Production of Phytol, an ACAT Inhibitor, from Callus Culture of Lettuce (Lactuca sativa L.) (상추 (Lactuca sativa L.) callus로부터 ACAT 억제 활성물질, phytol의 생산)

  • The possibility for mass production of phytol, inhibitory diterpene against ACAT (Acyl-CoA: Cholesterol acyltransferase) was investigated by using callus culture of lettuce. The callus were induced from lettuce cotyledon explants on MS medium containing 0.5 mg.L$^{-1}$ NAA after 4 week's culture. Adventitious roots were formed from the explants on MS medium containing 0.5 mg.L$^{-1}$ IBA or 1.0 mg.L$^{-1}$ NAA. Adventitious shoots and roots were emerged from the callus when the callus was transferred to MS medium containing auxin alone, or with cytokinin. The plant growth regulators and their concentrations for the organogenesis were 1.0 mg.L$^{-1}$ NAA, 0.1 mg.L$^{-1}$ BA, 0.5 mg.L$^{-1}$ NAA with 0.1 mg.L$^{-1}$ kinetin, or 0.5 g.L$^{-1}$ 2.4-D with 1.0 mg.L$^{-1}$ kinetin. Analyses of chlorophyll contents showed that chlorophyll contents were higher in morphogenic calli than in non-morphogenic calli. However, the chemical analyses of gas chromatography indicated that phytol contents were not proportionate to the chlorophyll contents of callus. The content of phytol was higher in callus than in lettuce cotyledon.ledon.

Analysis of the Microbiota on Lettuce (Lactuca sativa L.) Cultivated in South Korea to Identify Foodborne Pathogens

  • Lettuce (Lactuca sativa L.) is a major ingredient used in many food recipes in South Korea. Lettuce samples were collected during their maximum production period between April and July in order to investigate the microbiota of lettuce during different seasons. 16S rRNA gene-based sequencing was conducted using Illumina MiSeq, and real-time PCR was performed for quantification. The number of total bacterial was greater in lettuce collected in July than in that collected in April, albeit with reduced diversity. The bacterial compositions varied according to the site and season of sample collection. Potential pathogenic species such as Bacillus spp., Enterococcus casseliflavus, Klebsiella pneumoniae, and Pseudomonas aeruginosa showed season-specific differences. Results of the network co-occurrence analysis with core genera correlations showed characteristics of bacterial species in lettuce, and provided clues regarding the role of different microbes, including potential pathogens, in this microbiota. Although further studies are needed to determine the specific effects of regional and seasonal characteristics on the lettuce microbiota, our results imply that the 16S rRNA gene-based sequencing approach can be used to detect pathogenic bacteria in lettuce.

Effect of Temperture and Light on Growth of Leaf Lettuce(Lactuca sativa L. var. crispa cv. Cheongchima) in Hydroponics (배양액 온도 및 차광정도가 청치마 상추의 생육에 미치는 영향)

  • This experiment was conducted to develop a practical method to growth in hydroponics conditions. For this purpose, leaf lettuce (Lactuca sativa L., cv. 'Cheongchima') was cultivated under different temperature and light conditions in experimental fields of Sahmyook university from March, 1991 to May, 1996. The visual quality score(VQS), a quantitative parameter to evaluate harvested leaf lettuce quality. The leaf lettuce cultivated at $25^{\circ}C$ solution temperature showed the highest fresh weight pe, plant (59.7g/plant), but lowest visual quality score (5.2), while leaf lettuce cultivated at 15$^{\circ}C$ solution temperature showed lower fresh weight per plant (25.2g/plant) but highest visual quality score (7.2). In general, growth of leaf lettuce was retarded by low solution temperature, but quality was higher than those of other solution temperature. Different day/night temperature of nutrient solutions 20/2$0^{\circ}C$ and 25/2$0^{\circ}C$ was highest in yield, but leaf type was grow longer, and visual quality are inferior to low temperature. Yield was decreased as 10 and 25% at 60 and 80% shading treatment, respectively. Chlorophyll content was decreased by shading. but plant height and root length were increased of leaf lettuce growth with hydroponics.

Effect of Nutrient Solution Composition Modification on the Internal Quality of Some Leaf Vegetables in Hydroponics (수경재배시 양액 조성 처리가 몇가지 엽채류의 내적 품질에 미치는 영향)

  • This study was conducted to find out the change of infernal quality, such as vitamin C and nitrate contents in some leaf vegetables grown hydropoincally in different nutrient conditions. Pak-choi (Brassica camperistis L. spp. chinesis Jusl.), chungchima (Lactuca sativa L. var. crispa cv. Chungchima) and romaine (Lactuca sativa L. var. longifolia Lam.) lettuces were cultivated for 2 weeks in 4 different nutrient solutions, such as tap water; no-nutrient, added $NH_4$, discarded $NO_3$, and supplied Yamazaki' solution for lettuce as a control. The growth of leaf vegetables was not different among nutrient solution treatments except tap water. The nitrate content showed the highest in control, and followed by $+NH_4$ treatment, $-NO_3$ and tap water treatment, regardless of kind of vegetables. The vitamin C content in 3 different vegetables showed the opposite result against nitrate content so that the treatment that showing the highest vitamin C content was tap water in romaine and chungchima lettuces, and $-NO_3$ treatment in pak-choi. The vitamin C and the nitrate content showed high correlations; $r=-0.614^*$ in pak-choi, $-0.651^*$ in romaine lettuce, and $-0.804^{**}$ in chungchima lettuce.

Effect of Nitric Oxide on Paraquat-Tolerance in Lettuce Leaves (상추잎의 Paraquat 내성에 미치는 Nitric oxide의 영향)

  • The protective effect of nitric oxide (NO) on the antioxidant system under paraquat(PQ) stress was investigated in leaves of 8-week-old lettuce (Lactuca sativa L.) plants. PQ stress caused a decrease of leaf growth including leaf length, width and weight. Application of NO donor, sodium nitroprusside (SNP), significantly alleviated PQ stress induced growth suppression. SNP permitted the survival of more green leaf tissue preventing chlorophyll content reduction and of higher quantum yield for photosystem II than in non-treated controls under PQ exposure, suggesting that NO has protective effect on chloroplast membrane in lettuce leaves. Flavonoids and anthocyanin were significantly accumulated in the leaves upon PQ exposure. However, the rapid increase of these compounds was alleviated in the SNP treated leaves. PQ treatment resulted in lipid peroxidation and induced accumulation of hydrogen peroxide ($H_2O_2$) in the leaves, while SNP prevented PQ induced increase in malondialdehyde (MDA) and $H_2O_2$. These results demonstrate that SNP serves as an antioxidant agent able to scavenge $H_2O_2$ to protect plant cells from oxidative damage. The activities of two antioxidant enzymes that scavenge reactive oxygen species, superoxide dismutase (SOD) and catalase (CAT) in lettuce leaves in the presence of NO donor under PQ stress were higher than those under PQ stress alone. Application of 2-(4-carboxyphenyl)-4, 4, 5, 5-tetramethylimidazoline-1-oxyl-3-oxide (PTIO), a specific NO scavenger, to the lettuce leaves arrested SNP mediated protective effect on leaf growth, photosynthetic pigment and antioxidant systems. However, PTIO had little effect on lettuce leaves under PQ stress compared with that of PQ stress alone. The obtained data suggest that the damage caused by PQ stress is in part due to increased generation of active oxygen by maintaining increased antioxidant enzyme activities and SNP protects plants from oxidative stress. From these results it is suggested that NO might act as a signal in activating active oxygen scavenging system that protects plants from oxidative damage induced by PQ stress and thus confer PQ tolerance.

Expression of $\beta$-Glucuronidase (GUS) Gene in Transgenic Lettuce (Lactuca sativa L.) and Its Progeny Analysis (형질전환된 상추내에서 GUS 유전자의 발현 및 후대검정)

  • Agrobacterium tumefaciens LBA 4404 harboring binary vector pBI 121 was used for genetic transformation of lettuce(Lactuca Sativa L.). Optimal shoot regeneration from cotyledon explants was obtained in MS medium supplemented with 0.1mg/L NAA and 1.0 mg/L 2ip. In this condition, cotyledon explants were cocultivated with A, tumefaciens for 2 days, and then transferred to selection medium supplemented with 50 mg/L kanamycin and 500 mg/L carbenicillin. These explants were subsequently subcultured every 2 weeks on shoot induction medium. PCR analysis indicated that the GUS gene was stably integrated into the nuclear genome of lettuce. Histochemical analysis based on the enzymatic activity of the CUS protein showed that GUS activity was associated with vascular tissue in leaves and roots. Progenies of Ro plants demonstrated a linked monogenic segregation for GUS gene.

Effect of Bacillus subtilis S37-2 on Microorganisms in Soil and Growth of Lettuce (Lactuca sativa)

  • The present study evaluated the variations in soil microbial population of controlled horticultural land used for lettuce (Lactuca sativa) cultivation by their fatty acid methyl ester and chemical properties. We utilized four treatment groups, no treatment (NT), culture medium (CM), Bacillus subtilis S37-2 (KACC 91281P) ${\times}10^6CFU\;mL^{-1}$ (BS1), and Bacillus subtilis $S37-2{\times}10^7CFU\;mL^{-1}$ (BS2) and analyzed these variations throughout the before treatment and harvesting stage. The chemical properties such as pH, organic matter, available phosphate, and electrical conductivity in soils before treatment and harvesting stage showed no significant difference among the treatments. Total numbers of bacteria and microbial biomass C in soil treated with BS1 were larger than those of NT, CM, and BS2, whereas total number of fungi at the harvesting stage was significantly lower in the BS1 soil than in the NT and CM soils (P < 0.05). On basis of leaf length, leaf width, leaf number and leaf weight, the growth characteristics lettuce on the soil treated with BS1 and BS2 was faster than those of NT and CM soils. Yield of lettuce with treated BS1 and BS2 were 35% and 29% more than that of NT, respectively.

Effect of Short Term Cold Treatment to Rhizosphere on Nitrate Concentration in Lettuce Plant under Hydroponic Culture System (단기간 근귄 저온처리가 수경재배 상추의 질산태 질소 함량 미치는 영향)

  • Lettuce (Lactuca sativa L.) plants were grown under hydroponic system to characterize the diurnal change of nitrate concentration and nitrate uptake rate and to examine the effect of short term cold treatment to rhizosphere on nitrate concentration and uptake rate in lettuce plant. The nitrate concentrations in midrib were two times higher than those in leaf. Nitrate concentration in the shoot reached to minimum (8.7 mg-N/GDW) at 14:00 and, thereafter, increased continuously until 23:00. During 11:00$\sim$17:00, nitrate uptake by lettuce plant was maximum (4.8 mg-N/GDW-Root/hr). Short term cold treatment reduced nitrate concentration in the shoot by 14$\sim$18%, and nitrate uptake rate by 50$\sim$55%, respectively. These results showed that short term cold treatment before harvest could be applied for the purpose of reduction of nitrate concentration in the leaf under hydroponic culture.

Study on Development of Non-Destructive Measurement Technique for Viability of Lettuce Seed (Lactuca sativa L) Using Hyperspectral Reflectance Imaging (초분광 반사광 영상을 이용한 상추(Lactuca sativa L) 종자의 활력 비파괴측정기술 개발에 관한 연구)

  • In this study, the feasibility of hyperspectral reflectance imaging technique was investigated for the discrimination of viable and non-viable lettuce seeds. The spectral data of hyperspectral reflectance images with the spectral range between 750 nm and 1000 nm were used to develop PLS-DA model for the classification of viable and non-viable lettuce seeds. The discrimination accuracy of the calibration set was 81.6% and that of the test set was 81.2%. The image analysis method was developed to construct the discriminant images of non-viable seeds with the developed PLS-DA model. The discrimination accuracy obtained from the resultant image were 91%, which showed the feasibility of hyperspectral reflectance imaging technique for the mass discrimination of non-viable lettuce seeds from viable ones.
