INVESTIGATION OF EQUINE HEMATOLOGICAL CONSTITUENTS IN CENTRAL TAIWAN. I. DISTRIBUTION OF THE BLOOD CELL PARAMETERS AND THE BIOCHEMICAL COMPOSITIONS OF SERUM

  • A total of 109 heads of horses and ponies from eight horseback riding clubs nearby Central Taiwan were investigated to evaluate the blood parameters and the biochemical compositions of serum for the documentation of clinical pathological diagnosis and exercise physiology. Blood samples were collected from the jugular vein of resting horses. The sex difference in the blood traits were compared both in horses and in ponies. Results shows that total plasma proteins (PP) and hematocrit (PCV) were found higher in male horses than in females (p < 0.05). The sexual effect also exertes a significant influence on the leucocyte (WBC) count, but not on the erythrocyte (RBC) concentration. According to the differential counts of leucocytes, the number of monocytes and lymphocytes was higher in the male pony than that of in male horse. A close relationship Between the erythrocyte sedimention rate (ESR) and the other blood parameters were found especially in PCV, RBC concentration, and plasma protein level. The average ESR observed at 60 minutes were $21.80{\pm}21.87mm$, $39.50{\pm}18.90mm$ and $43.73{\pm}17.89mm$ in stallions, geldings, and mares, respectively. Most of the biochemical components of horse serum detected were distributed in normal ranges, although some of the items show a great variation in such a large sample size.

The Effects of Hominis Placenta Hydrate (HPH) on the Bone Marrow Failuer Induced by 5-Fluorouracil

  • Aim: To examine the efficacy of Hominis Placenta Hydrate (HPH) on the hemopoiesis in a myelosuppression model system. Methods: Mice were injected with 5-Fluorouracil (5-FU) intraperitoneally and were administered with 200 mg/kg and 1000 mg/kg of HPH. Peripheral blood was analyzed at 5, 9, and 13 days. Histopathologic examination of bone marrow was performed at 5 days after 5-FU injection. The expression of cytokine involved in hemopoiesis was examined by using ELISA kit. Results: The hematology data demonstrated that the treatment of all the agents augmented monocytes and leucocytes counts in the peripheral blood WBC and platelet in HPH treated group were significant increased compared with control group. Also, cell numbers of RBC and Hb were restored. In HPH treated group, expression of IL-3, GM-CSF was significant increased But not TPO. Conclusions: Based on the above results, it is suggested that Hominis Placenta Hydrate is an effective remedy for the bone marrow failure and myelosuppression occurring during chemotherapy.

Experimental Studies on Induced Traumatic Pericarditis and Myocarditis in Ruminants (반추수(反芻獸)의 창상성심낭심근염(創傷性心囊心筋炎)에 관(關)한 실험적연구(實驗的硏究))

  • This study was performed to obtain an accurate methods of diagnosis in early stage of traumatic pericarditis and myocarditis in ruminants. Korean native goats were used as experimental animal and traumatic pericarditis and myocarditis were induced by surgical intervention. The changes in clinical signs, hematological values, blood enzymes and electrocardigrams were observed for 14 days after operation. The results obtained were as follows: 1. Body temperature in experimental group of induced traumatic pericarditis and myocarditis was slightly increased than control group that was rumenotomized. Heart rate was increased in both groups but the increased rate was noticed remarkably in experimental group. There was no change in respiratory rate in experimental and control group. 2. Total leucocytes counts and neutrophil percentage maintained higher level after operation and lymphocytes percentage noticed reversed proportion to neutrophil in both groups. 3. Serum total protein concentration and alkaline phosphatase activities after operation showed a little lower levels than these values prior to operation. Serum glutamic oxaloacetic transaminase and lactic dehydrogenase activities in control group showed slight increase through all the experimental period, while in experimental group these activities were markedly elevated and an increase in serum lactic dehydrogenase activities was more remarkable comparing to serum glutamic oxaloacetic transaminase. 4. Electrocardiograms in control group did not show any abnormalities except for slight increase in heart rate. In experimental group the amplitude of QRS complex was increased in duration of experimental period. The transference from QRS complex to T wave was uncertain and S-T segment moved upward obliquely through electrical zero line.

Effect of Bupleurum falcatum extract on cellular immune responses (시호 추출물이 세포성 면역반응에 미치는 영향)

  • Bupleurum falcatum has been used for treatment of inflammation, jaundice, influenza and hepatitis as a traditional orient folk medicine. This experiment was carried out to evaluate the effect of B falcatum extract on cellular immune responses in vivo and in vitro. Antigen binding cell(ABC) assay, antibody production, Arthus and delayed-type hypersensitivity(DTH) reaction against sheep erythrocytes(SRBC) were very depressed in B falcatum extract treated group in vivo. The growth of Staphylococcus aureus in brain heart infusion(BHI) broth containing B falcatum extract was remarkably inhibited. Otherwise, that of Salmonella typhyimurium was not significantly increased in vitro. When B falcatum extract pretreated mice were intraperitoneally(IP) injected S typhimurium and S aureus, respectively, the number of bacteria in peritoneal exudates were time dependent declination compared with those of control, and the weight of spleen and the number of macrophage migration into peritoneal cavity have no difference from those of untreated control. B falcatum extract gradually increased phagocytic activities of peritoneal macrophage against Candida albicans time and dose dependently, and was not significant production of migration inhibiotory factor(MIF). But migration abilities of normal leucocytes in B falcatum extract pretreated group were decreased dose dependently. When B falcatum extract was IP administered, these data indicate that B falcatum extract increases level of serum coticosterone. Therefore, B falcatum extract was indirectly mediated in immune system by serum coticosterone having relation to immunosuppression. These results lead to the conclusion that B falcatum extract acts as a trigger or regulator of cellular immune responses in immune system.

Application of zebrafish as a model for evaluation of vaccine efficacy against Philasterides dicentrarchi (Ciliphora: Scuticociliatia)

  • Zebrafish was firstly applied to an experimental model for scuticociliatosis caused by Philasterides dicentrarchi, a facultative parasitic ciliate in cultured marine fish. The susceptibility of zebrafish to infection of P. dicentrarchi was assessed by intraperitoneal injection of the ciliates, which produced typical symptoms of scuticociliatosis and significant mortality. The potential use of zebrafish as a model to evaluate the vaccine efficacy against scuticociliatosis was analyzed by immunization of zebrafish with the ciliates lysate. Furthermore, the effect of different adjuvants, such as Quillaja saponin (QS), Montanide, and Freund’s incomplete adjuvant (FIA) on the protective efficacy of the vaccine was investigated. Groups of zebrafish injected with QS or Montanide alone showed higher survival of fish against challenge test compared to control fish. The results suggest that adjuvant-mediated enhancement of innate immune responses play important roles in protection of fish against scuticociliatosis. The considerably high survival in the fish immunized with the antigen alone indicates that the ciliate lysate itself is highly immunogenic to zebrafish, which can elicit protective immune responses. The protective potential of the antigen, ciliate lysate, was enforced through combined administration with adjuvants including QS, Montinide and FIA. No or low mortalities in the groups of fish immunized with the antigen plus adjuvants suggests that the adaptive immune responses of zebrafish might be accelerated by the adjuvants or the protective potential of the antigen and adjuvants might synergistically interact. In spite of several shortcomings such as difficulties in sampling of serum and leucocytes enough to routine immunological analyses, zebrafsih might be the most convenient experimental animal for scuticociliatosis.

Enzymatic Determination of Somatic Cells by Using Transparisation in Raw Milk

  • The transparisation technology for milk and milk products could be applied widely and very importantly to various determination because transparisation can economize the cost and increase with precision in the milk payment system. Component of butanone or Triton in transparisation solvent would inhibit the growth of bacteria and method. Enzymatic determination of leucocytes were proposed to evaluate milk quality as mastitis in the milk payment system, this can be easily applied to simplify automation of the determation with the lowest investment cost in milk pay system. The significance of this technique, it can be used in the quality control of raw milk and milk products, milk payment system, and programming of national dairy project. Transparisation technology is used in somatic cell counting by enzymic methods. The range of deviation for this method is 16% in 74 samples. But the deviation is increased to 20% when the Infoss method is used. It is affected by the percentage of epithelial cells and white blood cells in somatic cells from different animals and the stages of aging. NAgase activity has an obvious correlation with white-blood cells in milk. In the case of mastitis the white-blood cells is 90-95% in somatic cells in milk, it is showing greater precision in measuring the state of mastitis. In conclusion the enzymic method of somatic cell counting is a relatively simple and easy method of measurement and can be easily practiced. And the importance of this method is also worth utilizing for indirect counting of Somatic cells by use of synthetic substrates to NAgase. In the future, with the further development of the research in this field, it will b possible to automatize the measurement.

Analysis of Phagocytosis and Birefringence in the Peritoneal Cells of the Rat, with Special Regard to the Mast Cells (흰쥐의 복강내 세포, 특히 비만세포의 식작용 및 복굴절성에 관한 분석)

  • The phagocytic petencies and birefringences of peritoneal mast cells of rats treated with particular dyes (neutral red, toluidine blue, pyronin, methylene blue, alcian blue, trypan blue, carmine, orange G, aniline blue, Janus green B, and India ink) were analyzed by means of phase contrast microscopy and polarizing microscopy. In addition, cytomorphic effects of the dyes on the peritoneal mast cells were also discussed. Phagocytic activities or ingestion of the dye particles were not observed in most cases, except for the India ink group. Hardly a macrophage appeared without some dark particles which were ingested or phagocytosed. Trypan blue and aniline blue produced very weak birefringence in the cytoplasm of mast cells but the rest did not produce even in the acid medium (neutral red and toluidine blue). The short and slender ectoplasmic processes of the mast cells and the leucocytes were also found in certain groups. The cytomorphic effects of the dyes on the mast cell were slight and variable.

Biological and Physicochemical Properties of Canine Parvovirus lated from the Dogs with Diarrhea (설사증 나환견으로 부터 분리한 Canine Parvovirus의 성상에 관한 연구)

  • From 1988 to 1989, 8 strains of canine parvovirus-2 (CPV-2) were isolated from the fecal specimens from the dogs that were clinically diagnosed as canine parvoviral enteritis in the veterinary hospitals located in the regions of Taejon and Chungbuk province. Studios on biological and physicochemical properties for the isolates were carried out. The results obtained by experiments are summarized as follows. 1. Among 62 fecal samples collected from the dogs with enteric diseases, 24 (38.7%) showed the haemagglutinating activity against porcine erythrocyte, ranging from 16 to 16, 384 of HA titers. 2. When 8 fecal specimens with high HA titer over 1, 000 were inocultated into CRFX cells, intranuclear inclusion bodies were obseverd in all of eight specimens, of w)lick three specimens showed cytoplasmic inclusions concurrently with the intranuclear inclusion bodies. 3. In study on species-specificity of haemagglutinating activity of the isolates, TJ-89-1 and TJ-89-2, it was found that the isolates revealed the highest haemagglutinating activity with porcine erythrocytes, showing the relatively lower haemagglutination titers with the erythrocytes from cat and rabbit. None of erythrocytes from other animals reacted with the isolates. 4. By the cross-haemagglutination inhibition test of the Isolates with reference viruses and sera, the Isolates were evidently identified as the strains of canine parvovirus-2. 5. In Physicochemical property test, it was evident that the isolates were stable in, lipid solvent, pH and heat treatment at $56^{\circ}C$ for 30 min. and contain DNA genome. 6. When seven puppies were inoculated intraorally with the isolate at HA titer of 8, 192, all of the puppies showed the symptoms of anorexia, vomiting, diarrhea and died at the 5th to 10th days post inoculation(pi). The fecal samples from all of the puppies revealed significantly high HA titers afterward the 5th days pi. Body temperature and the number of total leucocytes were slightly increased at the early stage of infection. but extremely decreased at the stage of collapse. HI titers of the sera started to increase at the 2nd to 3rd days pi reaching 512 to 1, 024 at the 4th to 5th day pi.

  • Inasmuch as a great variety of methods for the leukocyte differential counting have been described, it may be expected that actual counts obtained considerably vary according with the particular methods applied to the enumeration of differentiated leukocyes. An attempt, therefore, was undertaken to compare the values measured by five authentic methods commonly in use with those obtained by thirteen methods devised by the author. The results seemed to indicate that relatively reliable method is to count, end to end, all leucocytes on fields looted on the central horizontal axis of the film and two adjacent lines parallel to the central line and quartering the film.

A Study about the Effect of Antigrowth Virus (Third Virus of Swine) on Small Animals (Antigrowth Virus (Third Vhird Virus of Swine)의 소동물에 대한 연구)

  • Durlng various experiments and investigations with antigrowth virus, I studied especially the infective and antiinfective process of it in small animals, and have got following results: 1. The rabbits are easily infected with this virus, having following five stages; incubation period of 2 to 3 days, stage of fever for 2 to 4 day, transient anemic stage, transient fcteric stage and stage of lasting iron-precipitation in the spleen. 2. According to the appearance of icteruis, I have divided the rabbits into following three groups, the first gorup, weighting less than 1000g, died after appearance of icterus, the second group, weighting 1000 to 1500 g. recoy ered after appearance of icterus, and the third group, weighting more than 1500 g. recovered without the appearance of jaundice. 3. In hematological study, marked decrease in R. B. C. and Hb were recognized but various leucocytes except lymphocytes slightly increased. 4. In pathological study, the spleen exhibited marked swelling with abundent blood and indistinct border between trabeculae and pulps at the initial stage of icterus and notable atrophy at the end stage of it. There are few iron-cells in the spleen histologically at the initial stage, but they increased in number with granuar appearance at the end stage. In the first group of rabbits above mentionel, the iron-cells increase suddenly with the death of animal and show diffuse precipitation of iron. The liver and indey have defferent aspects in proportion to icterus. If icterus appears, they show markel changes with turbidity and necrosis, but without icterus they present only slight changes. 5. The neutralining antibody is easily proved in the immunized rabbits for about one year. 6. The passive immunity has the defense immunability until 10\ulcorner 7. Gunia pigs are easily infected with this virus and get the immunity. On fowls this virus can be kept in the blood for about ten days by various inoculating methods, and the fowls obtain the defensive ability by reinoculation, but are unable to succeed this ability from generation to generation. 8. when young swine are infected with this virus, their growth are disturbed and result in atrophied swine. It is therofor presumed that the etiology of the antigrowth of these atrophied swine is similar morphological and functional changes observed in the above rabbits.
