The present study investigated the effects of triacontanol (TRIA) on plant growth and peroxisomal enzyme activities in radish seedlings. The optimum concentration of TRIA with respect to radish seedling bioassay was decided to 1.0mg $1^{-1}$. In comparison to untreated controls (including Tween 20 treatment), 1.0mg $1^{-1}$ TRIA treatment caused an increase in seed germination rate and root growth, but no stimulation in hypocotyl growth. Chlorophyll accumulation in cotyledon during carly development stage increased rapidly, and degradation of chlorophyll in later stage due to the cotyledon senesence was noticeably retarded. Increase of soluble protein content in cotyledon at early period of development was observed. Isocitrate lyase and catalase activity was not significantly different in both the treated and the untreated plants. But, glycolate oxidase activity was inhibited by TRIA down to 20% against controls. Also, the increase of the activity of peroxidase, a leaf-senescence marker enzyme, was continuously retarded over control for 8 days of development. Based on above results, TRIA was found to be active in both the growth and the peroxisomal enzyme activities of radish seedlings.
A total of 192 Korean landraces were investigated for the comparison of each other as useful germplasm to organic farming and examined correlation between quantitative traits. Almost Korean landraces were classified into medium-heading variety and average days from seeding to heading was 111.1 days. Plant height and culm length were longer about 20∼40cm than general Japonica cultivar. Average width of leaf blade and average length of panicle were 1.3cm and 22.4cm, respectively, Average number of panicles per plant was 10.0 and almost varieties showed low tilling habit. Average length/width ratio of brown rice was 1.7 and several varieties were long grain type and average 1,000 grain weight was 21.6g. In correlation coefficient among quantitative traits, positive significant correlations were observed between culm length and leaf width, days from seeding to heading, but negative correlations were observed between days from seeding to heading and 1000 grain weight. 27.1% of Korean landraces showed weak and lodging degree in culm strength and 3.6% of total varieties showed purple margin on leaf blade and leaf sheath. In leaf blade angle and flag leaf angle, erect type was 46.9% and 10.9%, respectively. 24.0% of total varieties showed the slow and late degree of leaf senescence. The rate of awned type, waxy type and brown seed coat were 74.5%, 20.3% and 4.2%, respectively. The color of stigma, spikelet, apiculus and awn showed diverse color such as white, brown, red and purple.
Six rice cultivars showing various types of resistance or susceptibility to Pyricularia oryzae in the field were used to study the relationship between susceptibility and contents of carbohydrates, amino acids and phenolics at different plant maturities and leaf ages under controlled environmental conditions. No consistent differences between the susceptible cultivars and adult-plant-resistant cultivars were found in terms of the contents of individual carbohydrates, total amino acids and phenolics in rice leaves throughout the plant development. Only the adult-plant-resistant cultivar Dobong had lower contents of inositol throughout the plant development than the other cultivars. The amounts of sucrose, inositol, glucose, and fructose increased in all tested cultivars at eight leaf stage than those at five leaf stage, but slightly decreased at twelve leaf stage. In contrast, total amino acids and phenolics gradually decreased during plant development. With increasing age of rice leaves, the amounts of total soluble amino acids and phenolics gradually increased in healthy fifth leaf tissues, although there were no significant differences between the cultivars. In particular, a high level of phenolics existed in old fifth leaves of the cultivar Dobong. These results suggest that increased resistance to blast of matured rice plants and old leaves may be the two different phenomina derived from the physiological changes occurring during plant development and leaf senescence, probably functioning differently in P. oryzae development.
We have investigated the effects of abiotic and biotic stresses on leaf senescence using transgenic tobacco plants, in which cellular contents of polyamines were increased by introducing the genes of polyamine and ethylene biosynthesis in sense or antisense orientation. These transgenic plants showed accumulations of polyamines at higher levels than were found in wild-type. Stress-induced senescence was attenuated in transgenic plants cpmpared with wild-type plants, in terms of total chlorphyll loss and phenotypic changes after oxidative stress of hydrogen peroxide($H_2O_2$), high salinity, acid stress (pH3.0), ABA and fungal pathogen(phytophothora parasitica pv.Nicotianae). Transcripts for antioxidant enzyme, glutathionine-S-transferase and catalase, were also more abundant in transgenic plants than wild-type plants. These result suggested that higher expression of those genes caused a broad-spectrum resistance to abiotic stress/biotic stress. These phenomena indicate that polyamines may play an important role in contributing to the antioxidant defense function in plants. Our findings suggest that facilitate the improvement of stress tolerance of crop plants.
The genotypic (2n, 3n, 4n) response of watermelon in vitro shoot tip culture was evaluated. Different genotypes had similar response in terms of shoot formation and growth. Shoot formation was better at lower concentration of 0.3 mg/L BA and higher concentration of 5-10.0 mg/L 2iP and kinetin, but growth of newly formed shoot was inhibited. With further subculture, kinetin did not promote shoot formation Better shoot formation was observed at 0.3-0.5 mg/L BA. Combination of 0.3 mg/L BA and 0.3-0.5 mg/L BA was effective in shoot multiplication, growth and induction of more internodes. Varrying levels of light intensity and agar concentration did not affect the performance of tetraploid plants. Higher light intensity and agar concentrations decreased the number of shoot formed in triploid plane. Growth in both genotype, however was inhibited. Higher light intensity was found to promote leaf senescence in all genotypes. All growth inhibitors decreased the number of shoots formed and slowed plant growth there by prolonging duration of cultures. Growth inhibitors were to observed to decrease incidence of hyperhydricity in culture. No difference in shoot formation was observed in each of the concentrations used in Ancymidol, TIBA, CCC and PP333. Shoot formation and growth was more inhibited in ABA treatments. Leaf expansion and growth was poor in all treatments.
Five soybean varieties of two early maturing Karikei73 and SS 79168, and three late maturing Tohoku76, Baegunkong and Jangbaegkong were used and evaluated in the study. Of the varieties examined, Karikei73 was characterized by the delayed leaf senescence. The varieties were planted in the pots of 1/3500 a filled with volcanic ash soil at the experimental fields of the National Institute of Agrobiological Resources in Japan. Major agronomic characteristics including the activity of nitrogen fixation for root nodules during the grain filling period were measured. Measurements during the stages were followed by the stage of development descriptions for soybeans made by Fehr and Carviness (1977). The acetylene reducing activity (ARA) per dry weight of root nodule measured using acetylene reduction assays was the highest at R4-R4.5 with decreasing trends thereafter for the early matruing varieties, while it continuously increased up to R6 but decreased rapidly thereafter for the late maturing varieties. The dry weights of root nodules and all parts of the host plant at each stage checked were greater in the late maturing varieties being the same in ARA per pot.
This study was conducted to evaluate the effect of Zn and Cu on physiological responses of tobacco at the different leaf ages for the purpose of phytoremediation of heavy metal contaminated field. Plant height was significantly decreased at Zn and Cu concentrations of 1.0 mM or higher. Especially, root dry weight decreased more (50% reduced) than shoot dry weight. Zn and Cu accumulation in the shoot and root was dramatically increased after 2.0 mM treatment. The accumulation pattern was different by leaf aging in that Zn accumulated more in senescence leaf and Cu accumulated more in growing leaf. The amounts of heavy metal accumulations per plant were 146,000 mg/kg for Zn and 20,500 mg/kg for Cu at 2.0 mM treatment. The proper harvesting time would be suggested as vegetative growth stage for Zn and reproductive stage for Cu. The concentrations of other inorganic ions (Mg, Mn, Fe) were decreased when Zn and Cu concentrations were increased. Cu concentration and Zn concentration of both shoot and root were decreased in Zn treatment and Cu treatment, respectively.
In Ulrung floral community the growing environments and nutritive values of wild garlic was investigated. The wild garlic, grown for $3{\sim}4\;years$ in community area, were able to develope bulbs. The bulbs with 2 leaves had the highest ratio of propotion as 75.1% of share among the distribution of bulbs with completely developed leaves and 1 leaf-bulbs and 3 leaves-bulbs had 19.6% and 5.3%, respectively. Also the 2 or 3 leaves-bulbs indicated the better growth states in the part of shoot and bulbs than 1 leaf-bulbs, and there was a significant defferences in each growth between 2 leaves-and 3 leaves-bulbs. On the wild garlics we cannot find out insect injuries. However, there was deseases with symptoms of leaf-spots and lesion from May to June. At first it made a small spots on the leaves, followed by senescence at arrounding area of spots, and then it expanded to whole leaf. This pathogenic bacterium was supposed to be a member of Aristastoma. The contents of crude protein, fat, and carbohydrate in each portion of wild garlics decresed in the order of shoot, expanded leaves and bulbs. On the contrary, crude fibers increased in the same order and so it is not suitable for human consumption nutritional value. However, because the shoots of wild garlics have more than the other Allium sp., the shoots will be a good quality food stuff.
This study was conducted to elucidate the shelf-life and quality changes in relation to biochemical components in leaf lettuce (Lactuca sativa L.). The shelf-life of leaf lettuce at room temperature was 2 to 3 days. But it was extended to 3 weeks by packaging in a 0.01 mm thick polyethylene film sack when stored at $3^{\circ}C$. The ascorbic acid contents of fresh leaf lettuce was 25 mg per 100 gram fresh weight. The acid at room temperature was almost destroyed after 4 days storage. But the contents of ascorbic acid at $3^{\circ}C$ maintained about 50 to 60% of the initial level in packaging of polyethylene film sack after 8 days storage. The content of chlorophyll was greatly decreased at room temperature but no significant changes were found at $3^{\circ}C$. The changes of total sugar and reducing sugar contents during storage were not very different between treatments. The contents of alkali soluble protein and free amino acid gradually increased in the treatments of polyethylene film sack packaging during storage in general, but the contents decreased after the increase in control treatment. Nucleic acid content, peroxidase and polyphenoloxidase activities were measured and discussed in relation to the leaf senescence.
Thirty-six $F_3$ hybrid poplar (Populus trichocarpa${\times}$P. deltoides) clones were fumigated with ozone to select for ozone sensitive and resistant clones. Fumigation was applied for 6 to 8 hours each day for approximately 3 months at ozone concentrations of 90 to 115 ppb using by open-top chambers. Height, diameter, number of leaves, total biomass, biomass components, root/shoot ratios, leaf drop and stomatal response were investigated. In summary, ozone generally reduced height, diameter, number of leaves, total biomass, and root/shoot ratios. Ozone stress induced leaf drop and foliar senescence in trees. This study showed very low relationship between total biomass and stomatal conductance. Increased plant resistant to ozone is not always correlated with stomatal behaviour. Probably, characterization of biochemical and other physiological responses to ozone exposure can provide a better understanding of tree response to ozone environment.
