Antioxidant Activities and the Effect of Reducing Serum Alcohol Concentration of Lentinus edodes (표고버섯의 항산화능과 알코올분해능에 미치는 영향)

  • Objectives : The purpose of this study is to evaluate antioxidant activities and reducing serum alcohol concentration of extract of Lentinus edodes on the alcohol administered rats. Methods : Antioxidant effect was measured by total phenolic compound and DPPH-radical scavenging activity of extract of Lentinus edodes in vitro. Blood alcohol concentration, aldehyde concentration, malondialdehyde concentration, glutathion concentration were measured in vivo. Results : The extract of Lentinus edodes increased DPPH-radical scavenging activity dose-dependently. The water extract with boiling water showed lower antioxidant activity and phenolic content than 70% ethanol extract in vitro. Blood alcohol concentration was significantly reduced by pre-treatment of ethanol extract of Lentinus edodes. The effect was more significant than commercial product used as a positive control. Conclusions : This study suggest that Lentinus edodes can be a potential nature resource for the management of ethanol toxicity although the mechanism of action involved in the treatment remains to be explored.

  • The aqueous extract from the fruit body of Lentinus edodes was evaluated for inhibitory activities against $\alpha$-glucosidase isolated from Spargue-Dawley male rats. A aqueous extract of Lentinus edodes exhibited 13.8% inhibitory activity on using 2 mM p-nitrophenyl $\alpha$-D-glucopyranoside as a substrate ($IC_{50}$ 75.3 mg/ml). The aqueous extract of Lentinus edodes inhibition type on $\alpha$-glucosidase was determined to be competitive inhibition. When it was oral administered to increase of blood glucose levels after STZ-induced in a dose dependent dietary. These results suggest that aqueous extract of Lentinus edodes effect a metabolism of intestine, and thereby reducing the increase of blood glucose after STZ-induced.

Development of natural Seasoning Based on Mushroom (버섯 천연조미료 제조에 관한 연구)

  • This study was conducted to provide basic data for the development of natural seasoning. The quality chatacteristics of water exacts from Pleurotus ostreatus and Lentinus edodes were determind. Also, savory taste, fragrance, color and overall preference was evaluated. The preference test on the seasonings prepared from Pleurotus ostreatus and Lentinus edodes with the addition of sea tangle was also performed. The crude protein content of dried, Lentinus edodes and Pleurotur ostreatus was 20.4% and 30.4% respectively. The total organic acid contents were higher in Plourotus ostreatus than in Lentinus edodes extracts. The total free amino acid adn total nucleotide contents were higher in Pleurotus ostreatus than in Lentinus edodes extracts. Three nucleotide, adenosine monophosphate(AMP) , guanosine monophosphate(GMP), xanthine monophosphate(XMP) were found in mushroom seasonings while inosine monophosphate(IMP) was not detected. Pleurotus ostreatus extract showed higher score in savory taste and overall acceptability than Lentinus edodes extract, Pleurotur ostreatus seasoning added with 1% sea tangle and Lentinus edodes seasoning added with 1.5% sea tangle were rated as high in overall quality In a sensory test of foods cooked with mushroom seasoning. It was shown that panels in their twenties well used to the taste of monosodium glutamate while the people in their thirties preferred the specific flavor of the mushroom seasoning.

  • The objective of this study was to evaluate the in vivo effect of Lentinus edodes on the harmful enzymes of mouse intestinal bacteria. When mouse intestinal microflora were cultured in the anaerobic media containing Lentinus edodes water extract or trehalose (LD) isolated From its extract, final pH of the cultured media was significantly decreased and the activities of harmful enzymes, particulary ${\beta}-glucuronidase$ and tryptophanase, were significantly inhibited. By orally administering Lentinus edodes water extract or LD, mouse fecal ${\beta}-glucuronidase$ and tryptophanase were also signifcantly inhibited.

  • Alkali extract obtained from mycelia of Lentinus edodes IY105 was shown to have potent anticomplementary activity and alternative complementary activity in vitro. It was also shown to activate reticuloendothelial system of ICR mice in the carbon clearance. Amount of carbohydrates and protein of the extract were 17% and 42% respectively. It was found that carbohydrates were consisted of 5 monosaccharides and protein was consisted of 16 amino acids. Antitumor activity was observed in the alkali extract treated group of sarcoma 180 bearing ICR mice.

  • This study was aimed to reduce the period of bed log through the protection of harmful fungi and to investigate the effects of physiologically activating substance on the yield increase of Lentinus edodes. Extracts of Allium fistulosm, Hordeum vulagare var. hexastichon, Lentinus edodes. Daucus carota var. sativa, and Citrus junos were used as a physiologically activating substances. The degree of mycelial growth tested by color change after Benzene - azo - ${\alpha}$ - naphthylamine. One percent extract of Album fistulosm was most highly effective as a physiologically activating substance on mycelial biomass growth of Lentinus edodes. Best effects of application of physiologically activating substance was observed in Quercus variabilis bed logs. These results suggested that supplement with physiologically activating substance to the bed log would be beneficial for the production of Lentinus edodes.

Optimization of Media Composition and Culture Conditions for the Mycelial Growth of Coriolus versicolor and Lentinus edodes (Coriolus versicolor와 Lentinus erodes의 영양배지 조성 및 배양조건의 최적화)

  • The media compositions and culture conditions were optimized for mycelial growth of Coriolus versicolor and Lentinus edodes. Media composition for optimal growth of Coriolus versicolor was 2.0% glucose 0.4% peptone and 0.6% yeast extract. Media composition for optimal growth of Lenttnus edodes was 2.0% glucose 2.0% starch 0.4% bacto-soytone and 0.6% yeast extract. The media supplemented with KH2PO4, 0.046% KH2PO4 0.1% and MgSO4, .7H2O 0.05% supported better mycelial growth than the media without mineral salts. The optimum temperature for mycelial growth ranged from $25^{\circ}C$-28$^{\circ}C$. The optimum pH range for mycelial growth of Coriolus versicolor was 5.2~5.6 while that of Lentinus edodes appeared to be 5.75.

  • The effect of mushroom extracts from Pleurotus ostreatus and Lentinus edodes on carbon tetrachloride(CCl4)-induced hepatotoxicity was investigated. Rats were administered orally each mushroom extract at the dose of 150mg/kg, foolwed by treatment with CCl4. Liver damage was produced in male Sprague-Dawley rats, after 21hrs from dosing with CCl4(0.25ml/kg) which were given intraperitoneally. Liver damage without renal injury was confirmed by measuring plasma enzyme, creatinine and blood analysis and liver analysis. Plasma aminotransferase activity, and levels of cholesterol and triglyceride were analyzed. Plasma alanine aminotransferase and aspartate aminotransferase activities were decreased by 34% and 61.5% in pretreatment group of Lentinus edodes compared with CCl4 treated group, respectively. The adminstration of all mushroom extracts led the plasma cholesterol and triglyceride levels decrease more than the CCl4-treated rats. These results suggest that Lentinus edodes extract protect liver from damage induced by CCl4.

  • Anticomplementary activity of 61 strains of Korean higher fungi was screened for immunostimulation. Extracts from 11 of 61 strains including 5 of Ganoderma lucidum, 3 of Lentinus edodes, 2 of Cordyceps sp. and 1 of Agaricus campestris, showed higher anticomplementary activity than krestin which was immunopotent extract from Japanese Coriolus versicolor. The most potent anticomplementary activity was found with extract from Lentinus edodes IY105 whose complement consumption was 31.7%.

  • To develope natural food preservatives, ethanol and water extracts were prepared from the Lentinus edodes and antimicrobial activities were examined against 10 microorganisms which were food borne pathogens and / or food poisoning microorganisms and food-related bacteria and yeasts. Ethanol extract exhibited antimicrobial activities for the microorganisms tested, but not on lactic acid bacteria and yeast Especially, minimum inhibitory concentration(MIC) for Escherichia coli were as low as 0.5 mg/mL. Antimicrobial activity of the ethanol extract was stable by the heating at 121$^{\circ}C$ for 15 min and not affected by pH. The ethanol extract of Lentinus edodes exhibiting high antimicrobial activity. The highest antimicrobial activity adjust bacteria tested was found in the ethylacetate fraction.
