR 형별 분석법 개발)

  • Background: Research on next-generation sequencing (NGS)-based HLA typing is active. To resolve the phase ambiguity and long turn-around-time of conventional high resolution HLA typing, this study developed a NGS-based high resolution HLA typing method that can handle large-scale samples within an efficient testing time. Methods: For HLA NGS, the condition of nucleic acid extraction, library construction, PCR mechanism, and HLA typing with bioinformatics were developed. To confirm the accuracy of the NGS-based HLA typing method, the results of 192 samples HLA typed by SSOP and 28 samples typed by SBT compared to NGS-based HLA-A, -B and -DR typing. Results: DNA library construction through two-step PCR, NGS sequencing with MiSeq (Illumina Inc., San Diego, USA), and the data analysis platform were established. NGS-based HLA typing results were compatible with known HLA types from 220 blood samples. Conclusion: The NSG-based HLA typing method could handle large volume samples with high-throughput. Therefore, it would be useful for HLA typing of bone marrow donation volunteers.

Selection of Vaccinia Virus-Neutralizing Antibody from a Phage-Display Human-Antibody Library

  • Although smallpox was eradicated in 1980, it is still considered a potential agent of biowarfare and bioterrorism. Smallpox has the potential for high mortality rates along with a major public health impact, eventually causing public panic and social disruption. Passive administration of neutralizing monoclonal antibodies (mAbs) is an effective intervention for various adverse reactions caused by vaccination and the unpredictable nature of emerging and bioterrorist-related infections. Currently, vaccinia immune globulin (VIG) is manufactured from vaccinia vaccine-boosted plasma; however, this production method is not ideal because of its limited availability, low specific activity, and risk of contamination with blood-borne infectious agents. To overcome the limitations of VIG production from human plasma, we isolated two human single-chain variable fragments (scFvs), (SC34 and SC212), bound to vaccinia virus (VACV), from a scFv phage library constructed from the B cells of VACV vaccine-boosted volunteers. The scFvs were converted to human IgG1 (VC34 and VC212). These two anti-VACV mAbs were produced in Chinese Hamster Ovary (CHO) DG44 cells. The binding affinities of VC34 and VC212 were estimated by competition ELISA to $IC_{50}$ values of $2{\mu}g/ml$ (13.33 nM) and $22{\mu}g/ml$ (146.67 nM), respectively. Only the VC212 mAb was proven to neutralize the VACV, as evidenced by the plaque reduction neutralization test (PRNT) result with a $PRNT_{50}$ of ~0.16 mg/ml (${\sim}1.07{\mu}M$). This VC212 could serve as a valuable starting material for further development of VACV-neutralizing human immunoglobulin for a prophylactic measure against post-vaccination complications and for post-exposure treatment against smallpox.

A Survey and Study on Operator's Awareness for a Suggestion of Policy for Revitalization of Small Libraries in Gwangmyeong City (광명시 작은도서관 활성화 정책 제안을 위한 운영자 인식조사 연구)

  • Small libraries have continued to be extended since 2013 enactment of regulations requiring establishment of small libraries in multi family housing complex not less than 500 households and the 'Living SOC Project' of national government. Gwangmyeong city designated the year of 2020 as the 'year of citizen autonomy' sharing the authority with citizens and publicly announced its plan to rapidly extend the living and cultural spaces as a part of that extension. Likewise, as the social environment and the need of users are expected to change, it seems that the small libraries in Gyangmyeong City must prepare a plan for practicing a long and mid term practices in responding to that change. Thus, this Study tries to make a suggestion for a plan for the policy for revitalization of small libraries in order to handle those changing social environment and the need of users. For this, a survey and in-depth group interview were conducted for the operators of the small libraries in Gyangmyeong City. As a result of this Study, the following policies are suggested: first, improvement of support system for improving operational system, strengthening of cooperative system of public libraries, and strengthening of role of public small libraries, strengthening of public characters of private small libraties and differentiated support through evaluation based on abilities; and second, it is necessary for utilization of circulating librarians and professional volunteers, sharing of materials through mutual lending of books, associate librarians having term of office and revitalization of small library conference by regions.

VNTR Polymorphism of Tyrosine Hydroxylase Gene Intron 1 in Schizophrenics (정신분열병 환자의 Tyrosine Hydroxylase 유전자 Intron 1의 VNTR 다형성)

  • Until recently, the etiology of schizophrenia was generally attributed to abnormalities in dopaminergic neurotransmission. Specifically, an excess of dopaminergic activity in the mesolimbic system has been postulated to produce the positive symptoms, while decreased dopaminergic activity in the mesocortical system has been suggested to cause negative symptoms. Accordingly, we performed an association study of schizophrenia with TH gene. Three hundred and seventy four biologically unrelated schizophrenic patients meeting DSM-III-R criteria from Kangnam St. Mary's Hospital affiliated with Catholic university of Korea were recruited for our study. The 393 healthy controls were volunteers for DNA library of Kangnam St. Mary's Hospital without personal or family history of psychiatric and neurologic illness. DNA was extracted from peripheral mononuclear cells and polymorphic region was amplified by polymerase chain reaction. TH intron 1 VNTR polymorphism was typed by silver staining. The allele distributions of TH gene were not different between schizophrenics and controls. However, the frequency of allele A was significantly higher in positive group than that of negative group of schizophrenics. These findings suggest that poitive schizophrenia may be associated with allele A of TH gene.
