The esterase isozyme of several legume plants were separated and visualized by horizontal starch gel electrophoresis using enzyme-specific staining. Extracts used were prepared from fully expanded young leaf and stem of six legume species which were red clover(Trifolium Pretense L.), ladino clover(Trifolium repense L.), wild white clover(Trifolium repense L.), alfalfa(Medicage sativa L.), mimosoides(Cassia mimosoides var nomame Makino), and amoena(Vicia amoena Fisch). The number of band, Phenotype and staining intensity of esterase isozyme in leaf and stem varies depending on the plant species. However, there are little difference between leaf and stem esterase isozyme in same species except alfalfa. And in the leaf and stem of mimosoides and amoena showed not any esterase(Fig. 2). Among the examined plants, the highest staining intensity and the rapidest migrating esterase isozyme was Est 1.

Starch gel electrophoresis was used to examine the banding pattern of Esterase isozyme in the leaf, root-nodule and seedling of four wild legume species, Trifolim repense, Glycine soja, Phaseolus nipponensis and Vigna uexillata. The number of band, enzyme activity and migrating rate of esterase isozyme varies depending on the species and tissues of legume plants. The isozyme banding pattern in the cotyledon and radicle of T. repense showed same pattern, however, the number of band were varible among the cotyledon of G. soja, P. nipponensis and V. vexzllata, respectively. Est-1 in the leaf of G. soja, V, vexillata. root-nodule of G. soja and seedling of V. vexillata expreesed the highest enzyme activity. The Est-1 showed the rapidest migrating rate among the isozymes.
In germinating pea, contents of enodgenous polyamine in the leaf and stem were determined, and protein content, peroxidase activity and pattern of isozymes were examined in the leaf treated with polyamines. During growth of the pea for 14 days in light condition, the polyamines in leaf and stem showed the highest level at the 5th day, and were decreased rapidly at the 7th day, kept almost constant level since then. The putrescine level was relatively higher than those of spermidine and spermine, and cadaverine was also detected. On the other hand, in the leaf treated with spermine (0.01 mM) protein content increased about 250% than that of the control, the peroxidase activity increased ore than 100% in spermine of 0.01 mM and 0.1 mM. In treating with putrescine of 0.1 mM the pattern of peroxidase isozyme appeared 4 new cathodic bands (pI 4.8, 5.6, 5.9 and 6.8) compared with the control, the clear cathodic bands (pI 5.6, 5.9, 6.4 and 6.6) were also observed in spermine of 0.1 mM. These results suggest that polyamines were important factor in the differentiation of pea at the early stage of germination.
The plantlet was regenerated on MS medium containing BAP (2 mg/I) and 1M (1 mg/I) from leaf discs of pepper after 3 weeks of culture. And then, we investigated the activity of peroxidase and esterase and the pattern of their isozymes from leaf, stem and root in order to observe physiological and biochemical changes on the developemental stage, respectively. The peroxidase was expressed with tissue specificity because peroxidase activity according to the developemental stage of the tissue was not only highest in the leaf of the pepper at 10 days after it germinated but also 2 new bands of its isozyme were found in pI 7.2 and pI 5.2. However, a new pI 3.4 band was found in the leaf and root of the pepper after 14 days of germination, and in the stem was found out pI 5.2 band. As regeneration of leaf dises was progressed, its peroxiase activity was increased about 80% more than that of control after 14 days of culture and new pI 3.2 and 6.5 bands of it isozyme were found. The results suggested that peroxidase would be connected with regeneration of pepper. Also, esterase activity was increased about 50% more than that of control after 14 days of culture, the pattern of esterase isozyme was shown to be 3 cathodic bands and 1 anodic band after 7 days of culture.ulture.
A electrophoretic study on seveal important enzymes was conducted with seed, leaf and stem of Benincasa hipida (Thunb) Cogn and Commelina communis L. Isozyme patterns of esterase were different from the parts and fruit of wax gourd appeared only one band. In the peroxidase isozyme, fruit of wax gourd appeared four bands, but stem of common dayflower no appeared. In the amylase isozyme, seed and fruit of 'gourd were appeared clearly one band, respectively.

The variation of the esterase isozyme, germination rate, chemical composition and digestibility of field bean(G1ycine soja S. and Z.) were estimated. The results are as follows; 1. The banding patterns of the esterase isozyme in field bean were varied with the tissue and habitat. 2. The enzyme activity of the Est-I, Est-2, Est-3 and Est-4 in field bean showed a high value compared with the other enzyme. 3. The range of germination temperature in field bean was 10-40C and the optimum germination temperature was 25- 38^{\circ}C.$. 4. The crude protein(CP) contents was 19.9% in the whole plant, 27.8% in the leaf and 45.9% in the seed, the cellulose contents was 29.5% in the whole plant, 23.0% in the leaf and 13.8% in the seed, the neutral detergent fiber(NDF) was 62.6% in the whole plant, 47.9% in the leaf and 47.9% in the seed and the acid detergent fiber(ADF) was 44.5% in the whole plant, 28.4% in the leaf and 28.4% in the seed, respectively. 5. The digestibility of the field bean was 44.1% in the whole plant, 49.6% in the leaf and 75.1% in the seed, NDF was 26.2% in the whole plant 46.2% in the leaf, ADF was 29.0% in the whole plant, 47.7% in the leaf and 58.0% in the seed and Cellulose was 48.7% in the whole plant, 58.0% in the leaf and 70.2% in the seed, respectively. 6. Total digestible nutrients(TDN) of the field bean was 47.4% in the whole plant, 51.5% in the leaf and 70.2% in the seed, respectively. The digestible energy(DE) value was 2.1 kcal/g in the whole plant, 2.27 kcal/g in the leaf and 3.10 kcal/g in the seed and the metabolizable energy(ME) value was 1.72 kcal/g in the whole plant, 1.86 kcal/g in the leaf and 3.23 kcal/g in the seed, respectively.

The esterase isozyme in tissue of wild legume plants were separated by horizontal starch gel electrophoresis. Extracts used in this study were prepared from fully expanded young leaf, cotyledon and radicle of seedling and root-nodule of Glycine sola, Vigna vexillata var. tsuscmensis and Trifoliwn repens. The results are as follows; 1. Each tissue examined had a characteristic banding pattcrn. Number of bands in each species, G. soja, V. vexillata, and T . repens, were 14, 8 and 1 1 bands, respectively. And difference in esterase isozyme bands were greater from tissue to tissue than difference between habitat. 2. Est-I, Est-2. Est-3 and Est-4 in G. soja, Est-I in V. vexillata and Est-l and Est-2 in T. repens showed strong cnzyme activity than other enzyme. 3. Esterase isozyme variation in G. soja and T . repens showed more variety than V. vexillata. This is resulted from many genotypic differences within species. 4. The main enLyme among thc esterase isozyme were Est-I. Est-2, Est-3 and Est-4.
The present study performed the isolation of cytosolic ascorbate peroxidase (APX) isozymes and analyzed the pattern of their activity development and also investigated the change in some other enzyme activities related to the ascorbate-glutathione pathway from the senescing wheat leaves. The aim of this work is to examine the possibility that in the cytoplasm of wheat leaves the ascorbate-glutathione pathway p!ays a significant role in relation to leaf senescence involving an $H_2O_2$ accumulation and then to show the effect of benzyladenine (BA) on that pathway. During the leaf senescence characterized by increases in ChI breakdown and H202 accumulation under the 4-day dark incubation of matured leaf segments; i) no significant increase of total cytosolic APX was observed, ii) a dehydroascorbate reductase (DHAR) activity was decreased rapidly, iii) a slight increase of glutathione reductase (GR) activity occurred. In the BA-treated leaves; however, i) the total activity of APX increased conspicuously, ii) the decrease of DHAR activity was relatively inhibited, iii) the GR activity increase was more enhanced, and iv) the decrease of ascorbate content and the increase of H202 content were retarded as compared with those of control leaves. Three isozymes of cytosolic APX were found by using a native-electrophoretic gel in senescing wheat leaves and two of them occurred with major activity. In the developmental patterns of cytosolic APX isozymes, only two isozyme bands ("a" and "b") appeared with almost constant activity through 4 days of incubation in the control leaves, while one additional weak isozyme band ("c") and a little increase of "b" isozyme activity were detected in the BA-treated leaves. EspeciaUy, the development of "a" isozyme activity increased remarkably compared with that of control leaves. The increased capacity for peroxide scavenging due to the enhanced activity of all 3 enzymes (APX, DHAR, GR) participating in the ascorbate-glutathione pathway in BA-treated leaves suggested that this pathway might playa significant role in the processes related to the wheat leaf senescence.scence.
In this study, we calculated the genetic relationships of Pestalotiopsis species collected from various places in southern part of Korea through isozyme analyses. As a result, EST showed the largest number of band, and the number of bands were ranged from 5 to 7 on the average. All the isozymes used in this study showed distinctive band patterns for each isolates. Similarities among the compared isolates ranged from 48 to 93%. Isolates SP7, SP19 and SP23 showed more than 90% similarities, and most isolates showed similarities ranging from 65 to 82%.
This study was carried out to obtain the basic information for the clarification of spring growth habits mechanism of naked barleys. The isozyme patterns and activities of peroxidase in the young spike and leaf blade were analyzed during the differentiation and development of young spike. The characteristic differences between the normal and rosetted type were in c and g isozymes in young spike, and in i isozyme in the leaf blade. In the normal type, c and i isozymes disappeared at the stage of spi-kelet differentiation, g isozyme at the stage of flolet differentiation. But, in the rosetted type, those three isozymes remained in dark stained condition until the time of final sampling. Especially, those three isozymes were higher in the rosetted type than those in the normal type even at the stage of bract differentiation(BDS), just prior to the reproductive stage. The activities of peroxidase decreased slowly after BDS in the young spike and leaf blade in the normal type, While, in the rosetted type, increased linearly, and the degree of increasing was remarkable in the young spike. It was interesting that the degree of activities in young spike was higher in the rosetted type than that in the normal type even at BDS. From the above results, the remarkable differences of the isozyme patterns and activities at BDS between the normal and rosetted type were considered to be the physiological expression of the varieties concerned with the degree of spring growth habits.
