Lactic acid bacteria (LAB) are widespread in a variety of environments including fermented dairy products, gastroinstetinal and urogenital tracts of human and animals, plant, soil and water. Leuconostoc mesenteroides DB3 was detected by the strongest antibacterial activities among 24 Leuconostoc strains isolated from Camellia japonica flowers. Acid tolerance of L. mesenteroides DB3 existed up to pH 2.5, but the resistance did not show at pH 2.0, which relatively excellent acid resistance existed. Bile acid tolerance was very stable within the test range to 1.2%. L. mesenteroides DB3 exhibited the optimal growth at 30℃, and showed a slight slow growth when compared with L. mesenteroides KCTC3505, which reached a stationary phase at 18 hr. The pH was changed along with the growth curve, but was maintained above pH 3.98. L. mesenteroides DB3 had higher initial antibacterial activities when compared to L. mesenteroides KCTC3505, but it showed similar activities with the standard strain after the latter part of the logarithmic growth phase. Although lactic acid production in L. mesenteroides DB3 was induced by lower amount in the initial part to the standard strain, it was exhibited by similar amounts after the late logarithmic growth phase. Muicin adhesion of L. mesenteroides DB-3 maintained superior to L. mesenteroides KCTC3505. Both strains showed excellent emulsification ability for kerosene. In summary, we evaluate that L. mesenteroides DB-3 has a high potential for application as probiotics owing to its excellent antibacterial activity, acid resistance, bile acid resistance, and muicin adhesion.

This study was focused on developing and obtaining a kimchi starter for use in commercial kimchi production. Kimchi varieties made with selected starters are of high quality, have high levels of mannitol, and extended shelf life. The starters were screened for properties such as mannitol production, low gas/acid production, and acid resistance. Finally, kimchi fermentation testing was performed using selected LAB starters. Kimchi samples were prepared with lactic acid bacteria (LAB) starters, including Leuconostoc mesenteroides PBio03 and Leuconostoc mesenteroides PBio104. The LAB starters are isolated from kimchi and can grow under pH 3.0 and low temperature conditions of 5℃. Four kimchi samples were fermented and stored for 28 days at 5℃. The kimchi samples made with starters (PBio03 and PBio104) had better quality (production of mannitol and maintenance of heterofermentative LAB dominance) than the non-starter kimchi samples. In the starter kimchi, Leu. mesenteroides was the dominant LAB, comprising 80% and 70% of total LAB counts at 7 and 21 days, respectively. Mannitol content of the kimchi with Leu. mesenteroides PBio03 was 1,423 ± 19.1 mg/100 g at 28 days, which was higher than that of the non-starter kimchi sample (1,027 ± 12.2 mg/100 g). These results show the possibility of producing kimchi with improved qualities using Leu. mesenteroides PBio03 and PBio104 as starters.
To investigate the effects of Lactobacillus curvatus and Leuconostoc mesenteroides on suan cai (pickled Chinese cabbage) fermentation, L. curvatus and/or Ln. mesenteroides were inoculated into suan cai. Physicochemical indexes were measured, and the microbial dynamics during the fermentation were analyzed by Illumina MiSeq sequencing and quantitative polymerase chain reaction (qPCR). The results showed that inoculation with lactic acid bacteria (LAB) lowered the pH of the fermentation system more rapidly. The decrease in water-soluble carbohydrates in the inoculated treatments occurred more rapidly than in the control. The LAB counts in the control were lower than in other inoculated treatments during the first 12 days of fermentation. According to the Illumina MiSeq sequencing analyses, Firmicutes, Proteobacteria, Bacteroidetes, Actinobacteria, Cyanobacteria, Fusobacteria, and Verrucomicrobia were present in the fermentations, along with other unclassified bacteria. Generally, Firmicutes was predominant during the fermentation in all treatments. At the genus level, 16 genera were detected. The relative abundance of Lactobacillus in all inoculated treatments was higher than in the control. The relative abundance of Lactobacillus in the treatments containing L. curvatus was higher than in the Ln. mesenteroides-only treatment. The relative abundance of Leuconostoc in the Ln. mesenteroides-containing treatments increased continuously throughout the fermentation. Leuconostoc was highest in the Ln. mesenteroides-only treatment. According to the qPCR results, L. curvatus and/or Ln. mesenteroides inoculations could effectively inhabit the fermentation system. L. curvatus dominated the fermentation in the inoculated treatments.
Studies on the optimum conditions for dextran production and the properties of dextransucrase (DS) were performed with Leuconostoc mesenteroides from Sikhae and Leuconostoc mesenteroides NRRL B-512(F). Dextransucrases were partially purified by lyophilization of the culture supernatant and subsequent gel chromatography on Bio-Gel A-5(m). The storage stabilities of Sikhae DS and B-512(F) DS were decreaed by the addition of dextranase. The optimum conditions for the enzyme stability were pH 5 and below $30^{\circ}C$. The B-512(F) DS lost the activity at pH 4, while Sikhae DS had 30% of the activity at the same pH. The activity of DS was decreased by EDTA. confirming the metalloprotein character of the enzymes, and was restored by the addition of calcium ions. Concanavalin A completely removed the activity of DSs, confirming the glycoprotein character of the enzymes.
Two lactic strains, Leuconostoc mesenteroides Lu5 and Pediococcus pentosaceus P1 isolated from Kimchi, were used to determine the optimum conditions for protoplast formation and regeneration. The maximum protoplast formation rate was obtained with both strains at early exponential growth phase and decreased rapidly during growth phase. For P. pentosaceus P1, 30 $\mu$g/ml of lysozyme treatment was sufficient to obtain over 90% of protoplast formation and 300 $\mu$g/ml for L. mesenteroides Lu5, showing great difference in sensitivity of these strains to lysozyme. For both strains, best results were obtained at pH 7, using 0.5 M sucrose as osmotic stabilizer. For regeneration of protoplast, the highest regeneration rate was obtained after 15 minutes of lysozyme treatment and declined drastically with prolonged digestion.
Four strains of lactic acid bacteria which produced bacteriocins inhibitory to Listeria species were isolated from Kimchi, and were identified as Leuconostoc mesenteroides subsp. mesenteroides (2 strains), Leuconostoc paramesenteroides and Pediococcus pentosaceus. The bacteriocins produced by the isolates inhibited all of the Listeria monocytogenes strains tested, but L. denigrificans 28 and L. welchimeri 89 were not inhibited by the bacteriocin produced by the Leu. paramesenteroides isolate. The bacteriocin produced by the P. pentosaceus isolate was more inhibitory against sensitive strains and showed broader spectrum of antimicrobial activity than those produced by other isolates. The bacteriocins produced by Leuconostoc isolates were sensitive to pronase E treatment, but that produced by the P. pentosaceus isolate was not completely inactivated. The bacteriocins produced by all of the isolates were not sensitive to catalase, ${\alpha}$-amylase and lysozyme and heat (30 min at $100^{\circ}C$) treatments.

In order to study the addition effect of Leuconostoc mesenteroides and nisin on Escherichia coli and lactic acid bacteria during fermentation and storage of Kimchi, Kimchi was stored at 4$^{\circ}C$ for 7 days and then in-creased the temperature to $25^{\circ}C$. Lactic acid content on Kimchi fermentation at 4$^{\circ}C$ was maintained initial content which was increased upto 0.9% and lactic bacteria was also increased after switching to $25^{\circ}C$. E. coli, on the other hand, was a little decreased from the initial level, but a significant decrease was found for the those Kimchi of Leuconostoc muenteroides added and nisin treated group when the fermentation temperature was switched to $25^{\circ}C$.
숙성 중인 김치에서 생육과 산 생성이 우수한 유산균을 분리하여 API 50 CHL kit로 확인하고 16S rDNA의 염기 서열을 분석한 결과 Leuconostoc mesenteroides로 동정되어 Leuconostoc mesenteroides KC51로 명명하였다. 대두에 10배의 증류수를 가하여 제조한 두유에 KC51 균주를 접종하고 $30^{\circ}C$에서 정치 배양한 결과, 생균수는 접종 후 9시간까지 급격히 증가하였으며 그 이후 완만하게 증가하여 배양 12시간에는 $2.70{\times}10^9$ CFU/g까지 증가하였다. 적정산도는 배양 12시간에 0.35%까지 증가하였으며,pH 는 pH 4.52까지 감소하였다. 이때 유기산 농도는 젖산과 초산의 함량이 각각 0.56%와 0.10%로 분석 되었다. 두유 발효액에서 Leuconostoc mesenteroides의 증식에 의한 점도의 증가는 관찰되지 않았으며, 관능적으로도 신맛을 제외한 항목에서 두유와 유사하였다. 또한 $4^{\circ}C$에서 두유 발효액은 2주일의 저장기간 동안 pH, 적정산도와 생균수의 변화는 미미하였다.

Thirty four Leuconostoc strains originated from kimchi were subjected to analyse their genetical similarity by using RAPD-PCR. At the 0.5 level of similarity, L. mesenteroides subsp. mesenteroides and L. mesenteroides subsp. dextranicum. formed 3 and 2 clusters, and each L. lactis and L. paramesenteroides strain was clustered in one, respectively. Every type strain tested as a control, belongs to the cluster or one of the clusters of the corresponding species or subspecies as the isolates from kimchi, respectively. However the similarity between a and b cluster was less than 0.2 and c cluster is even less similar to both of them (a and b). L. mesenteroides subsp. mesenteroides strains from kimchi have shown a varaiety of RAPD patterns in spite of morphological and physiological similarities.
Leuconostoc mesenteroides can be used to produce mannitol by fermentation, but the mannitol productivity is not high. Therefore, in this study we modified the chromosome of Leuconostoc mesenteroides by genetic methods to obtain high-yield strains for mannitol production. In this study, gene knock-out strains and gene knock-in strains were constructed by a two-step homologous recombination method. The mannitol productivity of the pat gene (which encodes phosphate acetyltransferase) deletion strain (${\Delta}pat::amy$), the fk gene (which encodes fructokinase) deletion strain (${\Delta}fk::amy$) and the stpk gene (which encodes serine-threonine protein kinase) deletion strain (${\Delta}stpk::amy$) were all increased compared to the wild type, and the productivity of mannitol for each strain was 84.8%, 83.5% and 84.1%, respectively. The mannitol productivity of the mdh gene (which encodes mannitol dehydrogenase) knock-in strains (${\Delta}pat::mdh$, ${\Delta}fk::mdh$ and ${\Delta}stpk::mdh$) was increased to a higher level than that of the single-gene deletion strains, and the productivity of mannitol for each was 96.5%, 88% and 93.2%, respectively. The multi-mutant strain ${\Delta}dts{\Delta}ldh{\Delta}pat::mdh{\Delta}stpk::mdh{\Delta}fk::mdh$ had mannitol productivity of 97.3%. This work shows that multi-gene knock-out and gene knock-in strains have the greatest impact on mannitol production, with mannitol productivity of 97.3% and an increase of 24.7% over wild type. This study used the methods of gene knock-out and gene knock-in to genetically modify the chromosome of Leuconostoc mesenteroides. It is of great significance that we increased the ability of Leuconostoc mesenteroides to produce mannitol and revealed its broad development prospects.
