만성 분절수면과 식이제한이 식욕조절 호르몬 및 심혈관 위험지표에 미치는 영향 (Effects of Chronic Sleep Fragmentation and Diet Restriction on Appetite-Regulating Hormones and Cardiometabolic Indicators)

  • 본 연구는 7주령의 백서 40마리를 이용하여 만성 분절수면과 식이제한이 식욕조절 호르몬을 포함한 심혈관 위험지표에 미치는 영향에 대해 실험하였다. 13일 동안의 만성적인 분절수면 및 식이제한의 조건에 노출된 백서의 체중 변화와 혈중 leptin, ghrelin, adiponectin, cortisol, epinephrine, norepinephrine 등의 호르몬 농도, 심혈관 위험지표인 혈중 총콜레스테롤, LDL-cholesterol, HDL-cholesterol, 중성지방, 유리지방산의 농도를 대조군 및 3군의 실험군(만성 분절수면 군, 식이제한 군, 분절수면과 식이제한 모두를 적용한 군)에서 비교하였다. 그 결과 실험기간 동안 만성 분절수면 군에서 백서의 체중이 증가하며, 혈중 leptin 및 adiponectin 농도가 감소하고 ghrelin 농도가 증가하여 결국 혈중 LDL-cholesterol 농도가 증가하였다. 분절수면과 식이제한을 동시에 적용한 백서에서는 체중이 감소하고 adiponectin 농도는 대조군과 유의적인 차이를 보이지 않았으며 ghrelin 농도는 분절수면만 했던 백서에 비해 감소한 것으로 나타나 식이제한이 식욕을 조절하는 것으로 나타났다. 하지만 이들 백서에서 혈중 leptin 농도가 현격히 감소하고 혈중 LDL-cholesterol 농도가 증가하는 양상을 나타내 만성 분절수면 환자들의 심한 식이제한이 심혈관질환의 위험을 더욱 증가시킬 수 있다는 결과를 보여주었다.

Leptin 유전자 결핍 동물모델에서 단호박분말 투여가 지방대사변화에 미치는 영향 (Effect of sweet pumpkin powder on lipid metabolism in leptin-deficient mice)

  • 본 연구에서는 ob/ob 마우스를 이용한 비만유발 동물모델에서 단호박분말 투여가 체지방 감소에 효과적인지 확인하기 위하여 6주간 식이를 통해 10% 단호박분말을 투여하였다. 그 결과, 단호박분말과 열처리 단호박분말을 섭취한 ob/ob 마우스에서 식이섭취량이 증가하였음에도 불구하고 체중과 지방조직 무게가 감소되었음을 확인하였으며 비만에 의한 인슐린 저항성과 이상지질혈증을 예방할 수 있음을 확인하였다. 체중 감소 효능의 기전을 확인하기 위해 지방합성과 지방분해에 관여하는 인자들의 mRNA와 단백질 발현을 관찰한 결과, 단호박 분말과 열처리 단호박분말을 섭취한 ob/ob 마우스에서 지방합성 인자들의 활성이 감소되었고, 지방분해에 관련하는 효소의 활성이 증가되었음을 확인하였다 (Fig. 6). 따라서 본 연구의 결과를 통해 단호박분말 섭취는 비만이 유도된 ob/ob 마우스에서 지방조직의 지방분해 증가와 지방합성 감소를 통해 체중 감소 효능 및 비만에 의한 대사질환 발생을 예방할 수 있다고 제안한다.

  • Antidiabetic effects of the acid hydrolysate of silk fibroin were investigated by oral administration to animal models for diabetes mellitus, Fibroin protein was extracted from cocoon and digested to peptides of low-molecular weight range (mainly below 3,000) and amino acids by acid hydrolysis, Feeding of the fibroin hydrolysate resulted in a significant recovering effect on reduction of body weight gain and a lowering effect on blood glucose gain in streptozotocin-induced diabetic Sprague Dawley rats (STZ rats) which were used as an insulin-dependent diabetic animal model. But the body weight and blood glucose level in C57BL/KsJ-db/db mice (db/db mice), an non-insulin-dependent diabetic animal model, were not changed significantly by the feeding, On the other hand, plasma leptin levels increased according to increased feeding amount of the hydrolysate in STZ rats and db/db mice in common, It was concluded from the results that the fibroin hydrolysate might stimulate the insulin secretion by recovering or activating pancreatic ${\beta}$ cells and result in the increased plasma leptin level. It was also deduced that the antidiabetic improvements in body weight and blood glucose gain in STZ were thought to be due to the increased insulin secretion, but in db/db mice of which the diabetic symptoms were caused by insulin resistance, the stimulated secretion of insulin was unlikely to be able to change body weight and blood glucose level significantly.

연잎-연근 복합 발효물이 흰쥐의 내당능 및 지질대사 관련 유전자 발현에 미치는 영향 (Effects of Fermented Lotus Extracts on Glucose Intolerance and Lipid Metabolism-related Gene Expression)

  • Objectives This study was performed to evaluate the effects of fermented lotus extracts on prediabetes and hyperlipidemia in high fructose diet rats. Methods Extracts of lotus leaf and lotus root were fermented using 4 different probiotics separately, including Lactobacillus plantarum, Lactobacillus rhamnosus, Bifidobacterium breve, and Bifidobacterium longum. Expressions of adipogenic transcription factors including Adiponectin, GLUT-4, Leptin, PPAR gamma, Resistin and Visfatin were analyzed by Real time PCR and Western blotting analysis. Results Fermented lotus extracts reduced blood glucose. Fermented lotus extracts inhibited adipogenic transcription factors by inhibiting preadipocytes differentiation. The level of gene expression of Adiponectin, GLUT-4, Leptin, PPAR gamma, Resistin and Visfatin in relation to that of GAPDH were increase or decrease significantly with the Fermented lotus formulation group. Conclusions Fermented lotus extracts showed hypoglycemic and hypolipidemic effects by inhibiting preadipocyte differentiation and controlling insulin sensitivity in high fructose diet rats.

Urinary 6-sulfatoxymelatonin level in girls and its relationship with obesity

  • Purpose: Short sleep duration is associated with obesity. Urinary 6-sulfatoxymelatonin (6-OHMS), the principal metabolite of melatonin, is closely related with sleep. We evaluated the difference in urinary 6-OHMS levels between obese girls and normal weight girls, and the relationship of urinary 6-OHMS with other hormones regulating body weight and metabolism. Methods: A total of 79 girls (6.3 to 12.4 years) were included in this study, of whom 34 were obese; 15, overweight; and 30, normal-weight. We examined their pubertal status and bone age. Fasting serum levels of total ghrelin, leptin, insulin, and first morning urinary 6-OHMS were measured. Homeostatic model assessment-insulin resistance (HOMA-IR) was calculated from the fasting insulin and glucose levels. Results: There was no significant difference in the creatinine adjusted 6-OHMS levels between the obese girls and the control group. Urinary 6-OHMS did not show any correlations with body mass index (BMI), BMI percentile, total ghrelin, leptin, and HOMA-IR. Negative correlations were found between urinary 6-OHMS levels and chronological and bone ages. Conclusion: Our results suggest that melatonin production is not reduced consistently in obese girls.

증포 횟수를 달리하여 만든 선식을 첨가한 고지방식이가 비만 쥐의 혈당과 염증지표에 미치는 영향 (Effects of Sunsik Prepared through Steaming-Drying Cycles on Blood Glucose and Inflammatory Marker in Rats Fed High Fat)

  • This study was conducted to evaluate the effects of Sunsik prepared by steaming -drying cycles- on blood glucose and inflammatory markers in high fat diet-induced obese rats. The experimental animals were divided into five groups and subjected to diets for 8 weeks; normal diet (control), high fat diet (HF), and high fat diet+10% supplementation of Sunsik (1HF, 3HF, 7HF, 9HF). The results showed body weight, food efficiency ratio, blood glucose and insulin levels of high fat diet groups were significantly higher than those of the control group. Increased glucose levels by high fat diet were reduced to normal levels by the four Sunsik supplementations. Insulin resistance (HOMA-IR) of the HF group was significantly higher than those of the control group, whereas supplementation with the four types of Sunsik reduced insulin resistance to similar levels as the control group. Increased insulin secretion and leptin levels of high fat diet-induced obese rats were significantly reduced by supplementation with the four types of Sunsik. C-reactive protein (CRP) of inflammation related components was significantly higher than the control group. However, increased CRP concentration by high fat diet was significantly reduced by supplementation with Sunsik.

Intrauterine diabetic milieu instigates dysregulated adipocytokines production in F1 offspring

  • Background: Intrauterine environment plays a pivotal role in the origin of fatal diseases such as the metabolic syndrome. Diabetes is associated with low-grade inflammatory state and dysregulated adipokines production. The aim of this study is to investigate the effect of maternal diabetes on adipocytokines (adiponectin, leptin and TNF-${\alpha}$) production in F1 offspring in rats. Methods: The offspring groups were as follows: F1 offspring of control mothers under control diet (CD) (CF1-CD), F1 offspring of control mothers under high caloric diet (HCD) (CF1-HCD), F1 offspring of diabetic mothers under CD (DF1-CD), and F1 offspring of diabetic mothers under HCD (DF1-HCD). Every 5 weeks post-natal, 10 pups of each subgroup were culled to obtain blood samples for biochemical analysis. Results: The results indicate that DF1-CD and DF1-HCD groups exhibited hyperinsulinemia, dyslipidemia, insulin resistance and impaired glucose homeostasis compared to CF1-CD (p > 0.05). DF1-CD and DF1-HCD groups had high hepatic and muscular depositions of TGs. The significant elevated NEFA level only appeared in offspring of diabetic mothers that was fed HCD. DF1-CD and DF1-HCD groups demonstrated low serum levels of adiponectin, high levels of leptin, and elevated levels of TNF-${\alpha}$ compared to CF1-CD (p > 0.05). These results reveal the disturbed metabolic lipid profile of offspring of diabetic mothers and could guide further characterization of the mechanisms involved. Conclusion: Dysregulated adipocytokines production could be a possible mechanism for the transgenerational transmittance of diabetes, especially following a postnatal diabetogenic environment. Moreover, the exacerbating effects of postnatal HCD on NEFA in rats might be prone to adipcytokine dysregulation. Furthermore, dysregulation of serum adipokines is a prevalent consequence of maternal diabetes and could guide further investigations to predict the development of metabolic disturbances.

Sasa borealis leaves extract improves insulin resistance by modulating inflammatory cytokine secretion in high fat diet-induced obese C57/BL6J mice

  • Obesity is considered a mild inflammatory state, and the secretion of inflammation-related cytokines rises as adipose tissue expands. Inflammatory cytokines, including tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interlukin 6 (IL-6) and monocyte-chemoattractant protein 1 (MCP-1), are modulated by adipose tissue and known to play an important role in insulin resistance which is the common characteristics of obesity related disorders. In this study we analyzed the effects of Sasa borealis leaves extract on inflammatory cytokines and insulin resistance in diet induced obese C57/BL6J mice. The obese state was induced by a high fat diet for 20 weeks and then the mice were divided into two groups; obese control group (OBC, n = 7) and experimental group (OB-SBE, n = 7). The OBC group was fed a high fat diet and the OB-SBE group was fed a high fat diet containing 5% Sasa borealis leaves extract (SBE) for 12 weeks. We also used mice fed a standard diet as a normal control (NC, n = 7). The body weight and adipose tissue weight in the OB group were significantly higher than those in the NC group. The effects of the high fat diet were reduced by SBE treatments, and the body weight and adipose tissue deposition in the OB-SBE group were significantly decreased compared to the OBC group. The OBC group showed higher serum glucose and insulin levels which resulted in a significant increase of incremental area under the curve (IAUC) and HOMA-IR than the NC group. Also, serum leptin, TNF-${\alpha}$, and IL-6 levels were significantly higher in the OBC group than in the NC group. In contrast, the OB-SBE group showed a reversal in the metabolic defects, including a decrease in glucose, insulin, IAUC, HOMA-IR, TNF-${\alpha}$, IL-6 and leptin levels. These results suggest that BSE can suppress increased weight gain and/or fat deposition induced by a high fat diet and theses effects are accompanied by modulation of the inflammatory cytokines, TNF-${\alpha}$ and IL-6 secretion resulting in improved insulin resistance.

'보건소 비만클리닉 시범사업'으로 실시한 Belly Dance와 영양교육이 비만여성의 신체조성 및 혈청지질 농도에 미치는 영향 (Effects of Belly Dancing and Nutritional Education on Body Composition and Serum Lipids Profiles of Obese Women in a Study, 'Obesity Clinic Projects at Community Healthcenter')

  • 본 연구는 비만중년여성 34명을 대상으로 벨리댄스와 영양교육 실시가 신체조성과 혈청지질 및 인슐린 저항성에 미치는 요인을 구명하기 위하여 벨리댄스와 영양교육을 12주간 실시한 후 운동전과 후의 신체조성, 혈청지질, 인슐린 저항성, leptin 및 adiponectin 호르몬, 그리고 항산화능을 측정하여 비교분석하였다. 벨리댄스 프로그램은 1회 1시간씩 주3회 실시하였으며, 운동 강도는 60~85% HRmax로 실시하였다. 영양교육프로그램은 영양 교육과 개별 상담으로 나누어 총 4회 실하였다. 신체조성중 체중 5.53%, 체지방률 9.51%, BMI 5.61% 유의적으로 감소하였으며, 골격근량은 0.81% 증가하였다. 혈청지질의 변화는 TG 13.50%, TC 9.51%, LDL-C 10.43% 유의적으로 감소하였으며, HDL-C는 3.24% 증가하였다. 인슐린 농도는 19.76% HOMA-IR 지수는 18.22% 유의적으로 감소였다. Leptin 호르몬의 분비는 32.35% 유의적으로 감소하였으나 adiponectin 농도는 변화가 없었다. 그러나 총 유리기 농도는 증가하는 경향을 보였고, 혈중 총 항산화능의 변화는 없었다. 본 연구의 결과를 살펴보면 보건소에서 실시하는 비만클리닉 프로그램은 비만중년 여성의 체지방률을 낮추어 혈청지질 및 인슐린 저항성을 개선함으로써 건강을 증진시키는 긍정적인 효과가 있을 것으로 사료된다.

Combined Trial of Fish Oil and Exercise Training Prevents Impairment in Insulin Action on Glucose Transport of Skeletal Muscle Induced by High-Fat Diet in Rats

  • The purpose of the present study was to determine the preventive effects of combined interventional trial of fish oil treatment and exercise training on insulin resistance of skeletal muscle in high-fat fed rats. Male Wistar rats were randomly divided into chow diet (CD), high-fat diet (HF), high-fat diet with fish oil (FO), high-fat diet with exercise training (EX), and FO+EX groups. The rats in control group were fed chow diet containing, as percents of calories, 58.9% carbohydrate, 12.4% fat, and 28.7% protein. High-fat diet provided 32% energy as lard, 18% as corn oil, 27% as carbohydrate and 23% as casein. The fish oil diet had the same composition as the high fat diet except that 100 g menhaden oil was substituted for corn oil. Insulin sensitivity was assessed by in vitro glucose transport in the soleus muscle after diet treatment and treadmill running for 4 weeks. While the FO or EX only partially prevented insulin resistance on glucose transport and visceral obesity induced by high-fat diet, these interventions completely corrected hyperinsulinemia and hyperglycemia from the high-fat diet. The rats in the FO+EX showed normalized insulin action on glucose transport, plasma chemicals and visceral fat mass. Insulin-mediated glucose transport was negatively associated with total visceral fat mass (r=-0.734; p<0.000), plasma triglyceride (r=-0.403; p<0.05) and lepin (r=-0.583; p<0.001) concentrations with significance. Multiple stepwise regression analysis showed that only total visceral fat mass was independently associated with insulin-mediated glucose transport (r=-0.668; p<0.000). In conclusion, combined interventional trial of FO+EX recovered insulin resistance on glucose transport of skeletal muscle induced by high-fat diet. Visceral fat mass might be more important factor than plasma TG and leptin to induce insulin resistance on glucose transport of skeletal muscle in high-fat fed rats.
