The Correlation Between Deltamethrin Exposure and Urinary 3-PBA Concentrations in Rats (Deltamethrin에 노출된 흰쥐의 뇨 중 3-PBA 검출 및 노출상관성)

  • BACKGROUND: Pyrethroids (PYRs) are a widely used insecticide in agriculture and household area. In mammals, PYRs such as deltamethrin is metabolized to 3-phenoxybenzoic acid (3-PBA) in liver that is mainly excreted in urine. This study is designed to single exposure of deltamethrin to rats in a dose-dependent manner and identify the correlation between deltamethrin exposure and its metabolite (3-PBA) in urine. METHODS AND RESULTS: Exposure levels of deltamethrin were control (0 mg/kg bw), low (0.0705 mg/kg bw), medium (0.705 mg/kg bw) and high (7.05 mg/kg bw) dose. Low concentration was derived by ussing Korea predictive operator exposure model (KoPOEM). Dermal exposure persisted for 6 h, and urine specimens were collected for 24 h. The urine matrix was removed after a series of procedures and 3-PBA was analyzed by gas chromatography/mass spectrometry. CONCLUSION: There was a strong correlation ($R^2=0.83$) between the amount of oral exposure to delta me thrin and urinary levelof3-PBAexcreted. In dermal exposure groups of deltamethrin except high-dose, also there was a good correlation between urinary 3-PBA and deltamethrin exposure, but not stronger than in oral deltamethrin exposure groups. Based on these results, therefore, the amount of 3-PBA in urine can be used as a good monitoring indicator that reflexing the exposure level of deltamethrin to human body.

Habitat Characteristics and Management of Abandoned Rice Paddy Field Wetlands in Mountain - In Case of the Uldae Wetland in Bukhansan National Park - (도시 내 묵논습지 생물서식 특성 및 관리방안 -북한산국립공원 울대습지를 대상으로-)

  • The purpose of this study is to identify the ecological characteristics and biological interactions between species of the abandoned rice paddy field in mountainous areas and to suggest a management strategy for stable food chain formation and biodiversity enhancement. The study site is located in Uldae wetland of Songchu district Bukhansan National Park, site characteristics and biological habitat characteristics were identified through site survey and literature survey. With regard to physical environment, among geographical features, the Uldae Wetland and the neighborhood inside the basin was a gently sloping area($5{\sim}15^{\circ}$). And 64.0% of basin faced the north. With regard to water environment, the Uldae Wetland was wetland of rainfed paddy field depending on precipitation and the system of stream flowing into the wetland from valley. According to the results of examining flora in plant ecology, in general, they were herbaceous wetland species. 88.6% of existing plants inside the Uldae Wetland basin was a forest in the mountain. And Quercus spp. community and Pinus densiflora community accounted for 64.6% of that, and was dominant. Except for that, Salix koreensis community was distributed. The existing vegetation of Uldae Wetland inhabited wetland species and terrestrialization indicator species, and it was thought that partial terrestrialization inside the Uldae Wetland was in progress after the discontinuation of paddy cultivation, such as the expansion of Salix koreensis distribution area. In the status of appearing faunae in the Uldae Wetland with regard to wildbirds of appearing principal species, The Uldae wetland was based on a abandoned rice paddy field various wildlife, and was a wildlife feeding, spawning, and resting place. The water environment was an important factor in maintaining the wetland living creatures function, habitat of waterbirds and benthic macroinvertebrates, amphibians and odonate are spawning ground and habitat, it was affecting the vegetation ecosystem based on wetlands. In order to maintain the diversity of wildlife, it was important to maintain smooth water supply and water level. A stable food chain will be formed and the Uldae wetland biodiversity will be abundant by establishing the relationship between the species of Uldae wetland, which is abandoned rice paddy field, and the habitat environment favored by species belonging to the ecosystem stepwise linkage. The ecological characteristics of the Uldae wetlands and the relation between the species were analyzed and the environmental conditions were reflected in the planning and management plan of Uldae wetland ecology.

Expression of the Heat Shock Protein 70 Gene and External Developmental Traits of Two Bivalvia Species, Crassostrea gigas and Mytilus galloprovincialis, under Aquaculture Environments (사육환경에 따른 이매패류 (Crassostrea gigas, Mytilus galloprovincialis)의 외부형질 성장과 Heat Shock Protein 70 유전자 발현)

  • Environmental changes exert harmful effects on organisms inhabiting coastal regions. These changes are also associated with reduced production in aquaculture farms. In this study, we investigated internal and external responses of two Bivalvia species (Crassostrea gigas and Mytilus galloprovincialis) in Gamak Bay under stressful environmental conditions in aquaculture farms. We investigated external responses such as weight, size, and environment exposure time, and analyzed the expression of the HSP70 gene. C. gigas HSP70 gene expression level was significantly high in the C3 aquaculture farm site, but the weight and size of C. gigas were high in the C2 aquaculture farm site. The response of C. gigas HSP70 mRNA was associated with the environmental exposure time in each aquaculture farm. Expression of M. galloprovincialis HSP70 gene was found to be significantly higher in the M2 aquaculture farm site than in the M1 site, whereas the weight of M. galloprovincialis was observed to be higher in the M1 site. The size and environmental exposure time of M. galloprovincialis were similar between M1 and M2 sites. In addition, HSP70 sequences of C. gigas and M. galloprovincialis showed high similarity with that of another marine species. According to our results, there were differences in internal responses following environmental stress in aquaculture farms, with respect to HSP70 gene expression. The results suggest that the HSP70 gene is a useful molecular indicator for monitoring stress responses in Bivalvia species in the field.

A Study on the Hand Hygiene of Food Handlers of Food Court and Cafeteria in University Campus (대학 구내 휴게음식점 종사자의 손 위생관리에 관한 연구)

  • This study was performed to investigate awareness of hand washing, hand washing behavior, and the levels of indicator microorganisms on hands of food handlers who work in the food court and cafeteria of a university campus. The three methods used were questionnaire survey by interview, direct observation in restrooms, and microbiological examination according to the Food Code of Korea. A positive attitude toward hand washing compliance was reported by the responded food handlers; however, improper hand washing and poor hand hygiene of the food handlers were recognized by the unnoticed direct observation. Significant differences were found between the questionnaire survey and the direct observation (p < 0.05) in hand washing compliance after using the toilet, duration of hand washing, use of hand washing agent, washing different parts of the hands, hand-drying method, temperature of water, and method of turning off the water. Samples taken from their hands before work showed higher level of standard plate count, total and fecal coliforms, and Escherichia coli than those taken after washing with water. After washing hands with antiseptic liquid soap, the bacterial populations including Staphylococcus aureus on hands were dramatically reduced. This study indicates that there is a remarkable difference between the food handlers' awareness of hand washing and their hand washing behavior. Poor hand washing compliance and hand hygiene were indicated by the positive results of total and fecal coliforms, E. coli, and S. aureus on hands of some food handlers. The findings of this study suggest that the hand hygiene of the food handlers need to be improved. More training/education on hand washing and hand hygiene of the food handlers should be necessary.

Analgesic effect of acupuncture applied to $SI_6$ in a rat model of neuropathic pain (흰쥐의 신경병증성(神經病症性) 통증(痛症) 모델에서 양로(養老) 자침(刺鍼)의 진통효과(鎭痛效果))

  • Objectives : The usage of acupuncture has gained popularity for certain chronic pain conditions. However, the efficacy of acupuncture in various diseases has not been fully established and the underlying mechanism is not clearly understood. In the present study, the effect of electroacupuncture (EA) applied to yangno$(SI_6)$ on the neuropathic pain was examined. Methods : A common source of persistent pain in human is a neuropathic pain. Neuropathic pain was induced by tight ligation of L5 spinal nerve. When rats developed pain behaviors, EA was applied for 30 min. under enflurane anesthesia with repeated train stimuli at the intensity of 10X of muscle twitch threshold. The foot withdraw latency of the hind limb was measured for an indicator of pain level after each manipulation. Results : EA increased the mechanical threshold of the foot in the rat model of neuropathic pain significantly for the duration of 1 hr. suggesting a partial alleviation of pain. EA applied to SI6 point produced a significant improvement of mechanical sensitivity of the foot lasting for at least 1 h. However, $ST_{36}$ point did not produce any significant increase of mechanical sensitivity. The improvement of mechanical threshold was interpreted as an analgesic effect. The analgesic effort was specific to the acupuncture point since the analgesic effect on the neuropathic pain model could not be mimicked by EA applied to a point, $ST_{36}$. In addition, this analgesic effect of EA is mediated by a adrenergic mechanism of descending control of spinal cord from the brain. Conclusions : The data suggest that EA produces a potent analgesic effect on the neuropathic pain model in the rat; and 2) that EA-induced analgesia is mediated by a adrenergic mechanism of descending control in a point specific manner.

Effects of Antibacteria and Adhesive Inhibition of Scutellaria baicalensis Extract on Streptococcus mutans (황금(Scutellaria baicalensis) 추출물에 의한 Streptococcus mutans의 항균 및 부착억제 효과)

  • The natural products are used to be development of new antibacterial substances against human pathogenic bacteria. Adherence to the tooth surface by S. mutans is an important step in initiation of dental caries. This study was to examine antibacterial activity and anti-adhesive effect of Scutellaria baicalensis extract against S. mutans. Extracts of S. baicalensis were tested for antimicrobial activities by paper disc methods and radial diffusion assay methods, and bacterial adherence assay using 3 type of hydroxyapatite. The antibacterial level of ethyl acetate extract, IPK-3 on the growth of S. mutans was 125 mg/ml of minimum inhibitory concentration (MIC). The maximum growth of S. mutans in medium added with IPK-3 extract (50 mg/ml) was delayed to 30 hr, while the highest at 24 hr in control medium. The pH values of the control medium was 5.63 at 18 hr, but the media supplemented with IPK-3 extract was pH 6.50 at 12 hr. In adhesive inhibition assay, S. mutans was labelled with the fluorescent indicator DAPI and measured with fluorescence microscope. Adhesion of S. mutans on hydroxyapatite beads was inhibited by IPK-3 extracts. These results suggest that S. baicalensis extract can be used as an effective material for antibacterial activity and adhesive inhibition against S. mutans.

Correlation between the Illuminance and the Flowering and Leaf Growth of Trees at Night - In Case of Downtown from Jamsil Station to Olympic Park, Seoul - (도심 내 야간의 조도와 봄철 수목 개화 및 잎 생장 간 상관관계 - 서울 잠실역에서 올림픽공원 구간을 대상으로 -)

  • In this paper, according to the illuminance at night with artificial lighting in downtown Seoul, trees flowering and leaf growth rate was performed as a case study to examine whether there is a difference. The illuminance was measured at 78 points on a total of 26 points, the range of illuminance were divided into three groups considering land use, Group A (plots 1-7) were 4.90 ~ 112.50 lx, Group B (plots 8 to 18) were 0.45 ~ 42.40 lx, group C (plots 19 to 26) were 0.28 ~ 22.10 lx. According to the One-way ANOVA on illuminance difference, the survey groups were classified into three groups. To survey the ratio of the flowering Prunus yedoensis and Rhododendron spp. and to survey the ratio of leaf growth Zelkova serrata, Ginkgo biloba, and Acer palmatum each 30 individuals were investigated. Rhododendron spp. to study the ratio of the flowering and Acer palmatum to study the ratio of leaf growth were determined to be useful as indicator woody species according to the correlation. The higher illuminance of night lighting is, the faster flowering and leaf growth of trees will be by correlation between the mean illuminance of each group and the flowering and leaf growth of trees. In the future, should be made a more detailed study of how much affect to the trees with some impact on the level of light pollution at night lighting.

A Landscape Ecological Model for Assessing the Korean Urban Forests (도시숲 평가를 위한 경관생태학적 모형 개발)

  • The purpose of this study is to verify the effectiveness of the biotope model in applying and developing Korean urban forests. We found that there are 17 biotope assessment indicators, including forest layer structure, site conditions, ratio of broad-leaved trees, species richness, etc. In terms of correlation analysis between indicators, the stand ages and the period of space formation have the highest relativity(coefficient 0.684). On the other hand, indicators that have negative relativity are layer structure and risk, with a coefficient of -0.412. Ten models were developed for the multiple regression analysis. 10 variables(site conditions(X2), ratio of broad-leaved trees(X3) and so forth except layer structure(X1), species richness(X4)) were found to have a 95% significance level The results from comparing the regression model and adding-up estimation matrix, the most accurate one was Model 3, which has a 91.7% out of the 10 models. However more monitoring will be needed to improve the accuracy of models for the Korean urban forests in future.

Studies on health management and nutritional evaluation by milk components analysis in dairy cows III.   • Milk urea nitrogen (MUN) determination is being used an indicator of the protein-energy balance in dairy herds. A faulty balance can be corrected to optimize milk production and animal health. This parameter is regarded as a potential tool to evaluate suboptimal feeding practices and reproductive disorders. Therefore, the purpose of this study was to investigate the response of milk composition by regular feeding analysis and to compared the relationship between MUN and milk protein(MP) and fertility at the insemination period in Holstein dairy cows. Total of 355 artificial insemination (AI) for 150 Holstein cows in the herd were used to examine the relationship between MUN and MP content and conception rate. The AI occured for the cows 50 to 150 day in milk, and MUN and MP concentration were determined using automated infrared procedures. The mean${\pm}$standard deviation of MUN and MP concentration in the herd were $15.6{\pm}2.1mg/dl$ and $3.23{\pm}0.38%$, respectively. MUN contents of bulk milk were increase by elevated crude protein intake. The conception rate was lower in the cows in which the level of MUN was lower than > 8.0mg/dl (10.0%) or > higher than 25mg/dl (15.4%) relative to the cows in MUN content of 12.0~17.9 mg/dl (36.7%) at the time of insemination. Also, lower MP than 3.0% or higher MP than 3.25% were associated with a lower conception rates. Consequently, MUN and MP analyses may be used serve as a monitoring tool of protein and energy nutritional balance to improve reproduction efficiency in Holstein dairy cows.

The Feasibility of Cathepsin B Level in Preoperatively Screening Patients with Thyroid Cancer and Nodular Hyperplasia (갑상선암 및 결절성 증식증 환자의 수술전 스크리닝을 위한 cathepsin B의 발현 양상)

  • To evaluate the feasibility of cathepsin-B levels in preoperatively screening patients with thyroid cancer, we assigned these patients to either the thyroid cancer group (n=32) or the nodular hyperplasia group (n=7). Five healthy volunteers served as controls (n=5). We quantified cathepsin-B expressions in cancerous lesions with follicular carcinoma and hyperplastic lesions with nodular hyperplasia, and compared the degrees to those of normal thyroid tissue, which was obtained from matched contralateral lobe. The activity of serum cathepsin B was significantly higher in patients with thyroid carcinoma ($284.87{\pm}79.32$, ${\times}10^{-2}\;mU$) and those with nodular hyperplasia ($255.45{\pm}95.68$, ${\times}10^{-2}\;mU$) than compared to normal control ($168.94{\pm}15.10$, ${\times}10^{-2}\;mU$) (p<0.05). Based on the results of immunoassay, the concentrations of cathepsin B in the thyroid cancer group ($15.50{\pm}7.86\;ng/ml$) and the nodular hyperplasia group ($17.64{\pm}7.49\;ng/ml$) were higher than those of the control group ($4.85{\pm}0.61\;ng/ml$). The degree of cathepsin-B mRNA expression was significantly higher in cancerous or hyperplastic lesions than normal thyroid tissues from matched contralateral lobe with follicular carcinoma or non-neoplastic thyroid disease. Our results indicate that the activity of serum cathepsin B is a useful indicator in screening patients with nodular hyperplasia or neoplastic thyroid disease and it may be involved in the abnormal proliferation of cells.
